Development of Gelatin Films with Designed Antimicrobial Peptide and Silver Nanoparticles

Gelatin films from cold water fish skin filled with designed antimicrobial peptide or silver nanoparticles were formulated for biomedical applications. Antibacterial activities of gelatin films containing silver nanoparticles and lytic peptides were evaluated to study the growth of a Staphylococcus species. The results suggest that inclusion of silver nanoparticles and lytic peptides into the composition of biocompatible films is feasible, with significant retention of the antimicrobial activity for both agents. The results presented here give evidence that both the silver nanoparticles and lytic peptides retain their antibacterial activity in such film matrices and the surrounding medium. Therefore, such biocompatible films may have potential use in biomedical applications such as wound dressing, superficial infection and contamination control. Evaluation of the mechanical properties for silver nanoparticles gelatin films revealed that the tensile strength appeared to be optimal at 0.3% silver nanoparticles loading. However, the strain-to-failure decreased with respect to the neat film.

Carissa carandas L. is traditionally used as antibacterial medicine and accumulates many antioxidant phytochemicals. Here, we expand this traditional usage with the green biosynthesis of silver nanoparticles (AgNPs) achieved using a Carissa carandas L. leaf extract as a reducing and capping agent. The green synthesis of AgNPs reaction was carried out using 1mM silver nitrate and leaf extract. The effect of temperature on the synthesis of AgNPs was examined using room temperature (25 °C) and 60 °C. The silver nanoparticles were formed in one hour by stirring at room temperature. In this case, a yellowish brown colour was developed. The successful formation of silver nanoparticles was confirmed by UV–Vis, Fourier transform infrared (FT-IR) and X-ray diffraction (XRD) analysis. The characteristic peaks of the UV-vis spectrum and XRD confirmed the synthesis of AgNPs. The biosynthesised AgNPs showed potential antioxidant activity through DPPH assay. These AgNPs also exhibited potential antibacterial activity against human pathogenic bacteria. The results were compared with the antioxidant and antibacterial activities of the plant extract, and clearly suggest that the green biosynthesized AgNPs can constitute an effective antioxidant and antibacterial agent.

Abstract Antimicrobial resistance not only increases the contagiousness of infectious diseases but also a threat for the future as it is one of the health care concern around the globe. Conventional antibiotics are unsuccessful in combating chronic infections caused by multidrug-resistant (MDR) bacteria, therefore it is important to design and develop novel strategies to tackle this problems. Among various novel strategies, Structurally Nanoengineered Antimicrobial Peptide Polymers (SNAPPs) have been introduced in recent years to overcome this global health care issue and they are found to be more efficient in their performance. Many facile methods are adapted to synthesize complex SNAPPs with required dimensions and unique functionalities. Their unique characteristics and remarkable properties have been exploited for their immense applications in various fields including biomedicine, targeting therapies, gene delivery, bioimaging, and many more. This review article deals with its background, design, synthesis, mechanism of action, and wider applications in various fields of SNAPPs. Graphic abstract

Peptide drugs hold great promise for the treatment of infectious diseases thanks to their novel mechanisms of action, low toxicity, high specificity, and ease of synthesis and modification. Naturally developing self-assembly in nature has inspired remarkable interest in self-assembly of peptides to functional nanomaterials. As a matter of fact, their structural, mechanical, and functional advantages, plus their high bio-compatibility and bio-degradability make them excellent candidates for facilitating biomedical applications. This review focuses on the self-assembly of peptides for the fabrication of antibacterial nanomaterials holding great interest for substituting antibiotics, with emphasis on strategies to achieve nano-architectures of self-assembly. The antibacterial activities achieved by these nanomaterials are also described.

Silver nanoparticles (Ag-NPs) have diverted the attention of the scientific community and industrialist itself due to their wide range of applications in industry for the preparation of consumer products and highly accepted application in biomedical fields (especially their efficacy against microbes, anti-inflammatory effects, and wound healing ability). The governing factor for their potent efficacy against microbes is considered to be the various mechanisms enabling it to prevent microbial proliferation and their infections. Furthermore a number of new techniques have been developed to synthesize Ag-NPs with controlled size and geometry. In this review, various synthetic routes adapted for the preparation of the Ag-NPs, the mechanisms involved in its antimicrobial activity, its importance/application in commercial as well as biomedical fields, and possible application in future have been discussed in detail.
