Background: An accurate measurement of the target volume is of primary importance in theragnostics of hyperthyroidism Objective: Our purpose was to evaluate the accuracy of a threshold–based isocontour extraction procedure for thyroid tissue volumetry from SPECT-CT. Methods: Cylindrical vials with a fixed volume of 99mTcO4 at different activities were inserted into a neck phantom in two different thickness settings. Images were acquired by orienting the phantom in different positions, i.e., 40 planar images and 40 SPECT-CT. The fixed values of the iso-contouring threshold for SPECT and SPECT-CT were calculated by means of linear and spline regression models. Mean, Median, Standard Deviation, Standard Error, Mean Absolute Percentage Error and Root Mean-Square Error were computed. Any difference between the planar method, SPECT and SPECT-CT and the effective volume was evaluated by means of ANOVA and post-hoc tests. Moreover, planar and SPECT-CT acquisitions were performed in 8 patients with hyperthyroidism, considering relevant percentage differences greater than > 20 % from CT gold standard. Results: Concerning phantom studies, the planar method shows higher values of each parameter than the other two methods. SPECT-CT shows lower variability. However, no significant differences were observed between SPECT and SPECT-CT measurements. In patients, relevant differences were found in 7 out of 9 lesions with the planar method, in 6 lesions with SPECT, but in only one with SPECT-CT. Conclution: Our study confirms the superiority of SPECT in volume measurement if compared with the planar method. A more accurate measurement can be obtained from SPECT-CT.

Abstract Background Patrinia scabra Bunge is a well-known herbal medicine for its favorable treatment on inflammatory diseases owing to its effective ingredients, in which iridoid glycoside plays an extremely significant role. This article aimed to improve the content of total iridoid glycosides in crude extract through a series optimization of extraction procedure. Moreover, considering that both pain and inflammation are two correlated responses triggered in response to injury, irritants or pathogen, the article investigated the anti-inflammatory and analgesic activities of P. scabra to screen out the active fraction. Method P. scabra was extracted by ultrasonic-microwave synergistic extraction (UMSE) to obtain total iridoid glycosides (PSI), during which a series of conditions were investigated based on single-factor experiments. The extraction process was further optimized by a reliable statistical method of response surface methodology (RSM). The elution fractions of P. scabra extract were prepared by macroporous resin column chromatography. Through the various animal experiment including acetic acid-induced writhing test, formalin induced licking and flinching, carrageenan-induced mice paw oedema test and xylene-induced ear edema in mice, the active fractions with favorable analgesic and anti-inflammatory effect were reasonably screen out. Results The content of PSI could reach up to 81.42 ± 0.31 mg/g under the optimum conditions as follows: ethanol concentration of 52%, material-to-liquid ratio of 1:18 g/mL, microwave power at 610 W and extraction time of 45 min. After gradient elution by the macroporous resin, the content of PSI increased significantly. Compared with other concentrations of elution liquid, the content of PSI in 30 and 50% ethanol eluate was increased to reach 497.65 and 506.90 mg/g, respectively. Owing to the pharmacology experiment, it was reasonably revealed that 30 and 50% ethanol elution fractions of P. scabra could relieve pain centrally and peripherally, exhibiting good analgesic and anti-inflammatory activities. Conclusion Patrinia scabra possessed rich iridoids and exhibited significant analgesic and anti-inflammatory activities.

Bisphenols are well-known endocrine disruptors that can easily migrate from plastic and can containers to food. Due to the complicated matrix and ultra-low concentrations of bisphenols in food, samples require extensive preparation before instrumental analysis. In this paper, an environmental sensitive microgel was employed as a sorbent for the preconcentration of four bisphenols, bisphenol A (BPA), bisphenol B (BPB), bisphenol E (BPE) and bisphenol F (BPF), from liquid food samples. Liquid chromatography with fluorescence detection (LC-FLD) was used for the quantification of bisphenols. By applying microgel solid-phase extraction procedure, the limits of detections achieved in liquid food samples can be lowered to 0.9 µg·L−1 for BPF and BPA, 2.3 µg·L−1 for BPE and 2.9 µg·L−1 for BPB. Only 5 mg of microgel was sufficient to achieve good recoveries (70.5–109%) with precision (RSD 0.21–5.01%, n = 3) for different analyzed liquid food samples spiked at concentration levels of 50 µg·L−1. In five out of twelve of the analyzed samples (pineapple, mandarin, peach, mushroom and pickles), they were contaminated with BPA, and the determined concentration was in the range of 6.2–22 µg·L−1; however, these results are below the specific migration limit (SML) set for BPA (50 µg·kg−1).

Objective: This study compared the effectiveness of auditory distraction and brief relaxation therapy for reducing anxiety in patients undergoing tooth extraction. Methods: A non-blind, three-armed, randomized control trial was carried out. The targeted study population were patients who needed extraction of a non-restorable and non-mobile molar tooth. Eighty-six patients were recruited; the brief relaxation therapy and auditory distraction groups had 32 participants each, while 10 of the 22 participants in the control group were excluded due to missing data. The Hierarchical Anxiety Questionnaire was used to assess the anxiety level. The Mann–Whitney U or Kruskal–Wallis test was performed to compare means between the groups. The before and after comparisons in each group were carried out using the Wilcoxon Signed-Rank test. The alpha value was set at 0.05, and data were analyzed using SPSS version 24. Results: The mean anxiety score after brief relaxation therapy and auditory distraction had significantly decreased (p < 0.001). Although not significant (p = 0.13), there was a slight increase in the anxiety score of the study participants in the control group just before the extraction procedure. Brief relaxation therapy was significantly effective in reducing anxiety scores in comparison to the control group (MD = 5.87, 95% CI = 2.58, 9.17; p = 0.001), but auditory distraction was not (p = 0.14). Conclusion: Both auditory distraction and brief relaxation therapy were effective in reducing patient anxiety before a dental procedure. Furthermore, it would be interesting to learn if these findings remain consistent for more complex dental procedures, such as surgical removal of an impacted third molar.

A β-cyclodextrin-decorated magnetic activated carbon adsorbent was prepared and characterized using various analytical techniques (X-ray diffraction (XRD), scanning electron microscopy–electron diffraction spectroscopy (SEM-EDS) and transmission electron microscopy (TEM)), and the adsorbent was used in the development of a magnetic solid-phase microextraction (MSPE) method for the preconcentration of estrone, β-estradiol, hydrocortisone and progesterone in wastewater and river water samples. This method was optimized using the central composite design in order to determine the experimental parameters affecting the extraction procedure. The quantification of hormones was achieved using high-performance liquid chromatography equipped with a photodiode array detector (HPLC-DAD). Under optimum conditions, the linearity ranged from 0.04 to 300 µg L−1 with a correlation of determinations of 0.9969–0.9991. The limits of detection and quantification were between 0.01–0.03 and 0.033–0.1 µg L−1, with intraday and interday precisions at 1.1–3.4 and 3.2–4.2. The equilibrium data were best described by the Langmuir isotherm model, and high adsorption capacities (217–294 mg g−1) were obtained. The developed procedure demonstrated high potential as an effective technique for use in wastewater samples without significant interferences, and the adsorbent could be reused up to eight times.

Herbal medicines and their preparations have been widely used from the thousands of years in developing and developed countries in the primary health care of society and community. They have great demand due its safety, efficacy with minimum side or adverse effects. Commiphora caudata. It’s known as konda mava in kannada, ikkata in Sanskrit and hill mango in English. It’s widely used in the management of various disorders. Hence the identification of bioactive fractions from various parts of selected medicinal plant is important. In the present research work an attempt has been made to screen and assess the antibacterial, antifungal and anti-inflammatory activities of Commiphora caudata barks. The barks of selected plant material were collected, authenticated, powdered and subjected for extraction procedure. The extracts were screened for presence of various phytoconstituents. The antibacterial activity of chloroform and methanolic extracts were performed against various strains of bacteria and fungi. The extracts also were investigated for its in-vivo anti-inflammatory activity. The result of investigation concludes that chloroform and methanolic extract of plant were potential to inhibit the growth of selected strains of microorganism and also produced potential anti-inflammatory effect.

Solvent front position extraction procedure was used to prepare biological samples containing selected antihypertensive drugs (ramipril, lercanidipine, indapamide, valsartan, hydrochlorothiazide, perindopril, and nebivolol). Substances separated from the biological matrix components (bovine serum albumin) were quantified by means of liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Sample preparation process was performed with the use of a prototype horizontal chamber with a moving pipette driven by a 3D printer mechanism enabling a controlled eluent flow velocity. Application of this device was advantageous for simultaneous preparation of several samples for further quantitative analysis, with a synchronized reduction of manual operations and solvent consumption. Quantitative results obtained for the majority of the investigated antihypertensive drugs in a complex biological matrix were satisfactory. The values of the %RSD were around 5% for six of the seven substances (with the exception of indapamide). The method exhibits a suitable accuracy (the relative error percentage was below 10% for most drugs). The values of LOD and LOQ were in the range of 1.19 µg/L–8.53 µg/L and 3.61 µg/L–25.8 µg/L, respectively.

Paper-ash is used for remediation of heavily contaminated soils with metals, but remediation efficiency after longer periods has not been reported. To gain insights into the mechanisms of immobilization of cadmium (Cd), lead (Pb), and znic (Zn), a study was performed in the laboratory experiment in uncontaminated, artificially contaminated, and remediated soils, and these soils treated with sulfate, to mimic conditions in contaminated soil from zinc smelter site. Remediation was performed by mixing contaminated soil with paper-ash to immobilize Cd, Pb, and Zn in the geotechnical composite. Partitioning of Cd, Pb, and Zn was studied over one year in seven-time intervals applying the sequential extraction procedure and complementary X-ray diffraction analyses. This methodological approach enabled us to follow the redistribution of Cd, Pb, and Zn over time, thus, to studying immobilization mechanisms and assessing the remediation efficiency and stability of newly formed mineral phases. Cd, Pb, and Zn were effectively immobilized by precipitation of insoluble hydroxides after the addition of paper-ash and by the carbonization process in insoluble carbonate minerals. After remediation, Cd, Pb, and Zn concentrations in the water-soluble fraction were well below the limiting values for inertness: Cd by 100 times, Pb by 125 times, and Zn by 10 times. Sulfate treatment did not influence the remediation efficiency. Experimental data confirmed the high remediation efficiency and stability of insoluble Cd, Pb, and Zn mineral phases in geotechnical composites.
