Обсуждаются результаты анализа последовательности 6 экзона гена CYP11B2 в 40 образцах ДНК пациентов с диагнозом артериальная гипертензия. Выявлена высокая частота мутации в сайте rs4538 (NC_000008.11:g.142913286G>T). Частота носителей аллелей G и Т составила 0,556 и 0,444 соответственно. Уровень содержания альдостерона в сыворотке крови у обследованных в среднем составил 206,1 пг/мл; обнаружена высокая вариация в концентрациях - от 18,18 пг/мл до 933,8 пг/мл. Предварительная оценка ассоциации полиморфизма показала, что при носительстве аллельного варианта Т уровень альдостерона выше (224,0 пг/мл), чем при G (191,7 пг/мл). The paper presents results of processing sequences of 6th exon CYP11B2 taken from 40 DNA samples of patients diagnosed with arterial hypertension. Site rs4538 (NC_000008.11:g.142913286G>T, NM_000498.3:c.1120C>A) revealed a high mutation frequency. The carrier frequencies of allelic variants G and T were 0.556 and 0.444 respectively. The aldosterone content in the blood serum samples averaged 206.1 pg/ml, with high variation in concentrations ranging from 18.18 pg/ml to 933.8 pg/ml. Preliminary evaluation of polymorphism association showed that aldosterone levels were higher for allele T variant (224.0 pg/ml) than for G (191.7 pg/ml).

ABSTRACT This paper describes a mutator system in the nematode Caenorhabditis elegans var. Bergerac for the gene unc-22. Of nine C. elegans and two C. briggsae strains tested only the Bergerac BO strain yielded mutant animals at a high frequency and the unc-22 IV gene is a preferred mutational target. The forward spontaneous mutation frequency at the unc-22 locus in Bergerac BO is about 1 × 10-4, and most of these spontaneous unc-22 mutations revert at frequencies between 2 × 10-3 and 2 × 10-4. Both the forward mutation frequency and the reversion frequency are sensitive to genetic background. Spontaneous unc-22 mutations derived in a Bergerac background and placed in a primarily Bristol background revert at frequencies of <10-6. When reintroduced into a Bergerac/Bristol hybrid background the mutations once again become unstable. The mutator activity could not be localized to a discrete site in the Bergerac genome. Nor did mutator activity require the Bergerac unc-22 gene as a target since the Bristol unc-22 homolog placed in a Bergerac background also showed high mutation frequency. Intragenic mapping of two spontaneous unc-22 alleles, st136 and st137, place both mutations in the central region of the known unc-22 map. However, these mutations probably recombine with one another, suggesting that the unstable mutations can occur in more than one site in unc-22. Examination of the phenotypic effect of these mutations on muscle structure indicates that they are less severe in their effect than a known amber allele. We suggest that this mutator system is polygenic and dispersed over the nematode genome and could represent activity of the transposable element Tc1.

Currently, one of the main economic policies of the Russian Federation in the field of food security is production of safe food and consumer protection. The aim of the research was to study the content of such toxic elements as Zn, Cd and Pb in milk and blood of cows. Experimental studies were carried out at stud farms Plemzavod Tayozhny LLC in Suhobuzimsky District and OPH Solyanskoe LLC in Rybinsk District, Krasnoyarsk Krai, Russian Federation. The milksamples were studied by atomic absorption method; the blood serum samples, by mass spectrometry. Blood serum samples were obtained by sedimentation of whole blood and retraction of the blood clot followed by centrifugation. The obtained data were processed by the method of variation statistics with the calculation of statistical indicators. The lowest contents of Zn, Cd and Pb were found in blood serum of the cows of OPH Solyanskoe LLC; it was 0.038, 0.001, and 0.002 mg/l, respectively. The milk samplescollected at Plemzavod Tayozhny LLCcontained less heavy metals than those collected at OPH Solyanskoe LLC: Zn by 2.069 mg/l, Cd by 0.01 mg/l, and Pb by 0.061 mg/l (P>0.999). When calculating the correlation coefficients, weak positive and negative correlationswere established between Zn and Pb content in blood and milk. The detected amounts of toxic elements did not exceed the maximum permissible concentrations, except for Pb content in milk sampled at OPH Solyanskoe LLC, which was higher than the standard content by 0.026 mg/l.

Aim: Although the levels of cardiac troponin I (cTnI) have proved to be a useful diagnostic biomarker of acute myocardial infarction, there are a wide variety of point-of-care (POC) analysers, which provide measurements of cTnI. The aim of this study was to compare the results obtained by the ADVIA Centaur ultra-assay cTnI assay (us-cTnI), ADVIA Centaur high-sensitive cTnI assay (hs-cTnI) and a POC high-sensitivity assay using PATHFAST. We also aimed to explore total turnaround time (TAT) for laboratory results using the POC PATHFAST analyser. Methods: Samples from 161 patients were taken. Of these samples, 129 were tested with all three assays (us-cTnI, hs-cTnI and PATHFAST), and 32 samples were tested on PATHFAST for the comparison of whole blood, serum and plasma. Results: Comparison of the POC testing methods in this study demonstrated that there are strong linear relationships between all three cTnI assays (us-cTnI, hs-cTnI and POC on PATHFAST). Furthermore, we also show there are strong linear relationships between the two high-sensitive cTnI assays (hs-cTnI and PATHFAST) for blood serum samples, as determined by Passing–Bablok regression analyses. In our comparison of our new data with our older study, the TAT went down. Conclusion: The timeliness of laboratory results is, in addition to accuracy and precision, one of the key indicators of laboratory performance, and at the same time has a significant impact on the course of the patient’s condition. It is therefore important that the laboratory strives to meet the expectations of clinicians regarding the time from the order to the result of the analysis.

It is known that both chronic obstructive pulmonary disease and arterial hypertension are multifactorial diseases, and develop as a result of a complex interaction of genetic and environmental factors. The purpose of this work was to study the metabolism of caspase-7 and caspase-9 in patients with chronic obstructive pulmonary disease (COPD) in combination with arterial hypertension (AH). Materials and methods. We examined 23 patients with a diagnosis of essential hypertension stage II and COPD stage II without a clinically significant concomitant pathology. The plasma level of caspase-7 and caspase-9 was determined using the appropriate ELISA test systems (manufacturer – Bender Medsystems, Austria) at the Medical and Laboratory Training Centre of Zaporizhzhia State Medical University in accordance with the instructions attached to the kit. The analysis of the dynamics of the studied cysteine ​​proteases indicates a statistically significant elevation of these apoptosis markers in the conditions of hypertension and COPD. The value of caspase-7 in the cohort of patients with COPD + AH significantly exceeded similar figures for the groups of patients with a single pathology presented by AH or COPD, and averaged 0.41 ± 0.09 ng / ml. A statistically significant elevation of the caspase-9 level in patients with COPD + AH up to 2.16 ± 0.29 ng / ml compared with other groups also indicates a more significant induction of apoptotic processes in comorbid pathology than in isolated AH or COPD. Conclusion. The obtained results require further clarification of the nature of the relationship between changes in the metabolism of cysteine ​​proteases and the primacy of pathogenetic processes in the mechanisms of risk formation for complications and progression of ventilation disorders in patients with COPD and comorbid essential hypertension.

The paper presents the results of research in which 50 women with essential hypertension illness (arterial hypertension I-II degree) have taken part. 20 patients received long-acting metoprolol therapy (Egilok - retard, EGIS AO) in the dose of 25-100 mg, if necessary in a combination with hydrochlorothiazide 25 mg. In group of comparison (30 subjects) treatment with enalapril (5-10 mg/сут) or by a combination of enalapril and hydrochlorothiazide was administarted. It is shown, that monotherapy by metoprolol-retard allowed to reach a «target» levels of blood pressure in 89,5 % of women, in 10,5 % cases the combined therapy was required. At 12-week of treatment significant improvement of parameters of concentration of attention and anxiety decrease was observed.

Out of 2942 blood serum samples from small ruminants of 10 flocks with a natural course of brucellosis caused by B. melitensis, 322 samples reacted with both antigens in the RID, of which 90 samples only with the O-PS M antigen (from B. melitensis), only with O-PS A-antigen (from B. abortus) reactive was not revealed. In healthy sheep immunized against brucellosis with the vaccine from strain 19 according to different schemes, only the O-PS M antigen was not found to react. Reaction with O-PS A- and M-antigens was observed in animals that were immunized twice subcutaneously at a dose of 40 billion mc. - after 2 months. after revaccination (60%), as well as in those reimmunized conjunctivally at a dose of 4 billion mc. according to the background of primary immunization subcutaneously at a dose of 40 billion mc. (10%) In animals immunized once or twice conjunctivally, reacting in RID with both antigens was not detected. Out of 2432 blood serum samples of small ruminants, 10 flocks with a brucellosis problem immunized against brucellosis with a vaccine from B.abortus strain 19 according to different schemes, 151 samples (6.2%) reacted positively with both O-PS antigens in RID with both O-PS antigens, of which only 86 samples (56.9%) reacted with O-PS M-antigen. The prevalence of indications of RID with O-PS M-antigen over RID with O-PS A-antigen (O-PS antigen made from Brucellae abortus) in small ruminants in one or another flock is characteristic of infection caused by brucellae melitensis at least in the absence, at least in the presence of the fact of immunization with a vaccine from the B. abortus 19 strain. RID with O-PS M-antigen is an objective indicator of epizootic danger and is able to differentiate brucellosis (B. melitensis) in small ruminants from vaccination-induced reactions (B. abortus 19).
