Introduction: Obesity creates health problems by increasing the risks of chronic diseases such as type 2 diabetes and cardiovascular disorders. Obesity leads to insulin resistance, higher blood glucose and cholesterol levels. Adipose tissues synthesize adiponectin which acts as anti-inflammatory, antidiabetic, and anti-atherogenic agent. Meanwhile, vitamin E is an antioxidant that acts as an anti-inflammation. Aim: The purpose of this study was to analyze the effects of vitamin E supplementation to metabolic markers on diet-induced obesity in mice. Materials and methods: Twenty-four mice (Mus musculus, L) aged four weeks were divided into six groups which were fed different diets and given vitamin E in different dosages or methods. The period of treatment was 18 weeks. The mice body weights were measured every week; blood sugar and cholesterol levels were measured every six weeks, and the adiponectin level measurement was done at week 18. Results: A repeated measures ANOVA showed that body weight and cholesterol level within groups were not significantly different [F(15, 54)=1.417, 0.173 and F(10, 36)=1.391, 0.224 respectively]. The glucose levels were found to be significantly different [F(7.646, 27.526)=2.625, 0.030]. There was no significant difference in the adiponectin levels. Conclusions: Vitamin E supplementation could not prevent the increase of body weight, the elevation of blood sugar and cholesterol levels, and also could not increase adiponectin level.

Serum vitamin E levels are reduced in newborn infants. It has been reported that this deficiency is responsible, in part, for the development of anemia in premature infants during the first 6 weeks of life. The efficacy of vitamin E supplementation for the prevention of anemia in premature infants has been studied in a randomized, controlled, and blinded trial. Premature infants whose birth weights were less than 1,500 g were given, by gavage, 25 IU of dl-α-tocopherol or a similar volume of the drug vehicle. Treatment was continued for the first 6 weeks of life. A total of 178 infants were studied. Vitamin E levels were significantly higher in a supplemented group by day 3 and for the remainder of the 6-week period. At 6 weeks of age, there was no significant difference between the supplemented and unsupplemented groups in hemoglobin concentration, reticulocyte and platelet counts, or erythrocyte morphology. It is concluded that there is no evidence to support a policy of administering vitamin E to premature infants to prevent the anemia of prematurity.

To evaluate the efficacy of four early intramuscular injections of vitamin E given in addition to continuous minimal oral vitamin E supplementation, 168 very low-birth-weight infants (≤1,500 g) have enrolled in a randomized, double-masked, clinical study. All infants received vitamin E orally, 100 mg/kg/d. In addition, on days 1, 2, 4, and 6, seventy-nine infants received vitamin E intramuscularly, 15, 10, 10, and 10 mg/kg, respectively. On the same days, 89 control infants received placebo intramuscular injections. Multivariate analysis of the 135 infants who survived ≥10 weeks showed no significant difference in the development of severe retrolental fibroplasia between these two supplementation schedules (P = .86). Plasma vitamin E levels never exceeded a mean of 3.3 mg/100 mL, and no toxicity was observed. Ultrastructural analyses of seven pairs of whole eye donations from infants receiving IM vitamin E demonstrated identical kinetics of gap junction formation between adjacent spindle cells as compared with 13 pairs of whole eye donations from control infants (P > .3). Therefore, oral vitamin E supplementation affords retinal protection against the development of severe retrolental fibroplasia when initiated on the first day of life and maintained continuously until retinal vascularization is complete.

Abstract Background Chemotherapy often causes chemotherapy-induced peripheral neuropathy (CIPN), but effective prevention measures are still lacking. Whether vitamin E can prevent peripheral neurotoxicity caused by chemotherapy is inconclusive. Therefore, we collected related randomized controlled trials (RCTs) and conducted a meta-analysis to examine whether vitamin E could prevent CIPN. Methods We searched PubMed, EMBASE and the Cochrane databases in November 2019 for eligible trials. Two reviewers conducted the analysis independently when studies were homogeneous enough. Results Eight RCTs, involving 555 patients, were identified. Upon pooling these RCTs, patients who received vitamin E supplementation of 600 mg/day had a significantly lower incidence of peripheral neuropathy (RR 0.31; 95% CI 0.14 to 0.65; P = 0.002) induced by chemotherapy compared with the placebo group. Vitamin E played a key role in decreasing the incidence of peripheral neuropathy in the cisplatin chemotherapy group (RR 0.28; 95% CI 0.14 to 0.54; P = 0.0001). With regard to improvements in sural amplitude, vitamin E supplementation significantly decreased patients’ sural amplitude after three rounds of chemotherapy (RR -2.66; 95% CI -5.09 to -0.24; P =0.03) in contrast with that of placebo supplementation, while no significant difference was observed when patients were treated with vitamin E after six rounds of chemotherapy. In addition, the vitamin E group had better improvement in the neurotoxicity score (RR -2.65; 95% CI -4.01to -1.29; P = 0.0001) and a lower incidence of reflexes and distal paraesthesias (RR 0.50; 95% CI 0.29 to 0.87; P = 0.01) compared to the control group. Conclusion Available data included in this meta-analysis showed that vitamin E supplementation can confer modest improvement in the prevention of CIPN. However, large-scale, well-designed RCTs are needed to confirm the exact role of vitamin E supplementation in the prevention of CIPN.

Objective: To evaluate the hyperglycemic effects of Kursi Ziyabet (KZ) tablets on the rat models of diabetes mellitus. Methods: In total, 58 male SD rats were assigned randomly to six groups. All except the normal group were transformed into experimental diabetes mellitus rat models by injecting streptozocin. The hyperglycemic effect and the mechanism of Ziyabet were evaluated by body weight, food and water intake, fasting blood sugar, and related parameters by measuring the oxidative stress-related factors and lipid metabolism indicator level by the corresponding kits using the rat experimental models. Results: Compared with the model group, body weight markedly increased after 3–6 weeks of intragastric administration of Ziyabet tablets ([Formula: see text]), while the water intake significantly decreased in the same period of time ([Formula: see text]). Food intake and fasting blood sugar level also decreased with the high dosage of Ziyabet tablets ([Formula: see text]). There is no significant difference in pancreas’ MDA content of the Ziyabet groups when compared to the model group ([Formula: see text]), while significant increase in SOD level was observed in high-dosage KZ group ([Formula: see text]). The blood serum insulin and free fatty acid level also decreased in the high-dosage KZ group compared with the model group ([Formula: see text]). Conclusion: We conclude that Ziyabet tablets demonstrated protective effects on the diabetic rat models.

9114 Background: Peripheral neurotoxicity is a well recognized effect of cisplatin chemotherapy that can result in severe disability and represents a major dose-limiting factor. Several phase II studies have recently investigated the role of vitamin E as neuroprotectant in the prevention of cisplatin induced peripheral neurotoxicity and ototoxicity. Methods: An Italian randomized, placebo controlled, double blind multicentric study is ongoing to confirm the role of vitamin E supplementation in the prevention of neurotoxicity and ototoxicity induced by cisplatin Patients candidates to cisplatin chemotherapy were randomised to either vitamin E supplementation (a-tocopherol 400 mg/day) or to placebo. Patients were evaluated with neurological and neurophysiological examination before and after treatment. Neurotoxicity was measured using the comprehensive clinical and neurophysiological Total Neuropathy Score (TNS). Ototoxicity was evaluated with audiometric test and acoustic evoked potential before and after treatment. Results: 81 patients have been enrolled in 3 italian oncologic centers. An interim analysis on the first 50 patients was carried out. 25 patients (11 in the vit E group and 14 in the placebo group) received a cumulative dose higher than 300 mg/mq and were evaluable for neurotoxicity. Statistical analysis showed a significant difference (p < 0.05) in median neurotoxicity score observed in vit E group (TNS=1) respect to the placebo group (TNS=5). Conclusions: This is the first randomised, placebo controlled, double blind trial exploring the efficacy of vitamin E in the neuroprotection of cisplatin neurotoxicity. The results of this study confirm the neuroprotective effect of vitamin E supplementation against cisplatin-induced neurotoxicity. No significant financial relationships to disclose.

High-intensity exercise (3000-meter runner) contributes to muscle fatigue. Fatigue can be determined by measuring blood lactate levels. Blood lactate levels are a product of anaerobic metabolism. Lactate accumulation due to anaerobic physical activity can inhibit the glycolytic enzyme that affects decreased ATP production, damage of the calcium and sodium pumps in the muscles and causes fatigue. This study aimed to analyze the effect of vitamin E on the level of fatigue through the response of blood lactate levels in the runner 3000 meters. This research was a quasi-experimental research with a pretest-posttest design. Research subjects were 9 volunteer athletes (3000 meters runners) Aceh province, male, and aged 15-20 years. The treatments were vitamin E at a dose of 1x400 IU per day and administered for 14 days. Blood lactate levels were examined using the method of calorimetry. The statistical analysis was using the homogeneity test of variance (Levene's test), the normality test (Kolmogorov-Smirnov test) and paired t-test with a significant level of 5% (p-value<0.05). The results showed that blood lactate levels decreased approximately 13.93% after vitamin E supplementation. There was no significant difference (p=0.27) between blood lactate levels before and after vitamin E supplementation in athletes. In conclusion, vitamin E supplementation did not significantly lower blood lactate levels therefore vitamin E did not significantly reduce muscle fatigue in men's athletics.

The objective of the study was to assess the variability in the physiological responses of growing pigs on melatonin and vitamin E supplementation during summer and winter seasons.36 nos. of weaned crossbred (Hampshire X Assam local) female pigs were selected for the study. Eighteen (18) animals were subjected to treatment separately during summer and winter. The selected animals were divided into three groups, with six pigs in each group, consisting of the control group (Treatment 1), a second group comprising of animals fed with melatonin (Meloset) @3 mg/animal (Treatment 2) and a third group in which the animals were fed Vitamin E (Evion) @100 mg (Treatment 3), for both the seasons. The rectal temperature, respiration and pulse rate differed significantly (P>0.01) between seasons. There was also significant difference (P>0.01) in the mean pulse rate between treatment.

Aim and Objective: The aim of the study was to evaluate the role of Vitamin E supplementation in Type II diabetes mellitus (DM), to determines whether people with Type II DM treated with hypoglycemic agents alone, with or without Vitamin E, to determines the drug interaction in such treatment regimen, and to evaluates the Safety of the regimen.Methods: Type II DM patients with or without complications were included in this study along with serum glycated hemoglobin (HbA1c) concentration between 7.5% and 9.5%. They are divided into test group (which received hypoglycemic agent along with Vitamin E 4000 IU) and control group. Body mass index (BMI) status, fasting blood sugar (FBS), and post-prandial blood sugar (PPBS) were noted once in a month, HbA1c percentage, total cholesterol level (TC), and serum Vitamin E level were estimated and noted for every 3 months at total 9 months of this study. Patients with other comorbid conditions were prominent in this study.Results: It is perceptible with the analysis of obtained data that FBS, PPBS, HbA1c percentage, TC level, and BMI status of the patients were declined gradually in test group (patients with Vitamin E supplementation along with their hypoglycemic agents). Thus, antioxidant therapy is highly propitious whereby delaying the onset of complications in patients with DM. This development would be highly helpful for diabetic patients.
