Analysis of street foods of Kathmandu for bacterial contamination was performed in 12 different street foods. The surveillance study was carried in 200 children of primary grade from public school and 12 street vendors for the health hygiene and hazards associated with street food. Poor hygiene practice in preparation and handling of street food has been observed in the vendors. The lack of the knowledge in vendors about the source of bacterial contamination and absence of surveillance on street food has subjected street food to the high potential for food borne illness. The inadequate safety measure adopted by the targeted consumers of street food, the children, has augmented the risk associated with street food. All the food samples analyzed were contaminated with bacteria. The mesophilic count was recorded highest in Panipuri while as coliform count was highest in Chana tarkari. The least count of both was observed in Aaloo chop . Highest number of Staphylococccus aureus was found in Kerau (1.5X103cfu/g) and lowest in Momo (8.3 cfu/g). The dominant bacteria contaminating the food was S. aureus followed by Bacillus alvei, Escherichia coli, Enterobacter aerogenes, Bacillus subtilis, Serratia sp., S. saprophyticus. The contaminated hand and clothing of the person who prepare food are the major source of S. aureus. Highest percentage of E. coli found in Panipuri must be due to the use of contaminated water. Chana chatpate and Chana tarkari were the foods found to be contaminated with Salmonella sp. The type of food and the degree of hygiene practice adopted by vendor refl ects the type and magnitude of bacterial contamination. Implementation of hygienic practices in vendors may reduce the contamination of street food and health education of the school children will curtail the incidences of food borne illness. Abstract Background: Worldwide, food borne illness is often associated with consumption of meat and poultry products sold at retail markets. A study on the bacteriological status of chicken carcass in Osogbo, Nigeria, was carried out to determine the prevalence of Arcobacter species, Escherichia coli and Staphylococcus aureus in chicken carcasses. Methodology: A total of 100 samples of chicken carcasses were collected from two major processing points in Osogbo, Nigeria. The samples were analysed for the presence of bacterial contaminants using standard microbiological isolation and identification procedures, with antimicrobial susceptibility test performed using the disk diffusion method. Result: Of hundred chicken carcasses sampled, 38% were positive for Arcobacter species and E. coli while 60% accounted for S. aureus isolates. Ninety percent of Arcobacter spp isolates were susceptible to ciprofloxacin, 85% to gentamicin, and pefloxacin, 70% to chloramphenicol and 90% were resistant to amoxicillin, 85% to augumentin and 80% to streptomycin. Hundred percent of E. coli isolates were susceptible to ciprofloxacin, pefloxacin, 95% to gentamicin and 100% were resistant to streptomycin, 85 % resistant to amoxicillin, augumentin, while 100% of S. aureus isolates were susceptible to trimethoprim sulphamethoxazole, 90% susceptible to gentamicin, 80% to streptomycin and 100% of the S.aureus isolates were resistant to ampliclox. Conclusion: The bacteriological status of chicken carcass revealed high contamination with Arcobacter, E coli and S. aureus with varying degree of antibiotic resistance therefore, improvement in meat processing procedures and strict hygiene measures towards reduction of these pathogens in food products should be encouraged.

Abstract Pathogenic biotypes of E. coli have been incriminated in food-borne illness. Sensitivity to elevated temperature and atypical behavior precluded assessment of their incidence. A tentative recovery procedure involves direct streak, preenrichment in MacConkey broth at 35°C and transfer to lauryl tryptose broth with incubation at 44°C, and pre-enrichment in nutrient broth at 35°C and transfer to enteric enrichment broth with incubation at 41.5°C. Serological examination of enrichment simplifies analysis. Levine's eosin methylene blue agar is recommended for rapid lactose fermenters; MacConkey's agar should be used for slow fermenters. Primary screening reactions include H2S, arabinose, urease, o-nitrophenyl-β-D-galactosidase, and indole. Secondary reactions comprise lysine decarboxylase, Voges-Proskauer (at 22°C), and other tests required to differentiate E. coli from Citrobacter diver sus, C. freundii, Enterobacter hafniae, Klebsiella pneumoniae, and Shigella species. Slide and tube agglutination reactions are necessary for identification of somatic and capsular antigens. Enhancement of motility by serial passage in a semisolid medium is essential for recognition of flagellar antigen. Confirmatory reactions include nitrate reduction, cytochrome oxidase, and Gram stain. Potentiality for quantitation, applicability to animal pathogens, and ecological samples are discussed.

Contaminated street food has been linked to food-borne illness and food-borne outbreak. Chotpoti is a popular street food in Bangladesh. This study was conducted to identify enteric bacteria in chotpoti sold in different parts of the capital city Dhaka. From July to October, 2012 we purposively selected 18 sites where chotpoti was sold in Dhaka city. From each site we randomly selected six chotpoti stalls for sample collection. At each stall we aseptically collected approximately 100 gm of a combined sample of chotpoti, chili sauce and water used for cleaning utensils. The combined sample was homogenized and was serially diluted in a sterile test tube with normal saline to obtain a dilution up to 10-5. From each test tube 0.1 ml of dilution was plated on to MacConkey and Blood agar plates under aerobic conditions at 37?C for 24 to 48 hours. Bacterial growth was identified by the colonial morphology, Gram stain, and biochemical tests. Of the 108 samples we analyzed, 84 (78%) were contaminated with bacterial pathogens. Among isolated organisms Acinetobacter (66%) was the most dominant followed by Klebsiella spp. (54%), E. coli (3%) and Proteus spp. (0.9%). Two different organisms were present in 47 (44%) samples. In half of the study sites, every sample was contaminated; in only one site bacterial contamination was absent in all samples. Contaminated street food like chotpoti may cause food borne illness and pose risk of food borne epidemics. Further studies are required for quantitative assessment of pathogens to detect potential sources of contamination and to develop interventions that can effectively reduce street food contamination.Asian J. ABSTRACT “Norwalk-like viruses” (NLVs) and hepatitis A virus (HAV) are the most common causes of virus-mediated food-borne illness. Epidemiological investigations of outbreaks associated with these viruses have been hindered by the lack of available methods for the detection of NLVs and HAV in foodstuffs. Although reverse transcription (RT)-PCR methods have been useful in detecting NLVs and HAV in bivalve mollusks implicated in outbreaks, to date such methods have not been available for other foods. To address this need, we developed a method to detect NLVs and HAV recovered from food samples. The method involves washing of food samples with a guanidinium-phenol-based reagent, extraction with chloroform, and precipitation in isopropanol. Recovered viral RNA is amplified with HAV- or NLV-specific primers in RT-PCRs, using a viral RNA internal standard control to identify potential sample inhibition. By this method, 10 to 100 PCR units (estimated to be equivalent to 102 to 103 viral genome copies) of HAV and Norwalk virus seeded onto ham, turkey, and roast beef were detected. The method was applied to food samples implicated in an NLV-associated outbreak at a university cafeteria. Sliced deli ham was positive for a genogroup II NLV as determined by using both polymerase- and capsid-specific primers and probes. Sequence analysis of the PCR-amplified capsid region of the genome indicated that the sequence was identical to the sequence from virus detected in the stools of ill students. The developed method is rapid, simple, and efficient.

Background: Millions of people are suffering from food borne illness and it has become a growing public health concern in the world. To reduce the burden of food borne illness, food handlers must have accurate knowledge of food safety. The objective of the study was to assess the knowledge and level of personal hygiene practice among food handlers of Jahangirnagar University.Methods: It was a descriptive cross sectional study, done among the food handlers in Jahangirngar University Campus, Dhaka, Bangladesh from May 2016 to September 2016. Data were collected from 119 food handlers by using face to face interview using a pre-tested questionnaire. Statistical Package for Social Sciences (SPSS 22.0) software was used for data analysis.Results: Study found that 65.55% of the food handlers did not have adequate knowledge regarding personal hygiene. The study also found that 71.4% of the food handlers had poor hygiene practice. The study revealed that age, education and sleeping place and knowledge regarding personal hygiene were significantly associated with the respondent’s personal hygiene practice.Conclusions: The study shows that knowledge and its practice among the food handlers is very poor. Implementation of training and awareness program on personal hygiene are required to improve their knowledge and personal hygiene practice.

Food borne diseases are an increasingly recognized problem involving a wide spectrum of illnesses caused by bacterial contamination of food.Microorganism poses potential human health problems and is mainly transmitted through consumption of contaminated foods .Bangladesh is one of the densely populated country where majority of the people consume cheap foods prepared in unlicensed food selling points. The objective of the present study was to investigate the microbiological quality of different street food (Amra, Fusca, Chanachur and Guava) sold by various street vendor at Dinajpur, Bangladesh. A total of 20 samples were collected randomly from street vendors and tested for the presence of bacteria following standard microbiological method used for isolation, enumaration and identification of bacreria. Among the samples all had bacterial contamination. The total viable count (TVC) in diferrent street food samples was ranged from 8.0 × 105 CFU/g to 6.7x107 CFU/g. Among samples Escherichia coli 7(35%), Staphylococcus spp. 6(30%), Klebsiella spp. 5(25%), Salmonella spp. 1(5%) and Shigella spp. 1 (5%) were isolated. Escherichia coli were found highest 3(50%) Amra samples and 2(40%) Guava samples, then another organisms Staphylococcus spp. was found highest 2(50%) Chanachur and 2(40%) Fusca samples and then Klebsiella spp. was found highest 2(40%) Fusca samples. Antibiotic sensitivity test showed that Shigella spp., Staphylococcus spp., Klebsiella spp., Salmonella spp. and Escherichia coli were sensitive to Ciprofloxacin and Salmonella spp. were resistance to Gentamycin. All isolates found resistant to Cefixime, Cefalexin, Erythromycin, Fusidic acid, Cefuroxime and Aztreonam. All of the sample harbor multidrug resistant food borne bacteria which might cause public health hazards if these antibiotic resistance transfer to human.Asian J. Med. Biol. Res. ABSTRACTAn assay to identify the common food-borne pathogensSalmonella,Escherichia coli,Shigella, andListeria monocytogeneswas developed in collaboration with Ibis Biosciences (a division of Abbott Molecular) for the Plex-ID biosensor system, a platform that uses electrospray ionization mass spectroscopy (ESI-MS) to detect the base composition of short PCR amplicons. The new food-borne pathogen (FBP) plate has been experimentally designed using four gene segments for a total of eight amplicon targets. Initial work built a DNA base count database that contains more than 140Salmonella enterica, 139E. coli, 11Shigella, and 36Listeriapatterns and 18 otherEnterobacteriaceaeorganisms. This assay was tested to determine the scope of the assay's ability to detect and differentiate the enteric pathogens and to improve the reference database associated with the assay. More than 800 bacterial isolates ofS. enterica,E. coli, andShigellaspecies were analyzed. Overall, 100% ofS. enterica, 99% ofE. coli, and 73% ofShigellaspp. were detected using this assay. The assay was also able to identify 30% of theS. entericaserovars to the serovar level. To further characterize the assay, spiked food matrices and food samples collected during regulatory field work were also studied. While analysis of preenrichment media was inconsistent, identification ofS. entericafrom selective enrichment media resulted in serovar-level identifications for 8 of 10 regulatory samples. The results of this study suggest that this high-throughput method may be useful in clinical and regulatory laboratories testing for these pathogens.

AbstractEscherichia coliO157:H7 is the most well-studied serotype of enterohemorrhagicE. coli(EHEC) class ofE. coliintestinal pathogens, and is responsible for many outbreaks of serious food-borne illness worldwide each year. Adherence mechanisms are a critical component of its pathogenesis, persistence in natural reservoirs, and environmental contamination.E. coliO157:H7 has a highly effective virulence operon, the Locus of Enterocyte Effacement (LEE), and its encoded intimate adherence mechanism is well characterized. However, factors involved in the preceding initial attachment are not well understood. In this study, we propose a mechanism of initial adherence used byE. coliO157:H7in vitro. We describe a bacterial protein not previously reported to be involved in adherence, Slp, and its interactions with the human host protein polymeric immunoglobulin receptor (pIgR). The human pIgR has previously been shown to act as an adherence receptor for some mucosal pathogens, and is highly expressed in the intestine. Following observation of significant colocalization betweenE. coliO157:H7 and pIgR location on Caco-2 cells, a co-immunoprecipitation (Co-IP) assay using a human recombinant Fc-tagged pIgR protein led to the identification of this protein. Disruption of Slp expression inE. coliO157:H7, through deletion of its encoding geneslp, produced a significant adherence deficiency to Caco-2 cells at early time points associated with initial adherence. Plasmid complementation ofslpfully restored the wild-type phenotype. Furthermore, immunofluorescence microscopy revealed evidence that this interaction is specific to the pathogenic strains ofE. colitested, and not the nonpathogenic strain E. coli K12. Additionally, deletion ofslpresulted in the absence of the corresponding protein band in further Co-IP assays, while the plasmid-encodedslpcomplementation of the deletion mutant strain restored the wild-type pattern. The relationship was first demonstrated as a significant colocalization between the locations ofE. coliO157:H7 bacterial cells and pIgR protein using immunofluorescence microscopy. TheE. coliO157:H7 Slp protein was identified, and disruption of theslpgene resulted in a severe adherence deficiency to Caco-2 cells during initial adherence. This effect was reversed upon complementation of the Δslpstrain with a plasmid-encodedslpgene, and the constitutive over-expression ofslpresulted in hyper-adherence exceeding that of the wild-typeE. coliO157:H7. These data support the proposition that Slp directly contributes to initial adherence, with the pIgR protein as its proposed receptor.

This study aimed at identifying chemical and microorganism contamination in foods of street vendors around Institut Agama Islam (IAIN) Syekh Nurjati Cirebon. The samples are meatball, cilok, grilled fish cake, nuggets, rolade, sauce, crackers, and iced water drinks. The sample tests are formalin test, borax test, Rodhamine B test, Methanyl Yellow test, and Coliform & Escherichia coli MPN test. The results showed that some food samples like meatball, cilok, grilled fish cake, nuggets,and rolade were positive of formalin & borax contaminated. The iced water drinks are contaminated by Coliform & E. coli. The other samples like sauce and crackers  were negative of Rhodamine B & Methanyl Yellow contaminated. In conclusion, according to this study, the foods of street vendors at IAIN Cirebon are not yet safe to be consumed.
