The combined effect of farm management practices, transport time, and ageing time on the electrophoretic changes of sarcoplasmic (SPP) and myofibrillar (MFP) protein fractions of goat kids was studied. A total of 64 suckling goat kids were withdrawn from two farms with “high” (GW) and “low” (DW) welfare-friendly management practices, and they were transported for 2 or 6 h immediately before slaughtering. Longissimus lumborum samples were obtained at 3, 8, and 21 days post-mortem, and muscle proteins were separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis SDS-PAGE. Both protein extracts displayed significant changes attributable to meat maturation. Managing conditions of kids in DW farms increased the post-mortem susceptibility of muscle proteins. Some MFP of Longissimus lumborum muscle, such as troponin T, as well as 26–30 and 35–37 kDa fractions were influenced significantly by deficient on-farm management, and therefore, these protein fragments might be considered as indicators of low-welfare on-farm management in goat kids.

The livestock sector can be a major contributor to the mitigation of greenhouse (GHG) emissions. Within the sector, beef production produces the largest proportion of the livestock sector’s direct emissions. The objective of this study was to assess the on-farm GHG emissions in semi-arid rangelands in Argentina and to identify the relationships between emissions and current farm management practices. A survey recorded detailed information on farm management and characteristics. Assessments of GHG emissions were based on the IPCC Tier 2 protocols [1]. The relationships between farm management and GHG emissions were identified using General Linear Models. Cluster analysis was used to identify groups of farms that differed from others in emissions and farm characteristics. Emissions per product sold were low on farms that had improved livestock care management, rotational grazing, received technical advice, and had high animal and land productivities. Emissions per hectare of farmland were low on farms that had low stocking rates, low number of grazing paddocks, little or no land dedicated to improved pastures and forage crops, and low land productivity. Our results suggest that the implementation of realistic, relatively easy-to-adopt farming management practices has considerable potential for mitigating GHG emissions in semi-arid rangelands of central Argentina.

Lamb mortality is a key factor influencing ewe productivity and profitability. The current study investigated risk factors associated with and management practices implemented on sheep farms to reduce lamb mortality. A survey consisting of 13 multiple-part questions (57 separate questions) was administered to all sheep farmers participating in the Teagasc National Farm Survey, representative of the Irish national population of sheep farms. A total of 60% of respondents identify mating or lambing date, and this practice tended to be associated with reduced lamb mortality (1.2%, p = 0.08). Individual lambing pens were used by 88% of farmers, but 26% did not clean or disinfect them. A total of 79% and 9.5% of farmers applied iodine to all lambs’ navels and administered antibiotics to all lambs to treat and/or prevent diseases, respectively. Most farmers vaccinated their ewes (86%) and lambs (79%) against clostridial diseases and/or pasteurellosis; 13% vaccinated against abortion agents. Lamb mortality tended to be lower (Kruskal–Wallis (KW) = 2.749; p = 0.09) on farms that used stomach tubing, heat box, iodine, hospital, and individual pens compared with farms that do not implement all those practices. Predators, lamb birth weight, and diseases were perceived by respondents to be the three main causes of live-born lamb mortality. The gross margin is significantly higher on lowland farms by €37 per ewe compared with hill farms (Kruskal–Wallis (KW) = 4.056; p < 0.001). The combination of full-time farming and the use of hospital and individual pens improved gross margin (€18/ewe, p = 0.028). It is concluded that on-farm management practices affect both lamb mortality and flock gross margin.

Context Fraudulent information about food is an old and widespread problem, particularly regarding products with high economic value, such as meat and meat products. The motivation for food fraud is economic, but it can have serious impacts on public health, thus creating a food security problem. Approximately 90% of buffalo meat is marketed as beef in various regions where the consumption of buffalo meat is considered unusual. Aims To determine the electrophoretic profile of the raw Longissimus dorsi of cattle and buffalo species and to test the hypothesis that electrophoresis techniques can be used to distinguish meat from cattle from buffalo meat. Methods Fourteen 10-g samples of Longissimus dorsi (12th and 13th rib) tissue were taken from each animal of both species after slaughter. The meat of each species was analysed by native polyacrylamide gel electrophoresis (NATIVE PAGE) and by denaturing and non-denaturing sodium dodecyl sulfate (SDS)–PAGE. Differences (P &lt; 0.05) were observed between water-soluble cattle and buffalo muscle proteins in both NATIVE PAGE (relative mobilities and percentages of protein bands) and non-denaturing and denaturing SDS–PAGE (molecular weights in kDa and optical density index). Key results With the NATIVE PAGE technique, 10 protein bands were observed in the gel, and three of these bands exhibited differences between species (P ≤ 0.05). The non-denaturing and denaturing SDS–PAGE techniques yielded significantly different protein bands in the gel. The electrophoretic profiles of some cattle and buffalo muscle proteins are distinct; therefore, raw meat flesh samples of these animal species can be distinguished using these electrophoresis techniques. Conclusions Each of the three electrophoresis techniques used can distinguish meat from different animal species; however, when there is doubt about the animal species, the use of more than one electrophoretic technique is recommended, so as to obtain more reliable results. Implications The use of electrophoresis techniques to differentiate cattle and buffalo meat is promising. This technique could be used in cases of suspected food fraud, such as the replacement of beef with buffalo or vice versa, with reliable results that will be accepted by supervisory bodies.

On-farm studies provide a realistic setting to examine the impact of interactions of management practices on weed communities within various farming systems. Clustering farm units into groups that use similar management practices enables the replication of farm management systems within on-farm studies. The goal of this study is to objectively classify farm units into management systems on the basis of quantitative variables describing aspects of cropping history and chemical input levels using multivariate techniques. Twenty-eight Saskatchewan farmers provided details of their management practices from 1990 to 1997 through a series of questionnaires. Twelve variables derived from the questionnaires were used to describe cropping history, pesticide, tillage and fertilizer use on each farm unit. These variables were used to cluster farm units using minimum variance classification and NMS ordination. Both techniques identified seven farm management systems. The greatest differences were observed between organic and non-organic systems. Farm management systems that used annual fallow and continuous annual cropping histories were most similar. The consistent results obtained by use of the two unrelated methodologies indicate the utility of this approach for the classification of farm management systems. Abstract The purpose of this study was to determine whether the two monoclonal anti-cardiac troponin T (cTnT) antibodies (MAbs) used in the second generation cTnT assay by Boehringer Mannheim (BM, capture Ab, M11.7; detection Ab, M7) would detect cTnT isoforms expressed in human skeletal muscle in response to chronic renal disease (CRD). cTnT expression was examined in skeletal muscle biopsies obtained from 45 CRD patients, as well as nondiseased human heart (n = 3) and skeletal muscle (n = 3). cTnT proteins were resolved by modified 7.5% sodium dodecyl sulfate-polyacrylamide gel electrophoresis, transferred to nitrocellulose, and probed with the following anti-cTnT MAbs: M11.7; M7; JS-2, Lakeland Biomedical; and 13–11, Duke University. All four antibodies detected the cTnT isoforms (Ta, Te) expressed in human myocardium. In 20 of 45 skeletal muscle biopsies, MAb M11.7 recognized its epitope in one to three proteins, molecular mass 34–36 kDa, designated Te, Td, and Tc; the strongest signal was that of Te. The same proteins were recognized by MAbs JS-2 and 13–11. The BM M7 antibody did not detect the cTnT isoforms in the molecular mass range of 34–36 kDa. However, MAb M7 did detect a cTnT isoform, molecular mass 39 kDa, in 2 of 45 biopsies. This isoform had an electrophoretic mobility similar to the predominant heart cTnT isoform, Ta. We conclude that cTnT isoforms are expressed in the skeletal muscle of CRD patients. However, given the epitopes recognized by the BM MAbs M7 and M11.7 and the variable presence of these cTnT isoforms in skeletal muscle, the second generation BM cTnT assay will not detect these isoforms if they are released from skeletal muscle into the circulation.
