Result from interview with Manado fish scale handicraft industry stakeholder states that wantex is commonly used as colorant for fish scales, resulting in rather vivid and plastic-like colors. Through a series of experiment using natural and synthetic colorants, turmeric (yellow), red yeast rice (red), and synthetic food colorant (Brilliant Blue FCF) are most applicable colorants, producing good results on fish scales.  Combinations of these three colorants produce many good secondary colors with new and mild gradations and translucency, very potential to be developed for fashion jewelry explorations which use snapper fish scale as the main material.   Key words : colorant, natural, synthetic, fish, scale, snapper   Dari hasil wawancara industri kerajinan sisik ikan Manado, wantex tekstil digunakan sebagai zat pewarna sisik ikan sehingga hasil warna yang didapat cenderung pekat dan seperti plastik. Melalui berbagai uji pewarna alami dan sintetis, kunyit (kuning), angkak (merah) dan pewarna makanan sintetis biru (Biru Brilliant FCF) merupakan pewarna yang paling mudah untuk diaplikasikan dan juga menghasilkan warna yang baik pada sisik ikan. Pencampuran ketiga warna tersebut menghasilkan banyak warna sekunder dengan gradasi dan translusensi sisik ikan yang baru dan berwarna lembut sehingga potensial digunakan untuk eksplorasi perhiasan mode berbahan dasar sisik ikan kakap. Comparative scanning electron microscopy studies of fish scales of different orders, families, genera and species within genera have demonstrated differences which warrant elaboration. These differences in detail appear to be sufficient to act as “fingerprints”, at least, for family differences. To date, the lateral line scales have been primarily studied. These demonstrate differences in the lateral line canals; the pattern of ridging with or without secondary protuberances along the edges; the pattern of spines or their absence on the anterior border of the scales; the presence or absence of single or multiple holes on the ventral and dorsal sides of the lateral line canal covers. The distances between the ridges in the pattern appear likewise to be important.A statement of fish scale structure and a comparison of family and species differences will be presented.The authors wish to thank Dr. Donald Marzalek and Mr. Wallace Charm of the Marine and Atmospheric Laboratory of the University of Miami and Dr. Sheldon Moll and Dr. Richard Turnage of AMR for their exhaustive help in these preliminary studies.

Scanning electron microscopy of fish scales has facilitated the application of micro-structures to systematics. Electron microscopy studies have added more information on the structure of the scale and the associated cells, many problems still remain unsolved, because of our incomplete knowledge of the process of calcification. One of the main purposes of these studies has been to study the histology, histochemistry, and ultrastructure of both calcified and decalcified scales, and associated cells, and to obtain more information on the mechanism of calcification in the scales. The study of a calcified scale with the electron microscope is complicated by the difficulty in sectioning this material because of the close association of very hard tissue with very soft tissues. Sections often shatter and blemishes are difficult to avoid. Therefore the aim of this study is firstly to develop techniques for the preparation of cross sections of fish scales for scanning electron microscopy and secondly the application of these techniques for the determination of the structures and calcification of fish scales.

Background: Oxysterol relationship with cardiovascular (CV) risk factors is poorly explored, especially in moderately hypercholesterolaemic subjects. Moreover, the impact of nutraceuticals controlling hypercholesterolaemia on plasma levels of 24-, 25- and 27-hydroxycholesterol (24-OHC, 25-OHC, 27-OHC) is unknown. Methods: Subjects (n = 33; 18–70 years) with moderate hypercholesterolaemia (low-density lipoprotein cholesterol (LDL-C:): 130–200 mg/dL), in primary CV prevention as well as low CV risk were studied cross-sectionally. Moreover, they were evaluated after treatment with a nutraceutical combination (Bifidobacterium longum BB536, red yeast rice extract (10 mg/dose monacolin K)), following a double-blind, randomized, placebo-controlled design. We evaluated 24-OHC, 25-OHC and 27-OHC levels by gas chromatography/mass spectrometry analysis. Results: 24-OHC and 25-OHC were significantly correlated, 24-OHC was correlated with apoB. 27-OHC and 27-OHC/total cholesterol (TC) were higher in men (median 209 ng/mL and 77 ng/mg, respectively) vs. women (median 168 ng/mL and 56 ng/mg, respectively); 27-OHC/TC was significantly correlated with abdominal circumference, visceral fat and, negatively, with high-density lipoprotein cholesterol (HDL-C). Triglycerides were significantly correlated with 24-OHC, 25-OHC and 27-OHC and with 24-OHC/TC and 25-OHC/TC. After intervention, 27-OHC levels were significantly reduced by 10.4% in the nutraceutical group Levels of 24-OHC, 24-OHC/TC, 25-OHC, 25-OHC/TC and 27-OHC/TC were unchanged. Conclusions: In this study, conducted in moderate hypercholesterolemic subjects, we observed novel relationships between 24-OHC, 25-OHC and 27-OHC and CV risk biomarkers. In addition, no adverse changes of OHC levels upon nutraceutical treatment were found.
