Many root fungal endophytes inhabiting forest trees have potential impact on the health and disease progression of certain tree species. Hence, the screening of root endophytes for their biocontrol abilities is relevant for their potential to protect their hosts against invaders. The aim of this research is to screen for the potential inhibitory effects of selected conifer root endophytes during interaction, in vitro, with the root rot pathogen, Heterobasidion parviporum. Here, we introduce a guideline that facilitates the use of root fungal endophytes as biocontrol agents. We isolated fungal root endophytes from eight different conifers. These root fungal endophytes were evaluated for their antagonism against the root rot pathogen, H. parviporum, by means of paired-culture antagonism assays. We determined the antagonism of the isolated root fungal endophytes to elucidate potential biocontrol applications. For the analysis, a software package in R was developed. Endophyte candidates with antagonistic potential were identified.

Abstract The goal of the present research was to ascertain the potential root endophytic fungi against Dematophora necatrix, Hartig, the causal pathogen of white root rot in apples; however, it has an expanded range of hosts across different fruit trees. Out of 36 endophytic fungi segregated from symptomless roots of apple, wheat, maize, marigold, cherry, and garlic plants, only 9 isolates showed inhibitory effect during preliminary screening and were further assayed under in vitro, pot, and field conditions against the white root rot pathogen/disease. Under in vitro conditions, maximum mycelial inhibition of 81.48% was obtained with the isolate Aspergillus aculeatus strain C2. Microscopic studies on interaction between fungal endophyte with hyphal tips of D. necatrix revealed various morphological abnormalities in the hyphae. In glasshouse conditions, seed treatment pursued by soil application with Crinipellis tabtim strain M8 isolate was highly effective and exhibited 93.55% disease control. Similarly, under field conditions, the overall maximum disease control was exhibited by Crinipellis tabtim strain M8 (84.95%). The most promising root endophytes that were identified rely on morphological and ITS sequence analysis. Root colonization assay was performed which revealed maximum endosphere and rhizosphere colonization with Crinipellis tabtim strain M8. Additionally, confocal microscopic illustrations of transverse sections of root cells tenanted by fungal endophytes as compared to untreated control suggested the persistence and establishment of endophytes in the endosphere of apple seedlings. These findings present the first report on colonization of apple roots by fungal root endophytes suggesting an alternative and sustainable approach for management of white root rot disease.

An increase in cork oak diseases caused by Biscogniauxia mediterranea and Diplodia corticola has been reported in the last decade. Due to the high socio-economic and ecologic importance of this plant species in the Mediterranean Basin, the search for preventive or treatment measures to control these diseases is an urgent need. Fungal endophytes were recovered from cork oak trees with different disease severity levels, using culture-dependent methods. The results showed a higher number of potential pathogens than beneficial fungi such as cork oak endophytes, even in healthy plants. The antagonist potential of a selection of eight cork oak fungal endophytes was tested against B. mediterranea and D. corticola by dual-plate assays. The tested endophytes were more efficient in inhibiting D. corticola than B. mediterranea growth, but Simplicillium aogashimaense, Fimetariella rabenhorstii, Chaetomium sp. and Alternaria alternata revealed a high potential to inhibit the growth of both. Simplicillium aogashimaense caused macroscopic and microscopic mycelial/hyphal deformations and presented promising results in controlling both phytopathogens’ growth in vitro. The evaluation of the antagonistic potential of non-volatile and volatile compounds also revealed that A. alternata compounds could be further explored for inhibiting both pathogens. These findings provide valuable knowledge that can be further explored in in vivo assays to find a suitable biocontrol agent for these cork oak diseases.

ABSTRACT Despite the favorable edaphoclimatic conditions for avocado production in Brazil, diseases such as root rot caused by the pathogen Phytophthora cinnamomi compromise the crop. With the aim of managing root rot in avocado, the present study aimed to evaluate chemical and biological control with isolates of Trichoderma spp. and Pseudomonas fluorescens. Thus, three assays were conducted to assess: (i) mycelial inhibition of P. cinnamomi by isolates of Trichoderma spp. and P. fluorescens from different crop systems; (ii) effect of autoclaved and non-autoclaved metabolites of P. fluorescens, and (iii) chemical or biological treatment of avocado seedlings on the control of root rot under field conditions. The isolates of Trichoderma spp. from maize cultivation soil and the commercial products formulated with Trichoderma presented greater antagonism (p <0.05) to the pathogen P. cinnamomi in the in vitro tests. Similarly, non-autoclaved metabolites of P. fluorescens presented antagonistic potential to control P. cinnamomi. Under field conditions, the fungicide metalaxyl and the bioagents showed effectiveness in controlling P. cinnamomi, as well as greater root length and mass. Results demonstrated potential for the biological control of avocado root rot with Trichoderma spp. and P. fluorescens.

Pea (Pisum sativum) is an important leguminous crop in many countries including India. Wilt and root rot of pea is an important and widespread disease that often causes significant reduction in the yield and quality of harvested peas throughout the production areas. It is the most important and widespread disease of pea grown in relatively dry and warm area. In-vitro effectiveness of various antagonistic fungal isolates namely T. harzianum (Th1, Th2, Th3, Th4 and Th5) was evaluated against Fusarium oxysporum f. sp. pisi, Rhizoctonia solani and Pythium ultimum by dual culture technique on potato dextrose agar. According to the observation recorded after 5 days, all the rhizospheric fungal isolates evaluated for their antagonistic potential against wilt and root-rot pathogens, exhibited significant effect on radial growth inhibition of pathogens in comparison to control. Among the fungal isolates, Th3 and Th5 of T. harzianum proved to be most effective in reducing the growth of F. oxysporum f. sp. pisi, R. solani and P. ultimum. It was worthy to note that all rhizospheric fungal isolates visualized an increase in their antagonistic potential over the period of time in subsequent hours of inoculation.

Amaryllidaceae family comprises many crops of high market potential for the food and pharmaceutical industries. Nowadays, the utilization of plants as a source of bioactive compounds requires the plant/endophytic microbiome interactions, which affect all aspects of crop’s quantity and quality. This review highlights the taxonomy, ecology, and bioactive chemicals synthesized by endophytic fungi isolated from plants of the Amaryllidaceae family with a focus on the detection of pharmaceutically valuable plant and fungi constituents. The fungal microbiome of Amaryllidaceae is species- and tissue-dependent, although dominating endophytes are ubiquitous and isolated worldwide from taxonomically different hosts. Root sections showed higher colonization as compared to bulbs and leaves through the adaptation of endophytic fungi to particular morphological and physiological conditions of the plant tissues. Fungal endophytes associated with Amaryllidaceae plants are a natural source of ecofriendly bioagents of unique activities, with special regard to those associated with Amarylloidae subfamily. The latter may be exploited as stimuli of alkaloids production in host tissues or can be used as a source of these compounds through in vitro synthesis. Endophytes also showed antagonistic potential against fungal, bacterial, and viral plant diseases and may find an application as alternatives to synthetic pesticides. Although Amaryllidaceae crops are cultivated worldwide and have great economic importance, the knowledge on their endophytic fungal communities and their biochemical potential has been neglected so far.

Terrestrial plants including forest trees are generally known to live in close association with microbial organisms. The inherent features of this close association can be commensalism, parasitism or mutualism. The term “microbiota” has been used to describe this ecological community of plant-associated pathogenic, mutualistic, endophytic and commensal microorganisms. Many of these microbiota inhabiting forest trees could have a potential impact on the health of, and disease progression in, forest biomes. Comparatively, studies on forest tree microbiomes and their roles in mutualism and disease lag far behind parallel work on crop and human microbiome projects. Very recently, our understanding of plant and tree microbiomes has been enriched due to novel technological advances using metabarcoding, metagenomics, metatranscriptomics and metaproteomics approaches. In addition, the availability of massive DNA databases (e.g., NCBI (USA), EMBL (Europe), DDBJ (Japan), UNITE (Estonia)) as well as powerful computational and bioinformatics tools has helped to facilitate data mining by researchers across diverse disciplines. Available data demonstrate that plant phyllosphere bacterial communities are dominated by members of only a few phyla (Proteobacteria, Actinobacteria, Bacteroidetes). In bulk forest soil, the dominant fungal group is Basidiomycota, whereas Ascomycota is the most prevalent group within plant tissues. The current challenge, however, is how to harness and link the acquired knowledge on microbiomes for translational forest management. Among tree-associated microorganisms, endophytic fungal biota are attracting a lot of attention for their beneficial health- and growth-promoting effects, and were preferentially discussed in this review.

Abstract. The goals of this study were to determine and evaluate the bioaccessibility of total anthocyanin and procyanidin in apple (Amasya, Malus communis), red grape (Papazkarası, Vitis vinifera) and cinnamon (Cassia, Cinnamomum) using an in vitro static digestion system based on human gastrointestinal physiologically relevant conditions. Also, in vitro inhibitory effects of these foods on lipid (lipase) and carbohydrate digestive enzymes (α-amylase and α-glucosidase) were performed with before and after digested samples using acarbose and methylumbelliferyl oleate (4MUO) as the positive control. While the highest total anthocyanin content was found in red grape (164 ± 2.51 mg/100 g), the highest procyanidin content was found in cinnamon (6432 ± 177.31 mg/100 g) (p < 0.05). The anthocyanin bioaccessibilities were found as 10.2 ± 1%, 8.23 ± 0.64%, and 8.73 ± 0.70% in apple, red grape, and cinnamon, respectively. The procyanidin bioaccessibilities of apple, red grape, and cinnamon were found as 17.57 ± 0.71%, 14.08 ± 0.74% and 18.75 ± 1.49%, respectively. The analyzed apple, red grape and cinnamon showed the inhibitory activity against α-glucosidase (IC50 544 ± 21.94, 445 ± 15.67, 1592 ± 17.58 μg/mL, respectively), α-amylase (IC50 38.4 ± 7.26, 56.1 ± 3.60, 3.54 ± 0.86 μg/mL, respectively), and lipase (IC50 52.7 ± 2.05, 581 ± 54.14, 49.6 ± 2.72 μg/mL), respectively. According to our results apple, red grape and cinnamon have potential to inhibit of lipase, α-amylase and α-glucosidase digestive enzymes.
