Acute liver injury (ALI) is a life-threatening syndrome with high mortality and lacks effective therapies. Rodents under LPS (lipopolysaccharide)/D-Gal (D-galactosamine) stress mimic ALI by presenting dramatically increased inflammation and cell death in the liver. Euglena gracilis, functioning like dietary fiber, is commonly used as a paramylon (Pa)-rich nutritional supplement that has various biological effects such as regulating immune system, anti-obesity, and anti-tumor. Here, we found that Pa or sonicated and alkalized paramylon (SA-Pa) alleviated the LPS/D-Gal-induced hepatic histopathological abnormalities in mice. Compared with Pa, SA-Pa had lower molecular weights/sizes and showed better efficacy in alleviating injury-induced hepatic functions, as well as the transcriptional levels of inflammatory cytokines. Moreover, SA-Pa treatment promoted M2 macrophage activation that enhanced the anti-inflammatory function in the liver, and downregulated STAT3 target genes, such as Fos, Jun, and Socs3 upon the injury. Meanwhile, SA-Pa treatment also alleviated apoptosis and necroptosis caused by the injury. Our results demonstrated that SA-Pa efficiently protected the liver from LPS/D-Gal-induced ALI by alleviating inflammation and cell death.

Conventional process schemes of municipal sewage water treatment, advantages, and disadvantages of the methods applied when removing biogenic elements were considered. It was shown that the existing shortcomings cause additional explicit costs and difficulties when disposing of the resulting waste. Low efficiency of the removal processes themselves causing residual concentrations of biogenic elements in the treated sewage water was also shown. A process scheme for treating municipal sewage water was proposed. It includes the use of a photobioreactor of a proposed design for the removal of biogenic elements due to the metabolism of microalgae. It was experimentally shown that the use of Euglena gracilis strain for removal of phosphates in initial concentrations of 4, 7, and 14 mg/dm3 from sewage water is the most efficient way. It makes it possible to reduce these concentrations to residual 0…0.55 mg/dm3 in four days. A 3.75…5.58 times increment of microalgae biomass during this period was also shown. A mathematical model was constructed for calculating the time of staying the sewage water and microalgae mixture in a photobioreactor to achieve the required degree of removal of biogenic elements. Based on the proposed model and experimental studies, the required time of staying in the rector working area was calculated. It was shown that with the use of Euglena gracilis strain but without removal of biogenic elements at previous purification stages (process schemes including only mechanical purification), the time of mixture staying in the working zone was 37.81 hrs. With partial removal of biogenic elements at the stage of biological treatment (0.55 mg/dm3 total nitrogen, 0.91 mg/dm3 ammonium nitrogen, 0.44 mg/dm3 phosphates), this time was reduced to 26.66 hrs. It was found that the use of Euglena gracilis strain instead of Chlorella vulgaris (FC-16) in the removal of phosphates results in a 2-time increase in the process efficiency and a 50 % decrease in time of mixture staying in the working zone. Recommendations for calculating geometric parameters of photobioreactors of the proposed design were given for use in the process schemes

Naturally occurring compounds isolated from the microalga Euglena gracilis, such as polysaccharide paramylon, exhibit antimicrobial, anti-viral, antitumor, and anti-inflammatory activities. Whether live E. gracilis cells and its aqueous extract accelerate burn wound healing remains to be investigated. In this study, live E. gracilis cells and its aqueous extract were mixed with chitosan-hyaluronic acid hydrogel (CS/HA) to form cell + CS/HA and extract + CS/HA, which were then smeared onto the deeply burned skin of mice. The efficacy of these mixtures in accelerating wound healing was assessed through wound size reduction measurement, histological and immunofluorescence analyses, and serum pro-inflammatory cytokine level (INF-γ, IL-1β, and IL-6) determination. The live E. gracilis cells and its aqueous extract were found to facilitate wound healing by enhancing re-epithelization and reducing fibroplasia without stimulating excessive inflammatory response. In conclusion, live E. gracilis cells and its aqueous extract can be potentially used to treat cutaneous wounds.

Carotenoids are photosynthetic pigments and hydrophobic antioxidants that are necessary for the survival of photosynthetic organisms, including the microalga Euglena gracilis. In the present study, we identified an uncharacterized gene encoding the E. gracilis β-carotene synthetic enzyme lycopene cyclase (EgLCY) and discovered a relationship between EgLCY-mediated carotenoid synthesis and the reactive oxygen species (ROS) scavenging system ascorbate-glutathione cycle. The EgLCY cDNA sequence was obtained via homology searching E. gracilis transcriptome data. An enzyme assay using Escherichia coli demonstrated that EgLCY converts lycopene to β-carotene. E. gracilis treated with EgLCY double-stranded RNA (dsRNA) produced colorless cells with hypertrophic appearance, inhibited growth, and marked decrease in carotenoid and chlorophyll content, suggesting that EgLCY is essential for the synthesis of β-carotene and downstream carotenoids, which are abundant and physiologically functional. In EgLCY dsRNA-treated cells, the ascorbate-glutathione cycle, composed of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDAR), and glutathione reductase (GR), was unusually modulated; APX and GR activities significantly decreased, whereas DHAR and MDAR activities increased. Ascorbate content was significantly increased and glutathione content significantly decreased in EgLCY dsRNA-treated cells and was correlated with their recycling enzyme activities. Fluorescent imaging demonstrated that EgLCY dsRNA-treated cells accumulated higher levels of H2O2 compared to wild-type cells. Taken together, this study revealed that EgLCY-mediated synthesis of β-carotene and downstream carotenoid species upregulates APX activity and increases glutathione pool size for H2O2 scavenging. Our study suggests a possible relationship between carotenoid synthesis and the ascorbate-glutathione cycle for ROS scavenging in E. gracilis.
