Lupins anthracnose is a destructive seed and airborne disease caused by Colletotrichum lupini, affecting stems and pods. Primary seed infections as low as 0.01–0.1% can cause very severe yield losses. One of the most effective management strategies is the development of a robust and sensitive seed detection assay to screen seed lots before planting. PCR-based detection systems exhibit higher levels of sensitivity than conventional techniques, but when applied to seed tests they require the extraction of PCR-quality DNA from target organisms in backgrounds of saprophytic organisms and inhibitory seed-derived compounds. To overcome these limitations, a new detection protocol for C. lupini based on a biological enrichment step followed by a PCR assay was developed. Several enrichment protocols were compared with Yeast Malt Broth amended with ampicillin, streptomycin, and lactic acid were the most efficient. A species-specific C. lupini primer pair was developed based on rDNA IGS sequences. The specificity was evaluated against 17 strains of C. lupini, 23 different Colletotrichum species, and 21 different organisms isolated from seeds of Lupinus albus cv. Multitalia, L. luteus cv. Mister, and L. angustifolius cv. Tango. The protocol described here enabled the detection of C. lupini in samples artificially infected with less than 1/10,000 infected seed.

Mangrove forests provide a large variety of ecosystem services (ES) to coastal societies. Using a case study focusing on the Ajuruteua peninsula in Northern Brazil and two ES, food provisioning (ES1) and global climate regulation (ES2), this paper proposes a new framework for quantifying and valuing mangrove ES and allow for their small-scale mapping. We modelled and spatialised the two ES from different perspectives, the demand (ES1) and the supply (ES2) side respectively. This was performed by combining worldwide databases related to the global human population (ES1) or mangrove distribution and canopy height (ES2) with locally derived parameters, such as crab catches (ES1) or species-specific allometric equations based on local estimates of tree structural parameters (ES2). Based on this approach, we could estimate that the area delivers the basic nutrition of about 1400 households, which equals 2.7 million USD, and that the mangrove biomass in the area contains 2.1 million Mg C, amounting to 50.9 million USD, if it were paid as certificates. In addition to those figures, we provide high-resolution maps showing which areas are more valuable for the two respective ES, information that could help inform management strategies in the future.

Survival and inoculum production of Gibberella zeae (Schwein.) Petch (anamorph Fusarium graminearum (Schwabe)), the causal agent of Fusarium head blight of wheat and barley, was related to the rate of wheat (Triticum aestivum L.) residue decomposition. Infested wheat residue, comprising intact nodes, internodes, and leaf sheaths, was placed in fiberglass mesh bags on the soil surface and at 7.5- to 10-cm and 15- to 20-cm depths in chisel-plowed plots and 15 to 20 cm deep in moldboard-plowed plots in October 1997. Residue was sampled monthly from April through November during 1998 and every 2 months through April to October 1999. Buried residue decomposed faster than residue placed on the soil surface. Less than 2% of the dry-matter residue remained in buried treatments after 24 months in the field, while 25% of the residue remained in the soil-surface treatment. Survival of G. zeae on node tissues was inversely related to the residue decomposition rate. Surface residue provided a substrate for G. zeae for a longer period of time than buried residue. Twenty-four months after the initiation of the trial, the level of colonization of nodes in buried residue was half the level of colonization of residue on the soil surface. Colonization of node tissues by G. zeae decreased over time, but increased for other Fusarium spp. Ascospores of G. zeae were still produced on residue pieces after 23 months, and these spores were capable of inducing disease. Data from this research may assist in developing effective management strategies for residues infested with G. zeae.

Climate change has been associated in phenological shifts for a variety of taxa. Amphibians, specifically the order Anura (frogs and toads), are considered particularly vulnerable due to their sensitivity to anthropogenic and environmental change. Previous research has documented shifts in the timing of anuran breeding that can be attributed, in part, to climate change, with potential implications for reproduction, survival, and development. This study aims to investigate how air temperature is associated with anuran calling phenology. I will examine the temporal trends in spring and summer air temperature in a lake in northern Ontario, Canada. and quantify seasonal patterns of calling anuran species using acoustic monitoring over a four-month period. I predict that there will be interspecific variation in peak calling associated with air temperature. Additionally, I expect to find asymmetrical association between air temperature and anuran species’ calling behaviour – wherein prolonged breeding species will have a larger optimal temperature range for calling compared to explosive breeding species. The findings of this research will aid in future conservation and provide insight for management strategies of anurans in Canada in response to anticipated climate warming.

Theobroma cacao is affected by viruses on every continent where the crop is cultivated, with the most well-known ones belonging to the Badnavirus genus. One of these, cacao mild mosaic virus (CaMMV), is present in the Americas, and is transmitted by several species of Pseudococcidae (mealybugs). To determine which species are associated with virus-affected cacao plants in North America, and to assess their potential as vectors, mealybugs (n = 166) were collected from infected trees in Florida, and identified using COI, ITS2, and 28S markers. The species present were Pseudococcus jackbeardsleyi (38%; n = 63), Maconellicoccus hirsutus (34.3%; n = 57), Pseudococcus comstocki (15.7%; n = 26), and Ferrisia virgata (12%; n = 20). Virus acquisition was assessed by testing mealybug DNA (0.8 ng) using a nested PCR that amplified a 500 bp fragment of the movement protein–coat protein region of CaMMV. Virus sequences were obtained from 34.6 to 43.1% of the insects tested; however, acquisition did not differ among species, X2 (3, N = 166) = 0.56, p < 0.91. This study identified two new mealybug species, P. jackbeardsleyi and M. hirsutus, as potential vectors of CaMMV. This information is essential for understanding the infection cycle of CaMMV and developing effective management strategies.

AbstractThe vermilion rockfish complex, which consists of the cryptic sister species vermilion and sunset rockfish, is one of the most valuable recreational fisheries on the U.S. West Coast. These species are currently managed as a single complex, and because of uncertainty surrounding the relative contribution of each species within existing data sources, the stock status of each species is not fully known. A reliable and cost-effective method is needed to disentangle these species that will allow for the development of abundance indices, life history profiles, and catch histories that may potentially support species-specific stock assessments. Using restriction-site associated DNA sequence (RADseq) markers we generated 10,003 polymorphic loci to characterize the vermilion rockfish complex. PCA and Bayesian clustering approaches based on these loci clearly distinguished between sunset and vermilion rockfishes and identified hybrid individuals. These loci included 203 highly differentiated (FST ≥ 0.99) single nucleotide polymorphisms, which we consider candidates in the planned development of a diagnostic assay capable of distinguishing between these cryptic species. In addition to clearly delineating to species, subsets of the interspecific markers allowed for insight into intraspecific differentiation in both species. Population genetic analyses for sunset rockfish identified two weakly divergent genetic groups with similar levels of genetic diversity. Vermilion rockfish, however, were characterized by three distinct genetic groups with much stronger signals of differentiation and significantly different genetic diversities. Collectively, these data will contribute to well-informed, species-specific management strategies to protect this valuable species complex.

Objective: To formulate a protocol for safe and effective management of an ophthalmologic practice in a hospital or a clinic during the Covid - 19 pandemic. Methods and analysis: Literature was reviewed regarding Covid-19 pandemic and ophthalmology. The following key words were used. Covid-19, telemedicine, challenges, management strategy. Result: An efficient protocol for ophthalmology practice during the Covid-19 pandemic was established which covered various protective measures that are essential and vital in the Covid-19 pandemic. Conclusion: An evidence-based protocol was formulated which may be modified in future as per the latest developments and trends in the ongoing pandemic.
