Erysipelas is a disease caused by the Erysipelothrix genus, whose main species is the E. rhusiopathiae, the causative agent of animal erysipelas and human erysipeloid. We isolated Erysipelothrix sp. strain 2 (ES2) from turkey’s organs during an outbreak in Brazilian commercial and breeder flocks with sepsis and high mortality levels. We studied 18 flocks, accounting for 182 samples, being eight flocks (84 samples) as ES2 positive with individuals demonstrating clinical symptoms and high mortality. We obtained the genetic variability of 19 samples with PFGE and found two clones, both from the same flock but different samples, and two clusters. Interestingly, we found 15 strains with high genetic variability among and within flocks. We have found a positive association between the proximity of ES2 positive turkey flocks and commercial swine sites through epidemiological analysis. We infected Vero cells with two different isolates and three distinct concentrations of ES2. After performing the morphometry, we recorded enlargement of the nucleus and nucleolus. Moreover, we performed fluorescence assays that resulted in apoptotic and necrotic cells. We demonstrated that ES2 could multiply in the extracellular medium and invade and survive inside Vero cells. For the first time, our finds show that ES2 may have similar behavior as E. rhusiopathiae as a facultative intracellular microorganism, which may represent a hazard for humans.

Toads in the family Bufonidae contain bufadienolides in their venom, which are characterized by their chemical diversity and high pharmacological potential. American trypanosomiasis is a neglected disease that affects an estimated 8 million people in tropical and subtropical countries. In this research, we investigated the chemical composition and antitrypanosomal activity of toad venom from Rhinella alata collected in Panama. Structural determination using mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectroscopy led to the identification of 10 bufadienolides. Compounds identified include the following: 16β-hydroxy-desacetyl-bufotalin-3-adipoyl-arginine ester (1), bufotalin (2), 16β-hydroxy-desacetyl-bufotalin-3-pimeloyl-arginine ester (3), bufotalin-3-pimeloyl-arginine ester (4), 16β-hydroxy-desacetyl-bufotalin-3-suberoyl-arginine ester (5), bufotalin-3-suberoyl-arginine ester (6), cinobufagin-3-adipoyl-arginine ester (7), cinobufagin-3-pimeloyl-arginine ester (8), cinobufagin-3-suberoyl-arginine ester (9), and cinobufagin (10). Among these, three new natural products, 1, 3, and 5, are described, and compounds 1–10 are reported for the first time in R. alata. The antitrypanosomal activity assessed in this study revealed that the presence of an arginyl-diacid attached to C-3, and a hydroxyl group at C-14 in the structure of bufadienolides that is important for their biological activity. Bufadienolides showed cytotoxic activity against epithelial kidney Vero cells; however, bufagins (2 and 10) displayed low mammalian cytotoxicity. Compounds 2 and 10 showed activity against the cancer cell lines MCF-7, NCI-H460, and SF-268.

We describe for the first time the genetic and antigenic characterization of 18 avian avulavirus type-6 viruses (AAvV-6) that were isolated from wild waterfowl in the Americas over the span of 12 years. Only one of the AAvV-6 viruses isolated failed to hemagglutinate chicken red blood cells. We were able to obtain full genome sequences of 16 and 2 fusion gene sequences from the remaining 2 isolates. This is more than double the number of full genome sequences available at the NCBI database. These AAvV-6 viruses phylogenetically grouped into the 2 existing AAvV-6 genotype subgroups indicating the existence of an intercontinental epidemiological link with other AAvV-6 viruses isolated from migratory waterfowl from different Eurasian countries. Antigenic maps made using HI assay data for these isolates showed that the two genetic groups were also antigenically distinct. An isolate representing each genotype was inoculated in specific pathogen free (SPF) chickens, however, no clinical symptoms were observed. A duplex fusion gene based real-time assay for the detection and genotyping of AAvV-6 to genotype 1 and 2 was developed. Using the developed assay, the viral shedding pattern in the infected chickens was examined. The chickens infected with both genotypes were able to shed the virus orally for about a week, however, no significant cloacal shedding was detected in chickens of both groups. Chickens in both groups developed detectable levels of anti-hemagglutinin antibodies 7 days after infection.

Abstract Genetics: The chromosome number reported for C. dactylon varies from 2n = 18 to 2n = 36 with diploid and polyploid populations (Cook et al., 2005). Ramakrishan and Singh (1966) and Sarandon (1991) have found differences in total biomass and biomass partition according to the origin of the population. Sarandon (1991) points out that characters are highly heritable, which means that high genetic variability for biomass production and variable architecture allows an ample base for selection, which in most cases is induced by herbicides, mechanical control or forage production. Reproductive Biology: C. dactylon is wind-pollinated and generally self-incompatible, suffering from inbreeding depression when genotypes are self-pollinated. Quantitative traits such as seed yield and forage yield can be dramatically negatively affected by inbreeding depression (Cook et al., 2005). In diploid populations, caryopses are formed after zygote formation. In polyploids, which are sterile, caryopses may be apomictic. Physiology: This C4 plant (Kissmann, 1991) has high rates of accumulation under adequate irradiance, water and nutrient supply and may consume 75 kg of N, 20 kg of P and more than 1,500,000 litres of water for 5000 kg/ha of biomass dry matter (Fernandez, 1991). In the south of Santa Fe province, Argentina, a maximum biomass of 8000 kg/ha may be generated under a summer crop of maize or sunflower with >75% located in the first 10 cm of the soil profile (Lombardo, 1973), whereas in Balcarce (Argentina) about 5000 kg/ha is commonly found in maize or sunflower stubble. Phenology: A photoperiod of 13 hours induces flowering. Low night temperatures coupled with high diurnal temperatures induces blooming (Nir and Koller, 1976). A reduction in irradiance drastically decreases inflorescence production (Moreira, 1975). In North America, annual plants reproduce during spring and perennial plants reproduce all year long (USDA-NRCS, 2014). Longevity: C. dactylon grows as both an annual and perennial grass. The annual growth-form becomes dormant and turns brown when nighttime temperatures fall below freezing or average daytime temperatures are below 10°C (Cook et al., 2005). Activity Patterns: Seeds may be the route of invasion in weed-free fields through the faeces of cows (Rodriguez, personal communication). Rhizome biomass exhibits an annual cyclic pattern and, as with any perennial weed, low temperatures reduce biomass and viability is lost as a consequence of the consumption of materials due to respiration and maintenance. The digestibility of stocked material is severely decreased, implying a loss in forage quality (Vaz Martins, 1989). This is a character that has largely improved in cultivated varieties. Each node has a physiological self-governing structure in relation to the apex, but is highly dependent on substances from other plant parts. The mother plant determines the runner growth pattern on the soil surface according to the sugar-gibberellin balance (Montaldi 1970). Node disconnection may be caused by natural decay and cultivation and produces damage in the breakdown zone and changes in hormone and nutrient relationships. It is widely demonstrated that rhizome or runner fragmentation induces the activation of buds. The proportion of activated buds increases as the number of buds per segment decreases (Moreira, 1980; Kigel and Koller, 1985; Fernandez and Bedmar, 1992). The cultivation method is mainly responsible for vegetative propagation fragmentation. The higher the cultivation intensity, the smaller the segments produced (Kigel and Koller, 1985). Population Size and Structure This weed produces an enormous number of small seeds (0.25-0.30 mg), the viability and dormancy of which are highly variable according to genotype and the conditions when formed. The seed is important because it confers high genetic variability on the population. Perez et al. (1995) recorded a very low germination rate. Uygur et al. (1985) obtained up to 15% germination at constant temperatures of 35-40°C, and 50% at temperatures alternating between 20 and 30°C. Moreira (1975) obtained up to 80% germination with the help of nitrate, chilling and alternating temperatures, and Elias (1986) recorded up to 96% germination from heavier samples of seed. Seeds remain viable in the soil for at least 2 years (Caixinhas et al., 1988). As a rule, cultivars have relatively high viability. Osmo-conditioning of Bermuda grass seeds with PEG followed by immediate sowing improved seed germination and seedling growth under saline conditions (Al-Humaid 2002). The probability of emergence and successful establishment of C. dactylon decreases with the depth of the fragment, but increases with the weight of the node and internode (Perez et al., 1998). Growth from plants originated from a runner may exhibit a different biomass partition than that from plants originated from a rhizome (Fernandez, 1986). From sprouting onwards, weed growth is controlled mainly by temperature (optimum 25-30°C) and radiation, but also by humidity and soil fertility. The efficiency of carbohydrate reserve usage during sprout growth is highly dependent on temperature and the type of vegetative structure; it is maximum at 20°C and is higher for rhizomes than for stolons (Satorre et al., 1996). Runners and rhizome growth begins 30 days after growth but only if soil temperature is >15°C. Rates of 15 g/g/day have been recorded in Argentina (Lescano de Ríos, 1982).

Introduction: Bovine ephemeral fever virus (BEFV) is an arthropod borne Rhabdovirus affects cattle and water buffalo causes acute febrile disease. Methodology: The clinical picture and epidemiological pattern of BEF were described among cattle in epidemics of 2007, 2009 and 2011 in four geographical regions of Kingdom Saudi Arabia (Eastern, Jizan, Qasim, and Riyadh). Serum samples were tested using VNT. Virus isolation and molecular characterization were carried out for the first time in KSA. Results: The main clinical symptoms were fever, stiffness, lameness, salivation and subcutaneous emphysema. The prevalence and the mortality rate of BEF have decreased from 70% and 4.6% in 2007 to 30% and 0.6% in 2011, respectively in the 4 studied areas. There was no region association with higher prevalence of BEF. The intracluster correlation (ICC) was estimated for the first time in KSA as 0.0034. BEFV had been isolated from 11 out of 20 samples (55%) and isolation was confirmed by VNT. The molecular detection of BEFV by RT-PCR and real- time RT-qPCR were found more sensitive for diagnosis of the disease than virus isolation; 80% and 90% for the former tests and 55% for the latter. Three isolates were sequenced, they showed 84.7% - 100% identities in between and shared 90.4%-96.5% sequence identity with a previously published sequence from Australia (KF679404). The generated sequences belonged to 3rd cluster of BEFV glycoprotein. Conclusions: BEF occurrence has cyclic nature and the efficacy of vaccines prepared from local strains has to be evaluated and considered in diseases control.

Interações droga-droga (IDDs) são responsáveis por problemas terapêuticos e as principais causas de reações adversas a medicamentos que levam a hospitalização. O objetivo do trabalho foi analisar a incidência de IDDs em pacientes admitidos em hospital público no Brasil, em 2015. Em um estudo transversal, 351pacientes foram selecionados com IDDs utilizando-se o banco de dados da Micromedex®, e as interações foram classificadas de acordo com a severidade e documentação. Os potenciais IDDs foram avaliados durante o acompanhamento farmacoterapêutico diário por anamnese farmacêutica e a análise dos exames laboratoriais foi realizada. Um total de 2.937 potenciais IDDs foram identificados e 28,42% deles tinham sinais e sintomas clínicos confirmados em 87 pacientes (29%). Entre os pacientes que apresentaram interações, 62,07% tinham mais que 60 anos. Neste grupo de idade, 61,27% de todos os sinais e sintomas clínicos observados foram também identificados, demonstrando uma associação positiva entre interações clínicas e idade. Além disso, uma correlação positiva entre o número de drogas prescritas e a ocorrência de sinais clínicos também foram observados. Muitos IDDs observados foram de severidade moderada e estavam relacionados a alterações das pressões arteriais e níveis glicêmicos. Este estudo demonstrou que IDDs estão diretamente relacionados a idade e número de drogas prescritas. E a grande frequência de IDDs com documentação fraca alertam para a necessidade de se analisar esse tipo de interação. Sendo assim, este estudo mostrou que potenciais IDDs e sinais e sintomas clínicos significantes em pacientes só reforçam a necessidade de se apoiar a farmácia clínica.   Palavras-chave: Interação Droga-Droga (IDD). Sinais e Sintomas.   Abstract Drug-drug interactions (DDIs) are responsible for therapeutic problems and the main causes of adverse drug reactions that require hospitalization. The aim of this study was to analyze the incidence of DDIs in patients admitted in a Public Hospital in Brazil, in 2015. In a cross-sectional study, DDIs were screened in 351 patients, using Micromedex® database, that classify interactions according to severity and documentary evidence. Potential DDIs were assessed at the daily pharmacotherapeutic follow-up through pharmaceutical anamnesis and analysis of laboratory tests were performed. A total of 2,937 potential DDIs were identified and 28.42% of these had confirmed signs and symptoms clinical in 87 patients (29%). Among the patients that presented interactions, 62.07% were older than 60 years. In this age group, 61.27% of all signs and symptoms clinical observed in the study were also identified, demonstrating a positive association between the occurrence of clinical interactions and age. In addition, a positive correlation between the number of drugs prescribed and the occurrence of signs clinicals was also observed. Most DDIs observed were of moderate severity and were related to imbalance of blood pressure and glycemic levels. This study demonstrated that DDIs are directly related to the age and number of drugs prescribed. And the greater frequency of DDIs with fair documentary evidence alerts to the need to consider all the possible interactions. Thereafter, this study showed that potential DDIs and sign and clinical symptoms  are significant in patients and reinforce the need to support Clinical Pharmacy.   Keywords: Drug-Drug Interaction (DDI). A goose hemorrhagic polyomavirus (GHPV) outbreak occurred in a Goose Farm in Hunan, China, between January and July 2021. Approximately 1,500 breeding goose died, and hatching rates dropped from the previous 85% to about 50% in this outbreak. GHPV HUN-01, isolated from the liver of infected Landes geese, shared a close evolutionary relationship with the Toulouse Goose 2000 and 14234 strain, isolated from geese in France and Hungary. The isolation of GHPV from the livers of dead embryos also demonstrates that the virus can be transmitted vertically. In conclusion, clinical and laboratory diagnostics of the diseased geese in this outbreak were consistent with GHPV being the causative agent. We learned that this is the first time that GHPV has been isolated from geese in mainland China.

Canine parvovirus (CPV) type 2 is the causative agent of acute hemorrhagic enteritis and high mortality in the affected dogs. Numerous studies have been done to understand the origin of the virus and to exhibit new variants and circulating strains. This report describes the detection and genomic characterization of CPV strains from indoor and outdoor dogs in Ankara, Turkey. Samples were sent to our laboratory due to clinical symptoms in puppies. We tested blood and swab samples to determine the presence of canine parvovirus (CPV) in three puppies and two adult dogs by reverse transcription-polymerase chain reaction (RT-PCR) using VP2 (capsid protein) region primers of canine parvoviruses. Following that, to provide molecular characterization data Maximum Likelihood (ML) method was used for phylogenetic analyses. Constructed phylogenetic trees from the aligned nucleotide sequences revealed that our CPV strains demonstrated high genetic similarities, with 100% identity match on nucleotide alignments with each other and classified in CPV-2b genotypes.They have placed on a monophyletic clade as a sister branch with CPV VAC S quantum with 98.9% nucleotide homology. Our findings suggest that CPV-2b is actual and frequently seen variant in Turkey and shows high similarities with other CPV variants and a bit less with FPVs in Turkey and around the world. CPV causes high mortality and morbidity in dogs and to develop effective vaccines for protection of dogs in Turkey where there are few numbers of studies that have been done, field strains should be isolated and characterised.
