Natural deep eutectic solvents (NaDESs) are recently developed green solvent alternatives to conventional fossil solvents. The present work systematically screened 22 different NaDESs for the ultrasonic-assisted extraction of bioactive components from Salvia miltiorrhiza (SM), a widely used traditional Chinese medical plant. The suitable solvent and extraction condition were optimized in a two-round screening. In comparison with fossil solvents, NaDESs, especially L-proline-lactic acid (L-Pro-Lac) showed significant advantages in the extraction of salvianolic acid B (SAB), tanshinone IIA (TIIA) and cryptotanshinone (CYT). The optimized yields of the three targeting compounds were 42.05, 1.485 and 0.839 mg/g, respectively. The present method was also applied to the pretreatment of SM samples from different geographic origins. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities of NaDES extracts were determined in the study to prove the feasibility of NaDES in bioactive component extraction. The application of NaDESs in the extraction of both hydrophilic and hydrophobic small molecules from SM is proved to be a green and efficient method for pretreatment of herbal materials.

The present study aimed at evaluating the effectiveness of different natural deep eutectic solvents (NADES) on the extraction of phenolic compounds from Lavandula pedunculata subsp. lusitanica (Chaytor) Franco, on the antioxidant activity, and acetylcholinesterase (AChE), butyrylcholinesterase (BChE) and tyrosinase (Tyr) inhibitory capacities. Ten different NADES were used in this research and compared with conventional solvents. Ultrasound-assisted extraction (UAE) for 60 min proved to be the best extraction condition, and proline:lactic acid (1:1) and choline chloride:urea (1:2) extracts showed the highest total phenolic contents (56.00 ± 0.77 mgGAE/gdw) and antioxidant activity [64.35 ± 1.74 mgTE/gdw and 72.13 ± 0.97 mgTE/gdw in 2.2-diphenyl-1-picrylhydrazyl (DPPH) and 2.2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods, respectively]. These extracts also exhibited enzymes inhibitory capacity particularly against Tyr and AChE. Even so, organic acid-based NADES showed to be the best extractants producing extracts with considerable ability to inhibit enzymes. Twenty-four phenolic compounds were identified by HPLC-HRMS, being rosmarinic acid, ferulic acid and salvianolic acid B the major compounds. The results confirmed that the combination of UAE and NADES provide an excellent alternative to organic solvents for sustainable and green extraction, and have huge potential for use in industrial applications involving the extraction of bioactive compounds from plants.

Natural deep eutectic solvents (NADESs), composed of natural primary metabolites, are now widely used as green and sustainable extraction solvents of bioactive components. In the present study, various NADESs were prepared to extract multi-components from different preparations of an herbal formula (Chinese name: Jinqi Jiangtang, JQJT) using ultrasound-assisted extraction (UAE). Results showed that most prepared NADESs provided more effective extraction of phenolic acids and alkaloids from JQJT preparations than conventional solvents. Among the tested NADESs, the solvent formed by choline chloride and laevulinic acid was selected to optimize the operational parameters using response surface methodology. The optimized extraction method was successfully applied to extract six major components in four commercial JQJT products, and quantification analysis was performed by the validated high-performance liquid chromatography with ultraviolet detection (HPLC-UV) method. The quantitative results indicated that preparations from different manufacturers showed different chemical profiles. In conclusion, NADESs-based UAE shows considerable potential as an efficient and green method for extraction of multi-bioactive components from commercial herbal preparations.

Deep eutectic solvents (DESs), a new group of ecofriendly solvent combined with the ultrasonic-assisted extraction (UAE) technique, were first successfully used for extraction of resveratrol from peanut roots. Resveratrol in the extract was analyzed and quantified using a HPLC-UV method. A series of DESs consisting of choline chloride (ChCl) and 1,4-butanediol, citric acid, and ethylene glycol were formulated, finding ChCl/1,4-butanediol was a most proper extraction system. The optimal extraction parameters were obtained using a Box–Behnken design (BBD) test combined with response surface methodology as follows: 40% of water in ChCl/1,4-butanediol (1/3, g/g) at 55°C for 40 min and solid/liquid ratio of 1:30 g/mL. The total extraction content and extraction yield of resveratrol from peanut roots could reach 38.91 mg/kg and 88.19%, respectively, under such optimal conditions. The present study will provide a typical example for using DESs to extract natural bioactive compounds from plants.

Herbs with antioxidant properties are usually preserved and extracted before being converted into commercial products. The main focus of this study was to determine the effect of preservation of A. sessilis red as well as extraction method on its antioxidant properties. A. sessilis red was preserved using two different methods; freeze drying and superheated steam drying, followed by extraction with 70% ethanol using conventional extraction and ultrasonic-assisted extraction. Drying in the superheated steam oven displayed shorter drying period of 1 hour compared to freeze drying (several days). Combination of superheated steam drying and ultrasonic-assisted extraction showed the highest extraction yields (12.99%). Results showed that superheated steam drying and ultrasonic-assisted extraction displayed an increase in the total phenolic content. In terms of antioxidant capacity, A. sessilis extracts obtained from superheated steam drying has higher radical scavenging activity (72.39% - 76.70%) than those freeze-dried (60.68% - 65.33%). Meanwhile, ultrasonic-assisted extraction had negatively impacted the radical scavenging activity of the extracts due to the formation of free radicals that are related to acoustic cavitation. As for ferric reducing antioxidant power, both superheated steam drying and ultrasonic assisted extraction yielded extracts with greater capacity. Present result shows that the combination of superheated steam drying and ultrasonic-assisted extraction enhanced total phenolic content by 60% and improved antioxidant activity based on ferric reducing antioxidant power assay

Ultrasonic-assisted extraction (UAE) method was used to polysaccharides from Radix Astragali (RAP), and the orthogonal test was applied to optimize extraction conditions. Furthermore, the antioxidant activities of RAP were evaluated by DPPH and superoxide anion radical scavenging activities assay. The results indicated that the optimal conditions for RAP extraction as followings: solid/liquid ratios of 1:20, extraction time of 10 min, ultrasonic power of 65 W and extraction temperature of 60°C. Meanwhile, RAP had relatively higher scavenging activities on DPPH and superoxide anion radicals.

Physalis pubescens L. is rich in natural pigments but has not yet been fully utilized. Ultrasound-assisted extraction of yellow pigment from Physalis pubescens L. was investigated by response surface methodology in this study. Optimal parameters were ultrasonic power of 29.21%, ultrasonic time of 14.41 min, and ultrasonic interval time of 10.55 s. The yield was 0.193% under optimal parameters. FRAP, ABTS, and superoxide radical scavenging activity of the yellow pigment were 6.11 ± 0.22 mmol/g, 2.80 ± 0.27 mmol/g, and 57281.5 ± 2749.5 U/g, respectively. The results showed that the yield of yellow pigment could be improved by ultrasonic-assisted extraction and the yellow pigment extracted by ultrasound had antioxidant activity.

Flaxseed is the chief oilseed crop that is grown in many regions of the world for medicinal and nutritional purposes. It has been used for oil and fiber since centuries. Flaxseed has an enormous role in functional foods for its nutritional and pharmaceutical values. Among the various components of flaxseed lignans, phenolic acids, proteins and oil are of the main interest. Oil and lignans have prime concentrations in the flaxseed. To study the extraction of these compounds, various extraction methods have been investigated. Oil has been extracted by mechanical pressing in ancient times but presently new techniques have been developed. These include microwave assisted extraction, supercritical CO2 extraction, ultrasonic assisted extraction, etc. Enzymes are also gaining importance in the extraction of oil, lignan and proteins as they give more yield of compounds and easily degrade the cell wall of the flaxseed. The need of these novel techniques lies in the fact that traditional methods have different shortcomings like low yield, more time, more energy and less environmental friendly. This review put on a view to different techniques which have been investigated for the extraction of different components of flaxseed. Quality evaluation and comparison of flaxseed oils and other bioactive components obtained by newer techniques with those produced by conventional extraction methods is also reported.
