Methicillin-resistant Staphylococcus aureus (MRSA) is a widespread pathogen that could cause different illnesses in both human and animals. Presence of MRSA in animals raises concerns of their capacity to act as reservoirs, particularly in wild animals. This study aimed to characterize the resistance and virulence patterns of S. aureus strains isolated from bat guano in Algeria. From March to May 2016, 98 bat guano samples from Aokas’s cave (Bejaia, Algeria) were collected. Swabs were taken for microbiological studies. Isolates were identified by Vitek® MS system, and antibiotic susceptibility was determined by disk diffusion method. The clonal origin, virulence and antibiotic resistance profiles of S. aureus isolates were characterized by whole genome sequencing. Eleven S. aureus strains were obtained from the 98 guano samples. Seven isolates were sensitive to all antibiotics tested and four (36.3%) were resistant to penicillin G, cefoxitin and fusidic acid. The four MRSA isolates were assigned to the sequence type ST149 and related to spa type t010. These isolates harbored a SCCmecIV element and the fusidic acid resistance element Q6GD50 (fusC). They carried different virulence genes including several enterotoxins (sea, egc enterotoxin locus, sec, sel), and the toxic shock syndrome toxin (tst). Our results highlight that bat guano may constitute an important reservoir of MRSA strains.

ABSTRACTThe epidemiology of community-acquired methicillin-resistantStaphylococcus aureus(CA-MRSA) differs from country to country. We assess the features of the ST80 European clone, which is the most prevalent PVL-positive CA-MRSA clone in Europe, and the TSST-1 ST5 clone that was recently described in France. In 2008, all MRSA strains susceptible to fluoroquinolones and gentamicin and resistant to fusidic acid that were isolated in 104 French laboratories were characterized usingagralleles,spatyping, and the staphylococcal cassette chromosomemecelement and PCR profiling of 21 toxin genes. Three phenotypes were defined: (i) kanamycin resistant, associated with the ST80 clone; (ii) kanamycin and tobramycin resistant, associated with the ST5 clone; and (iii) aminoglycoside susceptible, which was less frequently associated with the ST5 clone. Among the 7,253 MRSA strains isolated, 91 (1.3%) were ST80 CA-MRSA (89 phenotype 1) and 190 (2.6%) were ST5 CA-MRSA (146 phenotype 2, 42 phenotype 3). Compared to the latter, ST80 CA-MRSAs were more likely to be community acquired (80% versus 46%) and found in young patients (median age, 26.0 years versus 49.5 years) with deep cutaneous infections (48% versus 6%). They were less likely to be tetracycline susceptible (22% versus 85%) and to be isolated from respiratory infections (6% versus 27%). The TSST-1 ST5 clone has rapidly emerged in France and has become even more prevalent than the ST80 European clone, whose prevalence has remained stable. The epidemiological and clinical patterns of the two clones differ drastically. Given the low prevalence of both among all staphylococcal infections, no modification of antibiotic recommendations is required yet.

The present study was conducted to investigate the prevalence and antibiotic resistance among Campylobacter jejuni in ulam at farms and retail outlets located in Kuala Terengganu, Malaysia. A total of 526 samples (ulam, soil, and fertilizer) were investigated for the presence of C. jejuni and the gene for cytolethal distending toxin (cdt) by using a multiplex PCR method. Antibiotic susceptibility to 10 types of antibiotics was determined using the disk diffusion method for 33 C. jejuni isolates. The average prevalence of contaminated samples from farms, wet markets, and supermarkets was 35.29, 52.66, and 69.88%, respectively. The cdt gene was not detected in 24 of the 33 C. jejuni isolates, but 9 isolates harbored cdtC. Antibiotic resistance in C. jejuni isolates was highest to penicillin G (96.97% of isolates) followed by vancomycin (87.88%), ampicillin (75.76%), erythromycin (60.61%), tetracycline (9.09%), amikacin (6.06%), and norfloxacin (3.03%); none of the isolates were resistant to ciprofloxacin, enrofloxacin, and gentamicin. In this study, C. jejuni was present in ulam, and some isolates were highly resistant to some antibiotics but not to quinolones. Thus, appropriate attention and measures are required to prevent C. jejuni contamination on farms and at retail outlets.

The presence of Salmonella in food products and emergence of antibiotic resistance are the major challenges facing public health policies. A total of 2749 crocodile meat samples obtained from the Central Veterinary Laboratories in Zimbabwe were screened for Salmonella specieswere collected from three Zimbabwean commercial farms between the year 2012 and 2019 for a retrospective observational study to determine the prevalence and magnitude of antibiotics resistant Salmonella species in crocodile meat. The isolation of Salmonella was in accordance with the ISO 6579:2002 and the antibiotic susceptibility testing was carried out based on Clinical and Laboratory Standard Institute’s recommendations by means of the Kirby-Bauer disk diffusion method. SILAB Database was used to determine the prevalence of Salmonella species. Prevalence was stratified by year and farms. Twenty Salmonella isolates were identified using biochemical tests, and 15 were confirmed by polymerase chain reaction (PCR). Antimicrobial susceptibility profiles of the confirmed Salmonella isolates were examined using 14 antibiotics. The overall prevalence of Salmonella species in crocodile meat samples was 0.5%. The prevalence of Salmonella species ranged from 0.04% to 0.44% in the crocodile meat samples and annual prevalence ranged from 0.01% to 1%. The highest prevalence of Salmonella (4.4%) was recorded in the year 2012. Salmonella isolates from one of the three tested farms were resistant to Erythromycin (73.33%), Ampicillin (80%), and Penicillin G (100%). Generally, Salmonella isolates displayed lower resistance to Cefepime, Ceftriaxone, Amikacin, Tetracycline, Ertapenem, Florfenicol, and Erythromycin (0-53.33%) whereas all Salmonella isolates showed susceptibility to Cefepime, Ceftriaxone, Ertapenem, and Florfenicol. Although the study indicates low prevalence of Salmonella species in crocodile meat, there is a need for strict implementation of Hazard Analysis Critical Control Point (HACCP) to reduce contamination rates in meat and its products

ABSTRACTThe prevalence of resistance to fusidic acid in clinical isolates ofStaphylococcus aureus, including methicillin-resistantS. aureus(MRSA), has increased in the past 2 decades. However, there are limited data regarding the relative importance in this process of the different staphylococcal determinants that mediate resistance to fusidic acid. Furthermore, the roles played by clonal dissemination of fusidic acid-resistant strains versus horizontal transmission of fusidic acid resistance determinants have not been investigated in detail. To gain insight into both issues, we examined fusidic acid resistance in 1,639 MRSA isolates collected in Denmark between 2003 and 2005. Resistance to fusidic acid (MIC, >1 μg/ml) was exhibited by 291 (17.6%) isolates. For the majority of these isolates (∼87%), resistance was attributed to carriage offusBorfusC, while the remainder harbored mutations in the gene (fusA) encoding the drug target (EF-G). The CC80-MRSA-IV clone carryingfusBaccounted for ∼61% of the resistant isolates in this collection, while a single CC5 clone harboringfusCrepresented ∼12% of the resistant strains. These findings emphasize the importance of clonal dissemination of fusidic acid resistance within European MRSA strains. Nonetheless, the distribution offusBandfusCacross several genetic lineages, and their presence on multiple genetic elements, indicates that horizontal transmission of fusidic acid resistance genes has also played an important role in the increasing prevalence of fusidic acid resistance in MRSA.

Klebsiella pneumoniae is commonly found in environments, causing secondary infections in both human and animals, as well as antibiotic resistance problem. The objective of this study was to determine the prevalence of K. pneumoniae contaminated in the environments of surgical practice and laboratory dog husbandry rooms at Animal hospital, Rajamangala University of Technology Tawan-ok during 2019 - 2020. Two-hundred-swabbed samples were collected from these nearby environments and laboratory dogs themselves. Then, all samples were tested for K. pneumoniae and antibiotic susceptibility using disk diffusion method. From the results, the total prevalence was 3.5% and the high frequencies were found in three categories: laboratory dogs (35.7%), veterinarian belongings (3.3%) and building structure (2.5%). The antibiotic resistance was also detected including Cephalexin, Ceftazidime and Penicillin G (100%), Gentamicin and Colistin (85.71%), Amikacin (42.86%), and Ciprofloxacin (14.29%). Owning to the fact that laboratory dogs exposed to an external environment leading to bacterial contamination into themself and the husbandry room. Therefore, the hygiene of laboratory dogs and area in husbandry room should be primarily considered. Our study would be the preliminary baseline for the study of antibiotic resistant K. pneumoniae contamination in dogs, human, and their environments.

Background: Staphylococcus aureus is an opportunistic pathogen, which often causes asymptomatic diseases. This bacterium could cause several disorders, ranging from skin infections to life-threatening diseases. S. aureus could also develop resistance to a wide range of antibiotics. Objectives: The present study aimed to investigate the prevalence and pattern of antibiotic resistance in isolated S. aureus in Imam Reza Hospital of Kermanshah, Iran. Methods: This descriptive cross-sectional study was conducted on 2,228 patients with a positive S. aureus culture who were admitted to Imam Reza Hospital during 2016 - 2018. In accordance with the CLSI protocol, bacterial isolates were separated using laboratory tests, and antibiotic susceptibility was assessed using the standard disk-diffusion method. Data analysis was performed in SPSS version 24. Results: Most of the S. aureus-positive cases were isolated from the emergency ward (43.7%), blood samples (40.1%), and urine samples (23.9%). The highest antibiotic resistance was observed against erythromycin, penicillin G, ofloxacin, cefoxitin, clindamycin, and piperacillin. With 93.3% and 81.8% sensitivity, vancomycin and teicoplanin were respectively the most effective antibiotics against S. aureus. Conclusions: According to the results, the prevalence of resistant S. aureus strains could be due to the long-term hospitalization of patients and the overuse of antibiotics in infection treatment. Therefore, proper monitoring and development of effective infection control methods are essential in these healthcare settings.

<p><strong>BACKGROUND:</strong> Enteric fever has huge global burden. Surrogate markers may have a role in early diagnosis. <strong>AIM:</strong> Unselected retrospective analysis of 50 culture positive Salmonella enterica cases for epidemiology, laboratory<br />markers, antibacterial susceptibility, therapy, and outcome was done. This was a retrospective chart review of electronic medical records for 50 patients with Salmonella in blood cultures for in our hospital during May 2009-April 2010. <strong>MATERIALS AND METHODS:</strong> Blood culture was by automated Bactec™ system and antimicrobial susceptibility test was performed by disk diffusion method or automated system Phoenix 100™. Sensitivity of Widal, S. typhi IgM, C-reactive protein (CRP), total leukocyte count, erythrocyte sedimentation rate (ESR), differential eosinophil and lymphocyte counts, and liver enzymes, was determined for these cases. <strong>RESULTS:</strong> Salmonella cases were seen in all ages from 8 months to 59 years; serotype Typhi was the most common (72%). Sensitivity of S. typhi IgM immunochromatographic test was 78.9%, of Widal was 88.8%, and that of CRP, serum aspartate transaminase (AST), and serum alanine transaminase (ALT) ranged from 81.8 to 89.4%. Eosinophil count of zero and ESR were found to be 78.2% and 85.7% sensitive. Nalidixic acid resistance was seen in 96% cases and ciprofloxacin resistance/intermediate sensitivity in 26% cases. Resistance was not seen with ceftriaxone, while ampicillin, chloramphenicol, and co-trimoxazole (ACCo) resistance was 4%. Seven cases relapsed mostly due to improper treatment associated with choice/dose/duration of antibiotics. <strong>CONCLUSIONS:</strong> Surrogate laboratory markers can be utilized pending culture results. Though antibiotics and vaccines against Salmonella, and good sanitation facilities are available, it causes morbidity in all sections, gender, and ages of society. Strategies of prevention have not been very successful; therefore, early detection and effective treatment can prevent its complications and relapses.</p>

Ozone has a high wound healing capacity and antibacterial properties and can be used as a complementary treatment in infections. Methicillin-resistant S. aureus (MRSA) is the most common pathogen found in infected diabetic foot ulcers. Most of MRSA are resistant to several classes of antibiotics and, therefore, there is a need for new, effective, and well-tolerated agents. Thus, we aimed evaluate the antimicrobial and antibiofilm potentials of ozonated vegetable oils against MRSA strains isolated from diabetic foot ulcers. Six ozonated oils were produced with concentrations of ozone ranging from 0.53 to 17 mg of ozone/g of oil. The peroxide values were determined for each oil. Ozonated oils content on fatty acid was determined by gas chromatography equipped with a flame ionization detector. The antimicrobial susceptibility testing was performed by the Kirby–Bauer disk diffusion method and the effect of ozonated oils on biofilm formation ability and on established biofilms was investigated. In general, the content in identified unsaturated fatty acid in oils decreased with the increase of ozonation time and, consequently, the peroxide value increased. Most bacterial strains were inhibited by ozonated oil at a concentration of 4.24 mg/g. Ozonated oils had moderate to high ability to remove adhered cells and showed a high capacity to eradicate 24 h old biofilms. Our results show promising use of ozonated oils on the treatment of infections, in particular those caused by multidrug-resistant MRSA strains.

Methicillin-resistant Staphylococcus aureus (MRSA) are one of the main pathogens associated with purulent infections. MRSA clonal complex 97 (CC97) has been identified in a wide diversity of livestock animals. Therefore, we aimed to investigate the antibiotic resistance profiles of MRSA strains isolated from purulent lesions of food-producing rabbits. Samples from purulent lesions of 66 rabbits were collected in a slaughterhouse in Portugal. Samples were seeded onto ORSAB plates with 2 mg/L of oxacillin for MRSA isolation. Susceptibility to antibiotics was tested by the disk diffusion method against 14 antimicrobial agents. The presence of resistance genes, virulence factors and the immune evasion cluster (IEC) system was studied by polymerase chain reaction. All isolates were characterized by multilocus sequence typing (MLST), agr and spa typing. From the 66 samples analyzed, 16 (24.2%) MRSA were detected. All strains were classified as multidrug-resistant as they were resistant to at least three classes of antibiotics. All isolates showed resistance to penicillin, erythromycin and clindamycin. Seven isolates were resistant to gentamicin and harbored the aac(6′)-Ie-aph (2″)-Ia gene. Resistance to tetracycline was detected in 10 isolates harboring the tet(K) gene. The IEC genes were detected in three isolates. MRSA strains belonged to CC97, CC1, CC5, CC15 or CC22. The isolates were assigned to six different spa types. In this study we found a moderate prevalence of multidrug-resistant MRSA strains in food-producing rabbits. This may represent concern for food safety and public health, since cross-contamination may occur, leading to the spread of MRSA and, eventually, the possibility of ingestion of contaminated meat.

ABSTRACT We aimed to perform serotyping and the antimicrobial resistance profile of Salmonella spp. and Listeria monocytogenes strains isolated from raw meats imported illegally into Brazil along the borders of Argentina and Uruguay. Distinct isolates of Salmonella spp. (n = 6) and L. monocytogenes (n = 25) obtained from 270 of these food products of earlier work were serotyped and tested for antimicrobial resistance by agar disk diffusion method. For strains that were considered phenotypically resistant, antimicrobial resistance genes were investigated: strA, strB, floR, tetA, tetB, blaZ, blaTEM, ermB, ermC, and ereB to Salmonella sp. and blaZ and mecA to L. monocytogenes. All Salmonella isolates were identified as Salmonella Infantis; they were multidrug resistant and harbored the genes blaTEM (n = 6), strA (n = 1), strB (n = 1), floR (n = 1), ermB (n = 1), tetA (n = 3), and tetB (n = 3). L. monocytogenes isolates belonged to serovars 1/2a (n = 1), 1/2b (n = 14), 1/2c (n = 2), and 4b (n = 8), showed resistance only to penicillin G (n = 12), and did not show the blaZ and mecA genes. The results demonstrated that illegal foods that are commercialized in the Brazilian international border with Argentina and Uruguay may harbor foodborne pathogens, and some of them have multidrug resistance characteristics, such as Salmonella, emphasizing the need for greater control of international food transit in Brazil, especially in the region evaluated.
