The antioxidant activity, total phenolic and flavonoid contents of different extracts of the Clematis brachiata Thunb leaves were determined. The antioxidant activity was evaluated using spectroscopic methods against 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and 2,2′-azinobis [3-ethylbenzothiazoline-6-sulphonic acid] diammonium salt radical cation (ABTS). Folin Ciocalteu method was used to determine the total phenolics and Aluminium Chloride Colorimetric method was used to determine the flavonoids contents in these extracts. The results showed that the methanol extract of the leaf exhibited the highest antioxidant activity with the value of 180.45 ±2.4 μg mL-1 in DPPH and 60 ±0.80 μg mL-1 in ABTS assay among the extracts. The methanol extract contains more phenolic compounds (178±2.20 mg/g as galic acid equivalent per g dry matter) and the acetone extract contains more flavonoids (135.11±1.20 mg/g as quercitin equivalent per g dry matter) among the extracts. This study provides the evidence that the leaves of the Clematis brachiata Thunb could be a good source of natural antioxidant.Bangladesh J. Sci. Ind. Res.53(3), 185-192, 2018

Free radical is any molecular species that have unpaired free electrons in their outer orbital shell that make radicals highly reactive, resulting in pathogenesis conditions such as cellular injury, premature aging, cancer, hepatic disorders, neurodegenerative diseases, cardiovascular disease, and kidney disease. One source of natural antioxidant is jackfruit. The purpose of this research was to determine the phenolic and flavonoid contents in the extracts and fractions of jackfruit peel and their potential as antioxidants. Jackfruit peel powder was extracted from maceration. The total phenolic content was determined by the Folin-Ciocalteu method. Meanwhile, flavonoid content was determined using the aluminium chloride complex colorimetric method. Measurements of antioxidant activity were conducted using the 2,2-diphenyl-1-picrylhydrazyl (DPPH). The ethyl acetate fraction had high phenolic and flavonoid contents, which were 49.667±1.508 g GAE/100 g of sample and 70.199±0.374 g of quercertin equivalent/100 g. The ethyl acetate fraction had the strongest antioxidant activity with IC50 value of 4.539±0.201 µg/ mL and correlation value (R2 ) of 0.5881 for phenols and R2 of 0.7241 for flavonoids. Ethyl acetate fraction of jackfruit peel is very potential to be developed as a natural antioxidant and functional food.

Ocimum sanctum, commonly known as Tulasi in Nepal, is a pharmacologically important plant due to its active constituents. In this work, extraction was carried out in hexane, chloroform and methanol solvents and their phytochemical screening was performed. Total phenolic and flavonoid contents in the plant were measured by Folin-Ciocalteu colorimetric method and aluminum chloride colorimetric method respectively. Antioxidant activity of the extracts was evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Alkaloids, flavonoids, tannins, glycosides, polyphenols, terpenoids, tannins and steroids are mainly found in the extracts. Based on the result obtained, the plant posses a significant amount of total phenolic and total flavonoid content. Both phenolic and flavonoid contents were highest in methanol extract, followed by chloroform and hexane extract. Antioxidant activity of the extracts as ascorbic acid standard (IC50 value = 41.34 µg/mL) was in the order of methanol extract (IC50 value = 47.73 µg/mL) > chloroform extract (IC50 value = 79.46 µg/mL) > hexane extract (IC50 value = 94.68 µg/mL). The extent of the antioxidant activity of the plant is following the number of total phenolics and flavonoids present in it.

Purpose: To evaluate the antioxidant, anti-inflammatory and anti‐arthritic effects of the methanol extract of Tamus communis roots. Methods: The total phenolic and flavonoid contents were determined using Folin–Ciocalteu’s reagent and aluminium chloride assays, respectively while the antioxidant capacity was determined using DPPH and β-carotene/linoleate tests. For anti-inflammatory activity, carrageenan-induced paw edema in rat was used. Formaldehyde-induced paw edema and adjuvant-induced arthritis in rats were used to evaluate the anti-arthritic effect of the extract. Results: The total phenolic and flavonoid contents of T. communis showed 55.2 ± 0.036 mg GAE/g dry weight and 1.191 ± 0.002 mg QEq/g dry weight, respectively while the antioxidant activity (DPPH) showed a half-maximal inhibitory concentration (IC50 of 0.128 ± 0.011 mg/mL. Using βcarotene/linoleate assay, the extract showed 88.13 ± 4.03 % inhibition. In carrageenan-induced paw edema in rat, all doses of T. communis did not show inhibition of edema. In contrast, formaldehyde induced paw edema decreased at the dose of 150 mg/kg days 2 and 4. However, doses of 300 and 600 mg/kg did not show any activity. In adjuvant-induced arthritis, all used doses caused no reduction in arthritic scores and in paw thickness except a significant decrease with the dose 150 mg/kg at the last day. Conclusion: T. communis extract exhibits high antioxidant activity related to the phenolic compound contents. However, the results of the anti-inflammatory and anti‐arthritic studies did not support its use in folk medicine in the treatment of rheumatic ailment except on low doses

Euphorbia aegyptiaca is a herbaceous plant traditionally used in Sudan for treatment of various diseases, and the study of this plant is still limited. The aim of the present study was to screen the phytochemicals and to assess the Antioxidant activity, total phenolic, flavonoid contents and cytotoxic activity of Euphorbia aegyptiaca. The plant material was extracted successively by Soxhlet apparatus using n-hexane, chloroform and methanol. The chemical constituents of the extracts were carried out using the standard procedures. The Folin- Ciocalteu and Aluminium chloride method was employed to calculate the total phenolic and flavonoid content, respectively. The antioxidant activity, was assessed by measuring the scavenging activity of the DPPH (2.2Di (4-tert-octylphenyl)-1-picryl-hydrazyl) and Propyl Gallate as standard antioxidants. While cytotoxic activities were screened using brine shrimp. Phytochemical screening studies revealed that flavonoids, tannins, coumarins, saponins, sterols, terpenes, anthraquinones and alkaloids were the main phytochemicals present in extracts of E. aegyptiaca. The methanol extract showed the highest level of total phenolic contents (173.49±2.427 mg GAE/g) and flavonoid content (239.53±7.90 mg QE/g), and the highest antioxidant activity 89% with least (IC50 0.0449µg/ml), and the no toxicity against brine shrimp (LD50 3423.156). Furthermore, no toxicity in all extracts was observed. The present study is the first evaluation regarding the characterization of E. aegyptiaca and its safety, and the results demonstrate its antioxidant potential and suggest its safe therapeutic use. The results suggest that methanol extract is a rich source of phytochemicals and exhibits highest amount of and total phenolic, flavonoid content and significant antioxidant activity and it has no cytotoxic activity. E. aegyptiaca plant can be regarded as a promising Source of naturally occurring potential antioxidants. <p>To perform a qualitative and quantitative analysis of the phenolic and flavonoid contents and evaluate the antioxidant activity of ethyl acetate (EtOAc) and <em>n</em>-butanol (<em>n</em>-BuOH) extracts of the aerial parts of <em>Genista ulicina </em>Spach. from Algeria.<strong> </strong>The qualitative analysis of plant extracts was carried out by RP-HPLC using UV detector, whereas the quantification of total phenolic and flavonoid contents was completed according to the Folin-Ciocalteu procedure and aluminium chloride colorimetric method respectively. To evaluate the extract's antioxidant activity, Two in vitro antioxidant tests were employed: DPPH and β-carotene bleaching assay. The HPLC/DAD chromatogram showed several peaks indicating the presence of phenolic acids, flavonoids and isoflavonoids in both extracts. The total phenolic content (TPC) ranged from 62.56 and 50.45 mgGAE/g extract, while the total flavonoids content varied between 53.1 and 48.4 mgQE/g extract for EtOAC and <em>n</em>-BuOH respectively. EtOAc extract showed a maximum inhibition value (78.15%) at 150µg/mL using DPPH test and highest antioxidative power (82.42%) using β-carotene bleaching assay comparing with standards. The HPLC-UV analysis showed the richeness of both extracts in phenolic and flavonoid contents. The EtOAc<em> </em>extract exhibited good antioxidant activities comparing to the <em>n</em>-BuOH extract. Thus <em>Genista ulicina</em> could be indicated as a plant of phytopharmaceutical importance.<strong></strong></p>

Objective: To estimate the in vitro total phenolics, flavonoids contents, antioxidant and antimicrobial activities of various solvent extracts from the medicinal plant Physalis minima Linn.Methods: The crude bioactive were extracted from the dried powder of Physalis minima using methanol, ethyl acetate, chloroform and hexane solvents. Total phenolic content (TPC) and total flavonoid content (TFC) were estimated using Folin-Ciocalteu and aluminium chloride colorimetric methods respectively. 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and ferric reducing antioxidant power (FRAP) assays were used to determine the in vitro antioxidant capacity. The antimicrobial assay was done through agar well diffusion; minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using broth microdilution methods against the Gram-negative bacteria (Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, Proteus vulgaris) and Gram-positive bacteria (Staphylococcus aureus).Results: TPC expressed as gallic acid equivalents (GAE) ranged from 60.27±1.73-151.25±2.50 mg GAE/g dry weight, and TFC expressed as quercetin equivalents (QE) ranged from 56.66±0.80-158.84±2.30 mg QE/g dry weight. Methanol extract showed the highest antioxidant activity followed by ethyl acetate, chloroform, hexane extract and the IC50 values of methanol extract for scavenging DPPH and ABTS free radicals were 280.23±5.75-173.40±0.38µg/ml, respectively. All the extracts have shown potent antimicrobial activity for the zone of inhibition ranged from 9-35 mm; MICs and MBCs values ranged from 0.125-4.0 and 0.25-8.0 mg/ml, respectively towards tested pathogenic species.Conclusion: The comprehensive analysis of the present results demonstrated that Physalis minima possess high potential antioxidant properties which could be used as a viable source of natural antioxidants in treating infections caused by above-mentioned pathogens.

Paku atai plant (Angiopteris ferox Copel), specifically the tuber has been used empirically as an anticancer and antidote materials by Dayaknese in West Kutai, with limited scientific study. Thus, this research had been conducted to determine and evaluate the total phenolic, flavonoid content of this plant along with the antioxidant and toxicity profile. The research used several extracts’ solvents including extraction with ethanol (crude extract, CE) and subsequently partitioned with n-hexane (HF); ethyl acetate (EF) and ethanol aqueous (EAF). The resulted crude extract and fractions were then analyzed through colorimetric method to determine the phenolic and flavonoid total; with DPPH and FRAP to observe the antioxidant activity; and using BSLT method to evaluate the toxicity activity. The results showed that the EF fraction provided the strongest antioxidant activity with IC50 value of 13.79 μg/mL and iron reduction with FRAP value of 387.5±6.41 µM/g. However, a high correlation was observed between the antioxidant and the total phenolic content (r2 = 0.970 - 0.974) but little correlation in total flavonoid (r2 = 0.345 - 0.373). Furthermore, the EF (19.56±7.35) showed the highest toxicity activity followed by CE (22.42±2.10), HF (39.52±7.38), and EAF (41.75±5.10). Therefore, the paku atai tuber can be potentially developed as a natural antioxidant and anticancer material.

<p>Snake fruit (<em>Salacca zalacca</em> (Gaertner) Voss) is one of indigenous fruits from Southeast Asia that has been consumed for its antioxidant contents. Improving the fruit quality might increase its benefits for human health. This study aims to analyze fruit morphology, antioxidant activity, total phenolic and flavonoid contents of the fruit applied with goat manures and <em>Bacillus velezensis </em>B-27. The research used two-factor Randomized Complete Block Design (RCBD) with three blocks as replications. The first factor is the level of goat manure application i.e. 0 kg plant<sup>-1</sup>, 5 kg plant<sup>-1</sup> and 10 kg plant<sup>-1</sup>, while the second factor is the bacteria application i.e. applied with <em>Bacillus velezensis </em>B-27 and without<em> Bacillus velezensis </em>B-27. The research was conducted in June until December 2019 at Turi, Sleman, Yogyakarta, Indonesia. Fruit length, diameter and shape were measured as morphology parameters. Antioxidant activities were measured using 2,2-Diphenyl-1-picrylhydrazyl, whereas flavonoid and phenolic contents were determined by the aluminium-chloride colorimetric and Folin-Ciocalteu methods, respectively. Means of each parameter were analyzed using ANOVA, continued with the Tukey’s HSD test at a 5% significance level. The result showed that the application of 10 kg goat manure per plant with bacteria increased the fruit length (76.78 mm) and the diameter (62.72 mm). Addition of 10 kg goat manure per-plant combined with <em>Bacillus velezensis </em>B-27 gave the highest antioxidant (IC50 of 37.83 µg.mL<sup>-1</sup>), flavonoid (5.35 mgGAE.100 g<sup>-1</sup>) and total phenolic contents (44 mgQE.100 g<sup>-1</sup>).</p>

The total phenolics, flavonoids and in vitro antioxidant activity of petroleum ether, benzene, ethyl acetate, methanol and ethanol extracts of stem of Drypetes sepiaria were determined using various antioxidant model systems viz, DPPH, hydroxyl, superoxide, ABTS and reducing power. Total phenolic content was estimated by Folin-Ciocalteau method. Flavonoids were determined by Aluminium chloride method. The total phenolics and flavonoids contents were found to be 0.81 g 100 g-1 and 1.12g 100 g-1 respectively in the methanol extract. Among the solvent tested, methanol and ethanol extracts of D. sepiaria stem showed potent in vitro antioxidant activities. This investigation explored that D. sepiaria stem is a potential source of natural antioxidant.

Bilberries are known for their bioactive properties such as antioxidant activity. They contain a wide range of flavonoids and phenolic acids that show antioxidant activity. In the present work, the total phenolic and flavonoid contents, correlated with antioxidant activity of Vaccinium extracts were determined. The content of total phenolics in the extracts was determined spectrometrically according to the Folin-Ciocalteu procedure and calculated as gallic acid equivalents (GAE). The content of total flavonoids was measured also spectrophotometrically by using the aluminum chloride colorimetric assay. Effects of ethanol and methanol concentrations of the solutions on the contents of flavonoids and phenolic in extracts were investigated.
