Pregnancy rates and litter sizes following insemination of sows with fresh-extended and frozen-thawed semen averaged 71 vs. 53% and 10.6 vs. 4.4, respectively, and was not affected by the addition of 12.5 μg relaxin to semen. These results indicate no relaxin mediated effect on either pregnancy rate or litter size when sows are artificially inseminated with relaxin-supplemented semen. One hundred gilts and one hundred mixed-parity sows of Yorkshire and Landrace breeding were inseminated with semen with or without an additional 10 μg estradiol-17β. Pregnancy rates of gilts receiving estradiol-supplemented and control semen were 90 and 79%, respectively (P < 0.09). The estradiol supplementation of semen did not improve the pregnancy rates or litter size in sows. Key words: Swine, artificial insemination, estradiol

In all domestic species, acceptable pregnancy rate with frozen-thawed semen is 20-30% through simply cervical and vaginal insemination. Researchers have proved that achievable high results can be expected only from intrauterine insemination in ewes. Unlike the cow, and goat, the cervix of ewes is a formidable barrier to penetrate for transcervical intrauterine insemination. It is not only due to the physical size of the canal and a narrow luminal diameter (even at estrus), but also due to its caudally facing eccentric series of four to eight funnel-like rings. The major aim of this study was to evaluate a comparison of transcervical and laparoscopic intrauterine artificial insemination techniques with frozen –thawed semen on the reproductive performance of ewes.58ewes of 3-4 years of age, so called Altanbulag subspecies of breed were selected and equally divided into 2 groups for the AI methods from October 15-20, 2013 in the breeding station of Research Institute of Animal Husbandry in Hongor sum, Darhan uul province. The estrus was synchronized with EAZI-BREED TMCIDR in which the progesterone content was 0.3 g. After 12 days, the CIDR were removed, PMSG of 400 and 200 IU were administrated intramuscularly per ewe. The total success of synchronization rate was 93.3%. The results has shown the possibilities of laparoscopic and transcervical AI techniques in 55.17%, 41.37% (p>0.05) of pregnancy rates respectively, with frozen-thawed semen in ewes by ultrasonography diagnosis.Mongolian Journal of Agricultural Sciences Vol.13(2) 2014: 43-47

The reproductive performance of crossbred sheep maintained in total confinement was compared after artificial insemination with fresh or frozen semen. Estrus was synchronized with progestagen-impregnated vaginal sponges and pregnant mares’ serum gonadotropin. Inseminations were performed 54 and 60 h after sponge removal. The fertility of ewes inseminated with fresh semen was significantly higher than of ewes inseminated with frozen semen. Conception rates, lambing rates and litter size were 83%, 78% and 2.2 using fresh semen and 65%, 43% and 1.8 using frozen semen. In a group of similar ewes bred by natural service, the lambing performance was comparable to that obtained with fresh semen. The difference between conception and lambing rates suggests an increase in early embryonic mortality when breeding with frozen semen and confirms the need for improved frozen semen technology.

Research in the area of equine artificial insemination (AI) has led to its increased application in field trials (Loomis and Graham, 2008). However, procedures for equine semen collection, cooling and freezing of semen and artificial insemination need further improvement. This study examined the effect of semen preparation and ovulation on equine AI success. Stallion semen was collected via artificial vagina, diluted with skim-milk extender, and preserved by either cooling to 4°C or cryopreservation and storage in liquid nitrogen. Mares were examined by ultrasound for follicle development with every 12 h during oestrus. During AI, semen was inserted into uterine horn adjacent to the ovulating follicle sized over 4 cm and irregular shape. In experiment 1, mares were inseminated with either cooled, cooled-transported, or frozen–thawed semen. Cooled semen was used immediately after collection and cooling. Cooled-transported semen was used 4 h after semen collection and dilution. Semen freezing procedure was followed by Moore et al. (2006). In experiment 2, we examined the effect of buffer (skim-milk extender), which was infused into the uterus at the time of AI with frozen–thawed semen. In experiment 3, we compared AI pregnancy rates for mares ovulating spontaneously v. after treatment with hCG. Pregnancy data were analysed Chi square test where P-values < 0.05 were considered significantly different. In experiment 1, the pregnancy rates after AI using cooled, cooled-transported, and frozen–thawed semen were 60 (10/6), 50 (8/4), and 37.5% (8/3), respectively, and similar among treatments. In experiment 2, the pregnancy rate of mares infused with buffer at AI was 37.5% (8/3) which was higher than that with no buffer (0%; 6/0). In experiment 3, the pregnancy rates of mares were similar between ovulated spontaneously (25%; 8/2) and ovulated with hCG (50%; 8/4). The results suggest that equine semen that has been cooled, cooled and transported or frozen can be successfully used to establish AI pregnancy. Also, the pregnancy rates after AI can be increased by infusing buffer into the uterus at AI or by inducing ovulation with hCG. These experiments resulted in the first foal produced in Korea with frozen–thawed equine semen. In two different experiments crossbred gilts and(or) sows were used to investigate the effects of uterine exposure (prior to breeding) to nonviable semen (NS), seminal plasma (SP) or egg albumen (EA) on reproductive performance. Treatment solutions were infused via the cervix at estrus in gilts or within 2 d post-partum in sows. Females were artificially inseminated at the subsequent estrus. In the first experiment pregnancy rates for gilts infused with Beltsville Thawing Solution (BTS) as controls, NS or EA and subsequently inseminated, were 76, 85 and 54%, respectively, and were not significantly different among treatment groups. The number of total embryos (adjusted for number of corpora lutea) present at 38 d postinsemination was 9.6, 9.0 and 9.6, respectively, for BTS, ND and EA infused gilts and was not significantly different. Likewise, the number of live and dead embryos was not different among treatments. In the second experiment, the proportions of inseminated sows infused with BTS, NS or SP that subsequently farrowed were 87, 84 and 92%, respectively, and did not differ significantly among treatments. No significant differences were detected for the number of total, live or dead pigs born per litter among treatments. We conclude that uterine exposure to seminal proteins or egg albumen prior to breeding does not alter reproductive performance in gilts by 38 d of gestation. Also, the infusion of seminal proteins in sows shortly after parturition does not affect pregnancy rates or litter size at the subsequent farrowing. Equine AI with frozen semen has become more frequent in reproduction centers, but osmotic shock during thawing is still a major problem as it impairs gamete viability and fertility. To overcome this, multi ultrasonographic (US) exams must be performed to ensure AI at the closest time to ovulation possible. The objective was to evaluate, in vivo, the effects of dilution of the cryoprotectant dimethylphormamide on the success of a fixed timed (20 h post-ovulation) AI protocol, in an attempt to reduce osmotic shock and the intense labour usually required during equine AI with frozen semen. Thirty-five Mangalarga Marchador mares were used in this trial. When dominant follicles reached at least 35 mm in diameter, ovulation was induced with 6 mg of equine gonadotrophin extract (EGE) intravenously. Artificial insemination was performed 20 h after ovulation according to 1 of the following protocols: no dilution (n = 14): three 0.5-mL straws were thawed and the semen deposited by a flexible rod at the uterine tip in the ipsilateral horn where the pre-ovulatory follicle was detected; or dilution (n = 21): three 0.5-mL straws were thawed as described and centrifuged after addition of 1.5 mL of BotuSêmen® extender (1:1 dilution). Pregnancy rates were evaluated between treatments, age and presence of intrauterine liquid (IUL) classes and period between ovulation and AI. Data were analysed as a binomial distribution with the GENMOD procedure (SAS, Cary, NC, USA). Pregnancy rate was higher (P < 0.05) for mares inseminated with diluted semen (42.9%) compared to those inseminated with no semen dilution (4.8%). Age and IUL class did not influence pregnancy rates. It can be concluded that reduction of osmotic stress through the dilution of cryopreserved equine semen increased pregnancy rates after AI. Semen dilution, as tested in this trial, yielded good pregnancy rates in a fixed-time AI protocol in mares, simplified mare management greatly and allowed the use of a fixed-time protocol successfully. Peripheral progesterone (P4) concentrations as well as pregnancy rates (PR) between cows treated with Ovsynch or CIDR ovulation synchronization/TAI protocols were compared. Two hundred and twenty-seven postpartum lactating Holstein cows were randomly assigned to Ovsynch (n = 111) or CIDR (n = 116) ovulation synchronization/TAI protocols. Pregnancy was diagnosed by ultrasonography at day 35 after TAI and later confirmed by rectal palpation at day 60 post TAI. Milk samples were taken from the beginning of treatment (day -10) until day 35 after TAI (day 0) for P4 determination. Pregnancy rates for Ovsynch and CIDR groups of cows were 31 and 41%, respectively. The PR of ovulation synchronized cows for Ovsynch and CIDR groups were 49 and 69%, respectively. Progesterone levels during the sampling period were similar (P > 0.05) between Ovsynch and CIDR treated cows. Key words: Pregnancy rate, progesterone, Ovsynch, CIDR, cows

Four experiments were conducted to investigate the use of melengesterol acetate (MGA) and estradiol-17β (E-17β) for synchronizing estrus and improving reproductive performance in beef cattle. Synchronized pregnancy rates were higher (P < 0.05) in heifers fed MGA after AI than controls (4 of 9 versus 0 of 7, respectively), but were not different for cows (20 of 37, 54% versus 23 of 40, 58%). Estrus rate (percentage of cows detected in estrus) was higher (P < 0.05) for cows treated with E-17β + MGA compared with those treated with cloprostenol alone (85.0 versus 65.9%). Synchronized pregnancy rates were not significantly affected by feeding MGA or grain only after detected estrus and AI (20 of 31, 64% versus 17 of 30, 57%). For 20 cows not detected in estrus and bred 100 h after cloprostenol treatment, pregnancy rate was higher (P < 0.05) for those fed MGA after AI than controls (6 of 11, 54% versus 1 of 9, 11%). Reproductive performance and body weight were studied on 361 ewes, representing Finnsheep (F), DLS (a population of 1/2 Dorset, 1/4 Leicester, 1/4 Suffolk) and seven combinations ranging from 1/8 to 7/8 Finnsheep breeding. Conception rate in yearlings was 61.5% for DLS compared to 89.0% for F with the crosses being intermediate. Conception rate in older ewes was similar in the different genetic groups (avg. 94%). Ovulation rate and litter size at birth of DLS ewes were 1.72 and 1.44 lambs, which was less than half those of F ewes (3.51 and 2.86 lambs, respectively). Both traits increased progressively with an increase in F breeding in crosses and with advances in age. DLS ewes weaned 1.22 lambs compared to 2.03 lambs for F ewes and 1.84 lambs for 4/8 F ewes. The heaviest litters at weaning (31.7 kg) were raised by 4/8 F ewes, followed by 7/8 F (30.8 kg) while those raised by DLS ewes weighed 23.0 kg and F ewes 29.1 kg. Percentage of ova lost per ewe mated averaged 24% and ranged between 18% (DLS and 1/8 F) and 29% (6/8 F). About 3.6% of lambs were born dead and a further 13.8% died before weaning. Preweaning mortality rate was highest in F (22.9%) and lowest in 3/8 F (9.4%). Average kilograms of lambs weaned per ewe exposed was highest in 4/8 F (27.6 kg) followed by F (26.0 kg), whereas that of DLS was the lowest at 18.1 kg. The 4/8 F cross showed 25% heterosis in kg of lambs weaned per ewe exposed and 52.5% increase over DLS. Significant positive linear regressions were calculated for ovulation rate, litter size and preweaning mortality rate on proportion of Finnsheep breeding in crosses. The relation was quadratic for percent ova lost and lamb mortality at weaning. Yearling DLS females weighted 36 kg compared to 44 kg for F yearlings. However, at 5 yr of age DLS ewes weighed 62 kg, 5 kg heavier than F ewes. The heaviest ewes at all ages were the 4/8 F (45 kg at 1 yr, 65 kg at 5 yr). Abstract The present study aimed to determine the effects of progesterone-based gonadotropin-releasing hormone (GnRH), prostaglandin F2α (PGF2α), and equine chorionic gonadotropin (eCG) injections on progesterone profiles and pregnancy rates in cows with no estrus symptoms within 60 days after parturition. A total of 80 cows were included in the study. All animals had the progesterone-releasing device PRID®Delta placed intravaginally for nine days with an injection of GnRH. On the eighth day, PGF2α was injected, and PRID®Delta was removed from the vagina on day nine. Artificial insemination was carried out 60 hours after PRID®Delta removal. In half of the animals (n = 40), 600 IU of eCG was injected when PRID®Delta was removed on the ninth day before artificial insemination 60 hours later. Blood samples were taken from the tail vein on days 0 and 8 to determine progesterone levels. The pregnancy rate in the group that received eCG was 37.5%, while it was 27.5% in those that did not (P = 0.4). While the dominant follicle diameter was 15.5 mm in cows injected with eCG during timed artificial insemination, the follicle diameter was 12.4 mm in cows with no eCG injection (P <0.001). There were no differences in serum progesterone values in blood samples taken until the time of artificial insemination. However, progesterone values in the blood taken during artificial insemination were 0.94 ng/ml in the eCG- group and 0.72 ng/ml in the eCG+ group (P <0.05). As a result, it was determined that eCG injections, in addition to progesterone-based GnRH and PGF2α applications, increased the pregnancy rates in cows without symptoms of estrus. The dominant follicle diameter was larger in cows treated with eCG during artificial insemination; however, follicle size did not increase the pregnancy rate.
