Amiodarone low solubility and high permeability is the limiting step for its bioavailability, therefore new formulations are needed to improve the solubility of amiodarone either to increase its oral bioavailability or to reduce its toxic effects. Complexation of amiodarone with cyclodextrin results in improved dissolution rate, solubility, and allows for a more controlled drug release. We characterized the acute toxicity of a new amiodarone 2-hydroxypropyl-β-cyclodextrin complex (AMD/HP-β-CD) as powdered form and as a matrix based on Kollidon® and chitosan, administered intraperitoneally in laboratory animals. There were developed two formulations of matrix: one containing only pure AMD as a control sample (Fc) and one containing the inclusion complex with the optimal solubility (F). AMD was equitoxic with HP-β-CD after intraperitoneal administration (289.4 mg/kg for AMD and 298.3 mg/kg for AMD/HP-β-CD), with corresponding histopathological changes. The matrix based formulations presented higher LD50 values for acute toxicity, of 347.5 mg/kg for Fc and 455.6 mg/kg for F10, conducting to the idea of a safer administration because KOL and CHT matrix modified the solubility and controlled the AMD release. The LD50 is 1.5 higher for AMD/HP-β-CD included in a KOL and CHT based matrix compared to the pure AMD, administered intraperitoneally.

The research has been devoted to the study of acute toxicity of preparation based on derivative of indolocarba- zole - LCS-1208 in mice of both sexes and in rats of both sexes at intravenous and intraperitoneal administration of the drug. In the experiments the laboratory animals - outbred white rats and hybrid mice (C57Bl/6JxDBA2)Fl (B6D2F1) have been used. On the results of study has been obtained the calculated toxic doses of novel preparation based on derivative of indolocarbazole at intraperitoneal administration of the drug in mice of both sexes.

The article presents the results of the study of the acute toxic effect of the innovative disinfectant ‘Bondarmin’ (active substance — potassium peroxomonosulfate) on laboratory animals (mice, rats) are presented. Many scientific works of scientists in recent years have been devoted to the study of the toxicity of various disinfectants both in our country and abroad. However, today there are many topical issues regarding the toxicity and safety of some antimicrobials. Our work aimed to study the toxic effect on the laboratory animals and to establish the acute toxicity (LD50) of the developed disinfectant ‘Bondarmin’ when administered intraperitoneally. Experiments were carried out in the Laboratory of Pharmacology and Toxicology of the National University of Pharmacy (Kharkiv) and in the Educational and Scientific Laboratory of Genetic and Molecular Research Methods named after P. I. Verbitskiy in the Kharkiv State Zooveterinary Academy. Acute toxicity assessment (LD50) was carried out with intraperitoneal administration of the designed disinfectant to laboratory animals (mice, rats). The toxic effect of the newly developed disinfectant ‘Bondarmin’ for the intraperitoneal method of administration to laboratory animals (mice, rats) has been determined. For the intraperitoneal administration of the ‘Bondarmin’ disinfectant, the LD50 by Prozorovskiy method is 316.85 ± 19.26 mg/kg for mice, and 279.33 ± 19.80 mg/kg for rats. The disinfectant belongs to the IV toxicity class (low toxic substances). The results of toxicological studies allow us to recommend the use of ‘Bondarmin’ for disinfecting livestock facilities

Проведена сравнительная оценка эффективности антиоксидантного и гепатопротекторного действия пищевых добавок, полученных из выжимки винограда белых сортов по различным технологиям, в экспериментах на лабораторных животных. Установлено, что пищевая добавка «Порошок виноградный» (ПВ), полученная из выжимки винограда белых сортов по разработанной технологии, позволяет, по сравнению с контрольным образцом пищевой добавки, в большей степени снизить в сыворотке крови содержание продуктов перекисного окисления липидов – малонового диальдегида, диеновых коньюгатов и кетодиенов, что свидетельствует о более высокой эффективности антиоксидантного действия ПВ на организм животных, обусловленной большим содержанием микронутриентов с антиоксидантными свойствами. Добавка ПВ проявляет более высокую эффективность гепатопротекторного действия, заключающуюся в значительном снижении уровня активности аланинаминотрансферазы в сыворотке крови, по сравнению с контрольным образцом добавки. Comparative evaluation of the effectiveness of antioxidant and hepatoprotective action of food additives which obtained from grape pomace white varieties for various technologies, was carried out in experiments on laboratory animals. It is established that the food additive “Grape Powder” (GP), obtained from the pomace of white grape varieties by the developed technology, allows, in comparison with the control sample of the food additive, to reduce the content of lipid peroxidation products in the serum to a greater extent – Malon dialdehyde, diene conjugates and ketodienes, which indicates a higher efficiency of antioxidant action of GP on the organism of animals, due to the high content of micronutrients with antioxidant properties. The additive GP shows a higher efficiency of hepatoprotective action, which consists in a significant reduction in the level of alanine aminotransferase activity in the blood serum, compared to the control sample of the additive.

Abstract Oil-wetness and heterogeneity (i.e., existence of low and high permeability regions) are two main factors that result in low oil recovery by waterflood in carbonate reservoirs. The injected water is likely to flow through high permeability regions and bypass the oil in low permeability matrix. In this study, systematic coreflood tests were carried out in both "homogeneous" cores and "heterogeneous" cores. The heterogeneous coreflood test was proposed to model the heterogeneity of carbonate reservoirs, bypassing in low-permeability matrix during waterfloods, and dynamic imbibition of surfactant into the low-permeability matrix. The results of homogeneous coreflood tests showed that both secondary-waterflood and secondary-surfactant flood can achieve high oil recovery (>50%) from relatively homogenous cores. A shut-in phase after the surfactant injection resulted in an additional oil recovery, which suggests enough time should be allowed while using surfactants for wettability alteration. The core with a higher extent of heterogeneity produced lower oil recovery to waterflood in the coreflood tests. Final oil recovery from the matrix depends on matrix permeability as well as the rock heterogeneity. The results of heterogeneous coreflood tests showed that a slow surfactant injection (dynamic imbibition) can significantly improve the oil recovery if the oil-wet reservoir is not well-swept.

The present study was carried out to evaluate the acute toxicity and the effect of the aqueous extract of the roots from Pfaffia glomerata (Spreng) Pedersen (Amaranthaceae) (AEP) on the prevention of acetic acid-induced ulcer and on the healing process of previously induced ulcers. The acute toxicity was evaluated in Swiss mice after oral administration of a single dose and the chronic gastric ulcer was induced with local application of acetic acid. The results showed that the LD50 of the extract was 684.6 mg.kg-1 for the intraperitoneal administration and higher than 10 mg.kg-1by the oral route. The administration of the AEP did not prevent ulcers formation. However, the AEP increased of the healing process of previously induced ulcers. The results suggest that AEP chronically administered promote an increase of tissue healing, after the damage induced by acetic acid and the extract seemed to be destituted of toxic effects in the mice by the oral route.

Objective. To evaluate the activity of pyruvate kinase (PK) isoforms in normal conditions, in toxic damage of the liver and during its regeneration.Materials and methods. An experimental study was carried out on 45 Wistar rats. Toxic liver damage was induced by the intraperitoneal administration of carbon tetrachloride. Mechanical damage was simulated by the surgical resection of the liver. The levels of PK isoforms R/L and M in the blood serum and liver tissue of the laboratory animals were measured with an ELISA test.Results. It has been found that the level of PK isoform M signifcantly increases in chronic toxic liver damage, which may indicate the activation of the processes of liver cell proliferation in response to the damaging effect of hepatotoxin (Mann-Whitney U Test: Z = 2.143; p = 0.032). After liver resection, the level of PK R/L, which characterizes the activation of glycolysis, increased and the level of pyruvate kinase M increased signifcantly, which reflected the processes of reparative regeneration in the liver.Conclusion. The serum level of PK isoforms may be used as a laboratory criterion for the activity of reparative regeneration processes, which can be used to evaluate the reparative potential of the liver in case of toxic or mechanical damage, as well as in chronic diffuse diseases.

Dr. Dews writes: Dr. Milman, in her first paragraph on the dangers of marijuana, shows the dangers of jargon. "Acute toxicity" in the trade implies killing, usually within 24 hours. Large amounts of tetrahydrocannabinols are required to kill mice and other laboratory animals, hence "the acute toxicity is vanishingly small," a statement which says more in English than it should. My apologies. Dr. Milman in her penultimate paragraph quotes King and Cowen for the statement that following intravenous injection of marijuana, "the histopathology is real."

The objective of this study is to develop a highly permeable rejuvenating agent for the recycling of the asphalt pavement. The rheological properties and permeability of recycled asphalt after adding the self-developed rejuvenating agent, as well as two other agents, were compared and evaluated. An improved softening point method was devised to evaluate the permeability. In addition, the recycled asphalt was analyzed using Fourier transform infrared spectroscopy (FTIR). The results showed that the self-developed rejuvenating agent had high permeability, could effectively restore the performance of the aged asphalt, and could improve the aging-resistant property of the recycled asphalt. FTIR analysis showed that the matrix asphalt experienced oxygen absorption and dehydrogenation during the aging process. The aging of the SBS-modified asphalt was achieved via dual aging of the matrix asphalt and SBS-modified components. In addition, the rejuvenating agent CA had an inhibitory effect on asphalt aging, and its recycling efficiency was better than that of the rejuvenating agent A for the aged SBS-modified asphalt. Finally, a relationship between the microscopic functional group index and the macroscopic test index was established.
