The Effect of Kebar Grass (Biophtyum petersianum Klotzsch) Extract on the Number of Leydig Cells of Mice (Mus Musculus) Treated with TCDD

Methoxychlor (MXC) is an insecticide (DDT derivates) that has the potential for bioaccumulation in mammal and causes a disruptive effect on the hepar and reproductive system. This study was done to find out the benefits of curcumin as a natural ingredient to overcome the negative impact of Methoxychlor (MXC) on hepar and male reproductive organ of Balb’C mice (Mus musculus L). The study was carried out in a Completely Randomized Design (CRD) Posttest Only Control Group Design used four treatments and six replications. The curcumin treatment after administration of MXC was carried out by gavage with curcumin doses: 0.05; 0,1; and 0.2 mg/g body weight, every day for two weeks, respectively. Histological observations of the liver, and testis was performed using the paraffin method and Hematoxylin Eosin stained. The results showed that MXC exposure caused liver disruption by increasing the number of pycnotic necrotic hepatocytes and hydrophic degeneration hepatocytes. On the male reproductive organ, MXC caused testis impairment by reducing the number of Sertoli cells and Leydig cells, spermatogenic cell counts, and the diameter of seminiferous tubules. The administration of curcumin at doses of 0.1 mg/g bw in mice exposed to methoxychlor can reduce the number of hydrophic degeneration hepatocytes and tend to reduce the number of pycnotic hepatocytes; and also increase the number of Sertoli cells, the number of spermatogenic cells, and the diameter of the seminiferous tubules, and tend to reduce the amount of Leydig cells. Curcumin treatment tends to recover hepar dan testis disruption of mice that were exposed by MXC.

This study aims to determine the effect of red dragon (Hylocereus polyrhizus) fruit peel extract (RDFPE) on the parameters of Leydig cells number, seminiferous tubules diameter, and testicular weight of mice (Mus musculus) exposed to heat (40°C). Twenty adult male mice were divided randomly into five groups. The control group (C) mice only received a placebo. Meanwhile, the treatment groups mice were exposed to heat for 45 minutes daily for 36 days and oral administration of placebo, RDFPE of 250, 500, and 1000mg/kg BW for T0, T1, T2, and T3, respectively. The result showed that heat exposure on mice (T0 group) caused a lower of all of the parameters (p <0.05) than normal mice (control group, C). RDFPE administration at a dose of 250 mg/kg BW (T1 group) and 500 mg/kg BW (T2 group) resulted in a higher value of those parameters (p <0.05) compared to the T0 group. All those parameters of the T2 group (dose of 500 mg/kg BW) were not significantly different (p >0.05) than the control group (normal mice). However, the higher dose of RDFPE (1000 mg/kg BW, T3 group) resulted in the lower values of those parameters (p <0.05) than those of the T2 group. It could be concluded that 500mg/kg BW dose of RDFPE could return Leydig cells number, seminiferous tubules diameter, and testicular weight of mice (Mus musculus) exposed to heat.

This study was aim to examine the effect of kebar grass extract (Biophytum petersianum Klotzsch) to against number leydig cells of mice (Mus musculus) by exposed 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD). Twenty five male mice (Mus musculus) 4 months with avarage body weight 20 g were used. These animals were divided into five groups (K(-), K(+), P1, P2 and P3). K(-) was treated with placebo, K(+) was treated by exposed TCCD single dose injection intaperitoneal 7µg/KgBw, P1 was treated by exposed TCCD single dose injection intaperitoneal 7µg/KgBw and kebar grass extract 0,045 mg/g Bw/day P2 was treated by exposed TCCD single dose injection intaperitoneal 7µg/KgBw and kebar grass extract 0,080 mg/g Bw/day, P3 was treated by exposed TCCD single dose injection intaperitoneal 7µg/KgBw and kebar grass extract 1,350 mg/g Bw/day. This research has been conducted for 53 days. The data were compared using ANOVA and Duncan test by SPSS 22.4 for windows. The result showed that Kebar Grass Extract in all of groups can prevent the damage of leydig cells in testis that exposed by TCCD significantly (p<0,05) and kebar grass extract 0,135 mg/kgBw/day can increase amount of leydig cells maximaly.

The aim of this research is to know about the staging of spermatogenesis and leydig cells in mice that injected by Toxoplasma gondii after giving honey as a preventive substance. This research used 25 mice in age 2-4 month with 20-30 weight. Mice were divided into 5 groups (P0, P1, P2, P3 dan P4). P0 as a control teratment were given aquades in peroral, P1 was a group given honey in doses I (0.08ml/0.2ml/day) during 14 days in peroral, P2 was a group that given infection by T.gondii in doses 103 in peritonial, P3 was given honey in doses I 0.08ml/0.2ml/day) then injected by T.gondii, and P4 was given honey in doses II (0.12ml/0.2ml/day) then injected by T.gondii. This research conducted in 18 days, in day 14th mice were given honey and the day 15th–18th mice injected by T.gondii doses 103. The data result was analyzed with Analysis of Variant (ANOVA) and cotinued with Duncan. Comparision statistical showed with SPSS statistic 20 for Windows. The result of analysis, P0 and P1 were significantly different (p<0.05), with the increase number of spermatogenic and leydig cells. P0 and P2 also significantly different, with the decrease number of spermatogenic and leydig cells. In P2 and P3 group were not significantly different (P>0.05), also in P4 group were not significantly different with P0 and P1. The results of this research showed that honey in doses II can maintain the number of sprematogenic and leydig cells as a preventive substance against T.gondii.

Testicular biopsies were obtained from six young male subjects (age range 24-33) who complained of infertility and who had clinical evidence of oligospermia. This was confirmed on histological examination which showed a broad spectrum from profound hypospermatogenesis to relatively normal appearing germinal epithelium. Thickening of the tubular walls was noted in half of the cases and slight peritubular fibrosis in one. The Leydig cells were reported as normal or unremarkable.Transmission electron microscopy showed that the thickening of the supporting tissue of the germinal epithelium was caused more by an increase in the thickness of the layers of the lamina propria than of the tubular wall itself. The changes in the basement membrane of the tubular wall consisted mostly of a greater degree of infolding into the tubule and some reduplication which gave rise to a multilayered appearance.

The axolotl is a strictly aquatic salamander in which the larval external gills are retained throughout life. The external gills of the adult axolotl have been studied by light and electron microscopy for ultrastructural evidence of ionic transport. The thin epidermis of the gill filaments and gill stems is composed of 3 cell types: granular cells, the basal cells and a sparce population of intervening Leydig cells. The gill epidermis is devoid of muscles, and no mitotic figures were observed in any of its cells.The granular cells cover the gill surface as a continuous layer (Fig. 1, G) and contain secretory granules of different forms, located apically (Figs.1, 2, SG). Some granules are found intimately associated with the apical membrane while others fuse with it and release their contents onto the external surface (Fig. 3). The apical membranes of the granular cells exhibit microvilli which are covered by a PAS+ fuzzy coat, termed “glycocalyx” (Fig. 2, MV).

Smooth endoplasmic reticulum (SER) is involved in the biosynthesis of steroid hormones, and changes in the organisation and abundance of this organelle are regularly used as indicators of changes in the level of steroidogenesis. SER is typically arranged as a meshwork of anastomosing tubules which, with the transmission electron microscope, appear as a random mixture of cross, oblique and longitudinal sections. Less commonly the SER appears as swollen vesicles and it is generally suggested that this is an artefact caused during immersion fixation or during immersion of poorly-perfused tissue.During a previous study of the Leydig cells of a seasonally reproducing bat, in which tissue was fixed by immersion, we noted that tubular SER and vesicular SER often occured in adjacent cells and sometimes in the same cell, and that the abundance of the two types of SER changed seasonally. We came to doubt the widelyheld dogma that vesicular SER was an artefact of immersion fixation and set out to test the hypothesis that the method of fixation does not modify the ultrastructure of the SER.

ZusammenfassungEs werden die Fälle einer disseminierten letalen Toxoplasmose bei einer Farbmaus (Mus musculus) und einem Roten Panda (Ailurus fulgens) vorgestellt. Es handelte sich um eine als Haustier gehaltene Farbmaus und einen Roten Panda aus einem sächsischen zoologischen Garten. Die pathologische Untersuchung ergab bei beiden Tieren eine systemische Toxoplasmeninfektion. Eine hochgradige nekrotisierende Hepatitis stellte in beiden Fällen den histologischen Hauptbefund dar. Parasitenzysten fanden sich massenhaft in der Leber, in mäßiger Zahl im Gehirn und in geringer Zahl in anderen Organen. Mittels PAS-Reaktion waren diese Zysten bei der Farbmaus kaum darstellbar, beim Roten Panda dagegen sehr deutlich. PCR bzw. Immunhistologie bestätigten die Diagnose.

ABSTRACT By means of an indirect immunofluorescence technique, sera from 116 patients with Addison's disease, an equal number of age and sex matched controls and 97 patients with other endocrine diseases were examined for the occurrence of antibody to steroid-producing cells in ovary, testis and adrenal cortex. Fluorescent staining was observed in the theca cells of growing follicles, the theca lutein cells, testicular Leydig cells and adrenal cortical cells, i. e. cells which contain enzyme systems used in steroid hormone production. The "steroid-cell" antibody was present in 24 % of the patients with idiopathic Addison's disease, equally frequent in males and females, and in 17 % of the patients with tuberculous Addison's disease, but was rarely found in controls, including patients with other endocrine diseases. Female hypergonadotrophic hypogonadism made an exception, since the "steroid-cell" antibody was found in about half the cases with this condition.
