The protective effect of jasmonic acid (JA) was evaluated under stress (100 mM NaCl) condition. The investigations were carried on potato plants (Solanum tuberosum L.) of the mid-season variety Lugovskoy. Plant-regenerants were grafted and cultured in test tubes on modified Murashige-Skoog agar medium in the absence (control) or in the presence of JA at concentrations of 0.001; 0.1 and 10 M under optimal growing conditions or with the addition of NaCl. After 28 days of cultivation, growth (length of stem and root, number of tiers and leaves, plant mass) and physiological (proline content and photosynthetic pigments, determination of the osmotic potential of cell exudate) of the plants were assessed. For the first time it has been shown that jasmonic acid (0.1 and 10 M) manifests a pronounced protective effect on potato plants under salt stress condition. The protective effect based on the partial removal of the salt negative effect on the main photosynthetic pigments and the maintenance of the osmotic status of cell contents during salinization.

The localization of cysteine proteinase PLCP-2 was investigated in potato plants (Solanum tuberosum L.) cultivar Désirée by electron microscopy. Healthy and PVY<sup>NTN</sup> infected potato plants were grown in vitro on media with or without a supplement of jasmonic acid. We had already shown that PLCP-2 is present in leaves, stems, tips of shoots and tips of roots of healthy and PVY<sup>NTN</sup> infected plants. It was detected in various cell types in protein bodies in vacuoles, in cytoplasm and in cell walls. There were significantly larger amounts of PLCP-2 in plants grown on medium with a supplement of jasmonic acid in both healthy and virus infected plants. More protein bodies in vacuoles were found in plants grown on medium with addition of jasmonic acid.

Tujuan penelitian ini adalah mengidentifikasi toksisitas beberapa fungisida nabati dalam mengendalikan penyakit Layu Fusarium (F. oxysporum) pada tanaman kentang (Solanum tuberosum L.) secara In Vitro. Metode Penelitian yang digunakan yaitu metode umpan beracun. Data dianalisis dengan Rancangan Acak Lengkap (RAL) dengan Analisis Varian (ANAVA) yang dilanjutkan dengan menggunakan metode BNT (Beda Nyata Terkecil). Hasil Penelitian, diperoleh nilai toksisitas fungisida nabati tertinggi yaitu pada ekstrak daun sirsak dengan nilai HR (69,44%), kategori berpengaruh, ditandai dengan diameter koloni 2,75 cm (100ppm) dan yang terendah toksisitasnya yaitu pada ekstrak daun jeruk purut dengan nilai HR (49,81%), kategori cukup berpengaruh ditandai dengan diameter koloni 3,75 cm (25ppm). Semakin tinggi konsentrasi yang diujikan maka semakin tinggi toksisitas dari fungisida nabati yang diberikan.Kata Kunci: fungisida nabati, Fusarium oxysporum, tanaman kentang, In Vitro Abstract Potato plants (Solanum tuberosum L.) is one of the horticulture plants which often decreases in terms of production and productivity, due to the attack of wilt, one of which is caused by Fusarium oxysporum. The purpose of this study was to determine the toxicity of several biofungicides in controlling Fusarium wilt (F. oxysporum) in potato plants (Solanum tuberosum L.) in Vitro. The research method used was the In Vitro method with the poison bait method. Data were analyzed by Completely Randomized Design with Variant Analysis (ANAVA), followed by the BNT method. The results showed that the highest biofungicide toxicity value was soursop leaf extract with HR values (69.44%), influential categories, characterized by colony diameter 2.75 cm (100ppm) and the lowest toxicity, namely in kaffir lime leaf extract with a value of HR (49.81%), quite influential category was characterized by colony diameter of 3.75 cm (25ppm). The higher the concentration tested, the higher the toxicity of the biofungicide given.Keywords: biofungicides, Fusarium oxysporum, Potato Plants, In Vitro.

Salinity is a major problem affecting crop production all over the world. Excessive soil salinity can reduce the productivity of many agricultural crops including many vegetables and spices. Onion is one of the most important spices in the Asiatic region which is now in high demand. The experiment was conducted to observe in vitro regeneration of onion (Allium cepa L.) under salt stress condition from September 2016 to July 2017. The experiment was conducted as two factorial (genotype and treatment) Completely Randomized Design (CRD) with 3 replications for each treatment. Shoot tip segments of three genotypes namely Faridpuri, Taherpuri and Pusa red (Indian) were cultured in MS (Murashige and skoog, 1962) media supplemented with 25, 50, 75 and 100 mM NaCl. The genotype Faridpuri gave maximum salt tolerance upto 100 mM salinity level with 10.60 cm shoot length and 1.94 cm root length having the highest relative shoot and root growth. Pusa red was found to be salinity sensitive genotype which showing lowest shoot length of 7.03 cm and root length of 0.96 cm at 100 mM NaCl treatment. However, Taherpuri was tolerant up to 100 mM salinity level with 8.14 cm shoot length and 1.25 cm root length. Both the highest fresh weight of root (54.77 mg) and dry weight of root (41.36 mg) was from the genotype Faridpuri with 25 mM NaCl treatment. However, a convenient in vitro regeneration protocol of onion genotypes under different salinity level has been developed and the genotype Faridpuri can be used for further investigation in field condition to evaluate its performance at various salinity levels.

The aim of this study is to determine the effects of different concentrations and combinations of the phytohormones, 1-naphthaleneacetic acid (NAA), and 6-benzylaminopurine (BAP): M1 (0.5 mg / l +1 mg / l), M2 (1 mg / l + 0.5 mg / l) , M3 (2 mg / l +2 mg / l), M4 (0.5 mg / l + l mg / l, NAA), M5 (1.0 mg / l + l mg / l , NAA), and M6 (2.0 mg / l + l mg / l, NAA). This study was carried out in dark condition on callus induction of potato plants (Solanum tuberosum L.) cultivars from potato tuber bud so as to demonstrate the role of light. The callus initiation begins after 7 days of incubation for all studied media. After two months of incubation, the better development of callus was noted in Spunta variety by using medium M1, M2, M3, and M6. The calluses took a compact structure of brown-white color for both varieties with a callus induction rate of 20- 40%. This was collected with kondor variety for M2 and (M3, M4, M5) media respectively and 10-30% for M4 (M1, M2, M3) for Spunta variety also. The highest fresh weight was recorded on M2 medium with 0.26g for Kondor variety and 0.93g for Spunta variety.

Aim. Obtaining in vitro new lines of potato (Solanum tuberosum L.) cultivar Vernissage with genetically mediated resistance to the parasitic nematode H. schachtii on nutrient MS media with microbial bioregulators. Methods. In vitro conditions the process of organogenesis of potato plants on nutrient MS media containing microbial bioregulators used at concentrations 25-100 μl/l in combination with phytohormones 2 mg/l BAP and 0,1 mg/l NAA was investigated. Using dot blotting method the degree of hybridization between cytoplasmic si/miRNA isolated from cells of potato plants-regenerants, grown on the artificial invasive background, and nematode mRNA was studied. In the wheat seedlings cell-free system in vitro the silencing activity of si/miRNA isolated from cells of potato plants-regenerants on the template of nematode mRNA was investigated. Results. The experiments conducted in vitro conditions showed that the addition of microbial bioregulators at concentrations 25-100 μl/l in combination with phytohormones 2 mg/l BAP and 0,1 mg/l NAA into MS media increased the efficiency of regeneration of potato shoots to 43–47 % as compared with similar indices obtained on control MS media. The increase of the degree of hybridization to 19-38 % between cytoplasmic mRNA isolated from cells of nematode H. schachtii and si/miRNA isolated from cells of experimental potato plants-regenerants grown in vitro conditions on nutrient media containing bioregulators on the artificial invasive background was shown. Conclusions. Using microbial bioregulators in vitro conditions as components of nutrient MS medium increases potato shoot regeneration efficiency and enhances RNAi-mediated resistance of plants-regenerants to parasitic nematode H. schachtii. The present investigation was conducted with an aim to study the effect of salt stress (NaCl) on in vitro potato callus growth and the potential for exploiting this technique in improving salinity tolerance in potato. The results in the four varieties in the culture medium showed a decrease in all studied parameters like callus growth, relative average growth, water content and salinity sensitivity coefficient upon increasing the salinity of the medium in calluses. It has also been observed that the type of callus has a large effect on the above-mentioned parameters in which better callus growth adapted to 150 mmol NaCl is obtained than unsuitable calli in the same culture medium.

It has been previously shown that the chemically synthesized nanocomposite of selenium with arabinogalactan (NC Se/AG) is characterized by antibacterial effect upon the agent of ring rot – gram-positive bacterium Clavibacter sepedonicus (Cms), with the NC Se/AG having no negative effect on potato plants. In the present paper, it has been found that, 1 hour after the treatment of the NC Se/AG, a substantial elevation of lipid peroxidation products was observed in potato root tissues. This supports earlier results on the increase in reactive oxygen species (ROS) production in potato root tissues under the influence of NC Se/AG. It is proposed that the increased ROS content in potato may inhibit pathogen colonization of plants. This has been tested by seeding homogenised plant tissues of various potato zones (roots, stems, shoot apex zone) onto the nutrient medium. In plants infected with Cms and untreated with the NC, the number of colony forming units (CFUs) of Cms has been shown to be numerous both in potato culture medium and in root and stem tissues. In shoot apex zone of such plants, it has been revealed, bacteria also present, but in smaller quantities. Similar data have been obtained by seeding homogenised tissues from roots and stems of potato plants treated with the NC followed by infection with Cms. However, seeding from shoot apex zones of the plants has been given 4 times less CFUs than from potato plants not treated with the NC. The effect of the NC Se/AG upon the pathogen colonization of plants appears to depend on the titre of the microorganism. In shoot apex zone of plants, characterized with small number of CFUs of Cms, the pathogen growth has been decreased. For the first time, Cms bacteria in potato plant tissues in vitro have been visualized with the aid of scanning microscopy.
