Introduction: Dengue is one of the most serious mosquitos borne viral infection mainly affecting tropical and subtropical countries of the world. In absence of specific treatment and vaccine for dengue fever (DF); vector control is the only method by which spread of dengue can be prevented. The present study was conducted to determine the increasing dengue incidence in Bihar region. Material and Method: This study was done to report the sero prevalence of Dengue virus infection at Bihar, North India. The laboratory records of clinically suspected Dengue patients from jan 2018 to Nov 2018  were analyzed retrospectively for demographic features, seasonal variations, and results of IgM and IgG anti dengue antibodies tested by Dengue IGM capture enzyme linked immune sorbent assay (MAC ELISA). Results: A total of 1035 serum samples were analyzed. Out of which 209 samples (20.19%) were found positive for dengue virus infection. Maximum positive cases were seen in 2019 (53.11%). Seasonal trend showed that infection started appearing in august, peaked in October and slowly tapered by December. Conclusion: The most affected age group was 11 to 15 years of age, (57.3%) followed by 6 to10 year’s group (27.3 %)and least effective age group was 0 to 5 year (15.4%), majority of cases were found to be of secondary dengue virus infection (93.8%). Male to female ratio was 2:1.The present outbreak thus emphasizes the need for continuous sero epidemiological surveillance for the timely formulation and implementation of effective dengue control programme.  Keywords: Dengue, dengue haemorrhagic fever, IgM antibody capture enzyme linked immune sorbent assay (MAC ELISA), India, vector.

Abstract Background Dengue virus infection has been an important and serious public health concern in Taiwan, where local outbreaks of dengue fever occurred almost every year. To our knowledge, no nationwide investigation has been carried out to determine the actual extent of infection in the general population. Methods A total of 1308 random serum samples were collected from the general population in Taiwan in 2010. The antibody-captured enzyme-linked immunosorbent assays were used to detect DENV-specific IgM and IgG. Demographics data were used for risk analysis. Results The weighted overall seroprevalence was 1.96% for anti-DENV IgM, and 3.4% for anti-DENV IgG, respectively. A significant rise of DENV IgG seropositive rate had been noted since late adulthood stage, from 1.1% at the age group of 50–59 years to 7.6% at the age group of 60–69 years. For people aged over 70 years, the seropositive rate reached 19%. Age, nationality, and regions of residency were associated with the IgG seropositivity. There was no statistically significant difference in seroprevalence of anti-Dengue IgM, indicating recent infection, among univariate predictors we proposed, including gender, age, residency, nationality, and household size. Conclusions Our results indicated that the majority of population in Taiwan born after 1940 is naive to dengue virus and the prevalence of IgG antibody against dengue virus rises with age. Nationality, and regions of residency are associated with the exposure of population to infection by dengue viruses. Further studies are needed to realize the current situation of seroprevalence of dengue fever in Taiwan.

ABSTRACT We determined the usefulness of an immunoglobulin A (IgA) antibody-capture enzyme-linked immunosorbent assay for serodiagnosis of dengue virus infections. The results indicate that the presence of IgA and IgM in serum samples assures recent primary dengue virus infection even with a single serum sample.

Abstract Background: Because of the range and nonspecificity of clinical presentations of dengue virus infections, we felt there was a need to create diagnostic tests. We used artificial receptors for the virus to develop serologic assays to detect dengue virus infection. Methods: We coated a quartz crystal microbalance (QCM) with molecularly imprinted polymers specific for nonstructural protein 1 of flavivirus. These artificial receptors were specifically created on a QCM chip by polymerization of monomers and were cross-linked in the presence of the epitope site of nonstructural protein 1. We tested serum samples from patients with confirmed cases of dengue reported to the Center for Disease Control in Taipei. Samples were diluted 100-fold; no other sample pretreatment was used. The QCM response was compared with results of monoclonal ELISA. Results: QCM signals were >15 Hz in 18 of 21 (86%) of dengue samples and in 0 of 16 control samples. The correlation (r2) of the QCM response and the ELISA result was 0.73. Within-run and run-to-run imprecisions (CV) were 4%–28% and 10%–32%, respectively. Conclusions: The described assay offers a serologic technique for diagnosis of early viremia. The results illustrate the potential of well-organized polymers on the highly sensitive sensor system for diagnostic and biotechnological applications.

ABSTRACT There is an urgent need for accurate and simple dengue virus infection diagnostic assays in limited-resource settings of dengue endemicity, to assist patient management. Using a panel of reference samples (S. D. Blacksell, P. N. Newton, D. Bell, J. Kelley, M. P. Mammen, D. W. Vaughn, V. Wuthiekanun, A. Sungkakum, A. Nisalak, and N. P. Day, Clin. Infect. Dis. 42:1127-1134, 2006), we recently evaluated eihgt commercially available immunochromatographic rapid diagnostic tests (RDTs) designed to detect dengue virus-specific immunoglobulin M (IgM) and/or IgG. We found that 6/8 RDTs had sensitivities of less than 50% (range, 6 to 65%), but specificities were generally high. Here, in conjuction with dengue virus serotyping by reverse transcriptase PCR and in the limited-resource setting of Laos, where dengue virus is endemic, we evaluated the same eight RDTs against a previously validated dengue IgM/IgG enzyme-linked immunosorbent assay for diagnosis of acute dengue virus infection. Paired serum samples were collected from 87 patients, of whom 38 had confirmed dengue virus infections (4 had primary infections, 33 had secondary infections, and 1 had an infection of indeterminate status). RDT sensitivity was low, with 7/8 RDTs having admission sample sensitivities of less than 20% (range, 4 to 26%). The majority (6/8) of the RDTs, demonstrated high specificity (>95%). Kappa statistic values ranged from 6 to 54% for the RDTs, demonstrating poor to moderate variation between three operators. No RDT adequately differentiated between primary and secondary dengue virus infections. The findings of this study suggest that currently available RDTs based on the detection of IgM antibodies for the diagnosis of acute dengue virus infections are unlikely to be useful for patient management.

Dengue Virus infection is an emerging mosquito-borne disease. It is a global health problem and its expanding endemicity towards new territories is a serious concern. Relatively a new disease in Nepalese context, dengue abruptly appeared as massive outbreak in 2010, merely four years after its first introduction. It is a nagging public health problem in the low lands of Terai, expanding to new areas of Nepal in recent years.  A cross-sectional study was conducted to determine anti-Dengue IgM positive rate in Lumbini, Dhading and Chitwan district. The study was carried from June 2012 to November 2012. The total number of Serum samples was collected from 275 patients visiting hospitals with history of fever, headache and suspected DF. The samples were examined by ELISA. The anti-Dengue IgM positivity was found to be 29.09 %. The positive rate was highest in Dhading (70.37%) followed by Bharatpur (37.6%) and Lumbini (11.38%). The Dengue positive cases were higher in males (32.5 %) than female (24.8 %). The highest positive cases (41.6%) were from age group less than 15 years. Background: In India, dengue epidemics are becoming more frequent. Dengue viral infections are among the most important mosquito-borne disease of the Indian subcontinent. The study aimed to compare the clinical and haematological profile of all patients diagnosed with dengue viral infection. Patient with confirmed infection with dengue virus were prospectively evaluated for disease progression and to determine their final clinical outcome. The objective of the study was to study the clinico-haematological profile of dengue infection at rural tertiary care centre.Methods: This prospective study included 456 clinically suspected cases of dengue infection of ages more than 14 years of age. Dengue was confirmed by serological methods. Confirmed cases of dengue infection were analyzed according to age, sex and education. Clinical and hematological data were compared between dengue fever and dengue hemorrhagic fever.Results: Serologic dengue infection was positive in 220 (48%) out of 456 clinically suspected cases of the dengue infection. 127 (57.73%) patients were males and 93 (42.27%) were females. 149 (67.72%) patient had dengue fever while 71 (32.27%) had dengue hemorrhagic fever. The proportion of dengue cases was highest among male aged 15 to 24 years-old. A gradual decrease occurred in the percentage of dengue cases in the population aged 44 years-old or more. Common clinical symptoms were fever, headache, abdominal pain, myalgia, joint pain and retro-orbital pain. Common haematological abnormalities were thrombocytopenia, leukopenia and elevated alanine aminotransferase (SGOT).Conclusions: Differences in the clinical, biochemical and haematological profile of dengue virus infection, indicative of a variation in disease severity from DF to DHF. In this study, we present recent data on readily obtained clinical and laboratory data that can be used for early diagnosis and earlier appropriate treatment of dengue virus infection.

Aim: To analyze and quantify the 2018 dengue outbreak which occurred in district Shangla, Pakistan. Materials & methods: 964 suspected dengue samples were collected and examined for clinical manifestation and laboratory markers. Results: In all, 375 suspected cases were confirmed with dengue virus infection using nonstructural protein 1 (NS1) antigen, immunoglobulin M (IgM) & Immunoglobulin G (IgG) antibodies and real-time PCR whereas PCR was 92.2% sensitive. The most prevalent serotype was dengue virus 3 (60.26%). The male/female ratio was 1.84 and the most highly affected tehsil was Alpuri. The most affected age group was 16–40 years (70.4%). A significant number of cases were reported in September (48.54%). Conclusion: Recurrence of the dengue outbreaks in the study area could alarmingly increase the mortality rate, therefore, proper measures are essential to control dengue epidemics in the future.

This article reviews Dengue, a common viral disease in humans and is an emerging public health problem in Tarai Region of Nepal. The most affected are among the poorest populations living in remote, rural areas and urban slums who have even no access for medical treatment, acquired by bite of infected mosquito. Aedes Aegypti infected with dengue virus is the major source of infections for humans and cannot be transmitted from person-to-person because human are the dead end host. DENV-1 was first isolated by Ren Kimura and Susumu Hotta in Japan in 1943. An epidemic of DF involving at least 200,000 cases had occurred between 1942 and 1944 during World War II in Japanese port cities such as Nagasaki, Kobe, and Osaka. First case of dengue was reported in 2004 in Nepal. The seroprevalence study were done in different part of Nepal by IgM antibody capture ELISA and positive rate was highest (50.0%) in Biratnagar, and lowest (19.6%) in Chitwan male to female ratio was 2:1. IgM-positive rate was 29.0% at ages 21-30, 25.4% at ages 11-20 and 23.6% at ages 0-10, but 10.9% at ages 31-40, and ages over 40. There was not significant association between occupation of the patients and positive rate among farmer, labour, service, business and student. Abstract BACKGROUND: Dengue virus infection is an important emerging disease of the tropical and subtropical regions and is mainly diagnosed by serological detection of NS1 antigen and IgM antidengue antibodies. Since enzyme-linked immunosorbent assay (ELISA) facilities are not easily available at most diagnostic centers, so most of them use various commercially available rapid diagnostic tests (RDTs) kits. AIMS AND OBJECTIVES: This study was designed to access the diagnostic accuracy of four commercially available and widely used RDTs for serodiagnosis of dengue virus infection in Indian laboratories. SUBJECTS AND METHODS: The study was conducted at Department of Microbiology, G.S.V.M Medical College, Kanpur, India, to estimate the sensitivity and specificity of following RDTs: (1) Dengue Cassette (Panbio, Australia), (2) Bioline Dengue Duo (SD Diagnostics, Korea), (3) Dengue Day 1 test (J Mitra and Co., India), and (4) Dengucheck Duo (Tulip Diagnostics, India) on 72 confirmed dengue serum samples that were positive by dengue reverse transcription-polymerase chain reaction, dengue NS1, and IgM ELISA along with 80 serum samples from nondengue febrile illness patients. RESULTS: The majority of the RDTs demonstrated low sensitivity but good specificity for detecting NS1 antigen. Detection of antidengue IgM antibodies by RDTs demonstrated low sensitivity ranging from 27.8% to 77.7%. However, specificity was generally higher (50%–86.2%) and more consistent across the assays. CONCLUSION: The study results differed markedly from the RDTs manufacturers’ claimed performance characteristics. Therefore, the RDT results should be interpreted cautiously and ELISA should be performed as far as possible for serodiagnosis of dengue virus infection.
