Abstract. Rubber is known in Indonesia since the Dutch colonial period. Indonesia is the second largest natural rubber producer in the world after Thailand, rubber production contributes greatly to the Indonesian economy. Propagation of rubber seedlings to date is still done by grafting is by using the eyes of buds and seed plants, one of the alternatives to meet the demand for rubber seedlings that are increasing and not dependent on the season and to produce the clonal rootstock is homogeneous by tissue culture techniques. This study aims to find out the response of callus formation of young leaf rubber plant leaves (Hevea brasiliensis Muell, Arg) on MS medium with different concentration of ZPT (BAP and NAA). This research uses Cross Sectional type of observation research, with combination of MS and ZPT media used ie MS with BAP 1.5 ppm + NAA 0,05 ppm, BAP 2 ppm + NAA 0,1 ppm, BAP 2.5 ppm + NAA 0.2 ppm. The parameters observed were the forming of callus, callus texture and callus color for 21 days. Based on the results of the study showed that MS treatment with ZPT concentration (BAP 1.5 ppm + NAA 0.05ppm) gave the best result for callus formation response with highest explanation percentage of 0,06%.Keywords. Rubber plant (Hevea brasiliensis Muell Arg) is one of the important plantation commodities in Indonesia because of its role as a source of income. It stimulates the economic growth around the rubber plantation area. The propagation of rubber is still using conventional methods like grafting. The technique of tissue culture through callus induction is one of the alternatives of mass propagation of rubber seedling with quick and efficient time. The sterilization method is very important to determine the success of tissue culture techniques. Therefore, the aim of this research is to know the best method of sterilization and callus formation in rubber explants. The basic media used were WPM and MS, with BAP of 2 ppm and NAA 0.1 ppm. The best result of sterilization is by soaking 5% fungicide solution for 5 minutes, 5% Clorox solution for 15 minutes, betadin 10% solution for 5 minutes, and finally it rinsed with sterile water three times. The best medium uses WPM medium for callus induction, with 0.5 cm callus length and embryonic callus. In contrast, the MS medium has 0.4 cm callus length and non embryonic callus.

Garlic (Allium sativum L.) belongs to the Alliaceae family and originated from Asia and Mediterranean countries. Their bulblets are rich in starch and aromatic substances. The rate of garlic propagation in field conditions takes several years for the production of a certain number of seed bulbs for the release of a new variety. The use of tissue culture techniques is a useful tool for overcoming this problem. The aim of this work was to increase the mean number of shoots derived from the meristem isolation and to verify the percentage of callus formation and to analyze vigor of the material. The initial meristems were inoculated in a salt and vitamin B5 media except for the iron element, which was provided by MS medium added to in mg·L-1: myo-inositol (100.0), nicotinic acid (1.0), piridoxine (1.0), thiamine (10.0), sucrose (20.0 g·L-1), agar (6.0 g·L-1). BAP and TDZ were added at: 0.0; 1.0; 1.5; 2.0; and 2.5 μM This material remained in a growth room for a 16-h photoperiod, radiation of 20 μMol·m-2·s-1 and 25 °C for 40 days. Although `Sao Marcos' produced more vigorous shoots, no significant difference was found for the mean number of shoots. `Sao Valentim' cultivar shows more callus at the shoot base, making this cultivar more prone to somaclonal variation On the other hand, BAP estimulates the appearance of callus, but it has been shown that this is cultivar-dependent.

Plant tissue culture techniques were exploited for the micropropigation of Lisianthus (Eustoma grandiflora). Different concentrations of Benzyl adenine (BA), 6- Furfural amino purine (Kinetin), Indol butyric acid (IBA), were investigated in their effects at different micropropagation stages. Three explants (apical shoots, internodes, leaf discs) were used in this study. The effect of the interaction between BA and IBA on shoot multiplication was investigated in increasing the number of shoots on explants. Rooting was also studied after inclusion of IBA and NAA to Murashige and Skoog, 1962 culture medium (MS). During acclimatization stage, different ratios of river sand and peat moss as agricultural media were tested and plantlets survival was recorded. Maximum mean shoot number was recorded when leaf discs were cultured on MS medium supplemented with 0.3 mg/l of BA giving 6.8 shoots. Highest mean shoot length was obtained when BA at 0.1 mg/l was added to the culture medium giving 1.16 cm. Internodes achieved maximum mean shoot length reached 1.6 cm in the medium supplemented with 0.1 mg/l of BA, while the same concentration of KIN extended the mean shoot length up to 1.6 cm. Shoots that transferred to MS medium containing 1.0 mg/l of IBA achieved the highest root number (5.5) with mean length of 2.5 cm and achieved 70% rooting percentage. Plantlets were acclimatized achieving 90% survival when transferred to agricultural medium consisted of rivers and peat moss at 2:1 v/v ratio.

Epimedium elatum (Morren & Decne) of family Berberidaceace is a rare perennial medicinal plant, endemic to high altitude forests of Northwestern Himalayas in India. Ethnobotanically, it has been used as an ingredient for treatment of bone-joint disorders, impotence and kidney disorders in Kashmir Himalayas. Phytochemically, it is rich in Epimedin ABC and Icariin; all of these have been demonstrated to possess remarkable biological activities like PDE-5 inhibition (treatment of erectile dysfunction), anticancer, antiosteoporosis antioxidant and antiviral properties. The present investigation reports its traditional usage, comprehensive distribution and conservation status from twenty ecogeographical regions in Kashmir Himalayas, India. The species was reported from Gurez valley for the first time. Numerous threats like excessive grazing, deforestration, habitat fragmentation, tourism encroachment, landslides and excessive exploitation have decreased its natural populations in most of the surveyed habitats. Consequently, its existence may become threatened in near future if timely conservation steps are not taken immediately by concerned stakeholders involved in medicinal plant research. Moreover, use of plant tissue culture techniques is recommended for development of its in vitro propagation protocols. Therefore, introduction of this medicinal plant in botanical gardens, protected sites and development of monitoring programmes are needed for its immediate conservation in Northwestern Himalayas, India.

Chapter 3 analyzes English claims to a central role in a global network of indigenous and English people connected by faith around the world, claims made manifest in Of the Conversion of Five Thousand Nine Hundred Indians on the Island of Formosa, a 1650 publication by Baptist minister Henry Jessey, printed by radical bookseller Hannah Allen. It reports on Dutch missions in Taiwan, comparing them with evangelism efforts in New England. The coda considers the experiences of an Algonquian woman who is unnamed in Jessey’s tract but is identified as a basket maker, speculating on the meaning she may have encoded in her basket designs. Though we cannot “read” them directly, the fact that she made them, coupled with the provocative arguments offered by recent scholars about Native material culture in the colonial period, enables us to reconsider the print archive in which she appears.

The transformation that COVID-19 has brought upon the world is unparalleled. The impact on mental health is equally unprecedented and yet unexplored in depth. An online-based survey was administered to 413 community-based adults during COVID-19 confinement to explore psychological impact and identify high risk profiles. Young females concerned about the future, expressing high COVID-related distress, already following psychological therapy and suffering from pre-existing chronic conditions, were those at highest risk of psychological impact due to the COVID-19 situation. Findings could be employed to design tailored psychological interventions in the early stages of the outbreak to avoid the onset/exacerbation of psychopathology.
