Introduction: The immune-modulatory effects of synbiotics and their ability to reduce free radical levels may be useful for functional food that is able to be active throughout whole period of colonization of the gastrointestinal tract.The aim of the present study was to investigate the immune-modulatory and antioxidant effects of the synbiotic product "NАR," a probiotic beverage.Methods: The presence of IL-2, IL-4, IL-6, IL-8, IL-10, αTNF, γIFN, Ig A, Ig M, and Ig E was studied in vitro using a solid immunosorbent analysis. The total antioxidant activities of superoxide dismutase and glutathione reductase were determined by a spectrophotometry using the Sigma-Aldrich sets.Results: Studies of the immune-modulatory properties of the synbiotic product NAR showed 1.7 fold increase of γINF levels (p<0.01) in blood after consumption of the synbiotic product “NAR” in comparison to control values, whereas the concentrations of IL-4 and Ig E decreased 2.0 times (treatment: 9.3; control: 18.7; p<0.01) and 1.3 times (p<0.1), respectively. The consumption of the synbiotic product “NAR” caused an increase in the proportion of γINF/IL 4 (treatment: 15.4; control: 4.4; p<0.01), which indicates a reduction in functional activity of Th2-type lymphocytes in comparison with the function of Th1 cells.Our study showed a high level of the total antioxidant activity of the synbiotic product (67.4 mmol/ml). The antioxidant activity of the intact cells of consortium (15.3 mM/ml), which was the basis for the preparation of the symbiotic product, is several times lower than the activity observed in the symbiotic samples.Expression of SOD is one of the mechanisms of antioxidant stress radicals inactivation by bacteria. The analysis identified a superoxide dismutase activity of synbiotic product (1.42 U/mg protein). A glutathione reductase activity of the synbiotic product was elevated (0.06 U/ml). Conclusion: The majority of the inflammatory mediators found in the blood after the consumption of symbiotic product NAR were inflammatory mediators that activate a cellular component of the resistance. Moreover, the symbiotic product has a high antioxidant activity.

Introduction: Available evidence suggests that probiotics have different biological functions that depend on several mechanisms, such as antioxidant and DNA-protective activities. The probiotic consortium includes bacterial cultures such as Streptococcus thermophilus, Lactococcus lactis, Lactobacillus plantarum, and other bacterial cultures isolated from traditional Kazakh dairy products (ayran, kumys, shubat, and healthy clinical material). The aim of this study was to investigate the total antioxidant activity of the consortium of probiotic bacteria and to determine the activity of superoxide dismutase, glutathione reductase, and DNA-protective action.Material and methods: In vitro comet assay was used to determine the antigenotoxicity of the probiotic consortium. Total antioxidant activity was determined using a method of analysis with Trolox as the equivalent. The analysis method of superoxide dismutase activity assesses the inhibition rate of the nitroblue tetrazolium reduction to formazan by superoxide dismutase. Determination of glutathione reductase activity is based on the measurement of the NADPH oxidation speed.Results: A significantly high level of the total antioxidant activity of the probiotic consortium intact cells (15.3 mM/ml) was observed whereas the activity index of  lysate  was 11.1 mM/ml.The superoxide dismutase activity of probiotic consortium lysate was evaluated, with values that peaked at 0.24 U/mg protein. The superoxide dismutase activity of the consortium was lower in comparison to L.fernentum E-3 and L.fernentum E-18 cultures with values of 0.85 U/mg and 0.76 U/mg protein, respectively. SOD activity of probiotic consortium whole cells was not observed, which is typical for lactic acid bacteria.Glutathione reductase plays an important role in the optimal protection from oxidative stress. Glutathione reductase activity of the studied probiotic consortium was low; moreover, the activity of the lysate was two times higher than the activity of the cells reaching 0.01 units/ml. Investigations by Dr. Li have shown that the intracellular glutathione may give a significant protection of Lactococcus from the damaging action of H2O2, even at very low concentrations.The data from our study suggests that the co-incubation of the epithelial cells with probiotic bacteria reduces the percentage of damaged cells (damage index–0.60).Conclusion: The studied probiotic consortium has antigenotoxic and antioxidant activities. Preparations and products of this probiotic consortium may serve as a protective component in the intestinal microbial ecosystem.

<p>In the present work methanolic and water extracts of 19 medicinal plants, commonly used in Sudanese folk medicines against gastrointestinal tract, infectious diseases were investigated for their antioxidant activity. Antioxidant assays were carried out by using different in vitro models such as DPPH (2, 2-diphenyl-1picrylhydrazyl) radical scavenging and Iron chelating activity. Methanolic and water extracts of <em>Punica granatum </em>showed the highest total antioxidant activity of 71±0.03 and 73±0.01 respectively. The antioxidant activities of water extracts were poor when compared to the methanol extract. The iron chelating power of methanol and aqueous extracts of <em>Commiphora myrrha</em> 58±0.01 and <em>Azadirachta indica</em> 57±0.20 respectively.</p>

Background Amaranthus hybridus L. is an annual, erect or less commonly ascending herb that is a member of the Amaranthaceae family. Polysaccharides extracted from traditional Chinese medicines may be effective substances with antioxidant activity. Methods In this study, we isolated crude polysaccharides from A. hybridus (AHP-M) using microwave-assisted extraction. Then, the AHP-M was purified by chromatography with DEAE-32 cellulose, and two fractions, AHP-M-1 and AHP-M-2, were obtained. The structural characteristics of AHP-M-1 and AHP-M-2 were investigated, and their antioxidant activities were analyzed in vitro. Results We found that the monosaccharide composition of AHP-M-1 was different from that of AHP-M-2. The molecular weights of AHP-M-1 and AHP-M-2 were 77.625 kDa and 93.325 kDa, respectively. The results showed that the antioxidant activity of AHP-M-2 was better than that of AHP-M-1. For AHP-M-2, the DPPH radical scavenging rate at a concentration of 2 mg/mL was 78.87%, the hydroxyl radical scavenging rate was 39.34%, the superoxide anion radical scavenging rate was 80.2%, and the reduction ability of Fe3+ was approximately 0.90. The total antioxidant capacity per milligram of AHP-M-2 was 6.42, which was higher than that of Vitamin C (Vc). Conclusion The in vitro test indicated that AHP-M-1 and AHP-M-2 have good antioxidant activity, demonstrating that A. hybridus L. polysaccharide has immense potential as a natural antioxidants.

The present study aimed to determine the total polyphenol content and total antioxidant activities of ten selected seasonal fruit extracts grown in Sri Lanka. The aqueous refluxed fruit extracts (4h) were used at the initial concentration of 0.05 g/mL. The total polyphenol content was determined according to the Folin-Ciocalteu method. The total antioxidant activity was evaluated by DPPH (2, 2-diphenyl-2-picrylhydrazyl), FRAP (ferric reducing power) and TBA (thiobabituric acid assay) assays with L-ascorbic acid as the reference compound. The total polyphenol content of the fruit extracts varied from 0.81±0 to 17.54±0.50 mg GAE (gallic acid equivalents) per gram of dry weight. The antioxidant activities ranged in IC50 of 46.60±0.60 to 367.90 ± 4.90 μg/mL, 3.41±0.02 to 50.46±1.02 µM, 30.62±0.50 to 182.64±0.90 mg AAE (L-ascorbic acid equivalents) per gram of dry weight for DPPH, FRAP, TBA assays, respectively. All aqueous fruit extracts exert dose dependent in vitro antioxidant activities in different degrees. Among the selected fruit extracts S. carophylatum, P. zeylanica, A. comosus exert relatively high total antioxidant activity together with high total polyphenol content. Hence, the selected fruit extracts are deserved to be as potent sources for antioxidant lead compounds in the development of nutraceuticals using the above edible fruits.

Objective: The aim of this study was focussed on phytochemical analysis, in vitro antioxidant activities of different solvent extracts of Hypoestes phyllostachya leaf. Methods: The leaves were washed efficiently, shade dried and coarsely powdered. The powdered leaf material of Hypoestes phyllostachya was successively extracted with hexane, chloroform, methanol and water using Soxhlet apparatus. Preliminary phytochemical screenings for polyphenolics, proteins, phytosteroids, flavonoids, tannins, glycosides, alkaloids carbohydrates, polyphenolics, saponins were done by following standard procedure. In vitro antioxidant activities of various solvent extracts were assessed using DPPH, ABTS, Hydroxy radical, total antioxidant capacity and reducing power assay. Polyphenolics and flavonoids were also estimated. Results: The results showed that most of the phytochemical compounds such as carbohydrates, proteins, steroids, fixed oils and fatty acids, flavonoids, alkaloids, saponins, tannins, polyphenolics and glycosides present in the methanolic and aqueous extract of Hypoestes phyllostachya. The in vitro antioxidant activity of Hypoestes phyllostachya leaf clearly showed that it has better antioxidant activity, which was confirmed by different assays. Conclusion: From this work, it can be concluded that Hypoestes phyllostachya has the potential to be a good antioxidant but have only a lesser amount of flavonoids and polyphenolic contents. This was the first report on the pharmacological activity of the Hypoestes phyllostachya.

Background: Stilbene phytalexis (1,2-diphenyloethylen) and benzamide are beneficial for human health. To increase the stilbene ring activity, a new series of its derivatives containing benzamide structure was synthesized and evaluated for their in vitro antioxidant power. Methods: 1H nuclear magnetic resonance, mass spectroscopy, and chromatographic analyses were used to confirm the successful synthesis. The antioxidant properties were determined by the elimination of , HO , DPPH , ABTS+ radicals, total antioxidant status (TAS) and the ferric reducing antioxidant activities (TAC) measurements. Results: Stilbenebenzamide compounds showed a wide spectrum of antioxidant ability, however their total antioxidant power was weaker than those of butylated hydroxytoluene (BHT), ascorbic acid, and resveratrol. The highest antiradical activity towards and HO was shown by the compounds with structures containing amine group (SBEBA, SBA) ( : 37.7 – 38.0% and 40.8 – 43.5%, HO : 29.8%, 28.7% inhibition, respectively) at1.25 mM concentration. The antiradical power of SBEBA (0.29) in DPPH assay was lower than those of resveratrol (1.83), ascorbic acid (3.63) and BHT (4.09). The TAS values of the synthesized compounds ranged from 152.9±5.3 to 240.2±6.7µM trolox equivalent/gram (TE/g) and were much lower than those of BHT (1304±43.0), reservatrol (1360±29.0) and ascorbic acid (2782±39.7) µM TE/g. Similarly, the TAC values ranging from 29.7±0.9 to 41.5±1.6 µM TE were weaker than that of resveratrol (239.2 ±6.7 µM TE/g). Conclusion: The results suggest that the presence of hydroxyl group in stilbene ring should be considered in further design of stilbenebenzamide compounds to enhance their antioxidant activity.

Leafy vegetables are an excellent source of bioactive factors, traditionally used as important medical ingredients. Recently, some leafy vegetables are domesticated without the use of fertilizer, as well as the assessment of the effect of fertilizer on their nutritional value. This study aims at testing the effect of three mineral and organic fertilizers (Cowpat, NPK and NPK + cowpat) on three traditional leafy vegetables: Ceratotheca sesamoïdes, Sesamum radiatum and Justicia tenella. Their antioxidant activities were assessed at different harvesting times varying from six (6) to fourteen (14) Weeks After Transplantation (WAT) using three in vitro methods: Ferric reducing antioxidant power (FRAP), 2,2’-Azinobis3-ethylbenzothiazoline-6- sulfonic acid (ABTS) and 2,2-diphenyl-1-picryl hydrazil (DPPH). The FRAP, DPPH, ABTS assays were consistent and positively correlated (p < 0.001). Total antioxidant activities of leafy vegetables depended on species, harvesting time and methods. They ranged from 32.0 to 45.7 µmol Fe/g DW (for FRAP), with non fertilized J. tenella cut at 12 WAT giving the highest antioxidant activity. Percentage of inhibition using DPPH assay ranged from 11.4 to 87.2 % and showed that J. tenella fertilized with NPK and cowpat, and harvested at 9 WAT had the highest antioxidant activity. Regarding ABTS, the range of 17.6 to 28.9 µmol TE/g DW was recorded, and the leaves of C. sesamoïdes harvested at 10 WAT and fertilized with cowpat showed the highest level. Compared to other species, those studied here may best contribute to improve human health related to degenerative diseases. Moreover, significant and positive correlations were observed between the total phenolic compounds content and antioxidant activities of leaves regardless the methods used. The positive and significant correlations between the three assays (FRAP, DPPH, ABTS) allow to suggest the use of only anyone of them to check factors in the study. This paper highlights the potential of antioxidant capability of the leafy vegetables even fertilized.

Aim and Objective: Currently, the use of ingredients from natural sources has gained great attention in cosmetic field especially for the development of new photoprotective formulations. Therefore, the present study aimed to evaluate the cosmetic potential of the methanolic crude extract and the ethyl acetate fraction of the medicinal halophyte Tamarix gallica (Tg) growing in the area of Tebessa in eastern of Algeria, by assessing their phenolic and flavonoid contents, photoprotective and antioxidant activities. Methods: The research approach consisted of determination phenolic and flavonoid contents of aerial parts via FolinCiocalteu and aluminum chloride methods respectively. The antioxidant activity was measured through two in vitro methods DPPH radical scavenging activity and total antioxidant capacity test (TAC). The in vitro photoprotective effect was performed according to the parameter SPF (Sun Protection Factor) by using UV spectroscopic method in UV-B region (290-320 nm). Results: The methanolic extract (Tg-MeOH) and ethyl acetate (Tg-EtOAc) fraction showed good antioxidant activity with IC50’s 14,05±0,66, 27,58±1,98 µg/mL respectively in DPPH test. Furthermore, both extracts displayed strong total antioxidant capacity (287.01±7.85, 246.7±1.12 mg AAE/g, respectively) in TAC test. Both extracts exhibited high photoprotective activity, with sun protection factor (SPF) values 37.03±0.22 and 36.08±0.03. The antioxidant and photoprotective activities of these extracts is probably related to polyphenols content (190.27±0.74 mg AGE /g and 121.77±1.29 mg AGE /g, respectively) , and flavonoids (78.75±0.06 mg QE /g and 58.67±1.19mg/g). Conclusion: Our finding show that extracts of Tamarix gallica L could be a promising source to be mixed as natural sunscreens and antioxidants agent into photoprotective cosmetic formulations.

<p>The objective of the work reported in this paper was to evaluated effects of temperatures extraction on <em>in vitro</em> antioxidant activities of polysaccharides from the <em>Ophiocordyceps sobolifera. </em>The antioxidant capacity of <em>Ophiocordyceps sobolifera </em>polysaccharide was evaluated to use  2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and total antioxidant activity methods. From this study, it could be concluded that the highest antioxidant activity of the polysaccharides could be obtained from extraction temperature 80 <sup>o</sup>C. Total antioxidant capacity of polysaccharides from <em>Ophiocordyceps sobolifera </em>  contained from 0.1183 ± 0.0011 to 0.1454 ± 0.0021 mg GA/g or from 0.1105 ± 0.0004 to 0.1215 ± 0.0006 μmol AS/g. Polysaccharides has the most potential antioxidant activity with the lowest half maximal inhibitory concentration (IC<sub>50</sub>) value range 0.97 mg/mL to 1.06 mg/mL.</p><p> </p>

<p><strong>Abstract</strong><strong></strong></p><p>The present work examined the <em>in vitro</em> antioxidant activity of sequentially extracted, solvent extracts (Petroleum ether, chloroform and Methanol) of stem bark, leaves and callus from <em>Amoora rohituka</em>. The antioxidative capacity of the extracts were examined by using DPPH (1,1-diphenyl,2-picryl hydrazyl), ABTS (2,2’-azinobis-3-ethyl-benzothiozoline-6-sulphonic acid) and NOS (Nitric oxide scavenging) assays by using Gallic acid as standard control. Among the tested extracts, methanol extract of stem bark showed better antioxidative capacity. The total antioxidant activity was expressed as µg equivalent of Gallic acid per gram on dry weight basis. IC<sub>50</sub> values for methanol extract of stem bark were 7.53 µg/ml; 34.92 µg/ml and 56.31 µg/ml in ABTS, DPPH and NOS assays respectively. Whereas, leaves and callus extracts showed moderate antioxidative properties. </p>
