A COMPARISON OF FRESH AND FROZEN SEMEN IN THE INSEMINATION OF CONFINED SHEEP

The reproductive performance of crossbred sheep maintained in total confinement was compared after artificial insemination with fresh or frozen semen. Estrus was synchronized with progestagen-impregnated vaginal sponges and pregnant mares’ serum gonadotropin. Inseminations were performed 54 and 60 h after sponge removal. The fertility of ewes inseminated with fresh semen was significantly higher than of ewes inseminated with frozen semen. Conception rates, lambing rates and litter size were 83%, 78% and 2.2 using fresh semen and 65%, 43% and 1.8 using frozen semen. In a group of similar ewes bred by natural service, the lambing performance was comparable to that obtained with fresh semen. The difference between conception and lambing rates suggests an increase in early embryonic mortality when breeding with frozen semen and confirms the need for improved frozen semen technology.

ABSTRACTNinety-nine indigenous Greek goats (Capra prisca) on natural pasture were used to study reproductive performance as affected by method of breeding. At the beginning of the breeding season oestrus was induced with 45 mg fluorogestone acetate-impregnated vaginal sponges and 300 to 500 i.u. PMSG, given on day 15. The animals were separated into groups of 33 for mating: (i) by natural service (natural mating); (2) by artificial insemination with fresh semen (AI fresh); and (3) by insemination with frozen semen (AI frozen). Alpine bucks and Alpine semen were used for natural mating and AI of does.No effect of method of breeding was found on gestation length and sex ratio. Reproductive performance was significantly affected by method of breeding; at 21 days after insemination, fertility of does bred with fresh semen (69·7%) was comparable with that obtained with natural mating (72·7%) but the fertility of does bred with frozen semen (48·5%) was much lower. This difference was statistically significant at kidding (P < 0·05). Litter size was 2·14 and 1·86 (P < 0·05) for the AI fresh and AI frozen groups, respectively, and was 1·96 for the natural mating group. More multiple litters were obtained with the AI fresh group (P < 0·05). It is concluded that, induction and synchronization of oestrus and ovulation at the beginning of the breeding season gave reproductive performance with fresh semen comparable to that obtained with natural service, but use of frozen semen gave unsatisfactory results.

Pregnancy rates and litter sizes following insemination of sows with fresh-extended and frozen-thawed semen averaged 71 vs. 53% and 10.6 vs. 4.4, respectively, and was not affected by the addition of 12.5 μg relaxin to semen. These results indicate no relaxin mediated effect on either pregnancy rate or litter size when sows are artificially inseminated with relaxin-supplemented semen. Artificial insemination of ewes has become commercially available in Britain on a small scale over the last few years. It allows the farmer to benefit from MLC recorded rams which have been selected for growth rate and more recently for carcass quality. However, proximity to an A.I. centre is an important consideration as A.I. of ewes requires the use of fresh semen. Frozen semen has not vet achieved the quality and reliability necessary on a commercial scale. Insemination is carried out at a fixed time after the removal of Intra-vaginal pessaries and an injection of Pregnant Mare Serum Gonadotrophin (PMSG). Synchronisation of oestrus using sponges and PMSG adds to the cost of using a commercial A.I. service. If A.I. is not used then one ram per ten ewes is necessary for a synchronised mating which either requires a ram sharing group or considerable financial investment if rams of equivalent quality to those used in A.I. centres are to be purchased.

The use of liquid semen for artificial insemination program of Etawah crossbreed goat (PE) is an alternative to replace frozen semen which is constrained by limited and expensive facilities. Production of liquid semen is faster than frozen semen, but the viability of liquid semen which preserved with a standard extender such as tris egg yolk is very short. The purpose of this study was to determine the viability of PE goat semen in egg yolk tris substituted with energy sources such as glucose, galactose, and mannose and to determine the most efficient energy source for semen preservation. This research was conducted from August to September 2018 at the Artificial Insemination Center in Lembang, West Java. This study was designed in a randomized block design (RBD) consist of three experimental groups divided into five groups. Fresh semen of PE goats were preserved using extender which energy source has been modified. Results showed that using glucose in PE goat semen extender produced the best motility among other groups (64.29 ± 9.2%). The highest viability was found in extender with fructose substitution (86.76 ± 2.3%). The longest viability of liquid semen was found in the extender with glucose substitution. It lasted for six days.

The purpose of this research was to analysis the value of repeatability and correlation among the traits affecting the production of frozen semen from Holstein’s bull in Indonesia. Repeatability and correlation were calculated based on the data of frozen semen production of 15.699 records from 44 Holstein bulls at Singosari Artificial Insemination Center (SAIC) and 8.935 records from 39 Holstein bulls at Lembang Artificial Insemination Center (LAIC). Repeatability for volume, motility, fresh semen concentration and frozen semen production was evaluated by intraclass correlation method. The repeatability values of LAIC for volume, motility, fresh semen concentration and frozen semen production were 0.60; 0.54; 0.37 and 0.47. The repeatability values of SAIC for volume, motility, fresh semen concentration and frozen semen production were 0.54; 0.30; 0.43 and 0.29. The linear correlation value between volume, motility and fresh semen concentration with the amount of semen produced per collections were 0.41, 0.36, and 0.58. Concentration was the most factors influencing the number of frozen semen produced. The effectiveness of the selection of Holstein's frozen semen producing could be determined by the value of repeatability and the phenotypic correlation among semen quality traits such as volume, motility, concentration and frozen semen production.

One hundred gilts and one hundred mixed-parity sows of Yorkshire and Landrace breeding were inseminated with semen with or without an additional 10 μg estradiol-17β. Pregnancy rates of gilts receiving estradiol-supplemented and control semen were 90 and 79%, respectively (P < 0.09). A study was made of reproductive performance and embryonic mortality in XO and XX females. In the stock used, the mean litter size of XO females (4·46) was greatly below that of XX ones (8·17). One series of pregnant females of both karyotypes was dissected after 15 days' gestation, and another series after 3½days' gestation. In the former, there was a significantly greater amount of embryonic mortality in XO females both before implantation and at the small and large mole stages. By far the greater amount occurred before implantation. The data from dissections after 3½ days' gestation concerned pre-implantation embryos, since normal embryos at this point are at the late morula or early blastocyst stage. The embryos from XO females contained a large group of obviously and characteristically abnormal ones; they comprised 60/280 of the embryos from XO females, compared with 4/189 of the XX ones. They appeared to have developed abnormally from a very early stage, probably the two-cell stage, and were considered to represent the missing OY class of zygotes. In addition, it was concluded that there was probably an abnormally low segregation of nullo-X gametes from XO females.

ABSTRACTSix experiments involving the artificial insemination (AI) of a total of 300 female red deer (hinds) with frozen-thawed red deer semen (collected by electro-ejaculation) were conducted over 3 years. Insemination took place at fixed times following various oestrous synchronization procedures using progesterone withdrawal and treatment with pregnant mare's serum gonadotropin (PMSG). In the 1st year, the experiments evaluated basic AI techniques in which pregnancy rates were 45% in 20 hinds receiving two inseminations per vaginam (PV) and 56% in 27 hinds inseminated by the laparoscopic intrauterine method (IU). In the 2nd year, the experiments involved comparisons of the progesterone regime, one or two PV inseminations, and the timing of a single PV insemination. There was no effect of replacement of the progesterone device after 9 days and withdrawal 3 days later compared with the use of one device for the whole period in two experiments. The pregnancy rate for a double PV insemination was significantly higher than for a single PV insemination (58 and 34%; P < 0·05) and there was also a small effect of timing of insemination relative to the synchronization treatment. In the 3rd year all hinds were inseminated by the IU method. The experiments involved a comparison of various times of AI following progesterone withdrawal and a comparison of the progesterone regime. The overall pregnancy rate for 63 hinds inseminated was 56% with no difference between three times of insemination (48, 52 and 55 h). In the second experiment, the difference in pregnancy rate between treatment with progesterone for 15 days and 12 days (44 and 72% for 18 hinds per group) was not significant, but the interaction between the length of progesterone treatment and insemination time was significant (P < 0·05), with the 12-day progesterone/55 h insemination giving a much higher pregnancy rate than the 15-day/55 h insemination (89 and 20% respectively). Although no experiments involved direct comparisons of the routes of insemination, overall pregnancy rates were 56% for IU, 53% for double PV and 35% for single PV inseminations.

The objectives of this study were to examine the effects of several factors affecting fawning rate, litter size, litter weight and neonatal fawn mortality in white-tailed deer inseminated either transcervically or by means of laparoscopy. Oestrus synchronisation with a controlled internal drug release (CIDR)-based protocol and fixed-time artificial insemination (FTAI) was conducted in 130 white-tailed deer (Odocoileus virginianus texanus) during three reproductive seasons (2007–2009; 271 services) in a game-hunting ranch in a hot–arid environment (26°4′ N, 101°25′ W). Ninety additional non-treated does were exposed to bucks for natural mating. Fawning rate did not differ between AI methods (40.0 vs 45.0% for transcervical and laparoscopic AI, respectively). Overall fawning rate (proportion of all does fawning after FTAI and a subsequent period of buck exposure) did not differ between transcervical (89.5%), laparoscopic (80.3%) or natural (88.9%) insemination. Litter size per fawning doe was higher (P&lt;0.05) in naturally-served does (1.65±0.48) than in transcervically-inseminated does (1.40±0.51) or in laparoscopically-inseminated does (1.48±0.50). The main conclusion was that no enhancement of fawning rate or litter size occurred as a result of intrauterine deposition of semen by laparoscopy compared with the transcervical insemination technique.
