Edamame milk (Glycin max. L var. edamame) is one product with the advantages; easily produced and highly nutritious. This study aims was to determine the fat content of edamame milk prepared by edamame bean. The method used in this study is the experiment using a completely randomized design with 4 treatments and 5 replications. Fat analysis was done using the Gerber method. The fat levels of edamame milk at the first treatment, the average value of Edamame milk was 5.74% , the second 5.28%, the third 4.56% and the last treatment or the the fourth is fat at 2.0%. The results are compared with the other fat content of the other varieties, then it can be compared the fat content of Edamame varietes lower than the variants of Malika and Grobogan which are variants of Edamame fat content which is 5.74%, Malika fat content was 13.2%, and the Grobogan variance averaged 12.83%. The results of the analysis show that there are significant differences between treatments. For the average edamame fat content that is good is equal to 2.02% and the results close to the standard of soy milk that is equal to 2.0%.Keywords: fat content, edamame milk, Gerber method.ABSTRAKSusu edamame (Glycin max. L var. edamame) merupakan salah satu produk dengan kelebihan; mudah diproduksi dan bergizi tinggi. Penelitian ini bertujuan adalah menentukan kadar lemak susu edamame olahan biji Edamame. Metode yang digunakan dalam penelitian ini yaitu eksperimen dengan Rancangan Acak Lengkap dengan 4 perlakuan dan 5 kali ulangan. Analisis lemak dilakukan menggunakan metode Gerber. Hasil penelitian kadar lemak susu edamame pada perlakuan pertama, nilai rata-rata ulangan lemak susu edamame yaitu sebesar 5,74%, kedua 5,28% ketiga 4,56% dan keempat 2,02% Jika hasilnya dibandingkan dengan kadar lemak varietes lainnya maka bisa di bandingkan kadar lemak varietes edamame lebih rendah dari varietes malika dan grobogan yang mana varietes edamame kadar lemaknya yaitu sebesar 5,74%. Malika kadar lemaknya sebesar 13,2%, dan varietes Grobogan rata-ratanya sebesar 12,83%. Hasil analisis menunjukkan bahwa terdapat perbedaan yang signifikan diantara perlakuan. Rata-rata kadar lemak edamame yang baik yaitu sebesar 2,02%  dan hasilnya mendekati standar susu kedelai sebesar 2,0%Kata kunci: kadar lemak, susu edamame, metode Gerber.

<p><strong>Colour alternation and et/tylene changes during ripening tobacco leaves cultivar Madura and its effect to the quality of slice Madura tobacco</strong></p><p>The colour change and elhylene production in the leaves of Madura tobacco (Nicotiana tabacum) during ripening was studied at "Labora¬ torium Biokimia Pusat Antar Universitas Pangan dan Gizi Universitas Gadjahmada Yogyakata" and "Laboratorium Inti dan Atom Jurusan I'isika, FMIPA Universitas Gadjahmada Yogyakata". The change of colour was measured with Chrommameter CR-200. while the production of ethylcne, ACC (1-aminocyclopropanc-l-carboxylic acid) and MACC (malonyl-ACC) were measured with Photoaccuslic Spectroscopy, from the sample of 100 kg tobacco leaves cv. Jepon grown from April to September 1995 in Plakpak, Pcgantenan, Pamekasan, East Java. The experiment was designed as a completely randomized design in three replicates. The leaves samples were taken randomly from the mature middle leaves Results showed that endogenous ethylcne production al the irst day al a level of 169 ppb. was able to initiate ripening. Conversion percentage of ACC to ethylcne for 8 days ripening increased from 7.7 up lo 61.4 %. F tests least signiicantly different (1%). showed that yellow intensity signiicantly increased on the second day. This indicated thai ripening process was stated on the second day. On the other hand green intensity significantly decreased at the ifth day, thus no chlorophyl degradation. Brightness was signiicantly observed on Ihe third day of ripening, meaning that ripening stated on the third day. Based on the three criteria, the best ripening period was from the third lo the ilth day. This implies that the farmers will have three day periods of ripening which can be arranged to overcome the shotage of man power for slicing the leaves.</p>

The present study was conducted to polyploidy possibility induction of two Iranian endemic mint (Mentha mozaffarianii) ecotypes. For this purpose, three experiments were done. The first experiment was factorial, based on completely randomized design with three factors and three replications that rhizomes were used for treatment. The first factor including different colchicine concentrations (0, 0.025, 0.05, 0.1 and 0.2% that 0 as control). The second factor including two Mentha ecotypes (Ecotype A: Kamarej region and Ecotype B: Pirmohlat region) and the third factors consist of two soaking time (6 h and 12 h). In second experiment, apical meristem was treated. The factorial experiment based on randomized completely design with two factors and five replications. The factors including different colchicine concentrations (0, 0.035, 0.07, 0.15, 0.3 and 0.7%) and two ecotypes. In the third experiment, colchicine as combined with irrigation was applied. For this purpose, a factorial experiment in randomized complete design with two factors (colchicine concentrations: 0, 0.025, 0.012, 0.006% and two mint ecotypes) and three replications was conducted. At the end of all experiments, survival rate and tetraploidy percentage (by morphological change, stomata traits, flow cytometry and chromosome counts) were measured. The results showed that different treatment had significant effects on survival percent on all experiments and increasing of colchicine concentration caused decreasing plants survival. On the other hand, tetraploidy changes only in the first experiments were observed. Only in 0.025% colchicine treatment with 6 h soaking time on ecotype A, 12.66% tetraploidy was identified. Totally, it is seems that Mentha mozaffarianiihardly response to colchicine for tetraploidy induction.

ABSTRAK  Penelitian bertujuan untuk mengetahui pengaruh pemberian bubuk daun Moringa oleifera (MO) terhadap kuantitas dan kualitas semen sapi peranakan ongole (PO). Metode penelitian menggunakan percobaan lapang dengan rancangan acak lengkap dengan tiga perlakuan pemberian MO sebesar 0 kg/ekor/hari (P1); 0,05 kg/ekor/hari (P2) dan 0,1 kg/ekor/hari (P3). Analsis data menggunakan one way analysis of variance. Masing-masing perlakuan terdiri atas empat ekor dengan tiga periode pengamatan sebagai ulangan, yaitu periode pertama dan ketiga tanpa MO, dan periode kedua diberikan MO pada P2 dan P3. Hasil penelitian menunjukan bahwa volume dan pH semen perlakuan P1, P2 dan P3 tidak berbeda semua periode pengamatan. Motilitas massa spermatozoa P2 dan P3 periode kedua dan ketiga menunjukan nilai +++ berbeda nyata (P<0,05) dibandingkan dengan P1 (++); motilitas individu  perlakuan P1 menunjukkan nilai terendah 60,56±2,94 % (P<0,05) dibandingkan P2 dan P3 pada periode kedua dan ketiga (75,50±2,29 dan 72,50±2,34%). Konsentrasi spermatozoa P2 dan P3 periode kedua dan ketiga menunjukkan rata-rata  >1.350 juta/ml berbeda nyata (P<0,05) daripada P1 (876±152juta/ml), sedangkan total spermatozoa motil menunjukkan nilai tertinggi 5.647±829 juta/ml pada P3 periode kedua.Viabilitas spermatozoa P2 dan P3 menunjukkan nilai >88-89 % beda nyata (P<0,05) daripada P1 (<84 %) pada periode kedua dan ketiga, sedangkan nilai abnormalitas spermatozoa yang terbaik adalah P2 sebesar 4,30% pada periode kedua dan P3 sebesar 5,33% pada periode ketiga. Disimpulkan pemberian bubuk daun MO dengan dosis 0,1 kg/ekor/hari dapat memperbaiki kuantitas dan kualitas semen (viabilitas dan total spermatozoa motil) sapi pejantan PO.Kata kunci:  kuantitas dan kualitas semen, Moringa oleifera, sapi pejantan ABSTRACTThe research aimed to determine the effect of Moringa oleifera (MO) leaf powder on the quantity and quality of semen of ongole crossbreed. The research method used was a field experiment with a completely randomized design with three treatments giving MO of 0 kg/head/day (P1); 0.05 kg/head/day (P2) and 0.1 kg/head/day (P3). Each treatment consisted of four heads with three periods of observation as replications, namely the first and third periods without MO, and the second period was given MO on P2 and P3. Data analysis used a one-way analysis of variance. The results showed that the volume and pH of semen treated P1, P2, and P3 didn’t different in all observation periods. The second and third period spermatozoa mass motility of P2 and P3 showed that the value of +++ was significantly different (P<0.05) compared to P1 (++); Individual motility of P1 treatment showed the lowest value 60,56±2,94 % (P<0.05) compared to P2 and P3 in the second and third periods (75,50±2,29 and 72,50±2,34%). The second and third periods of P2 and P3 spermatozoa concentrations showed an average of >1,350 million/cc significantly different (P<0.05) than P1 (876±152 million/cc), while the total motile spermatozoa showed the highest value of 5,647±829 million/cc in the second period P3. The spermatozoa viability of P2 and P3 showed values >88-89% significantly different (P<0.05) than P1 (<84%) in the second and third periods, while the best spermatozoa abnormalities were P2 at 4.30% in the second period and P3 at 5.33% in the third period. It was concluded that the giving of MO leaf powder at a dose of 0,1 kg/head/day could improve the quantity and quality of semen (viability and total motile spermatozoa) of Ongole Crossbreed bulls. Saccharomyces cerevisiae is a yeast strain often used to improve the feed quality of ruminants. However, S. cerevisiae has limited capacity to provide biomass when inoculated with carbon sources and a low ability to produce cellulase enzymes. Here, we hypothesized that yeast in the rumen produces a large amount of biomass and could release cellulase enzymes to break down fiber content. Therefore, the aim of this study was to screen, isolate and identify yeast from the rumen fluids of Holstein Friesian steers and measure the efficiency of biomass production and cellulase activity. A fermentation medium containing sugarcane molasses as a carbon source and urea as a nitrogen source was optimized. Two fistulated–crossbred Holstein Friesian steers averaging 350 ± 20 kg body weight were used to screen and isolate the ruminal yeast. Two experiments were designed: First, a 12 × 3 × 3 factorial was used in a completely randomized design to determine biomass and carboxymethyl cellulase activity. Factor A was the isolated yeast and S. cerevisiae. Factor B was sugarcane molasses (M) concentration. Factor C was urea (U) concentration. In the second experiment, potential yeasts were selected, identified, and analyzed for 7 × 4 factorial use in a completely randomized design. Factor A was the incubation times. Factor B was the isolated yeast strains, including codes H-Khon Kaen University (KKU) 20 (as P. kudriavzevii-KKU20), I-KKU20 (C. tropicalis-KKU20), and C-KKU20 (as Galactomyces sp.-KKU20). Isolation was imposed under aerobic conditions, resulting in a total of 11 different colonies. Two appearances of colonies including asymmetric colonies of isolated yeast (indicated as A, B, C, E, and J) and ovoid colonies (coded as D, F, G, H, I, and K) were noted. Isolated yeast from the rumen capable of providing a high amount of biomass when inoculant consisted of the molasses 15% + urea 3% (M15 + U3), molasses 25% + urea 1% (M25 + U1), molasses 25% + urea 3% (M25 + U3), and molasses 25% + urea 5% (M25 + U5) when compared to the other media solution (p < 0.01). In addition, 11 isolated biomass-producing yeasts were found in the media solution of M25 + U1. There were 4 isolates cellulase producing yeasts discovered in the media solution of M25 + U1 and M25 + U5 whereas molasses 5% + urea 1% (M5 + U1), molasses 5% + urea 3% (M5 + U3), molasses 5% + urea 5% (M5 + U5), molasses 15% + urea 1% (M15 + U1), molasses 15% + urea 3% (M5 + U3), and M25 + U3 were found with 2, 3, 1, 2, 1, and 2 isolates, respectively. Ruminal yeast strains H-KKU20, I-KKU20, and C-KKU20 were selected for their ability to produce biomass. Identification of isolates H-KKU20 and I-KKU20 revealed that those isolates belonged to Pichia kudriavzevii-KKU20 and Candida tropicalis-KKU20 while C-KKU20 was identified as Galactomyces sp.-KKU20. Two strains provided maximum cell growth: P. kudriavzevii-KKU20 (9.78 and 10.02 Log cell/mL) and C. tropicalis-KKU20 (9.53 and 9.6 Log cells/mL) at 60 and 72 h of incubation time, respectively. The highest ethanol production was observed in S. cerevisiae at 76.4, 77.8, 78.5, and 78.6 g/L at 36, 48, 60, and 72 h of incubation time, respectively (p < 0.01). The P. kudriavzevii-KKU20 yielded the least reducing sugar at about 30.6 and 29.8 g/L at 60 and 72 h of incubation time, respectively. The screening and isolation of yeasts from rumen fluids resulted in 11 different yeasts being obtained. The potential yeasts discovered in the rumen fluid of cattle were Pichia kudriavzevii-KKU20, Candida tropicalis-KKU20, and Galactomyces sp.-KKU20. P. kudriavzevii-KKU20 had higher results than the other yeasts in terms of biomass production, cellulase enzyme activity, and cell number.

The increased rate of sugarcane harvest without previous burn has provided a very favorable environment to the froghopper Mahanarva fimbriolata (Stal, 1854), with high moisture and low temperature variation. Few works have studied the response of sugarcane to this pest, so little is known about resistant cultivars. Plant phenolics are widely studied compounds because of their known antiherbivore effect. This research aims to determine if the attack of M. fimbriolata nymphs stimulates the accumulation of total phenolics in sugarcane. The experiment was carried out in greenhouse and arranged in completely randomized design, in a 3 X 2 X 4 factorial with three replications. Second instar nymphs of M. fimbriolata were infested at the following rates: control, 2-4 and 4-8 nymphs per pot (first-second infestations, respectively). Pots were covered with nylon net and monitored daily to isolate the effect of leaf sucking adults. Leaf and root samples were collected and kept frozen in liquid nitrogen until analyses. Infested plants showed higher levels of phenolics in both root and leaf tissues. In roots, the cultivar SP80-1816 accumulated more phenolic compounds in response to the infestation of M. fimbriolata. On the other hand, higher levels were found in leaves and roots of control plants of SP86-42, which might be an indication of a non-preference mechanism. The increase of total phenolics in sugarcane infested with root-sucking froghopper nymphs does not seem to be useful to detect the resistance to this pest.

One of the entomopatogenous fungus types commonly observed and showed potency as myco-insectiside is Beauveria bassiana(Bals.) Vuill.In order to support effectiveness and patogenous activity of B. bassiana, it is necessary to add a carrying agent that protects its spores from ultra violet ray. This study aims to investigate the effect of storage temperature on viability of B. Bassianaspores on the carrier material. The observation was carried out in the Laboratory of Plant Diseases, Indonesian Coffee and Cocoa Research Institute. The research was arranged in completely randomized design by three factors. The first factor was carrier (C), that consists of C1 = rice flour, C2 = maize flour and C3 = tapioca flour. The second factor was dosage (D), that consists of D1 = 1 g B. bassiana+ 1 g carrier; D2 = 1 g B. bassiana+ 5 g carrier and D3 = 1 g B. bassiana+ 10 g carrier. The third factor was temperature of the storage (T),that consists of T1 = 5oC; T2 = 23oC and T3 = 29oC. Viability of B. Bassiana spores was examined by observing development of 100 blastopores randomly and determined under light microscope with 400 times magnification. Observation was conducted in two replicates after the spores of B. bassiana were kept in the storage for 2, 4, 8 and 16 weeks. The result showed that by adding 1 g tapioca flour and temperature of storage of 5oC was potentiall in keeping viability of B. bassianaspores at least for 2 months. It was due to that tapioca flour gave better effect than rice and maize flours in keeping the storage and appropriate low temperature. Viability of B. bassianaspores decreased with increasing carrier dosage, temperature and duration of the storage. Whereas, storage at 5oC was found to be a better condition in keeping viability of dry pure B. bassianaspores longer than conditions of 23o and 29oC. This research is about effectivity analysis of two kinds of fungi i.e. Beauveria bassiana and Metharrizium anisopliae as Aedes aegyptilarvacide, it has been conducted at laboratories Zoology in Biology Department, Mathematics and Natural Sciences Faculty, Syiah KualaUniversity. Aims of this research were estimating maximum time storage of fungi powder as effective larvacide and the number of bacteriacolonies were found in water used for experiment. This research was used Completely Randomized Design (CRD). The experiments consistof two factors i.e kind and saving time of fungi. The result show that the longer the storage time of Beauveria bassiana the more ineffectiveit was as larvacide and the least fungi colonies were found. Metharrizium anisopliae on the other hand, the longer it was kept, the moreeffective it was as larvacide and the more fungi colonies were found.

<p><em>Coffee is a beverage that is widely consumed around the world. Proper packaging and storage temperature may extend shelf life of ground coffee. The study aimed to analyze the shelf life of ground Arabica coffee stored in different packaging types and temperature, conducted at smallholder coffee plantations in Garut Regency and the Integrated Laboratory of Indonesian Industrial and Beverage Crops Research Institute, Sukabumi, from June to August 2018. A completely randomized design in factorial was used with 3 factors and 2 replications. The first factor was the packaging type  which consisted of 3 types: thick alumunium  foil 65</em><em>m</em><em> (AF65), thick alumunium  foil 130</em><em>m</em><em> (AF130), and thick lamination 114</em><em>m</em><em> (L144). The second factor was the storage temperature which consisted of 3 levels: 25 <sup>o</sup>C, 35 <sup>o</sup>C, and 45 <sup>o</sup>C, while the third factor was the storage period which consisted of 5 levels: coffee unstored, and coffee stored for 2 weeks, 4 weeks, 6 weeks, and 8 weeks. The variables observed were the water and fat content, and the analysis of shelf life was carried out using the ASLT (Accelerated Shelf Life Test) method. The results showed that during storage, the water content increased, whereas the fat content decreased. Fat content is a critical variable in determining the shelf life of coffee. The coffee in AF130 packaging has longer shelf life than in AF65 and L144. To extend the shelf life of coffee packaged in AF130 and L144 is best kept at 45<sup> o</sup>C whereas coffee in AF65 packaging  is ideally at 25<sup> o</sup>C.</em></p>

Red yeast rice is one of fermented rice product by Monascus purpureus. Traditionally, the substrate used to produce red yeast rice is rice. Usually, the rice with high amylose content is proper to produce red yeast rice than low amylose. The other substrates that be used to produce red yeast rice are corn and dried cassava. The purposes of this research were to determine the effect of substrates variation (rice, corn and dried cassava) on antioxidant activity and anti-cholesterol content in red yeast rice, and to determine the substrate(s) that produce the highest antioxidant activity and anticholesterol content. The design of this research was a Completely Randomized Design with one factor, i.e. the kind of substrates: rice, corn and dried cassava, with three replications. Then, the data were analyzed with ANOVA at a level of significance α=0.05, and continued with DMRT at the same level. This results showed that the effect of substrates kind to antioxidant activity and anti-cholesterol content on red yeast rice. The rice substrate had higher antioxidant activity and anti-cholesterol content than corn and dried cassava substrates. The antioxidant activity and the anti-cholesterol content on red yeast rice from rice substrate were 45.6100% and 0.026600%, respectively. The antioxidant activity and the anti-cholesterol content on red yeast rice from corn substrate were 44.0500% and 0.022833%, respectively, while the antioxidant activity and the anti-cholesterol content on red yeast rice from dried cassava substrate were 42.8333% and 0.013200%, respectively.

Salah satu faktor yang menyebabkan penurunan produksi kakao yaitu serangan hama penggerek buah kakao. Semut hitam (Dolichoderus thoracicus) diketahui sebagai agen hayati pengendali hama penggerek buah kakao. Penelitian ini bertujuan untuk menghitung kemampuan memangsa semut hitam pada larva penggerek buah kakao, dan menghitung persentase larva penggerek buah kakao yang dimangsa serta mengukur potensi semut hitam sebagai agen hayati penggerek buah kakao. Penelitian ini di laksanakan di Desa Jaya Makmur Kecamatan Nuhon Kabupaten Banggai. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 3 ulangan, dengan masing-masing perlakuan sebagai berikut : P1 (20 larva:10 predator), P2 (20 larva:15 predator), P3 (20 larva:20 predator), P4 (20 larva:25 predator), P5 (20 larva:30 predator), P6 (20 larva:35 predator). Hasil penelitian menunjukkan bahwa perlakuan semut hitam pada larva penggerek buah kakaoSalah satu faktor yang menyebabkan penurunan produksi kakao yaitu serangan hama penggerek buah kakao. Jumlah larva penggerek yang dimangsa tertinggi pada perlakuan P6 (20 larva:35 predator) dua minggu setelah aplikasi dengan nilai rata-rata 16,3 kemudian satu minggu setelah aplikasi dengan nilai rata-rata 14,3. Persentase jumlah larva penggerek yang dimangsa tertinggi pada perlakuan P6 yaitu 81% pada saat dua minggu setelah aplikasi dan 0,71% pada saat satu minggu setelah aplikasi. (Conopomorpha cramerella) berpengaruh sangat nyata terhadap larva penggerek buah kakao. Jumlah larva penggerek yang dimangsa tertinggi pada perlakuan P6 (20 larva:35 predator) dua minggu setelah aplikasi dengan nilai rata-rata 16,3 kemudian satu minggu setelah aplikasi dengan nilai rata-rata 14,3. Persentase jumlah larva penggerek yang dimangsa tertinggi pada perlakuan P6 yaitu 81% pada saat dua minggu setelah aplikasi dan 0,71% pada saat satu minggu setelah aplikasi. One of the factors that caused the decline in cocoa production was the attack of cocoa pod borer. Black ants (Dolichoderus thoracicus) are known to be biological agents to control cocoa pod borer. This study aims to calculated the ability to prey on black ants in the cocoa pod borer larvae, the percentage of cocoa pod borer larvae that are eaten and to measured the potential of black ants as biological agents of cocoa pod borer. This research was conducted in Jaya Makmur Village, Nuhon District, Banggai Regency. The research used a completely randomized design consisted of 6 treatments and 3 replications, with each treatment as follows : P1 (20 larvae: 10 predators), P2 (20 larvae: 15 predators), P3 (20 larvae: 20 predators), P4 (20 larvae: 25 predators), P5 (20 larvae: 30 predators), and P6 (20 larvae : 35 predators). The results showed that the treatment of black ants on cocoa pod borer larvae (Conopomorpha cramerella) had a very significant effected on the cocoa pod borer larvae. The highest number of dead borer larvae was in the P6 treatment (20 larvae: 35 predators) two weeks after application with an average value of 16.3 then one week after application with an average value of 14.3. The highest percentage of borer larvae preyed on the P6 treatment was 81% at two weeks after application and 0.71% at one week after application.
