Due to increased frequency of cyanobacterial blooms and emerging evidence of cyanotoxicity in biofilm, reliable methods for early cyanotoxin threat detection are of major importance for protection of human, animal and environmental health. To complement the current methods of risk assessment, this study aimed to evaluate selected qPCR assays for detection of potentially toxic cyanobacteria in environmental samples. In the course of one year, 25 plankton and 23 biofilm samples were collected from 15 water bodies in Slovenia. Three different analyses were performed and compared to each other; qPCR targeting mcyE, cyrJ and sxtA genes involved in cyanotoxin production, LC-MS/MS quantifying microcystin, cylindrospermopsin and saxitoxin concentration, and microscopic analyses identifying potentially toxic cyanobacterial taxa. qPCR analyses detected potentially toxic Microcystis in 10 lake plankton samples, and potentially toxic Planktothrix cells in 12 lake plankton and one lake biofilm sample. A positive correlation was observed between numbers of mcyE gene copies and microcystin concentrations. Potential cylindrospermopsin- and saxitoxin-producers were detected in three and seven lake biofilm samples, respectively. The study demonstrated a potential for cyanotoxin production that was left undetected by traditional methods in both plankton and biofilm samples. Thus, the qPCR method could be useful in regular monitoring of water bodies to improve risk assessment and enable timely measures.

Jammu division of Jammu and Kashmir is a region of high biological diversity and endemicity of fish fauna, but very little is known about the distribution of freshwater indigenous ornamental fish. The study was carried out across the tributaries of three rivers viz., Chenab, Tawi and Basantar to study the diversity of ornamental fish. Regular collection trips were carried out at 13 different stations for a period of one year from December 2017 to December 2018 and a total of 30 species were reported belonging to 8 families and 18 genera. In all the study stations Cyprinidae family was found to be most prevalent (85%) and Osphronemidae (1%) was the least. The present work is a preliminary study on the distribution and abundance of ornamental fishes from freshwater bodies of Jammu.

Abstract Toxic cyanobacterial blooms frequently develop in eutrophic freshwater bodies worldwide. Microcystis species produce microcystins (MCs) as a cyanotoxin. Certain bacteria that harbor the mlr gene cluster, especially mlrA, are capable of degrading MCs. However, MCs-degrading bacteria may possess or lack mlr genes (mlr+ and mlr− genotypes, respectively). In this study we investigated the genotype that predominantly contributes to biodegradation and cyanobacterial predator community structure with change in total MCs concentration in an aquatic environment. The two genotypes co-existed but mlr+ predominated, as indicated by the negative correlation between mlrA gene copy abundance and total MCs concentration. At the highest MCs concentrations, predation pressure by Phyllopoda, Copepoda, and Monogononta (rotifers) was reduced; thus, MCs may be toxic to cyanobacterial predators. The results suggest cooperation between MCs-degrading bacteria and predators may reduce Microcystis abundance and MCs concentration.

ABSTRACT Objective: to determine the diagnosis of the situation regarding documentation of falls and risk of falls in people older than 75 years in basic health units in Spain and Portugal. Method: mixed exploratory study in two stages: (i) quantitative descriptive of randomly selected fall records produced in one year (597 records; 197 Spanish and 400 Portuguese); and (ii) qualitative, with the purpose of knowing the perception of health professionals employing semi-structured interviews (72 professionals, 16 Spanish and 56 Portuguese). The study areas were two basic health units in southern Spain and northern Portugal. Results: in the fall records, the number of women was higher. The presence of fall was associated with the variables age, presence of dementia, osteoarticular disease, previous falls and consumption of antivertiginous medication. Health professionals perceived an absence of risk assessment instruments, as well as lack of prevention programs and lack of awareness of this event. Conclusion: falls are perceived as an area of priority attention for health professionals. Nonetheless, there is a lack of adherence to the registration of falls and risk assessment, due to organizational, logistical and motivational problems.

Cyanobacteria are photosynthetic microorganisms that are able to produce a large number of secondary metabolites. In freshwaters, under favorable conditions, they can rapidly multiply, forming blooms, and can release their toxic/bioactive metabolites in water. Among them, anabaenopeptins (APs) are a less studied class of cyclic bioactive cyanopeptides. The occurrence and structural variety of APs in cyanobacterial blooms and cultured strains from Greek freshwaters were investigated. Cyanobacterial extracts were analyzed with LC–qTRAP MS/MS using information-dependent acquisition in enhanced ion product mode in order to obtain the fragmentation mass spectra of APs. Thirteen APs were detected, and their possible structures were annotated based on the elucidation of fragmentation spectra, including three novel ones. APs were present in the majority of bloom samples (91%) collected from nine Greek lakes during different time periods. A large variety of APs was observed, with up to eight congeners co-occurring in the same sample. AP F (87%), Oscillamide Y (87%) and AP B (65%) were the most frequently detected congeners. Thirty cyanobacterial strain cultures were also analyzed. APs were only detected in one strain (Microcystis ichtyoblabe). The results contribute to a better understanding of APs produced by freshwater cyanobacteria and expand the range of structurally characterized APs.

The Norwegian Scientific Committee for Food Safety (Vitenskapskomiteen for mattrygghet, VKM) has, at the request of the Norwegian Food Safety Authority (Mattilsynet; NFSA), assessed the risk of "other substances" in food supplements and energy drinks sold in Norway. VKM has assessed the risk of doses given by NFSA. These risk assessments will provide NFSA with the scientific basis while regulating "other substances" in food supplements. "Other substances" are described in the food supplement directive 2002/46/EC as substances other than vitamins or minerals that have a nutritional and/ or physiological e ffect. It is added mainly to food supplements, but also to energy drinks and other foods. In this series of risk assessments of "other substances" the VKM has not evaluated any claimed beneficial effects from these substances, only possible adverse effects. The present report is a risk assessment of specified doses of L-threonine in food supplements, and it is based on previous risk assessments and articles retrieved from literature searches. According to information from NFSA, L-threonine is an ingredient in food supplements sold in Norway. NSFA has requested a risk assessment of 1000, 1200, 1500, 2000 and 2400 mg/day of L-threonine from food supplements.  L-threonine is an essential amino acid not known to cause any adverse health effects. Previous reports do not indicate a tolerable upper intake level, apart from an approval of a dose of 1150 mg/day by the Scientific Committee of the Spanish Agency for Food Safety and Nutrition (AESAN). Long-term studies in humans were not found. The only available human studies were: a small uncontrolled one-year pilot study with doses ranging from 0.5 to 2.5 g/day, one eight-week randomised controlled trial (RCT) using a dose of 7.5 g/day, and two 2-week RCTs using doses of 6 and 4.5 g/day. No adverse effects (diary method of registration of adverse effects) were reported in the eight-week clinical trial, and the only adverse effects observed in the two-week trials were one case of indigestion and one case of diarrhoea. A four-week rodent toxicity study indicated a no observed adverse effect level (NOAEL) of 854.3 mg/kg bw per day (only dose tested, no adverse effects observed).  The value used for comparison with the estimated exposure in the risk characterisation is the NOAEL defined in an 8-week randomised placebo controlled study in humans, 7500 mg/day. For a 70-kg individual, this corresponds to 107 mg/kg bw per day. Two human two-week studies and a small one-year pilot study support the notion that this dose will be well tolerated. The overall mean threonine intake according to NHANES III (3 g/day) is slightly larger than the doses requested for evaluation in the present risk assessment. No studies in children (10 to <14 years) and adolescents (14 to <18 years) were identified. Based on the included literature there was no evidence indicating that age affects the tolerance for relevant doses of threonine. Therefore, in this risk characterisation a tolerance as for adults, based on body weight, was assumed for these age groups. VKM concludes that: In adults (≥18 years), the specified doses 1000, 1200, 1500, 2000 and 2400 mg/day L-threonine in food supplements are unlikely to cause adverse health effects. In adolescents (14 to <18 years), the specified doses 1000, 1200, 1500, 2000 and 2400 mg/day L-threonine in food supplements are unlikely to cause adverse health effects. In children (10 to <14 years), the specified doses 1000, 1200, 1500, 2000 and 2400 mg/day L-threonine in food supplements are unlikely to cause adverse health effects. Children younger than 10 years were not within the scope of the present risk assessment.

The present paper attempts to report a total of 118 fish species spread over 11 orders, 26 families and 64 genera inhabited indifferent water bodies viz, rivers, reservoirs, streams, ponds, lakes, canals, ditches, paddy fields of Morang district, collected during a one-year survey. The order Cypriniformes is the richest one among 11 orders that comprises 59 species followed by Siluriformes having 31 and Perciformes with 19 representatives, respectively. Orders Anguilliformes, Osteoglossiformes, Beloniformes, Cyprinodontiformes, Synbranchiformes and Tetraodontiformes have only one representative. Olyra longicaudata is the new report from Morang district and the second report from Nepal. Besides this, other hill-stream fishes viz., Pseudecheneis sulcatus, Schizothorax plagiostomus, Schistura savona and Neolissochilus hexagonolepis recorded during the survey, also have not been previously reported from this district.

Mitten crab aquaculture is prevalent in China, however, knowledge about the threat of cyanobacteria in mitten crab aquaculture-impacted water bodies is limited. Here, seasonal variations of cyanobacteria and their relationships with environmental factors were investigated for Lake Guchenghu area. Results suggested the changes of cyanobacteria community in crab ponds distinguished from the adjacent lake. In the lake, cyanobacterial biomass (3.86 mg/L, 34.6% of the total phytoplankton) was the highest in autumn with the dominance of Oscillatoria, Aphanocapsa and Pesudanabaena. By contrast, in crab ponds, cyanobacteria (46.80 mg/L, 97.2% of the total phytoplankton biomass) were the most abundant in summer when Pesudanabaena and Raphidiopsis were the dominant species. Of particular note was that obviously higher abundance of filamentous and potentially harmful species (e.g., Raphidiopsis raciborskii and Dolichospermum circinale) were observed in ponds compared to the lake. Specifically, water depth (WD), permanganate index (CODMn), total phosphorus (TP), N:P ratio, and NO 2 −-N were the key environmental variables affected cyanobacteria composition. For crab ponds, N:P ratio, water temperature (WT) and TP were the potential environmental drivers of cyanobacteria development. This study highlighted the fact that mitten crab culture had non-negligible influences on the cyanobacteria community and additional attention should be paid to the cyanobacteria dynamics in mitten crab culture-impacted water bodies, especially for those potentially harmful species.
