These findings showed that raw milk from dairy cattle farms and collector’s bulk tank in Sebeta town was contaminated with public health important bacterial species like E. coli and Salmonella species and the observed resistant to certain antimicrobial drugs also needs attention. To ensure the quality of raw milk, stakeholders engaged in milk and dairy production chain should be trained on hygienic practices.

Domiati cheese is the most popular brand of cheese ripened in brine in the Middle East in terms of consumed quantities. This study was performed to investigate the impact of the microbiological quality of the used raw materials, the applied traditional processing techniques and ripening period on the quality and safety of the produced cheese. Three hundred random composite samples were collected from three factories at Fayoum Governorate, Egypt. Collected samples represent twenty-five each of: raw milk, table salt, calf rennet, microbial rennet, water, environmental air, whey, fresh cheese, ripened cheese & swabs from: worker hands; cheese molds and utensils; tanks. All samples were examined microbiologically for Standard Plate Count (SPC), coliforms count, Staphylococcus aureus (S. aureus) count, total yeast & mould count, presence of E. coli, Salmonellae and Listeria monocytogenes (L. monocytogenes). The mean value of SPC, coliforms, S. aureus and total yeast & mould counts ranged from (79×102 CFU/m3 for air to 13×108 CFU/g for fresh cheese), (7×102 MPN/ cm2 for tank swabs to 80×106 MPN/ml for raw milk), (9×102 CFU/g for salt to 69×106 CFU/g for fresh cheese) and (2×102 CFU/cm2 for hand swabs to 60×104 CFU/g for fresh cheese), respectively. Whereas, E. coli, Salmonella and L. monocytogenes failed to be detected in all examined samples. There were significant differences in all determined microbiological parameters (p ≤0.05) between fresh and ripened cheese which may be attributed to different adverse conditions such as water activity, pH, salt content and temperature carried out to improve the quality of the product.

In Africa, the use of poor and unhygienic methods for animal milking and milk processing that leads to spoilage of milk by microbes affects the production of milk and dairy products, especially in the small scale and local processing plants. This study was conducted to analyze the quality and safety of raw milk collected from six different towns in Kwami local government area of Gombe State, Nigeria. The samples were serially diluted using ten-fold dilution and used aliquot 1 ml to inoculate the appropriate media using pour plate technique. The total viable count for bacteria in CFU/ml on plate count agar (PCA) was highest in sample E from U/Anchau with an average of total viable count of 3.8x104 CFU/ml, followed by 3.0x104 CFU/ml in sample B (Dirri), then 2.8x104 CFU/ml in sample D from Burakosuma, 2.5x104 CFU/ml in sample F from Dun urji, 2.3x104 CFU/ml in sample C from Zanbe with least count from sample A at Bele as 1.8x104 CFU/ml. Five (5) bacterial species of public health importance were isolated and identified using biochemical tests namely; Enterobacter sp., Yersinia enterocolitica, Escherichia coli, Staphylococcus aureus and Salmonella sp.. Out of the organisms isolated, Enterobacter sp. had the highest occurrence of 93% (n=280), then Yersinia enterocolitica 90% (n=270), E. coli 70% (n=210), S. aureus 57% (n=170), and finally Salmonella sp. 23% (n=70). Based on the microbiological outcomes, preventive measures for milking and processing that focus on training of farmers and dairy employees for the improvement of the hygiene of local milk and dairy production chain should be defined.

Milk is such a food which can meet almost all nutritional needs of human lives. Raw or unprocessed milk supports the growth of wide variety of microorganisms. The major interests of this study were examining the microbial quality of raw milk collected from different locations of Jessore city in Bangladesh and determining antibiotic susceptibility pattern of some isolated bacteria. To do so, 12 raw milk samples were collected from different areas of Jessore city. Microbial analysis comprised of enumeration of TVC (total viable count), TCC (total coliform count) and TSC (total staphylococcal count). The highest TVC, TCC and TSC were 1.95x109 CFU/ml, 2.5x107 CFU/ml and 1.02x107 CFU/ml respectively. Prevalent bacterial populations were Klebsiella spp., Enterobacter spp., Shigella spp. Staphylococcus spp., Escherichia coli and Citrobacter spp. In order to observe the antibiotic susceptibility pattern, the antibiotic sensitivity test was performed for some randomly selected isolates of E. coli and Klebsiella spp. More than 90% isolates of Klebsiella spp. were found to be resistant against Erythromycin whereas more than 90% isolates were sensitive against Imipenem. On the other hand, 100% E. coli isolates were observed as resistant against Erythromycin and in case of Trimethopreme 100% isolates were sensitive. Multidrug resistance pattern was also found. Summary An 8-month survey of the bacteriological quality of bulk tank produced fluid milk supplies for four major markets in Ohio, was made utilizing the Standard Plate Count (SPC), the preliminary incubation count (PI), the thermoduric (pasteurized milk) count, and the coliform count. In terms of maximum standards of 200,000 and 100,000 organisms/ml, the SPC would have eliminated 13% and 20% of the milk samples, respectively. A SPC of 50,000/ml, a PI count of 200,000/ml, a thermoduric count of 500/ml, and a coliform count of 100/ml would have eliminated 37%, 34%, 40%, and 40%, respectively, of the samples but not all of the samples eliminated by one test were eliminated by another test. All of the tests employed showed a seasonal trend especially in the high count categories but the trend was less noticeable in the results of the preliminary incubation count. A combination of two of the methods was superior to any single bacteriological method employed in detecting unsatisfactory milk. Of the tests used, the combination of the thermoduric count (500/ml) and the coliform count (100/ml) was the most effective in the detection of unsatisfactory milk samples.

ABSTRACT Zoonotic bacteria such as Campylobacter, Listeria, and Shiga toxin–producing Escherichia coli have been found in bulk tank milk in many countries, and the consumption of raw milk has been implicated in outbreaks of disease in New Zealand. Fecal contamination at milking is probably the most common source of pathogenic bacteria in bulk tank milk. Raw milk was collected from 80 New Zealand dairy farms during 2011 and 2012 and tested periodically for Campylobacter, E. coli O157, Listeria monocytogenes, and Salmonella. Milk quality data such as coliform counts, total bacterial counts, and somatic cell counts also were collected. By treating the total bacterial count as a proxy for fecal contamination of milk and utilizing farm and animal level prevalence and shedding rates of each pathogen, a predictive model for the level of pathogenic bacteria in bulk tank raw milk was developed. The model utilizes a mixture distribution to combine the low level of contamination inherent in the milking process with isolated contamination events associated with significantly higher pathogen levels. By simulating the sampling and testing process, the predictive model was validated against the observed prevalence of each pathogen in the survey. The predicted prevalence was similar to the observed prevalence for E. coli O157 and Salmonella, although the predicted prevalence was higher than that observed in samples tested for Campylobacter.

This study aims to determine the ability of laboratory animal bacteria to resist desiccation and inactivation by hydrogen peroxide vapour (HPV) on paper bedding pieces. Bedding pieces were saturated with bacterial suspensions in water or 2% (w/v) bovine serum albumin (BSA) in water, and held in a mouse facility. Viable counts showed variable survival rates over time for the bacterial species used ([ Pasteurella] pneumotropica, Muribacter muris, Pseudomonas aeruginosa, Acinetobacter redioresistens, Escherichia coli, Klebsiella oxytoca, Bordetella bronchiseptica, Bordetella hinzii, Enterococcus faecalis, β-haemolytic Streptococcus spp., Staphylococcus aureus and Staphylococcus xylosus). Overall, BSA increased bacterial survival in the bedding pieces. The survival rates of Bacillus safensis were not influenced by BSA but depended on sporulation. When bedding pieces and Petri dishes inoculated with E. coli, P. aeruginosa and S. aureus were subjected to HPV disinfection, all bacterial species on the bedding pieces inoculated with bacterial suspensions in water were readily inactivated. By contrast, S. aureus and P. aeruginosa, but not E. coli cells survived HPV treatment in high numbers when inoculated on bedding pieces as a BSA suspension. Notably, all three bacterial species were readily inactivated by HPV even in the presence of BSA when smeared on smooth surfaces. In conclusion, the suspension medium and the carrier can influence the environmental survival and susceptibility of bacterial species to HPV. Our results may help to develop standard protocols that can be used to ensure the microbiological quality of experimental rodent housing.

This study aimed to study the inhibition activity of purified bacteriocin produced from the local isolation Lactococcuslactis ssp. lactis against pathogenic bacteria species isolated from clinical samples in some hospitals Baghdad city. Screening of L. lactis ssp. Lactis and isolated from the intestines fish and raw milk was performed in well diffusion method. The results showed that L. lactis ssp. lactis (Lc4) was the most efficient isolate in producing the bacteriocin as well observed inhibitory activity the increased that companied with the concentration, the concentration of the twice filtrate was better in obtaining higher inhibition diameters compared to the one-fold concentration. The concentrated bacteriocin was purified using the gel filtration column and Sephacryl S-200. The results showed the high inhibitory activity of the purified bacteriocin after the purification against the positive and negative bacteria of the Gram stain under study compared to the one-fold concentration and two-fold before purification , The diameters of the inhibition zones after gel-filtering of the purified bacteriocin reached S. aureus, S. epidermidis, P. aeruginosa, E. coli, E. clocae and S. marcescens (23, 25, 26, 20, 22 and 28) Mm respectively. The carbohydrate content of purified bacteriocin from L. lactis ssp. lactis (Lc4) isolate was 6.02% with a molecular weight of 6310 Dalton. The results showed that purified bacteriocin retained its inhibitory activity at pH 2-10 and showed the highest inhibition at pH 4-6 and lost at pH 12. The purified bacteriocin was characterized by thermal stability. It retained its effectiveness when exposed to 40, 60, 80, 100°C for 30, 15, 5 minutes and 120°C for 15.5 minutes and lost 50% of its effectiveness when exposed to 120°C for 30 minutes. Results The purified bacteriocin was effectively retained when treated with enzyme pepsin and trypsin of 37°C for one hour and at pH 7.

Brucellosis is an infectious disease that affect human as well as animals. The disease is caused by exposure to a bacterial species belong to the genus Brucella. Brucellosis has wide distribution in sub-Saharan African countries, including Sudan in which the diagnosis remains a great challenge and basically depend on serology. The present study was carried out on five dairy cattle farms allegedly free from brucellosis, but with sporadic cases of abortion. As well-known Sudan has a many dairy industries with peri‑urban dairy establishments which built in order to improve milk availability to rural communities. The aim of this study is to estimate the prevalence of brucellosis in Sudan, using samples submitted to collect from different farms. Five farms were tested with Rose Bengal Test (RBT) and Enzyme-linked Immune-absorbent Assay (ELISA). Parallel testing was used to determine whether a farm was to be considered positive or not. The result showed that out of 70 cow’s Blood samples examine 22 (31.4%) had positive RBPT results while the rest (68.6%) had Negative RBPT results. Of the Five farms which were examined the positive cows were observed in only 2 farms. The results show the need to carry out surveillance of brucellosis in cattle in Sudan to understand the spatial distribution of the disease in the country. These surveillance could reduce the risk of Brucellosis spreading and consequently improvement will take place in the quality of livestock and their dairy product which consumed by the human in those areas.
