The 0–1 test distinguishes between regular and chaotic dynamics for a deterministic system using a time series as a starting point without appealing to any state space reconstruction method. A modification of the 0–1 test allows for the determination of a more comprehensive range of signal dynamic behaviors, particularly in the field of biological signals. We report the results of applying the test and study with more details the PhotoPlethysmoGraphic (PPG) signal behavior from different healthy young subjects, although its use is extensible to other biological signals. While mainly used for heart rate and blood oxygen saturation monitoring, the PPG signal contains extensive physiological dynamics information. We show that the PPG signal, on a healthy young individual, is predominantly quasi-periodic on small timescales (short span of time concerning the dominant frequency). However, on large timescales, PPG signals yield an aperiodic behavior that can be firmly chaotic or a prior transition via an SNA (Strange Nonchaotic Attractor). The results are based on the behavior of well-known time series that are random, chaotic, aperiodic, periodic, and quasi-periodic.

Abstract The traditional time series studies consider the time series as a whole while carrying on the trend detection; therefore not enough attention is paid to the stage characteristic. On the other hand, the piecewise linear fitting type methods for trend detection are lacking consideration of the possibility that the same node belongs to multiple trends. The above two methods are affected by the start position of the sequence. In this paper, the concept of overlapping trend is proposed, and the definition of milestone nodes is given on its base; these way not only the recognition of overlapping trend is realized, but also the negative influence of the starting point of sequence is effectively reduced. The experimental results show that the computational accuracy is not affected by the improved algorithm and the time cost is greatly reduced when dealing with the processing tasks on dynamic growing data sequence.

Abstract In examining a plot of a time series of a scalar climate variable for indications of climate change, an investigator might pick out what appears to be a linear trend commencing near the end of the record. Visual determination of the starting time of the trend can lead to an incorrect conclusion that the trend is significant when the assessment is based on standard linear regression analysis; in fact, a presumed level of significance of 5% can be smaller than the actual level by up to an order of magnitude. An alternative procedure is suggested that is more appropriate for assessing the significance of a trend in which the starting point is identified visually.

Aim and Objective: Antibiotic resistance is a serious constraint to the development of new effective antibacterials. Therefore, the discovery of the new antibacterials remains one of the main challenges in modern medicinal chemistry. This study was undertaken to identify novel molecules with antibacterial activity. Materials and Methods: Using our unique double-reporter system, in-house large-scale HTS campaign was conducted for the identification of antibacterial potency of small-molecule compounds. The construction allows us to visually assess the underlying mechanism of action. After the initial HTS and rescreen procedure, luciferase assay, C14-test, determination of MIC value and PrestoBlue test were carried out. Results: HTS rounds and rescreen campaign have revealed the antibacterial activity of a series of Nsubstituted triazolo-azetidines and their isosteric derivatives that has not been reported previously. Primary hit-molecule demonstrated a MIC value of 12.5 µg/mL against E. coli Δ tolC with signs of translation blockage and no SOS-response. Translation inhibition (26%, luciferase assay) was achieved at high concentrations up to 160 µg/mL, while no activity was found using C14-test. The compound did not demonstrate cytotoxicity in the PrestoBlue assay against a panel of eukaryotic cells. Within a series of direct structural analogues bearing the same or bioisosteric scaffold, compound 2 was found to have an improved antibacterial potency (MIC=6.25 µg/mL) close to Erythromycin (MIC=2.5-5 µg/mL) against the same strain. In contrast to the parent hit, this compound was more active and selective, and provided a robust IP position. Conclusion: N-substituted triazolo-azetidine scaffold may be used as a versatile starting point for the development of novel active and selective antibacterial compounds.

Abstract New determinations of ocean tides are extracted from high-rate Global Positioning System (GPS) solutions at nine stations sitting on the Ross Ice Shelf. Five are multi-year time series. Three older time series are only 2–3 weeks long. These are not ideal, but they are still useful because they provide the only in situ tide observations in that sector of the ice shelf. The long tide-gauge observations from Scott Base and Cape Roberts are also reanalysed. They allow determination of some previously neglected tidal phenomena in this region, such as third-degree tides, and they provide context for analysis of the shorter datasets. The semidiurnal tides are small at all sites, yet M2 undergoes a clear seasonal cycle, which was first noted by Sir George Darwin while studying measurements from the Discovery expedition. Darwin saw a much larger modulation than we observe, and we consider possible explanations - instrumental or climatic - for this difference.

AbstractInterferometric Synthetic Aperture Radar (InSAR) enables detailed investigation of surface landslide movements, but it cannot provide information about subsurface structures. In this work, InSAR measurements were integrated with seismic noise in situ measurements to analyse both the surface and subsurface characteristics of a complex slow-moving landslide exhibiting multiple failure surfaces. The landslide body involves a town of around 6000 inhabitants, Villa de la Independencia (Bolivia), where extensive damages to buildings have been observed. To investigate the spatial-temporal characteristics of the landslide motion, Sentinel-1 displacement time series from October 2014 to December 2019 were produced. A new geometric inversion method is proposed to determine the best-fit sliding direction and inclination of the landslide. Our results indicate that the landslide is featured by a compound movement where three different blocks slide. This is further evidenced by seismic noise measurements which identified that the different dynamic characteristics of the three sub-blocks were possibly due to the different properties of shallow and deep slip surfaces. Determination of the slip surface depths allows for estimating the overall landslide volume (9.18 · 107 m3). Furthermore, Sentinel-1 time series show that the landslide movements manifest substantial accelerations in early 2018 and 2019, coinciding with increased precipitations in the late rainy season which are identified as the most likely triggers of the observed accelerations. This study showcases  the potential of integrating InSAR and seismic noise techniques to understand the landslide mechanism from ground to subsurface.

AbstractTime series with a significant trend, as is now being the case for the temperature in the course of climate change, need a careful approach for statistical evaluations. Climatological means and moments are usually taken from past data which means that the statistics does not fit to actual data anymore. Therefore, we need to determine the long-term trend before comparing actual data with the actual climate. This is not an easy task, because the determination of the signal—a climatic trend—is influenced by the random scatter of observed data. Different filter methods are tested upon their quality to obtain realistic smoothed trends of observed time series. A new method is proposed, which is based on a variational principle. It outperforms other conventional methods of smoothing, especially if periodic time series are processed. This new methodology is used to test, how extreme the temperature of 2018 in Vienna actually was. It is shown that the new annual temperature record of 2018 is not too extreme, if we consider the positive trend of the last decades. Also, the daily mean temperatures of 2018 are not found to be really extreme according to the present climate. The real extreme of the temperature record of Vienna—and many other places around the world—is the strongly increased positive temperature trend over the last years.

Density functional theory is used to determine the possible crystal structure of the CaSiO3 perovskites and their evolution under pressure. The ideal cubic perovskite is considered as a starting point for studying several possible lower-symmetry distorted structures. The theoretical lattice parameters and the atomic coordinates for all the structures are determined, and the results are discussed with respect to experimental data.

Human dual-specificity phosphatase 7 (DUSP7/Pyst2) is a 320-residue protein that belongs to the mitogen-activated protein kinase phosphatase (MKP) subfamily of dual-specificity phosphatases. Although its precise biological function is still not fully understood, previous reports have demonstrated that DUSP7 is overexpressed in myeloid leukemia and other malignancies. Therefore, there is interest in developing DUSP7 inhibitors as potential therapeutic agents, especially for cancer. Here, the purification, crystallization and structure determination of the catalytic domain of DUSP7 (Ser141–Ser289/C232S) at 1.67 Å resolution are reported. The structure described here provides a starting point for structure-assisted inhibitor-design efforts and adds to the growing knowledge base of three-dimensional structures of the dual-specificity phosphatase family.
