Plant gums and mucilage have wide applications in pharmaceutical formulations as emulsifying agents. But these are quite costly, therefore there is always the need for cheaper alternatives. In this study, Okra mucilage from Abelmoschus esculentus (L.) Moench pods was used to formulate Spondias mombin L. leaf extract emulsion. The emulsion was assessed for the effect of the mucilage on antioxidant activity of the extract using four standard in vitro assays. Phytochemical screenings of the mucilage as well as physico-chemical and microscopic characterization of the emulsion were also carried out. The mucilage contained carbohydrates, reducing sugars, terpenoids and unsaturated lactones. The pH of the resulting emulsion was 6.88. The size of the oil globules were small with the average size of 4. 8μm. The extract had superior 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, hydrogen peroxide radical scavenging and ferric ion reducing capacity compared to the emulsion (p ≤ 0.05). In addition, the emulsion had a better metal chelating activity compared to the extract (p ≤ 0.05). Okra mucilage may have reduced the antioxidant activity of the extract suggesting its incongruity in this emulsion formulation. Bangladesh Pharmaceutical Journal 24(1): 17-25, 2021

Background: Quinoline derivatives have been attracted much attention in drug discovery and synthetic derivatives of these scaffolds present a range of pharmacological activities. Therefore, organoselenium compounds are valuable scaffolds in organic synthesis because their pharmacological activities and their use as versatile building blocks for regio-, chemio-and stereoselective reactions. Thus, the synthesis of selenium-containing quinolines has great significance, and their applicability range from simple antioxidant agents, to selective DNA-binding and photocleaving agents. Objective: In the present study we describe the synthesis and antioxidant activity in vitro of new 7-chloroN(arylselanyl)quinolin-4-amines 5 by the reaction of 4,7-dichloroquinoline 4 with (arylselanyl)-amines 3. Methods: For the synthesis of 7-chloro-N(arylselanyl)quinolin-4-amines 5, we performed the reaction of (arylselanyl)- amines 3 with 4,7-dichloroquinoline 4 in the presence of Et3N at 120 °C in a sealed tube. The antioxidant activities of the compounds 5 were evaluated by the following in vitro assays: 2,2- diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), ferric ion reducing antioxidant power (FRAP), nitric oxide (NO) scavenging and superoxide dismutase-like activity (SOD-Like). Results: 7-Chloro-N(arylselanyl)quinolin-4-amines 5a-d has been synthesized in yields ranging from 68% to 82% by the reaction of 4,7-dichloroquinoline 4 with arylselanyl-amines 3a-d using Et3N as base, at 120 °C, in a sealed tube for 24 hours and tolerates different substituents, such as -OMe and -Cl, in the arylselanyl moiety. The obtained compounds 5a-d presented significant results with respect to the antioxidant potential, which had effect in the tests of inhibition of radical’s DPPH, ABTS+ and NO, as well as in the test that evaluates the capacity (FRAP) and in the superoxide dismutase-like activity assay (SOD-Like). It is worth mentioning that 7-chloro-N(arylselanyl)quinolin-4-amine 5b presented excellent results, demonstrating a better antioxidant capacity when compared to the others. Conclusion: According to the obtained results 7-chloro-N(arylselanyl)quinolin-4-amines 5 were synthesized in good yields by the reaction of 4,7-dichloroquinoline with arylselanyl-amines and tolerates different substituents in the arylselanyl moiety. The tested compounds presented significant antioxidant potential in the tests of inhibition of DPPH, ABTS+ and NO radicals, as well as in the FRAP and superoxide dismutase-like activity assays (SOD-Like).

Green pruning residues (GPRs) and leaves from 16 red and white Vitis vinifera L. cultivars from Piedmont (Italy) were studied. The investigated samples were extracted by ultrasound-assisted extraction optimized by an experimental design, and quali- and quantitatively analyzed by HPLC-PDA-MS/MS. GPRs and leaves show a similar polyphenolic pattern, with quercetin 3-O-glucuronide, caftaric acid, and quercetin 3-O-glucoside as the main components, although in variable proportions. The HPLC results were related to the antioxidant activity, measured as total phenolic content and through DPPH and ABTS assays with similar results. Colorimetric in vitro assays, offline combined with HPLC-PDA analysis, determine which compounds contribute to the antioxidant activity in terms of radical scavenging abilities. Valorization of GPRs is a potential source of natural compounds that could be of interest in the health field, increasing their economic value together with a positive effect on the environment.

Antioxidants from the natural products are essential to prevent the progression of free radical mediated diseases. In the present study, ethanol extract of propolis collected from different geographical origin were evaluated for their free radical scavenging potential by employing different in-vitro assays such as DPPH, ABTS, Nitric oxide and Hydrogen peroxide. All the tested samples contained considerable amount of total phenols and vitamin C content. In the entire assay, the percentage of inhibition increased with the increase in concentration. Among the propolis samples collected, the highest activity was found in Tamil Nadu, Kerala, Karnataka and Haryana. The difference in the antioxidant activity level was obtained from the assay may reflect a relative difference in the ability of antioxidant compounds to scavenge different free radicals in the extract. Phenols and vitamin C are the major contributors to antioxidant activity in propolis. The propolis from these locations may be of considerable interest in preventing the ill effects of excessive free radical generation in the human body.

The activities of the crude acetonic extract of red bean and its two fractions were determined using a β-carotene-linoleate model system as well as the total antioxidant activity (TAA), the total phenolics content (TPC), the DPPH radical-scavenging activity, and the reducing power assays. Results from the in vitro assays showed the highest values when tannins (fraction II) were tested. Specifically, the TAA of the tannins fraction was 4.37 mmol Trolox eq./g fraction; whereas, the crude extract and fraction I were 0.481 and 0.093 μmol Trolox eq./mg extract or fraction, respectively. The content of total phenolics in fraction II was the utmost (612 mg/g); the tannins content, assayed by the vanillin method and expressed as absorbance units at 500 nm per 1 g, was 938. RP-HPLC-PAD-MS profiling revealed the presence of 33 compounds: quercetin arabinoglucoside, quercetin rutinoside, quercetin, p-hydroxybenzoic acid, and kaempferol rutinoside were the most abundant phenolics in the extract.

Muscari comosum L. bulbs are commonly used as food in South Italy and also in folk medicine. By evaluating in vitro antioxidant activity and biological activities of their aqueous and methanol extracts, we shed light on the potential role, including both the nutraceutical and health benefits, of this plant. Total polyphenol content (TPC) and total flavonoid content (TFC) were evaluated by the Folin–Ciocalteu method and by the aluminum chloride method, respectively. Antioxidant activity was investigated by three in vitro assays and relative antioxidant capacity index (RACI) was calculated to compare results obtained by different tests. The extracts were tested to evaluate their possible involvement in redox homeostasis, using the human hepatoma (HepG2) cell line used as model. The extracts exhibited concentration/solvent dependent radical scavenging activity, as well as dysregulation of some genes involved in redox pathways by promoting Nrf2, SOD-2, GPX1, ABCC6 and ABCG2 expression. NMR metabolomics analysis suggests that HepG2 cells treated with Muscari comosum extracts experience changes in some metabolites involved in various metabolic pathways.

Plants are adundantly and are very promisive to be used as source of drugs in many diseases or infections and also it is a main agents of antioxidants which prevents the oxidative stress that are caused by the free radicals. There are numerous studies based on the pharmaceutical and classification of medicinal plants throughout the world. Leaves, fruits, roots are most frequently plant parts used in many research and studies. Here the present study was aimed to evaluate the in vitro antioxidant activity of aqueous and ethanol leaf extracts of Andrographis paniculata Nees and Rhinacanthus nasutus Kurz. Antioxidant is a substance which is used to prevent some types of cell damage in the body. Determination of their in vitro antioxidant activity were carried out by using methods such as DPPH (2,2-diphenyl-1picrylhydrazyl assay), ABTS (2,2′-azinobis (3-ethylbenzothiazolin 6-sulfonic acid assay), FRAP (ferric reducing antioxidant power assay) and SOD (super oxide anion scavenging) assay, H2O2 (hydrogen peroxide radical scavenging) assay. Moreover the ethanolic leaf extracts showed best antioxidant activity than the aqueous leaf extracts. Experimental results reveals that the leaves of A. paniculata have potent antioxidant and free radical scavenging activity than R. nasutus. Further investigation must be done for these two medicinal plants for the discovery of the bioactive compounds.

In vitro antioxidant free radical scavenging capacities of aqueous and alcoholic extracts of herbal drug containing stem bark of Ficus glomerata Roxb. and Symplocos racemosa Roxb. were evaluated. IC50 (µg/ml) of alcoholic, aqueous extract and Ascorbic Acid was found to be 41.529 ±10.135, 39.654 ±4.022and 17.511 ±1.17 in Hydrogen Peroxide Radical Scavenging, 1825 ±122, 1035 ±090 and 207 ±006 in DPPH radical scavenging, and 42.024 ±16.816, 49.926 ±16.240 and 5.503 ±0.545 during FRAP essay. Similarly, during ABTS radical scavenging, IC50 (µg/ml) was assessed as 28.088 ±5.618, 23.731 ±4.870 and 6.728 ±0.213 for alcoholic, aqueous extract and Trolox respectively. Alcoholic and Aqueous extracts exhibit high antioxidant activity possibly due to higher phenolic and flavonoid content

Objectives: The objective of this study was to estimate the total phenolic and flavonoid contents and to explore the antioxidant potential of various leaf extracts (chloroform, ethyl acetate, and ethanol) of Canthium coromandelicum, which is considered traditionally as an important medicinal plant.Methods: Antioxidant properties of the extracts were assessed using 1, 1- diphenyl -2- picrylhydrazyl and hydrogen peroxide radical scavenging assays for ascorbic acid equivalents. The total phenolic and flavonoid contents were also investigated to determine their correlation with the antioxidant activity of the leaf extracts and expressed in Gallic acid and quercetin equivalents, respectively.Results: The results showed that the content of total phenols and flavonoids was found to be high in ethyl acetate extract which was recorded as 61.02±1.30 mg Gallic acid equivalent (GAE/g) and 81.72±0.61 QE/g, respectively. Compared to other extracts, ethyl acetate leaf extract was found to possess high antioxidant activity at p<0.05 level, with high percentage of inhibition at 100 μg/ml concentration (82.70%) toward hydrogen peroxide radical scavenging with IC50 value 62.94 μg/ml. Statistically, two-tailed Pearson’s correlation showed strong positive correlations between hydrogenperoxide radical scavenging activity and total phenolic contents (TPC) (r=1.000) at p<0.05 level.Conclusion: The results obtained in this study clearly signifies that the ethyl acetate leaf extract of C. coromandelicum has high content of total phenols which are correlated to its antioxidant activity and thus has the potential to use as a source of natural antioxidants and can be explored as a therapeutic agent in free radical induced diseases.Keywords: Canthium coromandelicum, In-vitro antioxidant activity, Ethyl acetate extract, Pearson’s correlation, 1, 1- diphenyl -2- picrylhydrazyl, Hydrogen peroxide, Nitric oxide, Superoxide.

The methanolic extract of Duabanga grandiflora (Roxb. ex DC.) Walpers leaves was investigated to explore in-vitro anti-arthritic, thrombolytic and antioxidant potential and subsequent detection of preliminary phytochemicals. It is used for treating various diseases by folk practitioners and rural people. Initially, BSA protein denaturation method was employed to evaluate in-vitro anti-arthritic potency, where the extract exhibited potent anti-arthritic activity with 73.52±1.01% inhibition of BSA denaturation at a dose of 1000 μg/ml compared to the standard dichlofenac sodium (83.66±1.03% inhibition at 1000 μg/ml dose). In-vitro thrombolytic efficacy was determined using streptokinase (positive control) and water (negative control), and the average lysis of blood clot was found to be 31.12±1.09%, which is indicative towards the thrombolytic effect of the extract. Antioxidant activity was evaluated by the DPPH radical scavenging method followed by the total phenolic and flavonoid content determination. The crude methanolic extract revealed the IC50 value of 129.31μg/ml and 204.89μg/ml against DPPH and standard ascorbic acid, respectively. The total phenolic content and total flavonoid content of the plant leaves were observed to be 52.81±1.74μg/ml and 25.77±1.01μg/ml, respectively. The results indicate that the leaves of D. grandiflora possess favorable anti-arthritic activity, moderate thrombolytic and excellent antioxidant activity, which is demonstrating the presence of alkaloids, phenols, flavonoids and many other biologically important phyto-constituents that needs further exploration. Background: Curcumin, as the substantial constituent of the turmeric plant (Curcuma longa), plays a significant role in the prevention of various diseases, including diabetes. It possesses ideal structure features as enzyme inhibitor, including a flexible backbone, hydrophobic nature, and several available hydrogen bond (H-bond) donors and acceptors. Objective: The present study aimed at synthesizing several novel curcumin derivatives and further evaluation of these compounds for possible antioxidant and anti-diabetic properties along with inhibitory effect against two carbohydrate-hydrolyzing enzymes, α-amylase and α-glucosidase, as these enzymes are therapeutic targets for attenuation of postprandial hyperglycemia. Methods: Therefore, curcumin-based pyrido[2,3-d]pyrimidine derivatives were synthesized and identified using an instrumental technique like NMR spectroscopy and then screened for antioxidant and enzyme inhibitory potential. Total antioxidant activity, reducing power assay and 1,1-diphenyl-2-picrylhydrazyl (DPPH• ) radical scavenging activity were done to appraisal the antioxidant potential of these compounds in vitro. Results: Compounds L6-L9 showed higher antioxidant activity while L4, L9, L12 and especially L8 exhibited the best selectivity index (lowest α-amylase/α-glucosidase inhibition ratio). Conclusion: These antioxidant inhibitors may be potential anti-diabetic drugs, not only to reduce glycemic index but also to limit the activity of the major reactive oxygen species (ROS) producing pathways.
