Maize (Zea mays dis.L.) is one of the most important staple crops in the world. Sindhupalchok, Kavre and Nuwakot were the three districts selected for the maize CBSP groups under the respective Agriculture Development Offices identified by CDD for the present study. Altogether 2x6x3 samples for the study were collected. Seed damage as broken seeds, damages on embryo, seed quality assessments like seed moisture content, pure seed percentage, seed vigor measured by root and shoot length, seed germination, and embryo damages were carried in seed testing laboratory in Seed Science & Technology Division (SSTD) Khumaltar following the ISTA Rules, 2007. For the embryo damage test, tetrazolium salt test (TZ) was conducted following the procedures prescribed for maize seed in ISTA Rules, 2007. All the samples possessed of 99 % of pure seed. It ranged from 99.51 % in Nuwakot to 99.96 % in Kavre for samples using corn sheller. They were found within the standards for pure seed and were of acceptable range. The corn sheller was found as efficient as that of hand shelling. There was not found the effect on the quality of seed that they were within the standard values. The germination percentage decreased from initial 96 to 85 (Kavre); 94 to 87 (Sindhupalchwok) and 91 to 83 (Nuwakot). This study concludes that the corn sheller techniques offer Wooden corn sheller is a simple but traditional device made locally for shelling the maize kernels and distributed to Community based seed production (CBSP) farmers contribute to minimizing the impacts of hand sheller on agricultural systems.DOI: http://dx.doi.org/10.3126/ijasbt.v1i2.8198 Int J Appl Sci Biotechnol, Vol. 1(2): 68-71

A laboratory experiment with three replicates was conducted at Plant Physiology Division research laboratory of Bangladesh Agricultural Research Institute (BARI) during November, 2015 to observe the seed quality of rapeseed-mustard by harvesting at different maturity stages of siliqua. Seeds were collected from siliqua of different rapeseed-mustard varieties and harvesting was made at different maturity stages based on their external color i.e., Green, pale yellow, Golden yellow and full maturity stage of siliqua. After harvesting of siliqua as per external color seeds were dried naturally about 48 hours and were packed in polythene and stored at laboratory environment for next season uses. Under laboratory condition seeds were evaluated in terms of moisture and germination percentage, speed of germination, root and shoot length, vigor index etc. Moisture content was found significantly at elevated level in respect of mustard varieties of BARI Sarisha-14 (V2), BARI Sarisha-6 (V3) and Tori-7 (V4).  The variety Tori-7 and BARI Sarisha-14 showed  the highest vigor index-II & vigor index-I, respectively. Among the harvesting stages, most of the parameters showed the highest standards in seeds harvesting at full maturity of siliqua stage (H4) followed by the golden yellow siliqua stage (H3) and pale yellow siliqua stages seeds (H2).Irrespective of rapeseed-mustard varieties seed collected from golden yellow and pale yellow siliqua stages could  be stored up to twelve month for next season without significant loss in terms of germination percentage and vigor followed by full maturity stages of siliqua harvested seeds (H4). In this study, a comparative analysis of seed quality indicators of 1196 hybrid maize seed samples from the main maize-producing areas in China from 2013 to 2018 was carried out. The results showed that the maize seed quality in China had changed obviously in the past six years, and was mainly as follows: The percentage of samples with coated seed in 2015–2018 was higher than 62.8% in 2013 and all exceeded 97%; the sample rate of packaging according to seed number was from 24.5% in 2013 to 58.6% in 2018, and the percentage of samples which met the prescribed quality standards was from 89.2% in 2013 to 98.4% in 2018. Principal component analysis indicated that standard germination energy (SGE), standard germination percentage (SGP), cold test germination percentage (CTGP), accelerated aging test germination percentage (AATGP), and mean field seedling emergence (FSE) were the primary predictors of seed germination and seedling emergence. Meanwhile, combining other statistical methods, regression models of SGE, SGP, CTGP, and AATGP were established to predict the field seedling emergence. Furthermore, seed bulk density and total starch content were correlated with seed vigor, which needs to be further studied. This study offered a theoretical basis and data support to better understand the changes of maize quality in China over the past six years, and provided an important reference to further improve the maize seed quality in the future.

Vegetables have a critical function in human health and nutritional security. Vegetables are considered an essential building block of any diet. Out of the leafy vegetables, Amaranthus is the most popular and salable vegetable consumed by people all over India. Seed processing experiment was undertaken in Amaranthus tricolor (CO 2) by using the sieves placed inside the mechanical seed shaker (Gyratory sieve shaker) to improve the quality of Amaranthus seeds. The seeds of Amaranthus were size graded with seed shaker attached with various sieve size of BSS 18 X18 (R), BSS 20 X 20 (R), BSS 22 X 22 (R) and BSP 22 X 22 (P). During processing, the machine could be adjusted for 2, 3, 4 and 5 minutes with an oscillating speed of 1440 rpm. The separated seeds were evaluated for seed quality characteristics such as seed germination percentage, shoot length, root length, dry matter production, 1000 seed weight and seed recovery percentage. The results revealed that the sieve size of BSS 22 X 22 (R=retained) mesh sieve had the better quality seeds with a maximum recovery of 67.41 g and when it is operated for a period of 5 minutes. The germination percentage was improved from 77 % to 95 % with  1000 seed weight of 73.21 mg, and the observed recovery was 56 per cent with the vigour index of 1145. Hence, BSS 22 X 22 retained mesh sieve with a duration of 5 minutes could be recommended as an optimum sieve size for grading Amaranthus seeds for improving the seed quality.

Improved open pollinated maize (Zea mays L.) varieties exhibit different emergence reaction as a result of either inferior seed quality or affected by agronomic practices of maize planting. To address problem of low emergence percentage of maize varieties, an experiment was laid out in factorial design under two types of soil (red clay and sandy loam), three level of depth of sowing (7, 14 and 21 cm) for Rampur Composite, Manakamana-1 and Arun-2 maize varieties in Rampur, Chitwan for representing main season (May-August). The result of the experiment revealed that emergence count was highly significant for soil types and depth of sowing among the maize varieties tested. Manakamana-1 expressed lowest percentage (20 and 29%) followed by Arun-2 (22 and 33%) and Rampur Composite (24 and 39%) of emergence under red clay and sandy loam soils, respectively. Likewise, emergence count at 10 days after sowing was recorded highly significant for depth of showing indicating that irrespective of varieties higher the planting depth lower the emergence count.. There was a positive relation between depth of planting and length of mesocotyle resulting loss of seed vigor which furnished low emergence of the maize varieties. It is suggested that depth of sowing affect emergence percentage which consequently have effects on plant population maintenance. Hence, it was concluded that sowing below 7cm deep is not feasible for the tested varieties under red clay and sandy loam soils. A number of important factors may affect seed priming response, including seed quality. Effects of seed vigor on seed priming response were investigated using seed lots of two muskmelon (Cucumis melo L.) cultivars. Seeds of muskmelon, cvs. Mission and Top Net SR were artificially aged at 43°C for 0, 20 and 40 hours. Seeds were primed for six days in darkness at 25°C in KNO3 (0.35 mol L-1) aerated solution. Aged seeds germinated poorly at 17°C. Priming increased germination rate at 17 and 25°C and germination percentage at 17°C. An interaction effect on germination performance between vigor and priming was observed, especially at low temperature. Priming increased germination performance in seeds of low vigor, and the response was cultivar dependent.

Abstract Seed priming or invigoration is presowing treatments of improving germination aimed to reduce the time from sowing to emergence and improving emergence uniformity. Research on seed priming was conducted Indonesian Cereal Research Institute (ICERI) seed laboratory from January until March 2021 to evaluate the effect of seed priming on sorghum seed vigor. Seed quality parameters includes germination percentages, germination rate, shoot and primary root length, seedling dryweight, Sorghum seed variety of Suri 4 were used as seed material. Results showed that among the priming treatments that give positive effect on germination percentage, germination rate, seedling dry weight were priming treatment with KNO3 1.5%.

One of the main causes of food insecurity is the timely unavailability of quality seeds for smallholder farmers in developing countries. Improved storage technologies are effective in reducing storage losses. Thus, the objective of this research is to assess the traditional and emerging seed storage structures/materials for the maintenance of seed qualities. The effect of different storage conditions (moisture content of seed at the time of storage, i.e. 12±0.15% and 13.5±0.18%; storage containers such as metal bin, earthen pot, Purdue Improved Crop Storage (PICS) bag, and jute bag) on seed qualities of maize were assessed. The seed was collected from Chitwan, Nepal and a laboratory experiment was conducted at the central seed testing laboratory, seed quality control center (SQQC), Hariharbhawan, Lalitpur. The experiment was done using a two-factor complete randomized design in a four replicates design. Data regarding the seed qualities (seed moisture percentage, germination percentage, root and shoot length, and vigor index) just before store and also at 45, 90, 135, 180, and 240 days after storage (DAS) was performed as per the standard followed by  International Seed Testing Association (ISTA). The germination percentage, root, and shoot length decreased with increasing storage duration. The seed stored in the higher moisture level had significantly higher seed moisture throughout the storage duration. Seed stored in the traditional structures (jute bag and earthen pot) had lower seed moisture at 120 and 180 DAS. The seed stored at lower moisture resulted in a higher germination percentage, long root, and shoot length. Up to six months of storage germination of maize seeds stored in the earthen pots, PICS bag, and Jute bags were statistically similar. The traditional storage structures are equally effective for the maintenance of seed quality of maize.

Three field pea genotypes (NS Junior, Jezero and Javor) have been assessed for effects of genotype and mechanical damage during harvest on most important parameters of the physical quality of seed. Four harvest treatments were examined (hand harvest, mechanized harvest at 500, 650 and 800 rpm). After harvest, purity of harvested seed and percentages of seeds damaged by insects, seeds with cracked seed coat and broken seeds were analyzed. Highest contents of seeds with damaged seed coat (9.0%) and broken seeds (11.3%) were found in Javor, the genotype with largest seeds. After seed processing, seed vigor, germination, proportion of atypical seedlings and 1000-seed weight were determined. Lowest values of seed vigor and germination (79.8% and 84.9%, respectively) and the largest proportion of atypical seedlings (11.4%) were found in the genotype Javor. Highly significant correlations were found between seed vigor, germination and atypical seedlings on one side and 1000-seed weight on the other (r=-0.53, r=-0.51, and r=0.60, respectively). Damages that increase the portion of atypical seedlings have the largest impact on the quality of pea seeds. This characteristic is determined by the genotype, which should be kept in mind when defining objectives of field pea breeding programs.

Knowledge about seed storage behavior is important to preserve plant species. Clitoria fairchildiana R.A.Howard is an endemic species with medicinal properties, it is used in the recovery of degraded land. The objective of this study is to evaluate the effect of drying on the physiological quality of C. fairchildiana seeds. Seeds were oven dried along with silica gel for 18 days and in intervals of 2 or 3 days, samples were removed to assess the seed moisture content. This was done in an oven at 105±3 oC for 24h, using four replicates with 10 seeds each. After undergoing the above-mentioned treatment, a sample of seeds was stored in the freezer (-18 ºC) for 3 months. Sowing was carried out in plastic trays in a sand and sawdust mixture (1:1), previously sterilized in hot water (100 oC) for two hours. Seeds were left to germinate in a laboratory with no temperature and relative humidity control. Germination tests were performed over 14 days with daily counts of the number of emerged seedlings. The percentage of seed germination, abnormal seedlings and dead seeds were obtained 14 days after sowing. The experimental design was completely randomized with four replications of 25 seeds. The reduction of seed moisture content from 18.5% to 5.6% affected physiological seed quality causing a reduction in the germination percentage and an increase in dead seeds and abnormal seedling percentage. C. fairchildiana seeds are classified as intermediate and they can be desiccated up to 8.6% with no reduction in physiology quality.

Kapok (Bombax ceiba) is a deciduous tree that can grow in the dry-hot valley of southwestern China where its natural regeneration by seedlings is difficult. As mature fruit split open and seeds disperse by wind, it is difficult to collect fully mature seeds. The effects of seed moisture content (MC) and storage temperatures on seed germination of dark-brown seeds collected from split fruit and light-brown seeds collected ≈10–15 days earlier than the time of fruit split were studied to determine the effective germplasm preservation via the seeds. Dark-brown mature seeds could tolerate desiccation to less than 5% MC and could tolerate −20 and −80 °C. Seeds of kapok showed orthodox storage behavior. They can be stored at subzero temperatures with low MC for a long time. For light-brown seeds, germination percentage (GP), germination index (GI), seedling fresh weight (SFW), and vigor index (VI) decreased significantly after seed desiccation. Germination percentage of light-brown seeds with different MC increased to a different extent after being stored at different temperatures for 1 year (76% to 99%), compared with the fresh seeds (73%). Storing fresh seeds at 4 °C was most favorable to keep seed viability and seed vigor of light-brown seeds. Seed collection could be done several days earlier than the time of fruit burst to ensure increased quantity of collected seeds.
