Abstract The parasitic status in the coypus (nutrias) Myocastor coypus, both farm-bred or free-living on river banks in the Czech Republic, was determined. Faecal samples were collected from 200 coypus originating from 11 farms (farm-bred animals) and from 20 individuals living at 14 natural localities (feral animals). Faeces were examined for nematode eggs and coccidian oocysts using the McMaster method. The evaluation of faeces from farm-bred coypus indicated infection with the following parasites: Trichuris sp., Strongyloides sp., Trichostrongylus sp., Eimeria seidelli, Eimeria nutriae, Eimeria coypi, and Eimeria myopotami. Free-living feral rodents harboured Eimeria nutriae, E. coypi, Strongyloides sp. and in one case also Trichuris sp. An additional visceral examination of 20 coypus originating from five farms revealed two nematode species in the gastrointestinal tracts, namely Trichuris myocastoris and Strongyloides myopotami. The study indicated that feral coypus are far less parasitized than their captive counterparts.

Cryptosporidium spp., common parasites of vertebrates, remain poorly studied in wildlife. This study describes the novel Cryptosporidium species adapted to nutrias (Myocastor coypus). A total of 150 faecal samples of feral nutria were collected from locations in the Czech Republic and Slovakia and examined for Cryptosporidium spp. oocysts and specific DNA at the SSU, actin, HSP70, and gp60 loci. Molecular analyses revealed the presence of C. parvum (n = 1), C. ubiquitum subtype family XIId (n = 5) and Cryptosporidium myocastoris n. sp. XXIIa (n = 2), and XXIIb (n = 3). Only nutrias positive for C. myocastoris shed microscopically detectable oocysts, which measured 4.8–5.2 × 4.7–5.0 µm, and oocysts were infectious for experimentally infected nutrias with a prepatent period of 5–6 days, although not for mice, gerbils, or chickens. The infection was localised in jejunum and ileum without observable macroscopic changes. The microvilli adjacent to attached stages responded by elongating. Clinical signs were not observed in naturally or experimentally infected nutrias. Phylogenetic analyses at SSU, actin, and HSP70 loci demonstrated that C. myocastoris n. sp. is distinct from other valid Cryptosporidium species.

Abstract This epidemiological study monitored the occurrence of the coccidia genus Eimeria and their species composition over a oneyear period at an organic sheep farm in the Czech Republic. Individual faecal samples were collected from thirty lambs and thirty ewes once a month throughout the whole survey. As a result, 348 and 333 samples from ewes and lambs respectively were evaluated using the faecal flotation technique. The overall prevalence of eimeriosis was 75.7% and 54.0% for lambs and ewes respectively. Four Eimeria species (E. ovinoidalis, E. crandallis/weybridgensis, E. parva, and E. intricata) were identified in both, lambs and ewes, during this survey. The most prevalent species was E. ovinoidalis, with an overall prevalence of 84% in ewes and 85% in lambs, followed by E. parva and E. crandallis/weybridgensis. The oocysts faecal output was seasonal. The highest oocysts per gram levels were detected in February (139 000) and May (250 000) in ewes and in February (1 949 900), March (326 000), and May (187 700) in lambs. The intensity of Eimeria infection differed significantly (P < 0.0001) between ewes and lambs during the monitored period.

AbstractComposition and vertical distribution of soil nematode communities within soil profile were investigated in eight hop gardens in Czech Republic. In total, the presence of 78 nematode genera was confirmed. Genus Drilocephalobus (Coomans & Coomans, 1990) is new for fauna of the Czech Republic. The highest abundance of soil nematodes was found at a depth of 0–10 cm and declined with increasing depth of soil profile. The most dominant genus was Bitylenchus, followed by genera Acrobeloides, Ditylenchus, Chiloplacus and Cervidelus. Ten genera of plant parasitic nematodes were recorded: Bitylenchus (with prevalence of B. dubius), Helicotylenchus, Heterodera (with absolute prevalence of H. humuli), Geocenamus, Longidorella, Longidorus (only L. elongatus), Merlinius (with prevalence of M. brevidens), Paratylenchus and Pratylenchus. Low population densities of predators and omnivores, low values of the community indices (MI, ΣMI, SI, and CI), and high values of NCR, EI, and PPI/MI ratio indicated disturbed nematode communities in hop gardens and bacteria-dominated decomposition pathways in the soil food web.

This study was carried out to determine the prevalence of strongyles infecting pigs in Ogun State , Southwestern Nigeria. Faecal samples were collected from 209 pigs which were initially screened by centrifugal sedimentation technique. Faeces positive nematode eggs were then cultured using a modified Baermann's technique. Out of the 36.8% (77/209) infected pigs, 84.4%, 58.4%, 57.1%, 19.5% and 6.5% were for Hyostrongylus rubidus, Trichostrongylus colubriformis, Oesophagostomum spp., Strongyloides ransomi and Dictyocaulus viviparus respectively. Mixed infection with various nematode species accounted for 94.8% of the prevalence. The infection rate in piglets (43.7%) was higher than in weaners and adults without any significant differences. Similarly no significant differences were observed between diarrhoeic and non-diarrhoeic pigs. The results of this study thus showed the economic importance of strongylid nematode infections in intensively reared pigs in Ogun state. Regular screening to ascertain worm load and species involved will facilitate appropriate control and prevention strategies.

Porcine sapoviruses, members of the family Caliciviridae, have been considered as an aetiological agent of gastroenteritis in pigs. In this study, we analysed 251 faecal samples obtained from 3 to 90 day-old diarrhoeic pigs in the Czech Republic between January 2005 and June 2010 and tested them by negative staining electron microscopy for the presence of sapoviruses. Only one sample showed the presence of viral particles with characteristic sapovirus morphology. The presence of sapovirus (SaV) was confirmed by an RT-PCR assay with primers specific for the sapoviral RNA polymerase and capsid genes. Phylogenetic analysis based on a partial sequence of the RNA polymerase gene placed the new Czech isolate into the GVII genogroup of porcine sapoviruses; however, analysis of a portion of the capsid gene sequence classified the isolate as GIII of the genus Sapovirus. These contradictory findings indicate that recombinant porcine sapovirus was identified. According to our knowledge this is the first description of porcine sapovirus in domestic pigs in the Czech Republic

A unique human case of alveolar echinococcosis was described in 1979 from the Klatovy district of the Czech Republic. However, there were no previous epidemiological studies in this area focusing on detection of the source of infection –Echinococcus multilocularisadults producing eggs. During the period June 1997 to April 1999, 29 out of a total of 46 (63.3%) red foxes (Vulpes vulpes) in the Klatovy district and one of four foxes (25.0%) in the Pilsen South district were found to be infected with adult worms ofE. multilocularis. NoE. multilocularisadults were found in other animals from the Klatovy district (i.e. three specimens ofMartes martes, twoMartes foina, oneMustela erminea, twoMeles melesand oneFelis catusf.domestica). An examination of faecal samples from 55 dogs (Canis familiaris) from the Klatovy district resulted in the detection ofE. multilocularisDNA in one (1.8%) sample. The present results support the possibility that human alveolar echinococcosis previously described in the Czech Republic had the character of an autochthonous infection. There are also indications of a potential risk of infection to humans.

AbstractDuring a 3-year period, from April 1991 to December 1993, in a flock with four lambing periods annually, faecal samples from lambs at weaning at the age of 3 months (no. = 1756), were examined for gastro-intestinal nematode eggs by the McMaster method. Faecal samples were also examined at the age of 9 (no. = 170), 12 (no. = 157), 15 (no. = 253) and 18 (no. = 85) months from the females only. From June to December 1993 packed cell volume (PCV) was also examined from lambs at weaning. Seven genotypes were used in this study.The faecal egg counts were affected by genotype, birth season, type of birth and rearing, sex and age. First generation cross of Sumatra × Barbados Blackbelly had lowest eggs per g of faeces (EPG) followed by Sumatra, St Croix, Sumatra × St Croix Fl and F2 and Sumatra × Java Fat tail. Lambs born in February-March when rainfall was low had lower EPG (V < 0·05) than those born in May-June or August-September. The EPG of lambs born and reared singly were significantly lower (P < 0·001) than those born twin, triplet or quadruplet. Female lambs had lower EPG than male lambs (P < 0·001). The EPG of lambs at 3 months were higher than at 9, 12, 15 or 18 months (P < 0·001).PCV was affected by genotype, sex, type of birth and rearing and season of birth. There was a positive correlation between PCV and weaning weight of lambs and a negative correlation between EPG and weaning weight. These results show that the crosses of St Croix and Barbados Blackbelly with local Sumatra sheep are at least as resistant or more resistant to gastro-intestinal nematodes than pure Sumatra breed and therefore these breeds can be used in cross breeding programmes to improve the body size of local Sumatra sheep.

Nutria (Myocastor coypus) is a large rodent native to South America which was introduced worldwide originally with the intention of fur farming. Three colour forms (Standard, Moravian Silver, and Prestice Multicolour) fall into the Farm Animal Genetic Resources of the Czech Republic protected by the National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture coordinated by the National Focal Point. The objective of our investigation in the Czech Republic was to establish microsatellite panel for nutria and determine microsatellite variability in the local nutria population, levels of genetic diversity within and among subpopulations with respect to colour form and the levels of inbreeding within subpopulations. The genetic variability was studied in 64 animals by investigating 11 microsatellite markers (McoD214, McoD217, McoD59, McoD69, McoC124, McoC203, McoD60, McoB17, McoC118, McoA04, and McoD228) analysed by multicoloured capillary electrophoresis. The whole population showed a moderate level of genetic variability in terms of number of alleles (5 alleles per locus) or heterozygosity (50.4%) and the Prestice Multicolour form exhibited the highest level of diversity. Particular attention should be paid to organizing mate to minimise inbreeding, especially in the Standard colour form (F<sub>IS</sub> = 0.238) which showed the highest level of inbreeding out of the three colour forms. High combined exclusion probability (CEP) values (99.5, 94.5, and 99.9%) implied that the panel of microsatellite markers established in this study was usable for individual identification or routine parentage testing in nutria population in the Czech Republic. &nbsp;
