Antibiotic-Resistant Bacteria and Antibiotic Residue Contamination in Fresh Raw Foods Sold at Wholesale Markets in Thailand

Objective: To investigate the extent of antibiotic-resistant bacteria and antibiotic residue contamination in fresh raw foods sold at wholesale markets in Thailand, which may be the important drivers of antibiotic-resistant bacteria colonization and antibiotic-resistant bacterial infection in Thai population. Materials and Methods: Fresh raw foods, including food from animal products, seafoods, vegetables, fruits, and honey were purchased from two large wholesale markets in Thailand. Food samples were cultured for antibiotic-resistant bacteria and tested for the presence and amount of antibiotic residue. Results: Among 521 samples for bacterial culture, 86.9% grew at least one kind of bacteria. Enterobacteriaceae were commonly isolated and were commonly resistant to ampicillin (76.7% to 100%). ESBL-producers and ceftriaxone-resistant Enterobacteriaceae were prevalent in swine and duck samples (56.7% to 91.7%). Some isolates were resistant to co-amoxiclav (13.3% to 60.0%) and cefoxitin (5.0% to 30.0%). Colistin-resistant Enterobacteriaceae were observed in pork meat (1.4%) and chicken offal (7.0%). Ertapenem-resistant Enterobacteriaceae were detected in cha-om (26.7%). Among 501 samples for antibiotic residue testing, 37.1% contained at least one antibiotic residue. Enrofloxacin was the most prevalent antibiotic residue, followed by doxycycline and tilmicosin. Although most samples contained less antibiotics than the maximum residue limit (MRL), 7.0% contained an amount of at least one antibiotic above the MRL. Conclusion: Many fresh raw foods sold at wholesale markets in Thailand were contaminated with antibiotic-resistant bacteria, and some contained antibiotic residues. Therefore, Thai people are at risk of being colonized with antibiotic-resistant bacteria and developing antibiotic-resistant bacterial infection due to consuming foods contaminated with antibiotic-resistant bacteria or containing antibiotic residues. Antibiotic resistance of microorganisms is a serious health problem for both humans and animals. Infection of these bacteria may result in therapy failure, leading to high mortality rates. During an early intervention program process, the Sea Turtle Conservation Center of Thailand (STCCT) has faced high mortality rates due to bacterial infection. Previously, investigation of juvenile turtle carcasses found etiological agents in tissue lesions. Further determination of sea water in the turtle holding tanks revealed a prevalence of these causative agents in water samples, implying association of bacterial isolates in rearing water and infection in captive turtles. In this study, we examined the antibiotic resistance of bacteria in seawater from the turtle holding tank for a management plan of juvenile turtles with bacterial infection. The examination was carried out in three periods: 2015 to 2016, 2018, and 2019. The highest isolate numbers were resistant to beta-lactam, whilst low aminoglycoside resistance rates were observed. No gentamicin-resistant isolate was detected. Seventy-nine isolates (71.17%) were resistant to at least one antibiotic. Consideration of resistant bacterial and antibiotic numbers over three sampling periods indicated increased risk of antibiotic-resistant bacteria to sea turtle health. Essentially, this study emphasizes the importance of antibiotic-resistant bacterial assessment in rearing seawater for sea turtle husbandry.

In the last 20 years, changes in world technology have occurred which have allowed for the rapid transport of people, food, and goods. Unfortunately, antibiotic residues and antibiotic-resistant bacteria have been transported as well. Over the past 20 years, the rise in antibiotic-resistant gene carriage in virtually every species of bacteria, not just oral/respiratory bacteria, has been documented. In this review, the main mechanisms of resistance to the important antibiotics used for treatment of disease caused by oral/respiratory bacteria-including β-lactams, tetracycline, and metronidazole-are discussed in detail. Mechanisms of resistance for macrolides, lincosamides, streptogramins, trimethoprim, sulfonamides, aminoglycosides, and chloramphenicol are also discussed, along with the possible role that mercury resistance may play in the bacterial ecology.

AbstractSlaughterhouse wastewater is considered a reservoir for antibiotic-resistant bacteria and antibiotic residues, which are not sufficiently removed by conventional treatment processes. This study focuses on the occurrence of ESKAPE bacteria (Enterococcus spp., S. aureus, K. pneumoniae, A. baumannii, P. aeruginosa, Enterobacter spp.), ESBL (extended-spectrum β-lactamase)-producing E. coli, antibiotic resistance genes (ARGs) and antibiotic residues in wastewater from a poultry slaughterhouse. The efficacy of conventional and advanced treatments (i.e., ozonation) of the in-house wastewater treatment plant regarding their removal was also evaluated. Target culturable bacteria were detected only in the influent and effluent after conventional treatment. High abundances of genes (e.g., blaTEM, blaCTX-M-15, blaCTX-M-32, blaOXA-48, blaCMY and mcr-1) of up to 1.48 × 106 copies/100 mL were detected in raw influent. All of them were already significantly reduced by 1–4.2 log units after conventional treatment. Following ozonation, mcr-1 and blaCTX-M-32 were further reduced below the limit of detection. Antibiotic residues were detected in 55.6% (n = 10/18) of the wastewater samples. Despite the significant reduction through conventional and advanced treatments, effluents still exhibited high concentrations of some ARGs (e.g., sul1, ermB and blaOXA-48), ranging from 1.75 × 102 to 3.44 × 103 copies/100 mL. Thus, a combination of oxidative, adsorptive and membrane-based technologies should be considered.

Pig manure may contain antibiotic residues, antibiotic-resistant bacteria or pathogens, which may reach the environment upon fertilization. During this study, 69 antibiotic residues belonging to 12 classes were quantified in 89 pig slurry samples. These samples were also studied for the presence of Salmonella and for E. coli resistant to meropenem, colistin, ciprofloxacin, or cefotaxim. The obtained isolates were further tested for antibacterial susceptibility. No antibiotic residues were detected in four samples, whereas in the other samples, up to 12 antibiotics were found. The most frequently detected antibiotic residues were doxycycline, sulfadiazine, and lincomycin. Doxycycline was found in the highest concentration with a mean of 1476 µg/kg manure (range: 18–13632 µg/kg). Tylosin and oxytetracycline were found with mean concentrations of 784 µg/kg (range: 17–5599 µg/kg) and 482 µg/kg (range: 11–3865 µg/kg), respectively. Lincomycin, had a mean concentration of 177 µg/kg manure (range: 9–3154 µg/kg). All other 18 antibiotic residues were found with mean concentrations of less than 100 µg/kg manure. Fifty-one slurry samples harbored Salmonella; 35% of the Salmonella isolates were sensitive to a panel of 14 antibiotics, whereas the other 65% were resistant up to five antibiotics. For E. coli, 52 manure samples contained E. coli isolates which were resistant to ciprofloxacin and 22 resistant to cefotaxime. All ciprofloxacin and cefotaxime-resistant isolates were multi-resistant, with resistance up to nine and eight antibiotics, respectively. This research indicates that pig slurry used for fertilization often contains antibiotic residues and antibiotic-resistant bacteria, including pathogens.
