Целью исследований было проведение серологического мониторинга инфекционного бронхита кур (ИБК), анализ технологических схем вакцинаций и изучение напряжённости гуморального иммунитета к вирусу ИБК. Исследования проводились в Республике Крым в течение 20172018 гг. Материалом для исследований служили образцы сывороток крови от кур разного возраста (молодняк, взрослая птица), поступивших из двух ведущих птицеводческих предприятий Крыма. Определение антител к вирусу ИБК проводили методом иммуноферментного анализа (ИФА). Всего было исследовано 1324 образца сывороток крови от птицы разного возраста. Поствакцинальные антитела обнаружены в 1317 пробах (99,4). Установлено, что применение в обследуемых хозяйствах для профилактики ИБК живых и инактивированных одно и многовалентных вакцин отечественного и импортного производства в соответствии с действующим регламентом вакцинопрофилактики и разработанными в хозяйствах технологическими схемами вакцинации создаёт полноценную специфическую защиту у вакцинированного поголовья на протяжении всего периода продуктивности. Отмечено, что постепенное нарастание средних титров антител у птиц каждой следующей возрастной группы с пиком в 140 и 150дневном возрасте соотносится с классической схемой формирования гуморального иммунитета при применении живых и инактивированных вакцин. В дальнейшем у кур старше 300суточного возраста появляются достаточно пёстрые титры антител к ИБК, что может свидетельствовать о возможной циркуляции полевого вируса ИБК. В связи с этим имеется необходимость систематического мониторинга уровня антител к вирусу ИБК, что позволит и дальше контролировать эпизоотическую ситуацию к данному заболеванию в птицеводческих хозяйствах Крыма.The aim of the research was to conduct serological monitoring of avian infectious bronchitis (AIB), the analysis of technological schemes of vaccinations and the study of the intensity of humoral immune response to the AIB virus. The researches were carried out in the Republic of Crimea during 20172018. The material for the researches were samples of blood serum from chickens of different ages (young birds, mature birds) received from two leading poultry farms of the Crimea. Determination of antibodies to AIB virus was conducted by enzyme multipled immunoassay (ELISA test). A total of 1324 blood serum samples from birds of different ages were examined. Vaccineinduced antibodies were found in 1317 samples (99.4). It has been established that the use of live and inactivate monovalent and multivalent vaccines of domestic and foreign production in the surveyed farms for the prevention of AIB in accordance with the current vaccinal prevention schedule and the vaccination technological schemes developed on the farms creates full specific protection for the vaccinated livestock throughout the entire production period. It is noted that a gradual increase in the average antibody titers in birds of each subsequent age group with a peak at 140 and 150 days of age correlates with the classical scheme for the formation of antibody mediated immunity with the use of live and inactivate vaccines. Further chickens older than 300 days of age appear to have rather variegated antibody titers to AIB which may indicate a possible circulation of the field AIB virus. In this regard there is a need for systematic monitoring of the level of antibodies to the AIB virus which will allow further control of the epizootic situation to this disease in the poultry farms of the Crimea.

Infection of pigs caused by porcine circovirus type 2 (PCV2) is present throughout the world. The aim of this study was to determine, based on the follow-up of the class IgG titers in piglets and fattening pigs, the duration of colostral immunity against infections in pigs caused by PCV2. The study included 28 piglets, whose titer was determined by using indirect ELISA test in intervals up to 110 days of age. The observed average values of IgG in piglets aged 21 days (8.47 log2) and 35 days (6.69 log2) in our surveys indicate very high titers of maternal antibodies, and that the piglets at this age are, to some extent, protected from infection caused by PCV2. Absence of the specific anti PCV2 antibodies was determined in 14.29% (21 days old) and 15.38% (35 days old) piglets at this age. The average titer of antibodies, specific for PCV2, in blood serum of piglets on the 50th day decreased to 3.74 log2, what indicates that in the majority of piglets (61.54%) catabolism of colostral antibodies occurred. On the 80th day of the piglets life there were no seropositive specimens, which unambiguously indicates that in this age, there was a complete catabolism of colostral antibodies and the disappearance of passive immunity. In all the 110 days? old fattening pigs, an average antibody titer recorded a sharp rise (12.78 log2). Possible reasons for this sudden increase in PCV2 antibody titers in the blood serum of fattening pigs is is a widespread presence of this infection in our region, as well as the fact that there exists a part of pigs population which is not immune to PCV2 infection. The results of this research are important for choosing the optimal moment for vaccination, considering that high titers of colostral antibodies of class G on the 21st and 35th day of piglets? life have been proven. This data points out to a possible interference with vaccine immunogens in the case of vaccination in this particular age.

ABSTRACTBrucella abortus, a smooth strain of the genusBrucella, is the causative agent of bovine brucellosis. To support the ongoing development of diagnostic tests for bovine brucellosis, the use of Protein Saver cards (Whatman) for bovine blood serum and plasma sample collection has been evaluated. These cards offer significant logistical and safety alternatives to transporting and storing liquid samples and may aid in diagnostic programs and validation studies. To evaluate the utility of these cards, 204 bovine blood serum samples fromBrucella-infected and noninfected animals were stored on and eluted from the Protein Saver cards. Anti-Brucellasmooth lipopolysaccharide (sLPS) antibody titers for the serum eluates were compared to those of the unprocessed original serum samples by indirect enzyme-linked immunosorbent assay (ELISA). The results showed a highly significant correlation between titers from the serum eluates and the unprocessed sera. Therefore, under these circumstances, serum eluates and unprocessed serum samples may be used interchangeably. Blood plasma from 113 mitogen-stimulated whole-blood samples was added to and eluted from the Protein Saver cards. The gamma interferon (IFN-γ) titers in the plasma eluates were compared to those of the unprocessed plasma samples obtained by IFN-γ ELISA. The results showed a significant correlation between the plasma eluates and the unprocessed plasma samples. To derive a signal in the plasma eluate, it was necessary to develop a novel and highly sensitive ELISA for the detection of IFN-γ. The serum samples stored on cards at room temperature over a 10-day period showed little variation in antibody titers. However, the plasma eluates showed a progressive loss of IFN-γ recovery over 10 days when stored at room temperature.

Background and Aim: Puerperal diseases influence fertility and should be diagnosed as soon as possible. This study aimed to evaluate the applicability of serum concentrations of substance P (SP), vasoactive intestinal polypeptide (VIP), and interleukin (IL)1β in the early diagnosis of uterine involution disturbances. Materials and Methods: Blood serum samples of 86 dairy cows from six different farms were harvested within the first 20 days after calving from cows with uterine involution disturbances and healthy controls, respectively. Serum concentrations for SP, VIP, and IL-1β were determined using commercially available ELISA test kits. Statistical analyses included timely changes in blood serum levels and group comparisons of healthy cows and cows with uterine disease. Results: SP concentrations increased significantly within 20 days after calving (p<0.04) with no significant difference observed between the groups. Moreover, no significant differences were found between VIP and log IL-1β. Conclusion: Results showed that none of the examined serum parameters seems suitable as indicator of uterine involution disorders. Due to the timely changes in serum concentrations of SP after calving, a correlation to diseases might not be precluded. Further research is needed as regards the establishment of normative values concerning this parameter.

The aim of the study was to analyze the occurrence of caprine arthritis encephalitis virus (CAEV) in white improved goats, colored improved goats, Saanen goats, Alpine goats and Boer goats, raised in three regions of Poland. The analysis of CAEV infection levels was carried out in goat herds from the provinces in mid-Eastern Poland. The analysis included four goat herds. In total, they encompassed 251 herds of the following breeds: white improved goat, colored improved goat, Saanen goat, Alpine goat and Boer goat. The identification of the infected animals and the assessment of the epidemiological condition of the herds was carried out using serological analysis of blood serum samples. The assay of CAEV-specific antibodies in blood serum was performed using ELISA test. As a result of the observations it was established that with regards to the provinces, the prevalence level ranged from 35.40% in Mazowieckie province to 81.10% in Lubelskie province, and the average infection rate of animals in the studied region was 51.60%. As far as the breeds are concerned, it was found that the observed infection rate of the Saanen goat population was significantly higher (p ≤ 0.05) than the rates for improved white goat (48.90%) and Alpine goat (50.00%). In relation to the colored improved goat, the calculated statistical difference was greater (p ≤ 0.01), and the percentage of infected animals was 32.10%. The analysis of the infection rate in the Boer goat herd revealed that 66.70% of animals are seropositive, and the statistical analysis did not show any significant differences in prevalence compared to other studied breeds. To sum up, it should be noted that the improvement of the situation may be attributed to consistent examination of goats and development of CAEV-elimination programs for herds. When creating programs aimed at the elimination of the virus it is also important to remember to raise the awareness of the farmers with regards to the detrimental effect of CAEV infections..

This study includes examination of dogs from the territory of Serbia during the period from 2004. To 2011. Most of the dogs were from the territory of Belgrade. The total of 193 blood serum samples of proprietary dogs and 120 blood serum samples of stray dogs were tested for the presence of antibodies against Brucella canis. For diagnostics there was used the method of slow serum agglutination in test tubes with 2- mercaptoethanol. During eight years? period of investigation, out of 193 tested serums taken from proprietary dogs, 29 serum samples, or 15.03%, had undoubtedly positive titre of 1/200. During 2011. Out of 120 tested blood samples taken from stray dogs from Belgrade territory, positive titre of 1/200 had 8 samples, or 6.67%. The results of this investigation point out to a very high seroprevalence of antibodies against B.canis in dogs population from the teritiry of the Republic of Serbia.

 Introduction. In connection with the Ebola epidemic in the West African countries, including the Republic of Guinea, a failure in implementing measles immunization program was noted. A proportion of measles seronegative subjects in 2017 was 52.4% of total examined individuals. In 2018, a high proportion of measles cases among children aged 1–5 years (61.6%) was identified. In order to stop the 2018 outbreak, the Supplemental Immunization Campaign was conducted in the Konakri and Nzerekore prefectures in the Republic of Guinea. The aim of this study was to examine the 2019–2020 measles epidemic situation and assess the measles population immunity in the Republic of Guinea. Materials and methods. Measles-specific antibodies were examined in 1697 blood serum samples collected from residents of different regions of the Republic of Guinea, aged from 7 months to 76 years, obtained in 2019–2020, and tested retrospectively. The ELISA test systems Anti-Measles Virus ELISA (IgM) Euroimmun and Anti-Measles Virus ELISA (IgG) Euroimmun (Germany) were used. The presence of serum IgM measles antibodies was considered as acute measles infection. Statistical analysis was performed using the software package Statistica 6.0. Results. Blood sera (n = 638) were tested for IgM-measles, and in 46.6% of cases the diagnosis was confirmed by laboratory tests. The biggest proportion of the total cases (61.6%) was found in children aged 1–4 years. The second most important age group was 5–9 years of age, the third is children under 1 year: 18.5% and 11.8% of the total number of patients, respectively. Measles infection was registered in vaccinated patients in 7.4% of the total number of laboratory-confirmed cases. 1059 subjects were examined for IgG measles antibody. The lowest seroprevalence rate was found among children under 4 years of age (47.8%). The highest (85.5%) was found among subjects of 40 years old and older. Conclusion. Measles in GR remains a poorly controlled infection. As in the previous years of observation (2017–2018), children under 5 years of age are the most vulnerable cohort of the population, despite the 2018 DI campaign conducted in a number of GR territories. More problems with the measles control in the Republic of Guinea are expected in the period from 2021, as along with the COVID-19 epidemic, Ebola is repeatedly registered in the country. The Republic of Guinea particularly requires assistance from the international community to implement the WHO measles elimination program on a global scale.

The sera from 120 boars from 3 farrow-to-finish swine herds in Serbia were examined for Salmonella spp. antibodies in a cross-sectional study using an ELISA test. A total of 120 blood serum samples from boars imported from Denmark were examined. All boars were seronegative to Salmonella during the import, while after one year Salmonella spp. seroprevalence ranging from 0% to 45% was found in 3 herds using two tests in the time span of one year. Only on one farm during the import in 2011, after a year boars were still seronegative. A year later on the same farm, in boars imported in 2012 the seroprevalence was 30%. Seroprevalence of Salmonella spp. in boars tested on all farms was 24,17%.

Abstract The transplacental transmission is the primary route of Neospora caninum infection in bovine herds around the world. This study aimed to determine the frequency of transplacental transmission of the parasite in dairy cattle of Agreste region of Pernambuco through serological tests (IFAT and ELISA). Three hundred sixteen serum samples from cows and heifers and their offspring were analyzed. The transplacental transmission rate was 72.22% (13/18) for cows and 69.23% (9/13) for heifers by IFAT. ELISA test showed transplacental transmission rate of 43.58% (17/39) for cows and 50% (9/18) for heifers. The transplacental transmission rates were similar, in both groups in test, but a higher seropositivity was found in cows by IFAT. Data were statistically analyzed using the chi-square and Fisher’s exact test. A significant relationship of dependence between seropositivity of mothers and their offspring was found. The more frequent IFAT antibody titers and ELISA levels for N. caninum were, respectively, 200 and between four (cows) and five (heifers and offspring). In the Spearman correlation, no association was found between the magnitude of antibody titers for N. caninum between mothers and their offspring. The kappa test showed an index of 0.35, indicating a mild correlation between the serological tests used. The study suggests that cows and heifers are the main transmitters of N. caninum in the studied region and that vertical transmission is the major form of transmission in dairy herds of the Agreste region of Pernambuco.

Aim. To study the level of anti-diphtheria antitoxic antibodies in blood serum of vaccinated individuals over the period of 2015-2017. Materials and Methods. The study was conducted on the base of the Center of Hygiene and Epidemiology of the Ryazan region. The evaluation of antitoxic immunity against diphtheria was performed on the basis of the results of annual serological examination of at least 100 samples of blood sera for the content of diphtheria antitoxin in each age indicator group. In the study reaction of passive hem agglutination was used with standardized erythrocyte diphtheria antigen. Blood serum samples of individuals of indicator groups of the population with a documented history of vaccination were investigated. On the basis of the results of serological monitoring, the analysis of the state of anti-diphtheria antitoxic immunity in children (3-4 years, n=300), teenagers (16-17 years, n=302) and adults (30-39 years, n=323; 40-49 years, n=305) of the Ryazan region in 2015-2017 was carried out. Results. The level of protection among children, teenagers and adults in the studied period was 97.0 and 95.6%. The intensity of anti-diphtheria immunity was also high: 93.3-95.6% of the examined persons had medium and high antibody titers. Protective level of anti-diphtheria antibodies in 2015 was revealed in 97.3%, in 2016 in 93.3% and in 2017 in 98.3% of the examined samples. Conclusion. Analysis of the results of serological monitoring showed a positive dynamics of growth of the number of seropositive individuals with intense anti-diphtheria immunity. The data of serological monitoring correspond to the official statistical data of Directorate of Rospotrebnadzor in the Ryazan region on the high level of coverage of the population with vaccination. According to previously published epidemiological studies, in such conditions the incidence of diphtheria in the population is sporadic.

AbstractBackgroundThe approach to diagnosing, treating and monitoring severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection relies strongly on laboratory resources, with serological testing representing the mainstay for studying the onset, nature and persistence of humoral immune response. This study was aimed at evaluating the analytical performance of the novel Beckman Coulter anti-SARS-CoV-2 IgG chemiluminescent immunoassay.MethodsThis analytical assessment encompassed the calculation of intra-assay, inter-assay and total imprecision, linearity, limit of blank (LOB), limit of detection (LOD), functional sensitivity, and comparison of anti-SARS-CoV-2 antibodies values obtained on paired serum samples using DiaSorin Liaison SARS-CoV-2 S1/S2 IgG and Roche Elecsys Anti-SARS-CoV-2 total antibodies. Diagnostic performance was also tested against results of molecular testing on nasopharyngeal swabs, collected over the previous 4 months.ResultsIntra-assay, inter-assay and total imprecision of Beckman Coulter anti-SARS-CoV-2 IgG were between 4.3-4.8%, 2.3-3.9% and 4.9-6.2%, respectively. The linearity of the assay was excellent between 0.11-18.8 antibody titers. The LOB, LOD and functional sensitivity were 0.02, 0.02 and 0.05, respectively. The diagnostic accuracy (area under the curve; AUC) of Beckman Coulter anti-SARS-CoV-2 IgG compared to molecular testing was 0.87 (95% CI, 0.84-0.91; p<0.001) using manufacturer’s cut-off, and increased to 0.90 (95% CI, 0.86-0.94; p<0.001) with antibody titers. The AUC was non-significantly different from that of Roche Elecsys Anti-SARS-CoV-2, but was always higher than that of DiaSorin Liaison SARS-CoV-2 S1/S2 IgG. The correlation of Beckman Coulter Access SARS-CoV-2 IgG was 0.80 (95% CI, 0.75-0.84; p<0.001) with Roche Elecsys Anti-SARS-CoV-2 and 0.72 (95% CI, 0.66-0.77; p<0.001) with DiaSorin Liaison SARS-CoV-2 S1/S2 IgG, respectively.ConclusionsThe results of this analytical evaluation of Beckman Coulter Access anti-SARS-CoV-2 IgG suggests that this fully-automated chemiluminescent immunoassay represents a valuable resource for large and accurate seroprevalence surveys.
