AbstractChromosome segregation is mediated by spindle microtubules that attach to kinetochore via dynamic protein complexes, such as Ndc80, Ska, Cdt1 and ch-TOG during mitotic metaphase. While experimental studies have previously shown that these proteins and protein complexes are all essential for maintaining a stable kinetochore-microtubule interface, their exact roles in this process remains elusive. In this study, we employed experimental and computational methods in order to characterize how these proteins can strengthen kMT attachments in both non load-bearing and load-bearing conditions, typical of prometaphase and metaphase, respectively. Immunofluorescence staining of Hela cells showed that the levels of Ska and Cdt1 significantly increase from prometaphase to metaphase. Our new computational model showed that, by incorporating binding and unbinding of each protein complex, coupled with a biased diffusion mechanism, the displacement of a possible complex formed by Ndc80-Ska-Cdt1 is significantly higher than that formed by Ndc80 alone or Ndc80-Ska. In addition, when we use Ndc80/ch-TOG in the model, rupture force and time of attachment of the kMT interface increases. These results support the hypothesis that Ndc80-associated proteins strengthen kMT attachments and that it is the interplay between kMT protein complexes in metaphase that ensures stable attachments.

The pellicular structure of Euglena gracilis consists of a series of relatively rigid strips (Fig. 1) composed of ridges and grooves which are helically oriented along the cell and which fuse together into a common junction at either end of the cell. The strips are predominantly protein and consist in part of a series of fibers about 50 Å in diameter spaced about 85 Å apart and with a secondary periodicity of about 450 Å. Microtubules are also present below each strip (Fig. 1) and are often considered as part of the pellicular complex. In addition, there may be another fibrous component near the base of the pellicle which has not yet been very well defined.The pellicular complex lies underneath the plasma membrane and entirely within the cell (Fig. 1). Each strip of the complex forms an overlapping junction with the adjacent strip along one side of each groove (Fig. 1), in such a way that a certain amount of sideways movement is possible between one strip and the next.

SummaryTotal removal of a knee joint meniscus is followed by osteoarthritis in many mammalian species. Altered load-bearing has been observed in the human knee following meniscectomy but less is known about biochemical effects of meniscectomy in other species. Using pressure sensitive paper in sheep knee (stifle) joints it was found that, for comparable loads, the load-bearing area on the medial tibial condyle was significantly reduced following medial meniscectomy. Also, for loads of between 50 N and 500 N applied to the whole joint, the slope of the regression of contact area against load was much smaller. Following medial meniscectomy, the ability to increase contact area as load increased was markedly reduced.The load bearing area on the medial tibial condyle was reduced following meniscectomy.

The article describes the features of construction and technical expertise of structures of agricultural warehouse of mineral fertilizers. A draft program for visual and detailed instrumental inspection of building load-bearing structures has been developed. In accordance with the recommendations of the regulatory and technical documentation, measures have been developed to repair and restore the functional state of damaged structures.

The article considers an algorithm for analyzing the results of field strain-measurement studies of machine structures, which allows obtaining data for the modernization of elements in the form of coefficients of parameter changes. As the object of application of the method, the design element of the header was selected, which had failures due to insufficient endurance under cyclic bending stresses.
