Identification of secondary metabolites, antibacterial and antioxidant activities assay of methanol extract of sea cucumber Stichopus hermanii were conducted from May to July 2011 at the Natural Products Laboratory, Research Center for Oceanography, Indonesian Institute of Sciences. Sea cucumber used in this study comes from South Lampung waters. Objective of the study was to get information of secondary metabolites, antibacterial and antioxidant activities of methanol extract of sea cucumber S. hermanii. The extraction method which used in this experiment was the maceration method using methanol solvent. Identification of secondary metabolites was performed through observing reaction of colors, precipitation, and foaming. Antibacterial and antioxidant activities of methanol extract from sea cucumber were tested using agar diffusion method and reducing of free radicals 1.1-Diphenyl-2-Picrylhydrazyl (DPPH) respectively. The results showed that secondary metabolites identified in the methanol extract of sea cucumber S. hermanii were saponin and steroids. Both secondary metabolites had antibacterial activities against Staphylococcus aureus, Vibrio eltor and Bacilus subtilis. The analysis of antioxidant activity showed that the IC50 value of methanol extract of sea cucumber S. hermanii was about 65.08 ppm. It indicated that S. hermanii is having potency as antibacterial and antioxidant. Keywords: antibacterial, antioxidants, sea cucumber Stichopus hermanii, secondary metabolites

Bohadschia sp. is one of the sea cucumber species that has potential to be developed as a source of antibacterial from the sea. Samples of sea cucumber Bohadschia sp. used in this study collected from the Ratai bay waters, Lampung. This study aims to determine the type of secondary metabolites, antibacterial activity and compound composition analysis containing in the sea cucumber extract. Identification of secondary metabolites by observation of color reactions, precipitation and foam. The method used to antibacterial activity test was the agar diffusion method, while identification of the composition of compounds performed with Gas Chromatogaphy-Mass Spectroscopy (GC-MS) method.Top of FormThe results showed that the type of secondary metabolites contained in the extract of sea cucumber Bohadschia sp. were steroids and saponins. The extract of sea cucumber Bohadschia sp. showed antibacterial activity against Bacillus subtilis and Vibrio eltor. Results of GC-MS were 12 compounds and have a similarity index same or more than 90%. All compounds consist of organosilicon cyclic, fatty acid, steroid, cyclo alkene and alkena. The compound with biggest abundance was cholest-5-en-3-yl nonanoate (4.89%) and retention time was 37.370 minutes.

The antibacterial activity of three Cupressaceae plants (Thujaoccidentalis,ThujaorientalisandChamaecyparisobtusa) was tested against three bacteria using the agar diffusion method. The ether and ethylacetate fraction of crude methanol extract from the three plants showed potent antibacterial activity against the tested microorganisms. The result showed that Staphylococcus aureus revealed the most sensitivity among the tested bacteria. Thujaoccidentalisether fraction and Thujaorientalis hexane fraction exhibited the highest antibacterial activity against Staphylococcus aureus. E. coli was shown the highest MIC values compared to the other two tested bacteria, which indicates the lowest antibacterial activity against the bacterium. This study promises an interesting future for designing a potentially active antibacterial agent from the three Cupressaceae plants.

The aim of this study is to investigate in vitro antioxidant and antibacterial activities of the aqueous and hydroethanolic extracts for aerial parts of Herniaria hirsuta. Extracts were screened for their possible antioxidant activities by three tests: DPPH free radical-scavenging, reducing power, and molybdenum systems. The screening of antibacterial activity of extracts was individually evaluated against sixteen bacteria species using a disc diffusion method. Flavonoids, total phenols, and tannins content were performed for both extracts. It shows higher content in the hydroethanolic extract. The hydroethanolic extract showed a significant antioxidant activity for the three methods studies to the aqueous extract, but nonsignificant results compared to the reference (BHT). However, both extracts have negative effect on the strains studies for the antibacterial activity.

Abstract Identification of sea cucumbers from Benteng Inong Balee, Aceh Besar and their phytochemistry screening were conducted in December 2020 to January 2021 at Laboratory of Marine Chemistry and Fisheries Biotechnology, Universitas Syiah Kuala. The purpose of this study was to identify the species of sea cucumbers and its secondary metabolite content using phytochemistry screening and column chromatography. The species of sea cucumbers that were identified was Holothuria atra. The extraction method used in sea cucumber extraction was maceration method, while the separation of secondary metabolites used column-chromatography with eluent of n-hexane : ethyl acetate (8:4). The results showed that secondary metabolites obtained from phytochemical tests were flavonoids, saponins and triterpenoids.


Annona muricata is one of the important herbal plant that are widely used to treat antidiabetic, anti-inflammatory, insecticidal, antimalarial, anticancer, antibacterial and antioxidant activities. In the present study, extraction procedure was carried out in few step processes method. The extract contains high percentage of steroid, alkaloid, flavonoid, phenolic and saponin. The extract was used to study the antioxidant activity; antibacterial activities and anticancer activity by standard methods. The antioxidant activity was studied by using radical scavenging DPPH, FRAP and H2O2 method and disc diffusion method. The results of antibacterial activity of the following bacteria’s such as Pseudomonas, Staphylococcus, Bacillus, E.coli shows the maximum zone of inhibition against Staphylococcus aureus which is around 1.7 cm. The anticancer activity was carried out by MTT assay using Hep-G2 as cell line and results are reported in the paper.

Purpose The purpose of this paper is to extract, characterise and quantify curcumin from selected Zingiberaceae of “kunyit” or turmeric (Curcuma longa), “temu lawak” or Javanese turmeric (Curcuma xanthorrhiza), “temu pauh” (Curcuma mangga), “lempoyang” (Zingiber zerumbet) and “bonglai” (Zingiber cassumunar) using alkaline and chemical-based extraction method for antimicrobial and antioxidant activities. Design/methodology/approach Through the alkaline-based extraction method, all parts of rhizome samples were freeze-dried for 72 h before grounded into a fine powder and kept at −20°C. The powdered sample (0.1 g) was weighed and placed in a 50 mL tube. About 20 mL of 2 M NaOH solution was added into the tube. The solution was allowed to stand for 30 min. Then, 20 mL of ethyl acetate was added into the tube. The solution was mixed well then centrifuged at 13,500 rpm for 3 min. The upper layer was collected using a pipette. The process was repeated until the upper layer became almost colourless. The collected ethyl acetate solution was concentrated using a rotary evaporator to remove the ethyl acetate from the extracted compound. The concentrated curcumin was placed in a universal bottle, which was then dried from the remaining ethyl acetate using nitrogen drying process. The dried curcumin was then stored inside the freezer at −20ºC. The antimicrobial activities were using agar diffusion method against bacterial and fungi, while the antioxidant activity was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging assay. Findings All the samples successfully showed a single peak (curcumin) that gained from the high-performance liquid chromatography (HPLC) chromatogram analysis (at 425 nm) using the alkaline-based extraction method and the highest curcumin content was in turmeric (12.95 ± 1.07mg/g DW). At 10.0 mg/mL curcumin concentration, the best antibacterial activity was against on methicillin-resistant staphylococcus aureus (MRSA) with 7.50 ± 0.71 mm inhibition zone, while the best antifungal activity was against on Aspergillus niger with 8.00 ± 0.41 mm inhibition zone. The DPPH antioxidant test resulted in the highest inhibition (110.41 per cent) was at 0.25 mg/mL curcumin concentration. Originality/value Through HPLC analysis, all samples successfully showed a single peak of curcumin at 425 nm. The total carotenoid determination from turmeric revealed that the samples content was substantially higher using alkaline-based extraction (18.40 ± 0.07 mg/g DW) compared to chemical-based extraction (9.42 ± 0.20 mg/g ± SD).

Red galangal (Alpinia purpurata) is a plant that belongs to the Zingiberaceae family. Red galangal rhizome is traditionally used to treat phlegm and ringworm. Red galangal rhizome contains secondary metabolites of essential oils, eugnol, sesquiterpenes, pinen, kaemferida, galanagn and galangol which have anti-fungal properties. This study aims to determine the antifungal activity of red galangal rhizome ethanol extract against Malasezia furfur and Microsporum canis using agar diffusion method. Red galangal extract is made with a concentration variation of 2%, 5% and 10%. The results showed the antifungal activity of red galangal ethanol extract in the form of inhibiting diameters at an average concentration of 2%, 5% and 10% against Malasezia furfur was 16.77 mm, 17.91 mm and 19.72 mm. The average inhibitory diameter of Microsporum canis is 10.58 mm, 13.57 mm and 14.51 mm. Based on the results of this study it can be concluded that the ethanol extract of red galangal has antifungal activity against Malasezia furfur and Microsporum canis.

Antioxidant activity tests of methanol extracts of velvet beans (Mucuna pruriens) and velvet beans (Mucuna pruriens) in coffee preparations have been previously carried out by several researchers. This present study aims to examine the phytochemical and antioxidant activities of methanol extracts of Mucuna pruriens and Mucuna pruriens in coffee preparations. The methanol extract was tested for the phytochemical and antioxidant activities by applying DPPH (1,1-Dhiphenyl-2-picrylhydrazil) method. The results of the phytochemical test showed that the methanol extracts of the Mucuna pruriens and Mucuna pruriens in coffee preparations contained secondary metabolites of flavonoids, alkaloids, saponins, terpenoids, and tannins. The outcomes of the antioxidant activity test revealed that the methanol extracts of the Mucuna pruriens and Mucuna pruriens in coffee preparations had antioxidant activities, as indicated by the IC50 values of 42.09 ppm and 37.23 ppm. It was also revealed that the antioxidant content of Mucuna pruriens in coffee preparations was stronger than the content in the velvet beans alone.
