Annotation. The aim of the study was to improve the diagnosis of myocardial diastolic function in men with essential hypertension and chronic heart failure with different variants of the cardiotrophin-1 gene. 100 people aged 40–60 were examined: 50 men with asymptomatic essential hypertension and 50 men with essential hypertension complicated by chronic heart failure. Genotyping of the cardiotrophin-1 gene was performed by polymerase chain reaction. Determination of the concentration of cardiotrophin-1 was performed by enzyme-linked immunosorbent assay. Structural and functional parameters of the myocardium were evaluated using ultrasound of the heart. Mathematical processing of the material was performed on a personal computer using the standard statistical package SPSS 10. It was found that in persons with essential hypertension and chronic heart failure, carriers of the GA + AA variant of the cardiotrophin-1 gene (rs8046707) have significantly higher plasma peptide levels with diastolic dysfunction (p <0.05).

Recent studies indicate that 2-4% of the world population is sensitive to the presence of allergens in foods. It is estimated that there are 5-8% allergic children in Europe.This paper presents an overview of the analytical methods which are used in risk analysis and for making decisions in the management of foods that cause allergies in humans.Depending on the purpose of analysis, a variety of analytical techniques can be used: Enzyme-linked immunosorbent assay (ELISA) is recommended for quantitative determination of the composition of ingredients and final products, Lateral flow test (LFD) can be used during routine control of the efficiency of purification and determination of the composition of final products, PCR (polymerase chain reaction)-ELISA and real-time PCR are used to confirm the ELISA results for samples with a low content of allergens or where it is not possible to apply other tests. Mass spectroscopy (MS) can be used as a method for confirming the results of the routine tests and in detecting of small amounts of the allergens. This paper gives an overview of these techniques by listing the advantages and disadvantages of routine food analysis.

Annotation. In recent decades, the number of studies on the role of known and new biomarkers in the diagnosis of cardiovascular problems, among which your place is in the family of natriuretic peptides, has been growing for a long time. However, one of its representatives, vascular natriuretic peptide (CNP), has been restored, and the question of determining its final role still remains open. The aim of the study was to supplement the data on the possibility of using CNP as a biomarker of structural and functional changes in the myocardium in uncomplicated and complicated essential hypertension (EH) in postmenopausal women with polymorphic variants of the BNP gene. We examined 180 women aged 40-65 living in the Podillia region of Ukraine: 62 women with uncomplicated EH with left ventricular hypertrophy (LVH), 51 women with EH complicated by chronic heart failure (CHF), and 67 women without symptoms cardiovascular pathology, which formed the control group. All patients underwent a standardized general clinical, instrumental and laboratory examination. Genotyping of the BNP gene was performed by polymerase chain reaction. Plasma concentrations of natriuretic peptide were determined by plate solid-phase enzyme-linked immunosorbent assay. Mathematical processing was performed using the standard statistical package Statistica 10.0, using parametric and nonparametric statistical methods. The frequency of distribution of polymorphic gene variants in the population was checked according to the Hardy-Weinberg equilibrium. It was found that among the polymorphic variants of the BNP gene in women aged 40-65 years, the frequency of carriers of the T381C genotype and C allele significantly prevails, while inheriting a certain type of polymorphic variant of the BNP gene does not affect the concentration of CNP in blood plasma in all examined groups. Significantly the highest level of plasma concentration of CNP, which was 6.63±0.13 pmol/ml, was determined in members of the group with EH complicated by CHF. In groups of women with both uncomplicated EH and EH complicated by CHF, BMI is inversely correlated with plasma CNP levels.

<b><i>Background:</i></b> Foot-and-mouth disease (FMD) is an infectious and highly contagious disease of cloven-hoofed domestic and wild animals, causing heavy economic losses to the livestock industry. Rapid and reliable diagnosis of the disease is essential for the implementation of effective control measures. This study compared sandwich enzyme-linked immunosorbent assay (S-ELISA) and conventional reverse transcription polymerase chain reaction (RT-PCR) for the diagnosis of FMD. <b><i>Methods:</i></b> A total of 60 epithelial samples from suspected cases of FMD were tested using both S-ELISA and RT-PCR assays. The level of agreement between the assays was assessed by calculating the Kappa value. <b><i>Results:</i></b> S-ELISA detected 38 (63%) samples positive for FMD virus (FMDV). Being predominant, serotype O was detected in 22 (57.9%) of the total samples tested positive, whereas 9 (23.7%) and 7 (18.4%) samples were found positive for serotypes A and Asia-1, respectively. RT-PCR detected viral genome in 51 (85%) of the samples using pan-FMDV primers set, 1F/1R. Thirty-six samples were found positive and 7 negative by both the tests. The level of agreement between the tests was assessed by calculating the Kappa value, which was found to be fair (Kappa value = 0.303 and 95% CI = 0.089; 0.517) and significant (<i>p</i> = 0.009). However, 2 samples, which were found positive on S-ELISA tested negative on RT-PCR. This may be attributed to the presence of nucleotide mismatch(es) in the primer-binding sites that may have resulted in failure of amplification of the viral genome. The serotype-specific RT-PCR assays not only confirmed serotyping results of S-ELISA but were also able to establish serotype in 9 S-ELISA-negative but pan-FMDV RT-PCR-positive samples. <b><i>Conclusions:</i></b> The RT-PCR assay contributes significantly to establishing a quick, sensitive, and definitive diagnosis of FMD in resource-constrained countries. Samples giving negative results in S-ELISA should be tested in RT-PCR for the disease detection and virus typing.

Background:Objectives:to evaluate the effect of antirheumatic therapy according to the “treat to target” strategy on the course of chronic heart failure (CHF) in patients with early RA.Methods:The study included 22 patients CHF with valid diagnosis of RA (criteria ACR / EULAR, 2010), 17 (77%) of women, median (Me) age - 60 years, Me disease duration - 7 months; IgM seropositive for rheumatoid factor 10 (45%) and / or antibodies to the cyclic citrulline peptide 22 (100%), DAS28-5.6 [4,8;6,5]. CHF verified in accordance the recommendations for the diagnosis and treatment of CHF Society of Specialists in Heart Failure (2013). The concentration of NT-proBNP was determined by electrochemiluminescence. For all patients was started methotrexate (MT) therapy with a rapid increase in the dose to 30 mg per week subcutaneously. If the MT was not effective enough, after 3 months a biological Disease-Modifying Anti-Rheumatic Drug (bDMARDs) was added to the therapy, predominantly TNF-alpha inhibitors. After 18 months, 10 (45%) patients were in remission and low disease activity, 6 (60%) of patients underwent MT therapy in combination with bDMARDs.Results:In baseline CHF with preserved EF was revealed in 21 (95%) patients, in 1 patient - CHF with reduced EF. After 18 months there was a positive dynamics of improvement of clinical symptoms, echocardiographic indicators (decrease the size of the left atrium (LА) and the index of end-systolic volume of LА, IVRT, E’ LV), diastolic function of the left ventricle (LV). There was no decompensation of CHF. LV diastolic function normalized in 7 (32%) patients who reached the target level of blood pressure, remission (n = 5) and low (n = 2) disease activity, mainly in the treatment of MT and bDMARDs. In patients with RA and CHF, the level of NT-proBNP decreased from 192.2 [151.4; 266.4] to 114.0 [90.4; 163.4] pg / ml (p <0.001), normalized in 16 of 22 (73%) patients (p <0.001) with remission or low RA activity. In 5 (22%) patients, the clinical manifestations of CHF regressed, LV diastolic function and NT-proBNP level normalized.Conclusion:In patients with early RA and CHF anti-rheumatic therapy improves the clinical course of CHF. There were an improvement in the clinical course of CHF, diastolic function of the left ventricle and a decrease in NT-proBNP.Disclosure of Interests:None declared

Abstract The aim of the present study was to investigate the occurrence of N. caninum associated with abortions of dairy cattle from Santa Catarina state, southern Brazil by using enzyme-linked immunosorbent assay (ELISA), immunohistochemistry (IHC), and polymerase chain reaction (PCR). Blood from dairy cows that aborted along with intrathoracic fluid and tissue samples (brain, heart, liver, and lung) from their fetuses were collected and used for serology; PCR, histopathological, and immunohistochemistry (IHC) evaluations were also conducted. Twenty-one cows (51.2%) out of 41, and eight fetuses (26.7%) out of 30 were ELISA (HerdCheck, IDEXX) positive for N. caninum. Dams > 36 months of age had a higher risk of being serum positive than younger animals. PCR and IHC revealed that 38.8% (14/36) and 25.0% (9/36) of the fetuses were positive for N. caninum, respectively for each of the tests. Seropositive cows had a higher frequency of fetuses that were also positive by either intrathoracic fluid, PCR, or IHC. In summary, the present study observed a high frequency of N. caninum in abortions from dairy cows from southern Brazil, with a higher N. caninum prevalence found in cows that were older than 36 months. In addition, serology, PCR, and IHC should be used all together for better diagnosis of neosporosis in cattle.
