In the publication, a technological system of traditional whistling reed pipes produced by folk artists, widespread in Western Polissia, is revealed for the first time in a step-by-step process. The proposed exploration is based on its expeditionary records and material collected by various folklorists and employ- ees of Kyiv and Lviv research institutions, as well as some literary sources. 131 The existence of three types of nozzle instruments, the names of which correspond to the methods of their production: a pipe made from bark, a vykrutka and kolianka are established. Western Polissian folk reed pipes belong to the type of longitudinal whis- tle flutes with an internal cleft formed by the insertion of the whistling device at the upper end of the tube, and with holes placed on the front, sometimes on the back of the instrument. Whistle airphones have specific production features, according to the age group of masters: shepherd children, teens and adults. The easiest to manufacture among the discovered pipe instruments are ones made of bark, which had been previously removed from the branches of bush trees. In the process of removing the bark shepherds told special spells, which were recorded by the author on the researched area. The following two types of tools were made of wood: a kolianka in the form of a longitudinal splitting of the branch into two equal parts, a vykrutka - a screwing out the core of pine wood, which required a masters knowledge of certain empirical and professional skills. The folk pipe instruments played the role of a musical instrument, which corresponded to the individual artistic and aesthetic test of the flute maker and served as an integral part of the Polisian shepherd's everyday life

The genus Stachybotrys produces a broad diversity of secondary metabolites, including macrocyclic trichothecenes, atranones, and phenylspirodrimanes. Although the class of the phenylspirodrimanes is the major one and consists of a multitude of metabolites bearing various structural modifications, few investigations have been carried out. Thus, the presented study deals with the quantitative determination of several secondary metabolites produced by distinct Stachybotrys species for comparison of their metabolite profiles. For that purpose, 15 of the primarily produced secondary metabolites were isolated from fungal cultures and structurally characterized in order to be used as analytical standards for the development of an LC-MS/MS multimethod. The developed method was applied to the analysis of micro-scale extracts from 5 different Stachybotrys strains, which were cultured on different media. In that process, spontaneous dialdehyde/lactone isomerization was observed for some of the isolated secondary metabolites, and novel stachybotrychromenes were quantitatively investigated for the first time. The metabolite profiles of Stachybotrys species are considerably influenced by time of growth and substrate availability, as well as the individual biosynthetic potential of the respective species. Regarding the reported adverse effects associated with Stachybotrys growth in building environments, combinatory effects of the investigated secondary metabolites should be addressed and the role of the phenylspirodrimanes re-evaluated in future research.

Cognitive–behavioural therapy (CBT) is an effective treatment for a number of psychiatric disorders in adults of all ages. With the proportion of the population aged 65 or over increasing steadily, it is important to be aware of how the CBT needs of this age group can be best met. This article provides an overview of CBT and the historical context of using it with older people. Although an understanding of the individual, irrespective of age, is at the core of CBT, potential modifications to the procedure and content aimed at optimising its effectiveness for older people are discussed.

The role of the dialectical behaviour therapy (DBT) team lead is rarely discussed in the DBT academic or clinical literature. However, much implementation research and clinical experience in training and supporting teams new to DBT indicates the importance of the team lead to the correct and efficient functioning of the team itself. This chapter outlines the role of the team lead in relation to two of the functions of DBT; structuring the environment, and enhancing therapists’ capabilities and motivation. It outlines and discusses the core tasks of each of these functions for the team lead. Additionally, it describes the skills and strategies team leads need to learn and deploy to their team in the individual therapeutic and consultation team settings. Lastly, it outlines common dialectical tensions that can arise for team leads, and offers strategies for their management.

The paper advances a relational perspective to studying creativity at the individual level. Building on social network theory and techniques, we examine the role of social networks in shaping individuals' ability to generate a creative outcome. More specifically, we argue that individuals who occupy an intermediate position between the core and the periphery of their social system are in a favorable position to achieve creative results. In addition, the benefits accrued through an individual's intermediate core/periphery position can also be observed at the team level, when the same individual works in a team whose members come from both ends of the core/periphery continuum. We situate the analysis and test our hypotheses within the context of the Hollywood motion picture industry, which we trace over the period 1992–2003. The tragic case of Mayra Cabrera who died as a result of wrong route drug administration is notable as it was the first time a verdict of unlawful killing was recorded against an NHS Trust. Error within medicine is a significant cause of patient morbidity and mortality. We explore the costs of error, the dynamics of error causation, the role of both the individual and institution in accountability for error, as well as transferrable lessons from other industries to reduce error.

ABSTRACT Vipoxin is a heterodimeric neurotoxin isolated from the venom of the Bulgarian long-nosed viper Vipera ammodytes meridionalis. Vipoxin represents a noncovalent association of two subunits - a basic and toxic phospholipase A2 enzyme, and an acidic nonenzymatic component (vipoxin’s acidic component). It was postulated that the phospholipase A2 subunit was more toxic than the whole vipoxin complex and the function of the acidic component was to reduce the enzymatic and toxic activities of the basic phospholipase A2. In the present study, we report new data on the acute toxicity (LD50) of vipoxin and its individual separated components. Vipoxin LD50 (mice, i.p. and i.v.) values were found to be 0.7-1.2 mg/kg b.w. (i.p.) and 0.9-1.3 mg/kg b.w. (i.v.). The established LD50 values for the separated pure phospholipase A2 subunit are higher - 10.0-13.0 mg/kg b.w (i.p.) and 2.2-3.0 mg/kg b.w. (i.v.), i.e. the individual phospholipase A2 subunit displays less toxic activity than vipoxin, contrary to the data published in the literature. The reconstituted vipoxin complex (obtained after preliminary incubation of pure separated phospholipase A2 and acidic component showed enzyme activity and toxicity comparable to that of the native vipoxin complex. Addition of acidic component to the phospholipase A2 subunit showed a positive effect on the enzymatic activity, reaching maximal enzyme reaction rate of acidic component to phospholipase A2 molar ratio of 0.8:1 on using 4-nitro-3-octanoyloxy-benzoic acid as substrate. For the first time we showed that the acidic subunit was absolutely required for the toxic activity of vipoxin. Based on the obtained results, we assume that the function of the acidic component is to stabilize the neurotoxin’s quaternary structure, required for its toxic and enzymatic activities, similarly to the role of the acidic component of crotoxin.

Since 2010, outbreaks of haemorrhagic septicaemia (HS) caused by Pasteurella (P.) multocida capsular type B (PmB) emerged in Germany. In 2017, we noticed a close spatiotemporal relationship between HS outbreak sites and wolf (Canis lupus) territories. Thus, the main objectives of our study were to investigate the molecular epidemiology of German PmB-HS-isolates and to assess the role of wolves as putative vectors of this pathogen. We collected 83 PmB isolates from HS outbreaks that occurred between 2010 and 2019 and sampled 150 wolves, which were found dead in the years 2017 to 2019, revealing another three PmB isolates. A maximum-likelihood-based phylogeny of the core genomes of 65 PmB-HS-isolates and the three PmB-wolf-isolates showed high relatedness. Furthermore, all belonged to capsular:LPS:MLST genotype B:L2:ST122RIRDC and showed highly similar virulence gene profiles, but clustered separately from 35 global ST122RIRDC strains. Our data revealed that German HS outbreaks were caused by a distinct genomic lineage of PmB-ST122 strains, hinting towards an independent, ongoing epidemiologic event. We demonstrated for the first time, that carnivores, i.e., wolves, might harbour PmB as a part of their oropharyngeal microbiota. Furthermore, the results of our study imply that wolves can carry the pathogen over long distances, indicating a major role of that animal species in the ongoing epidemiological event of HS in Germany.

The purpose of this article is a dynamic analysis of the transformations taking place in the speech culture of bilingual students of the post-Soviet space. Russian-foreign bilingualism, as our survey shows, has undergone a certain reconfiguration over the past decade. Previous experiments convincingly proved that the Russian language is dominant in the speech culture of bilingual students, which was due to a number of estralinguistic and linguistic-functional factors. Autochthonous languages in the cognitive structure of students occupied a less stable position and were communicatively limited even in conditions of microdiscursive functioning. It was safe to say that Russian was the core of the linguistic consciousness of bilingual students. Today the situation has changed. The role of autochthonous languages is signified. The Russian language still remains a communicative dominant, providing the basic communicative needs of the individual (including the need for training and the formation of professional competencies), however, it is gradually shifting from the core towards the center, which may indicate that linguo-constructive functions in the future may be lost, and new ones associated with the ethnically primary language have not yet been formed (taking into account the small historical time), which will entail the phenomenon of mass semilingualism.

Core–shell based nanostructures are attractive candidates for photocatalysis owing to their tunable physicochemical properties, their interfacial contact effects, and their efficacy in charge-carrier separation. This study reports, for the first time, on the synthesis of mesoporous silica@nickel phyllosilicate/titania (mSiO2@NiPS/TiO2) core–shell nanostructures. The TEM results showed that the mSiO2@NiPS composite has a core–shell nanostructure with a unique flake-like shell morphology. XPS analysis revealed the successful formation of 1:1 nickel phyllosilicate on the SiO2 surface. The addition of TiO2 to the mSiO2@NiPS yielded the mSiO2@NiPS/TiO2 composite. The bandgap energy of mSiO2@NiPS and of mSiO2@NiPS/TiO2 were estimated to be 2.05 and 2.68 eV, respectively, indicating the role of titania in tuning the optoelectronic properties of the SiO2@nickel phyllosilicate. As a proof of concept, the core–shell nanostructures were used as photocatalysts for the degradation of methyl violet dye and the degradation efficiencies were found to be 72% and 99% for the mSiO2@NiPS and the mSiO2@NiPS/TiO2 nanostructures, respectively. Furthermore, a recyclability test revealed good stability and recyclability of the mSiO2@NiPS/TiO2 photocatalyst with a degradation efficacy of 93% after three cycles. The porous flake-like morphology of the nickel phyllosilicate acted as a suitable support for the TiO2 nanoparticles. Further, a coating of TiO2 on the mSiO2@NiPS surface greatly affected the surface features and optoelectronic properties of the core–shell nanostructure and yielded superior photocatalytic properties.

AbstractIn recent years, our understanding of the pathophysiology of chronic lymphocytic leukemia (CLL) has advanced significantly. It is now clear that CLL is a relatively proliferative disorder that requires the help of its microenvironment to be maintained and to progress. The stimulation of the CLL cell occurs in most, if not all, patients through antigen stimulation via the BCR. In addition, there is now a clearer appreciation of the role of the p53 pathway leading to chemoresistance. These insights are allowing a more targeted approach with the use of p53-independent drugs such as mAbs and high-dose steroids to overcome genetically poor-risk CLL. The elucidation of the molecular and intracellular signaling mechanisms of disease is just beginning to facilitate the development of several targeted small molecules that promise to revolutionize the treatment of CLL. The measurement of the level of minimal residual disease (MRD) in CLL is becoming more available, facilitating approaches in which the aim of therapy is the eradication of detectable MRD. This also promises to improve personalization of therapy to the individual. Recently, the addition of rituximab to fludarabine plus cyclophosphamide (FCR) has improved overall survival in CLL for the first time, and it appears that this will only be the first small step on the path to much more effective therapies and, hopefully, less toxic targeted therapies.
