Leaf scald (Xanthomonas albilineans) is an important pathogen of sugar cane in Indonesiaand cause significant losses at several main sugarcane plantations. Some control measures, e.i.bactericides smeared Lysol 20% or alcohol 70% on cutting knife seedling for disinfectant,and hot water treatment at 50 minutes on 52,5ᵒC, have not given satisfactory results inmanaging this leaf scald in sugarcane on the field. Biological control with filtrate ofendophytes bacterial were alternative to biocontrol agents Xanthomonas albilineans. Theobjectives of this research were to determine: influence of sterilization methods andendophytic bacterial filtrate dosages of sugarcane to control leaf scald (Xanthomonasalbilineans). This research was conducted in Phytopathology Laboratory at the Faculty ofAgriculture University of North Sumatra from April until December 2014. This research haddone with Completely Randomized Design (CRD) with three-factor: 1. type of endophyticbacterial filtrates: filtrate of B2, B7 and B9 Sterilization methods of endophytic bacterialfiltrate: control (no sterilization), Sterilization by autoclave and Sterilization by filtermembrane, 3. Dosage of endophytic filtrate with four-level: 0,1%, 0,01%, 0,001% and0,0001%. The result showed: 10 isolate endophytic bacteria had exploration were: B1, B2,B3, B4, B5, B6, B7, B8, B9, and B10. Three isolates had a clear zone on Petri dish ≥ 20 mm invitro testing is B9, B7, and B2. The results showed that the membrane filter sterilizationmethod was better when compared to autoclave sterilization, namely: the highest inhibitoryzone diameter of endophytic bacteria in the treatment of B9 filtrate with a membrane filtersterilization method 0,1% at 3 days after inoculation that was equal to 23.00 mm.

This study was intended to examine the pattern of interaction and mutant distribution of endophyticbacteria as a result of the transformation of the gfp gene in rice plant tissue in vitro. This study used 5isolates of rice endophytic gfp mutants namely IM-1, IM-3, IM-8, IM-24, IM-25. The study wasconducted in two stages, stage I using a simple Complete Complete Design (RAL) and stage II usingfactorial Complete Randomized Design (RAL) with a factor of 1: the type of plant tissue and factor 2:the type of mutant isolates.The results of the study of patterns of bacterial interactions in rice tissueculture showed that the percentage of infections of 5 gfp mutant endophytic bacteria isolates rangedfrom 4% -17%. The highest percentage was in mutant isolates IM-3 + gfp (17%) and the lowest was inmutants IM-8 + gfp (4%). The interaction pattern was not significantly different from the parametersof seed germination time. On the other hand, in observing the distribution of bacteria that infect ricetissue, they showed different pattern. Mutant IM-1 + gfp infection is higher in stem tissue, whereasIM-24 + gfp mutant infection is higher in root tissue and IM-25 + gfp mutant infection is higher inleaf tissue. Mutant infection of IM-25 + gfp in leaves had the highest number of colonies reaching1.64 x 109 cfu/ml.

Abstract This study aimed to identify the optimum rice stem cutting size combined with the application of Seprint liquid organic fertilizer (SLOF) to promote the growth and yield of ratoon rice. This research was conducted in a greenhouse at the Faculty of Agriculture, University of Muhammadiyah Sumatera Utara, North Sumatra Province, Indonesia, from January to August 2018. The experiment was set up as a completely randomized design factorial with four replications. The first factor consisted of the cutting sizes of the rice stem (R1 = 5 cm, R2 = 10 cm, and R3 = 15 cm) and the second factor involved the SLOF dosage (S1 = 5 mL L −1 , S2 = 10 mL L −1 , and S3 = 15 mL L −1 ). The parameters were analyzed using analysis of variance (ANOVA), and the means were determined by Duncan’s multiple range test (DMRT). The results showed that the R2 treatment significantly increased the ratoon rice height at 10 to 30 days after cutting (DAC), the number of tillers at 40 to 60 DAC, the number of productive tillers, and the yield per hectare. The S2 and S3 applications at significantly increased the yield per hill and ratoon rice height at 20 DAC. The R2S1 and R2S2 combination treatments significantly increased the number of filled grains per panicle and the ratoon rice height at 10 DAC. Cutting the stems at 10 cm combined with various dosages of SLOF resulted in greater growth and yield of ratoon rice for achieving an adequate food supply.

Water safety and management programs (WSMP) utilize field measurements to evaluate control limits and monitor water quality parameters including Legionella presence. This monitoring is important to verify that the plan is being implemented properly. However, once it has been determined when and how to sample for Legionella, it is important to choose appropriate collection and processing methods. We sought to compare processing immediate and flushed samples, filtration of different volumes collected, and sample hold times. Hot water samples were collected immediately and after a 2-min flush. These samples were plated directly and after filtration of either 100 mL, 200 mL, or 1 L. Additionally, unflushed samples were collected and processed immediately and after 1, 24, and 48 h of hold time. We found that flushed samples had significant reductions in Legionella counts compared to immediate samples. Processing 100 mL of that immediate sample both directly and after filter concentration yielded the highest concentration and percent sample positivity, respectively. We also show that there was no difference in culture values from time 0 compared to hold times of 1 h and 24 h. At 48 h, there were slightly fewer Legionella recovered than at time 0. However, Legionella counts were so variable based on sampling location and date that this hold time effect was minimal. The interpretation of Legionella culture results depends on the sample collection and processing methods used, as these can have a huge impact on the success of sampling and the validation of control measures.

Sugarcane can suffer severe yield losses when affected by leaf scald, a disease caused by Xanthomonas albilineans. This bacterial pathogen colonizes the vascular system of sugarcane, which can result in reduced plant growth and plant death. In order to better understand the molecular mechanisms involved in the resistance of sugarcane to leaf scald, a comparative proteomic study was performed with two sugarcane cultivars inoculated with X. albilineans: one resistant (LCP 85-384) and one susceptible (ROC20) to leaf scald. The iTRAQ (isobaric tags for relative and absolute quantification) approach at 0 and 48 h post-inoculation (hpi) was used to identify and annotate differentially expressed proteins (DEPs). A total of 4295 proteins were associated with 1099 gene ontology (GO) terms by GO analysis. Among those, 285 were DEPs during X. albilineans infection in cultivars LCP 85-384 and ROC20. One hundred seventy-two DEPs were identified in resistant cultivar LCP 85-384, and 113 of these proteins were upregulated and 59 were downregulated. One hundred ninety-two DEPs were found in susceptible cultivar ROC20 and half of these (92) were upregulated, whereas the other half corresponded to downregulated proteins. The significantly upregulated DEPs in LCP 85-384 were involved in metabolic pathways, the biosynthesis of secondary metabolites, and the phenylpropanoid biosynthesis pathway. Additionally, the expression of seven candidate genes related to photosynthesis and glycolytic pathways, plant innate immune system, glycosylation process, plant cytochrome P450, and non-specific lipid transfer protein was verified based on transcription levels in sugarcane during infection by X. albilineans. Our findings shed new light on the differential expression of proteins in sugarcane cultivars in response to infection by X. albilineans. The identification of these genes provides important information for sugarcane variety improvement programs using molecular breeding strategies.

Aims: The aim of this study was to evaluate the in vitro effect of different phosphite formulations and concentrations on the development of Colletotrichum musae. Sample: to evaluate the inhibition of germination, mycelial growth and sporulation of Colletotrichum musae. Study Design:  Treatments were conducted in a completely randomized design, with 4 replicates, each replicate consisting of 1 Petri dish. Place and Duration of Study:  Laboratory of Post-Harvest Pathology, State University of Montes Claros, between March and October 2017. Methodology: Three different phosphite formulations were used: FCu1 (4% Cu + 20% P2O5), FCu2 (4% Cu + 22% P2O5) at concentrations of 0.5;1.0; 1.5 and 2.0 mL L-1 and FK (42% P2O5 + 27.7% K2O) at concentrations of 0.5; 1.0; 1.5 and 2.0 mg.L-1. Products were incorporated into the respective culture media. Culture medium alone and culture medium + imazalil were used as controls. Petri dishes were housed in BOD chamber at 25°C under a 12 hours photoperiod. Results: Results were submitted to analysis of variance and regression, and means were compared by the Tukey test (P <0.05). Control was compared to the other treatments by the Dunnet's test (P <0.05). Among the tested phosphite formulations, copper and potassium phosphites were found to reduce the mycelial growth of Colletotrichum musae. FCu2 presents a fungicide-like effect from the concentration of 0.5 m.L-1 in the control of conidia production. As for the FCu1, a fungicide-like effect was observed in the control of germination from the concentration of 1.5 mL.L-1. Conclusion: A significant fungistatic effect was observed between the concentrations of the products in the mycelial growth, sporulation and germination obtaining control of up to 100% of the development of C. musae. Copper phosphites were as effective as fungicide in inhibiting fungal development.

Objective and Design.Legionellacontrol still remains a critical issue in healthcare settings where the preferred approach to health risk assessment and management is to develop a water safety plan. We report the experience of a university hospital, where a water safety plan has been applied since 2002, and the results obtained with the application of different methods for disinfecting hot water distribution systems in order to provide guidance for the management of water risk.Interventions.The disinfection procedures included continuous chlorination with chlorine dioxide (0.4–0.6 mg/L in recirculation loops) reinforced by endpoint filtration in critical areas and a water treatment based on monochloramine (2-3 mg/L). Real-time polymerase chain reaction and a new immunoseparation and adenosine triphosphate bioluminescence analysis were applied in environmental monitoring.Results.After 9 years, the integrated disinfection-filtration strategy significantly reduced positive sites by 55% and the mean count by 78% (P< .05); however, the high costs and the occurrence of a chlorine-tolerant clone belonging toLegionella pneumophilaST269 prompted us to test a new disinfectant. The shift to monochloramine allowed us to eliminate planktonicLegionellaand did not require additional endpoint filtration; however, nontuberculous mycobacteria were isolated more frequently as long as the monochloramine concentration was 2 mg/L; their cultivability was never regained by increasing the concentration up to 3 mg/L.Conclusions.Any disinfection method needs to be adjusted/fine-tuned in individual hospitals in order to maintain satisfactory results over time, and only a locally adapted evidence-based approach allows assessment of the efficacy and disadvantages of the control measures.
