The use of insects as a feasible and useful natural product resource is a novel and promising option in alternative medicine. Several components from insects and their larvae have been found to inhibit molecular pathways in different stages of cancer. This study aimed to analyze the effect of aqueous and alcoholic extracts of Vespa orientalis larvae on breast cancer MCF7 cells and investigate the underlying mechanisms. Our results showed that individual treatment with 5% aqueous or alcoholic larval extract inhibited MCF7 proliferation but had no cytotoxic effect on normal Vero cells. The anticancer effect was mediated through (1) induction of apoptosis, as indicated by increased expression of apoptotic genes (Bax, caspase3, and p53) and decreased expression of the anti-apoptotic gene Bcl2; (2) suppression of intracellular reactive oxygen species; (3) elevation of antioxidant enzymes (CAT, SOD, and GPx) and upregulation of the antioxidant regulator Nrf2 and its downstream target HO-1; (4) inhibition of migration as revealed by in vitro wound healing assay and downregulation of the migration-related gene MMP9 and upregulation of the anti-migratory gene TIMP1; and (5) downregulation of inflammation-related genes (NFκB and IL8). The aqueous extract exhibited the best anticancer effect with higher antioxidant activities but lower anti-inflammatory properties than the alcoholic extract. HPLC analysis revealed the presence of several flavonoids and phenolic compounds with highest concentrations for resveratrol and naringenin in aqueous extract and rosmarinic acid in alcoholic extract. This is the first report to explain the intracellular pathway by which flavonoids and phenolic compounds-rich extracts of Vespa orientalis larvae could induce MCF7 cell viability loss through the initiation of apoptosis, activation of antioxidants, and inhibition of migration and inflammation. Therefore, these extracts could be used as adjuvants for anticancer drugs and as antioxidant and anti-inflammatory agents.

The current study accentuates the significance of performing the multiplex approach of LC-HRESIMS, biological activity, and docking studies in drug discovery, taking into consideration a review of the literature. In this regard, the investigation of antioxidant and cytotoxic activities of Trigonella stellata collected from the Egyptian desert revealed a significant antioxidant capacity using DPPH with IC50 = 656.9 µg/mL and a moderate cytotoxicity against HepG2, MCF7, and CACO2, with IC50 values of 53.3, 48.3, and 55.8 µg/mL, respectively. The evaluation of total phenolic and flavonoid contents resulted in 32.8 mg GAE/g calculated as gallic acid equivalent and 5.6 mg RE/g calculated as rutin equivalent, respectively. Chemical profiling of T. stellata extract, using LC-HRESIMS analysis, revealed the presence of 15 metabolites, among which eleven compounds were detected for the first time in this species. Interestingly, in vitro testing of the antidiabetic activity of the alcoholic extract noted an α-glucosidase enzyme inhibitory activity (IC50 = 559.4 µg/mL) better than that of the standard Acarbose (IC50 = 799.9 µg/mL), in addition to a moderate inhibition of the α-amylase enzyme (IC50 = 0.77 µg/mL) compared to Acarbose (IC50 = 0.21 µg/mL). α-Glucosidase inhibition was also virtualized by binding interactions through the molecular docking study, presenting a high binding activity of six flavonoid glycosides, as well as the diterpenoid compound graecumoside A and the alkaloid fenugreekine. Taken together, the conglomeration of LC-HRESIMS, antidiabetic activity, and molecular docking studies shed light on T. stellata as a promising antidiabetic herb.

Objective: To screen α-amylase and α-glucosidase inhibitors from the different fractions of crude hydro-alcoholic extract of heartwood of Tecomella undulata Linn.Methods: Four fractions of crude hydro-alcoholic extract of heartwood of plant were used for in-vitro inhibitory assays against digestive enzymes: α-amylase and α-glucosidase. For assay, different concentrations (20, 40, 60, 80, 100 µg/ml) were used for all fractions. Standard protocol was used for preliminary phytochemical screening of different bioactive components present in all fractions.Results: The fractions have shown moderate to highest inhibitory activity against both enzymes. But, the strong inhibition was revealed by acetone fraction against α-amylase with very minimal inhibitory concentrations at IC50 values when compared with a standard drug acarbose. Several medicinally active phytocomponents such as flavanoids, saponin, anthraquinones, tannins, triterpenoids and phenols were observed in all studied fractions.Conclusion: The different fractions prepared from crude hydro-alcoholic extract of heartwood of plant are capable of inhibiting α-amylase and α-glucosidase and it can be concluded that heartwood of Tecomella undulata Linn. is partially active against postprandial hyperglycemia, thus diabetes mellitus.Keywords: Tecomella undulata Linn., Diabetes mellitus, α-Amylase, α-Glucosidase.

Objective: The important restraints in plant-derived medicine are the convenience of active composites which depends on diverse topographical situations, active compound build-up pattern, environmental circumstances and genetic makeup of the plant. The presented work was aimed to describe an efficient technique for fast propagation of Nothapodytes nimmoniana which is an imperative source of Camptothecin.Methods: Multiple shoots were successfully regenerated from the explants N. nimmoniana using Murashige and Skoog (MS) medium accompanied with IBA+KN+GA3 (1:3:2) mg/l. the hydro-alcoholic extract was obtained by soxhlet extraction for field grown roots (fgRE), shoots (fgSE) and in vitro plantlets (ivPL) and further subjected to phytochemicals analysis. HPLC analysis was performed to determine the percentage of camptothecin (CAM). Anti-proliferative studies were carried out followed by Acridine orange/Ethidium bromide staining. Extracts were also estimated for reactive nitrogen species (RNS) and reactive oxygen species (ROS).Results: Preliminary qualitative chemical analysis of the extracts displayed the presence of alkaloids, carbohydrates, steroids, saponins, terpenoids and phenolics compounds. HPLC analysis revealed the presence of CAM in all the extracts. ivPL exhibited lowest anti-proliferative activity in contrast to other extract and percentage of ROS and RNS was also less. Although ivPL did not exhibited prominent anti-tumour effect, but in vitro accumulation of CAM in 2-months old shoots provides an alternative resource to 5 y-old field grown plants.Conclusion: This work delivers a potential to lead to further increase the potency of CAM in in vitro shoots of two months old as an alternative to the field grown plants for treating cancers.

Objective: The objective of the study was to establish the anti-inflammatory and anti-arthritic potential of various fractions of Eclipta prostrata Linn. Methods: The four fractions, i.e., n-butanol, ethyl acetate, chloroform, and n-hexane from hydro-alcoholic extract were obtained. First, the fractions were evaluated through in vitro models, and then they were evaluated by in vivo anti-inflammatory model, i.e., carrageenan-induced paw edema model. Further, two active fractions were evaluated for the anti-arthritic activity using formaldehyde induced arthritis model. Results: The fractions at a dose of 100 and 200 mg/kg showed an anti-inflammatory activity, but the ethyl acetate and chloroform fraction will show maximum anti-inflammatory potential. Hence, they are further evaluated for anti-arthritic potential where they show significant activity. Conclusion: From the results, it is concluded that the ethyl acetate and chloroform fraction show significant anti-arthritic activity.

Pterygium is a relatively common eye disease that can display an aggressive clinical behaviour. To evaluate the in vitro effects of Curcuma longa on human pterygium-derived keratinocytes, specimens of pterygium from 20 patients undergoing pterygium surgical excision were collected. Pterygium explants were put into culture and derived keratinocytes were treated with an alcoholic extract of 1.3% Curcuma longa in 0.001% Benzalkonium Chloride for 3, 6, and 24 h. Cultured cells were examined for CAM5.2 (anti-cytokeratin antibody) and CD140 (anti-fibroblast transmembrane glycoprotein antibody) expression between 3th and 16th passage to assess cell homogeneity. TUNEL technique and Annexin-V/PI staining in flow cytometry were used to detect keratinocyte apoptosis. We showed that Curcuma longa exerts a proapoptotic effect on pterygium-derived keratinocytes already after 3 h treatment. Moreover, after 24 h treatment, Curcuma longa induces a significant increase in TUNEL as well as Annexin-V/PI positive cells in comparison to untreated samples. Our study confirms previous observations highlighting the expression, in pterygium keratinocytes, of nuclear VEGF and gives evidence for the first time to the expression of nuclear and cytoplasmic VEGF-R1. All in all, these findings suggest that Curcuma longa could have some therapeutic potential in the treatment and prevention of human pterygium.
