Plants and their associated microbiota share ecological and evolutionary traits that are considered to be inseparably woven. Their coexistence foresees the use of similar metabolic pathways, leading to the generation of molecules that can cross-regulate each other’s metabolism and ultimately influence plant phenotype. However, the extent to which the microbiota contributes to the overall plant metabolic landscape remains largely unexplored. Due to their early presence in the seed, seed-borne endophytic bacteria can intimately colonize the plant’s endosphere while conferring a series of phytobeneficial services to their host. Understanding the dynamics of these endophytic communities is a crucial step toward the formulation of microbial inoculants that can modulate the functionality of the plant-associated microbiota for improved plant fitness. In this work, wheat (Triticum aestivum) roots non-inoculated and inoculated with the bacterium Herbaspirillum seropedicae strain RAM10 were analyzed to explore the impact of inoculant–endophyte–wheat interrelationships on the regulation of tryptophan (Trp) metabolism in the endosphere environment. Root inoculation with H. seropedicae led to phylum-specific changes in the cultivable seed-borne endophytic community. This modulation shifted the metabolic potential of the community in light of its capacity to modulate the levels of key Trp-related metabolites involved in both indole-3-acetic acid (IAA) biosynthesis and in the kynurenine pathway. Our results support a mode of action of H. seropedicae relying on a shift in both the composition and functionality of the seed-borne endophytic community, which may govern important processes such as root growth. We finally provide a conceptual framework illustrating that interactions among roots, inoculants, and seed-borne endophytes are critical to fine-tuning the levels of IAA in the endosphere. Understanding the outcomes of these interactions is a crucial step toward the formulation of microbial inoculants based on their joint action with seed-borne endophytic communities to promote crop growth and health in a sustainable manner.

The food safety security (FSS) concept is perceived as an early warning system for minimizing food safety (FS) breaches, and it functions in conjunction with existing FS measures. Essentially, the function of FS and FSS measures can be visualized in two parts: (i) the FS preventive measures as actions taken at the stem level, and (ii) the FSS interventions as actions taken at the root level, to enhance the impact of the implemented safety steps. In practice, along with FS, FSS also draws its support from (i) legislative directives and regulatory measures for enforcing verifiable, timely, and effective compliance; (ii) measurement systems in place for sustained quality assurance; and (iii) shared responsibility to ensure cohesion among all the stakeholders namely, policy makers, regulators, food producers, processors and distributors, and consumers. However, the functional framework of FSS differs from that of FS by way of: (i) retooling the vulnerable segments of the preventive features of existing FS measures; (ii) fine-tuning response systems to efficiently preempt the FS breaches; (iii) building a long-term nutrient and toxicant surveillance network based on validated measurement systems functioning in real time; (iv) focusing on crisp, clear, and correct communication that resonates among all the stakeholders; and (v) developing inter-disciplinary human resources to meet ever-increasing FS challenges. Important determinants of FSS include: (i) strengthening international dialogue for refining regulatory reforms and addressing emerging risks; (ii) developing innovative and strategic action points for intervention {in addition to Hazard Analysis and Critical Control Points (HACCP) procedures]; and (iii) introducing additional science-based tools such as metrology-based measurement systems.

Background: Data suggest that having high expectations about hearing aids results in better overall outcome. However, some have postulated that excessively high expectations will result in disappointment and thus poor outcome. It has been suggested that counseling patients with unrealistic expectations about hearing aids prior to fitting may be beneficial. Data, however, are mixed as to the effectiveness of such counseling, in terms of both changes in expectations and final outcome. Purpose: The primary purpose of this study was to determine whether supplementing prefitting counseling with demonstration of real-world listening can (1) alter expectations of new hearing aid users and (2) increase satisfaction over verbal-only counseling. Secondary goals of the study were to examine (1) the relationship between prefitting expectations and postfitting outcome, and (2) the effect of hearing aid fine-tuning on hearing aid outcome. Research Design: Sixty new hearing aid users were fitted binaurally with Beltone Oria behind-the-ear digital hearing aids. Forty participants received prefitting counseling and demonstration of listening situations with the Beltone AVE™ (Audio Verification Environment) system; 20 received prefitting counseling without a demonstration of listening situations. Hearing aid expectations were measured at initial contact and following prefitting counseling. Reported hearing aid outcome was measured after eight to ten weeks of hearing aid use. Study Sample: Sixty new hearing aid users aged between 55 and 81 years with symmetrical sensorineural hearing loss. Intervention: Participants were randomly assigned to one of three experimental groups, between which the prefitting counseling and follow-up differed: Group 1 received prefitting counseling in combination with demonstration of listening situations. Additionally, if the participant had complaints about sound quality at the follow-up visit, the hearing aids were fine-tuned using the Beltone AVE system. Group 2 received prefitting counseling in combination with demonstration of listening situations with the Beltone AVE system, but no fine-tuning was provided at follow-up. Group 3 received prefitting hearing aid counseling that did not include demonstration of listening, and the hearing aids were not fine-tuned at the follow-up appointment. Results: The results showed that prefitting hearing aid counseling had small but significant effects on expectations. The two forms of counseling did not differ in their effectiveness at changing expectations; however, anecdotally, we learned from many participants that that they enjoyed listening to the auditory demonstrations and that they found them to be an interesting listening exercise. The data also show that positive expectations result in more positive outcome and that hearing aid fine-tuning is beneficial to the user. Conclusions: We conclude that prefitting counseling can be advantageous to hearing aid outcome and recommend the addition of prefitting counseling to address expectations associated with quality of life and self-image. The data emphasize the need to address unrealistic expectations prior to fitting hearing aids cautiously, so as not to decrease expectations to the extent of discouraging and demotivating the patient. Data also show that positive expectations regarding the impact hearing aids will have on psychosocial well-being are important for successful hearing aid outcome.

Anaerobic digestion is among the essential biological techniques used for stabilization of organic sludge from sewage and highly concentrated efflu-ents from food processing industries. It also recycles the municipal solid wastes into compost with simultaneous production of methane. The current study was performed to estimate the biomethanation potential of various agro- and industrial wastes like Jatropha de-oil cake, prawn shells, chicken feathers, bagasse, rice straw and wheat husk by mimicking the conditions in the biphasic Nisargruna biogas plant. A small volume of samples was chemi-cally characterized and allowed to decompose under aerobic and anaerobic conditions to determine the effect of aerobic predigestion (i.e. phase 1 of Nisargruna plant) on final methane production. The biogas produced was quantified by downward displacement of water. The observations indicated that approximately 60-80% methane was produced when Jatropha de-oil cake, prawn shells and rice straw was used. Conversely, the wheat straw and sugarcane wastes showed less methane formation, which may be due to the presence of complex polymers like lignocellulose and silica that considerably reduces the metabolic potential of microorganisms.

Reliable engine and emission models allow for an online monitoring of commercial gas turbine operation and help the plant operator and the original equipment manufacturer (OEM) to ensure emission compliance of the aging engine. However, model development and validation require fine-tuning on the particular engines, which may differ in a fleet of a single design type by production, assembly and aging status. For this purpose, Artificial Neural Networks (ANN) offer a good and fast alternative to traditional physically-based engine modeling, because the model creation and adaption is merely an automatized process in commercially available software environments. However, ANN performance depends strongly on the availability of suitable data and a-priori data processing. The present work investigates the impact of specific engine information from the OEM’s design tools on ANN performance. As an alternative to a strictly data-based benchmark approach, engine characteristics were incorporated into ANNs by a pre-processing of the raw measurements with a simplified engine model. The resulting ‘virtual’ measurements, i.e. hot gas temperatures, then served as inputs to ANN training and application during long-term gas turbine operation. When processed input parameters were used for ANNs, overall long-term NOx prediction improved by 55%, and CO prediction by 16% in terms of RMSE, yielding comparable overall RMSE values to the physically-based model.

Auxin, primarily indole-3-acetic acid (IAA), is a versatile signal molecule that regulates many aspects of plant growth, development, and stress response. Recently, microRNAs (miRNAs), a type of short non-coding RNA, have emerged as master regulators of the auxin response pathways by affecting auxin homeostasis and perception in plants. The combination of these miRNAs and the autoregulation of the auxin signaling pathways, as well as the interaction with other hormones, creates a regulatory network that controls the level of auxin perception and signal transduction to maintain signaling homeostasis. In this review, we will detail the miRNAs involved in auxin signaling to illustrate its in planta complex regulation.

Abstract. We introduce PARASO, a novel five-component fully-coupled regional climate model over an Antarctic circumpolar domain covering the full Southern Ocean. The state-of-the-art models used are f.ETISh1.7 (ice sheet), NEMO3.6 (ocean), LIM3.6 (sea ice), COSMO5.0 (atmosphere) and CLM4.5 (land), which are here run at an horizontal resolution close to 1/4°. One key-feature of this tool resides in a novel two-way coupling interface for representing ocean – ice-sheet interactions, through explicitly resolved ice-shelf cavities. The impact of atmospheric processes on the Antarctic ice sheet is also conveyed through computed COSMO-CLM – f.ETISh surface mass exchanges. In this technical paper, we briefly introduce each model's configuration and document the developments that were carried out in order to establish PARASO. The new offline-based NEMO – f.ETISh coupling interface is thoroughly described. Our developments also include a new surface tiling approach to combine open-ocean and sea-ice covered cells within COSMO, which was required to make this model relevant in the context of coupled simulations in polar regions. We present results from a 2000–2001 coupled two-year experiment. PARASO is numerically stable and fully operational. The 2-year simulation conducted without fine tuning of the model reproduced the main expected features, although remaining systematic biases provide perspectives for further adjustment and development.

AbstractMicroRNAs (miRNAs) are a group of small noncoding RNA molecules with significant capacity to regulate the gene expression at the post-transcriptional level in a sequence-specific manner either through translation repression or mRNA degradation triggering a fine-tuning biological impact. They have been implicated in several processes, including cell growth and development, signal transduction, cell proliferation and differentiation, metabolism, apoptosis, inflammation, and immune response modulation. However, over the last few years, extensive studies have shown the relevance of miRNAs in human pathophysiology. Common human parasitic diseases, such as Malaria, Leishmaniasis, Amoebiasis, Chagas disease, Schistosomiasis, Toxoplasmosis, Cryptosporidiosis, Clonorchiasis, and Echinococcosis are the leading cause of death worldwide. Thus, identifying and characterizing parasite-specific miRNAs and their host targets, as well as host-related miRNAs, are important for a deeper understanding of the pathophysiology of parasite-specific diseases at the molecular level. In this review, we have demonstrated the impact of human microRNAs during host−parasite interaction as well as their potential to be used for diagnosis and prognosis purposes.

NJ TRANSIT routinely sets service quality goals, monitors them, and uses the results to improve service. To support this business practice, NJ TRANSIT conducts customer satisfaction surveys of rail customers to understand customers’ needs and their level of satisfaction. On June 10, 1996, NJ TRANSIT initiated new rail service on its Morris and Essex rail lines called MidTOWN DIRECT. To assess the regional impacts of this major transit investment and to gain a better understanding of customers’ response to the new service, NJ TRANSIT conducted a survey of riders that included a customer satisfaction component. This study provided an opportunity for NJ TRANSIT to test the impact on customer satisfaction of introducing a new service option in a corridor that had well-established rail service. The study also provided insights into designing customer satisfaction measurement techniques for application during and after the implementation of significant new service offerings. To determine satisfaction levels of customers regarding the new service on the Morris and Essex Lines, and to determine whether the key drivers of satisfaction had changed, several statistical analyses were conducted. Several significant findings resulted from this customer satisfaction study. Several components of the MidTOWN DIRECT rail service required fine-tuning, especially with regard to frequency, on-time performance, and mechanical reliability. Although the satisfaction ratings were lower than initially expected, the availability of key driver satisfaction data can provide fast, targeted direction for service improvements that will pay the greatest dividends with customers.

Chronic obstructive pulmonary disease (COPD) is a chronic disease characterized by a progressive decline in lung function due to airflow limitation, mainly related to IL-1β-induced inflammation. We have hypothesized that single nucleotide polymorphisms (SNPs) in NLRP genes, coding for key regulators of IL-1β, are associated with pathogenesis and clinical phenotypes of COPD. We recruited 704 COPD individuals and 1238 healthy controls for this study. Twenty non-synonymous SNPs in 10 different NLRP genes were genotyped. Genetic associations were estimated using logistic regression, adjusting for age, gender, and smoking history. The impact of genotypes on patients’ overall survival was analyzed with the Kaplan–Meier method with the log-rank test. Serum IL-1β concentration was determined by high sensitivity assay and expression analysis was done by RT-PCR. Decreased lung function, measured by a forced expiratory volume in 1 s (FEV1% predicted), was significantly associated with the minor allele genotypes (AT + TT) of NLRP1 rs12150220 (p = 0.0002). The same rs12150220 genotypes exhibited a higher level of serum IL-1β compared to the AA genotype (p = 0.027) in COPD patients. NLRP8 rs306481 minor allele genotypes (AG + AA) were more common in the Global Initiative for Chronic Obstructive Lung Disease (GOLD) definition of group A (p = 0.0083). Polymorphisms in NLRP1 (rs12150220; OR = 0.55, p = 0.03) and NLRP4 (rs12462372; OR = 0.36, p = 0.03) were only nominally associated with COPD risk. In conclusion, coding polymorphisms in NLRP1 rs12150220 show an association with COPD disease severity, indicating that the fine-tuning of the NLRP1 inflammasome could be important in maintaining lung tissue integrity and treating the chronic inflammation of airways.
