Tempeh extract reduces cellular ROS levels and upregulates the expression of antioxidant enzymes

Tempeh is an Indonesian traditional food produced from soybeans through a mould fermentation involving Rhizopus oligosporus. It is rich in bioactive phytochemicals, including isoflavones that are known to exhibit antioxidant activities. This study aimed to investigate the ability of tempeh ethanol extract to reduce cellular reactive oxygen species (ROS) levels in HepG2 cells in vitro. Tempeh extract exhibited greater total phenolics, total flavonoids and free radical inhibition capacity than soybean extract. Both tempeh extract and soybean extract reduced the basal levels of cellular ROS in the cells, but tempeh extract induced higher expression of antioxidant enzymes [catalase, superoxide dismutase-2 (SOD2) and superoxide dismutase-3 (SOD3)] compared to soybean extract. This study provides novel evidence suggesting the ability of tempeh to tackle cellular oxidative stress by upregulating the expression of antioxidant enzymes. These findings would give an insight into the potential of tempeh to be developed as a functional food beneficial for human health.

Crab apple (Malus baccata (L.) Borkh.) was mainly distributed in Europe as an ornamental plant, but the nutritional properties of its edible fruits were not fully revealed. The aim of the current study was to characterize the phytochemical composition of ripen carb apple fruits and to evaluate their nutritional and antioxidant potentials. The fruits were assayed for moisture and ash content, proteins, lipids, carbohydrates, titratable acidity (TA), pH, total phenolic compounds and natural pigments. Among the analyzed carbohydrates cellulose was found in the highest content (6% dw), followed by sugars (sucrose, glucose and fructose) and 1.8 % dw uronic acids. The total chlorophylls and carotenoids contents in their fruits were 6.51 and 4.80 μg/g fw, respectively. Total monomeric anthocyanins were not detected. The highest content of total phenolic compounds (2.67 mg GAE/g fw) was found in 95 % ethanol extract from fruits, while the total flavonoids were relatively low – 0.1 mg QE/g fw. DPPH assay (17.27 mM TE/g fw) and FRAP assay (14.34 mM TE/g fw) demonstrated in vitro antioxidant activities of crabapple. Malus baccata fruits were evaluated as a rich source of dietary fibers and phenolic compounds with significant antioxidant potential that could be used in human nutrition.

Introduction: The immune-modulatory effects of synbiotics and their ability to reduce free radical levels may be useful for functional food that is able to be active throughout whole period of colonization of the gastrointestinal tract.The aim of the present study was to investigate the immune-modulatory and antioxidant effects of the synbiotic product "NАR," a probiotic beverage.Methods: The presence of IL-2, IL-4, IL-6, IL-8, IL-10, αTNF, γIFN, Ig A, Ig M, and Ig E was studied in vitro using a solid immunosorbent analysis. The total antioxidant activities of superoxide dismutase and glutathione reductase were determined by a spectrophotometry using the Sigma-Aldrich sets.Results: Studies of the immune-modulatory properties of the synbiotic product NAR showed 1.7 fold increase of γINF levels (p<0.01) in blood after consumption of the synbiotic product “NAR” in comparison to control values, whereas the concentrations of IL-4 and Ig E decreased 2.0 times (treatment: 9.3; control: 18.7; p<0.01) and 1.3 times (p<0.1), respectively. The consumption of the synbiotic product “NAR” caused an increase in the proportion of γINF/IL 4 (treatment: 15.4; control: 4.4; p<0.01), which indicates a reduction in functional activity of Th2-type lymphocytes in comparison with the function of Th1 cells.Our study showed a high level of the total antioxidant activity of the synbiotic product (67.4 mmol/ml). The antioxidant activity of the intact cells of consortium (15.3 mM/ml), which was the basis for the preparation of the symbiotic product, is several times lower than the activity observed in the symbiotic samples.Expression of SOD is one of the mechanisms of antioxidant stress radicals inactivation by bacteria. The analysis identified a superoxide dismutase activity of synbiotic product (1.42 U/mg protein). A glutathione reductase activity of the synbiotic product was elevated (0.06 U/ml). Conclusion: The majority of the inflammatory mediators found in the blood after the consumption of symbiotic product NAR were inflammatory mediators that activate a cellular component of the resistance. Moreover, the symbiotic product has a high antioxidant activity.

Consumers are interested in antimicrobial by-products from natural sources. The present study aims to estimate the antibacterial effects of different concentrations and solvent extracts such as methanol, ethanol, and water extract of pomegranate peels with the milk quality application in-vitro. Listeria monocytogens, E. coli, Salmonella, and Staphylococcus aureus were used as bacteria growth indicators in two pomegranate varieties (i.e., Wonderful and Francis). Besides, evaluating somephysical characterization, antibacterial and antioxidant activities. Results showed that the highest inhibition zone of 18.88 mm was observed with 90% ethanol extract of Wonderful variety against Listeria monocytogens, at 0, 12.5, 25, and 50 mg/mL concentrations, while the lowest inhibition zone of 11.29 mm was observed with 90% methanol extract of Francis variety, at 12.5 mg/mL concentration. The milk treated with WPE of Wonderful variety maintained the bacterial growth from 1.19 ±0.29 x 109 to 1.43 x 1011 CFU/mL at 4 °C. Punicalagin was the most abundant polyphenolic compound (165.05–190.43 µg/mL) followed by gallic acid (153.08–177.65 µg/mL), and p-Hydroxybenzoic acid (91.29–92.5 mg/mL). Acetaldehyde (23.09–27.15 PPM), followed by acetoin (9.65–15.33 PPM) were the most predominant volatile compounds. The WPE treated milk maintained the sensory evaluations such as taste, color, texture, and overall acceptance longer time longer time (i.e., 144 h) than FPE treated milk (i.e., 168 h) at 4°C. DPPH and ABTS radical scavenging of capacity (IC50 values) of FPE treated milk were found to be lower than WPE treated milk (i.e., 159.65 µg/mL and 131.87 µg/mL), respectively. While, FPE treated milk reported a higher viscosity content than WPE treated milk (13.11 cP). In conclusion, pomegranate peel extracts especially Wonderful variety may be used to prolong the shelf-life of milk in dairy products manufacturers.

Alzheimer's disease (AD) is linked to an extensive neuron loss via accumulation of amyloid-beta (Aβ) as senile plaques associated with reactive astrocytes and microglial activation in the brain. The objective of this study was to assess the therapeutic effect of WS-5 ethanol extract in vitro and in vivo against Aβ-induced AD in mice and to identify the extract’s active constituents. In the present study, WS-5 exerted a significant inhibitory effect on acetylcholinesterase (AChE). Analysis by transmission electron microscopy (TEM) revealed that WS-5 prevented Aβ oligomerization via inhibition of Aβ1-42 aggregation. Evaluation of antioxidant activities using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) demonstrated that WS-5 possessed a high antioxidant activity, which was confirmed by measuring the total antioxidant status (TAS). Furthermore, the anti-inflammatory properties of WS-5 were examined using lipopolysaccharide-stimulated BV-2 microglial cells. WS-5 significantly inhibited the lipopolysaccharide–induced production of nitric oxide and two proinflammatory cytokines, TNF-α and IL-6. The memory impairment in mice with Aβ-induced AD was studied using the Morris water maze and passive avoidance test. Immunohistochemistry was performed to monitor pathological changes in the hippocampus and cortex region of the mouse brain. The animal study showed that WS-5 (250 mg/kg) treatment improved learning and suppressed memory impairment as well as reduced Aβ plaque accumulation in Aβ-induced AD. HPLC analysis identified the extract’s active compounds that exert anti-AChE activity. In summary, our findings suggest that WS-5 could be applied as a natural product therapy with a focus on neuroinflammation-related neurodegenerative disorders.

SummaryIncreased intracellular reactive oxygen species (ROS) promote platelet activation. The sources of platelet-derived ROS are diverse and whether or not mitochondrial derived ROS, modulates platelet function is incompletely understood. Studies of platelets from patients with sickle cell disease, and diabetes suggest a correlation between mitochondrial ROS and platelet dysfunction. Therefore, we generated mice with a platelet specific knockout of superoxide dismutase 2 (SOD2-KO) to determine if increased mitochondrial ROS increases platelet activation. SOD2-KO platelets demonstrated decreased SOD2 activity and increased mitochondrial ROS, however total platelet ROS was unchanged. Mitochondrial function and content were maintained in non-stimulated platelets. However SOD2-KO platelets demonstrated decreased mitochondrial function following thrombin stimulation. In vitro platelet activation and spreading was normal and in vivo, deletion of SOD2 did not change tail-bleeding or arterial thrombosis indices. In pathophysiological models mediated by platelet-dependent immune mechanisms such as sepsis and autoimmune inflammatory arthritis, SOD2-KO mice were phenotypically identical to wildtype controls. These data demonstrate that increased mitochondrial ROS does not result in platelet dysfunction.

Chinese Jujube has long been used for human consumption and medicinal purposes in China. The objective of this study was to extract active substance from the powder of fermented Chinese Jujube and to detect their antioxidant capacities. Macroreticular resin was used to separate FJ-1 from 60% ethanol extract, and silica gel column was used for further separation of FJ-2. Precipitation of CPJ was conducted by addition of 95% ethanol to the water extract. The ability to eliminate DPPH•, O2• and •OH of this component was analyzed. Their activities show dose dependence character. FJ-1, FJ-2 show excellent antioxidant capacity on eliminating DPPH• and •OH, but CPJ shows lower capacity. The capacity on eliminating O2• of CPJ is higher than FJ-1, FJ-2 at the same concentration.

The purpose of this research work is to evaluates the in-vitro antioxidant and antioxidant enzymes inhibition potentials of Aframomum melegueta [Roscoe] K. Schum (Grains of Paradise) Leaf, Stem Bark, Seed Bark and Seed Extracts. Aframomum melegueta is a spice with a similar composition as Ginger, that belongs to the same Zingiberaceae family, used in Nigeria, West Africa and it is used for the treatment of infectious diseases such as urinary tract infections, cancer and diabetes. Antioxidants neutralize the effect of free radicals through different ways and may prevent the body from various diseases. The plant extract were collected from a rain forest in Akugba Akoko, Ondo state, Nigeria. The plant sample was dried, pulverized, filtered with Whatman No 1 filtered paper and the filtrates was concentrated in-vacuum using vacuum rotary evaporator at 40°C and was later concentrated to dryness in a hot-air oven at 40°C. Assessment of antioxidant potentials were performed using Singleton method Total flavonoids (mg/g), Phenol (mg/g), Ferric reducing (FRAP) potentials (mg/g), Free radical scavenging ability (DPPH) (1, 1- diphenyl-2-picryhydrazyl) potentials (%),2,2’-azino-bis(3-ethylbenthiazoline-6-sulphonic acid), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) scavenging potentials (mMol/g), Fe2+ chelation potentials (%) and OH Radical Scavenging potentials (%) were evaluated. Antioxidant enzymes of plant were assessed using Bentller method, Superoxide Dismutase (SOD) (%), Reduced Glutathione (GSH) (%), Catalase (CAT) and Glutathione Peroxidise (GPX) were evaluated. Seed extract among other extracts of Aframomum melegueta has the highest quantity of flavonoids, Phenol, FRAP, DPPH, ABTS, Fe2+ chelation and OH radical scavenging potential. The leaf extract of A. meleguata has the highest percentage of Superoxide Dismutase (SOD) and Catalase (CAT) while stem bark of Aframomum melegueta has the highest percentage of Reduced Glutathione (GSH) and Glutathione Peroxidise (GPX).it can be deduced from this research work that all parts of the A. melegueta has pharmacological and therapeutic activities judging from literature that A. melegueta can be used to prevent oxidative damage by ROS (Reactive oxygen species) reacting with free radical chelating and catalytic metals  which can be used to reduce and prevent various diseases like heart disease, cancer, DNA degeneration, pulmonary and neurological disorder. SOD, GPX, GSH help to control cytokins induced peroxidise level and mediate signal transduction in mammalian cells. It can be deduced that Aframomum melegueta should be the focal point in human diet, natural herbal drug supplement and even in addition to conventional drug to improve the potency and quality of natural drug system. The use of Aframomum melegueta should be encouraged. These results show the potential of Aframomum melegueta as a source of bioactive compounds to be used for pharmaceutical, nutraceutical, and cosmeceutical applications.
