CHEMICAL COMPOSITION AND PROTEIN CHARACTERISTICS OF LENTILS

Chemical and amino acid compositions, protein solubility (Osborn classification), and in vitro protein quality of six genotypes of lentils (Lens culinaris Medik.) were determined. Lentils contained an average of 28.6% protein, 3.1% ash, 4.4% fiber, 0.7% ether extract, 63.1% total carbohydrate (nitrogen-free extract), 44.3% starch and 4,186 kcal/kg of gross energy. Lentil starch contained 36.1% amylose. About one-half (47.1%) of the lentil proteins were soluble in 1.0 M sodium chloride, 3.8% soluble in water, 3.1% soluble in 70% ethanol, and 14.9% soluble in dilute hydrochloric acid. Only 24.0% of the lentil proteins were not solubilized by these solvents. The lentils contained 6.8% of the meal nitrogen as nonprotein nitrogen. The amino acid compositions of the six lentil genotypes were similar. The major amino acids were glutamic acid, aspartic acid, arginine, leucine and lysine; the minor amino acids were methionine, cystine and tryptophan. Because of the lower concentrations of the latter amino acids, lentil proteins gave a chemical score of 35, a protein score of 46, and an essential amino acid index of 63, relative to egg protein.

The amino acid composition, essential amino acid index (EAA-I), pepsin-pancreatin in vitro digestibility and pepsin-pancreatin-digest-residue-index (PPDR-I) of the rumen bacterial and protozoal protein of cows fed urea and ammonium salts as their sole source of nitrogen (0-cows) or as a partial source (ULP-cows), and of cows on normal protein-rich feed (NorP-cows), were determined. The amino acid compositions of the rumen bacteria showed very slight changes even though the diets were very different. The amino acid compositions of the pure bacterial strains isolated from the rumina differed slightly. The amino acid compositions of the rumen protozoa of the ULP- and NorP-cows differed only with respect to isoleucine and tyrosine. Protozoa could be found only occasionally in the rumen of the 0-cow, there being only two species. Their nutritional significance was obviously very small. When the nutritional quality of the microbial protein was studied on the basis of its amino acid composition it was found that the EAA-I of bacteria did not differ significantly. Neither did the EAA-I of protozoa differ. The pepsin-pancreatin in vitro digestibility of protozoa was higher on all the feeds than that of bacteria. The rumen bacterial in vitro digestibility with 0-cows differed from that of the ULP-samples but not from that of the NorP-samples. The digestibility of single amino acids, With the exception of diaminopimelic acid, glycine and alanine, did not differ from the digestibility of the total amino acids. The much larger number of bacteria in the rumen of 0-cows compared with those of ULP- and NorP-cows compensates in this way for the lower digestibility of bacterial protein in 0-cows. The PPDR-I of both bacteria and protozoa were well correlated with the in vitro digestibility.

Background: There is a constant search of new plant proteins, with adequate nutritional and functional properties, as well as bioactive properties and low-cost for utilization in various food formulations. Objective: The aim of this work was to access the nutritional and functional potential of protein from Irvingia gabonensis, for utilization as ingredient or supplement in food. Methods: Proximate composition and amino acid were analyzed. Nutritional parameters were calculated from amino acid composition. Physicochemical properties and secondary structure of protein were determined. Finally, effect of oil to water ratio (OWR), pH and concentration on emulsifying properties was analyzed. Results: The flour contained 22.26% protein, 5.30% ash and 60% carbohydrates. Proteins contained all essential amino acids, with high content of Leu, Ile, Val, Thr and sulfur-containing amino acids. Essential amino acid index (69%), protein efficiency ratio (2.39-2.63) and biological value (79.91%) were studied. The maximum protein solubility (61%) was noticed at pH 8, while high hydrophobicity was observed at pH 2. A transition from an irregular secondary structure to a more ordered structure was found from pH 2-4 to pH 6-10. pH, OWR and concentration significantly affected emulsifying properties of Irvingia gabonensis almonds. The maximum emulsifying capacity (EC) was observed under acidic pH and high flour concentration. EC increased with increasing OWR and concentration, while decreased with increasing pH. High ES (25-35%) was observed at pH 4-8 and OWR of 1/3 to 1/2 (v/v), at flour concentration of 3-4% (w/v). Conclusion: Irvingia gabonensis showed good potential as food ingredient or supplement.

Variation in the apparent ileal digestibility of amino acids in 19 meat and bone meal samples, obtained from commercial rendering plants in New Zealand, was determined using 5-week-old broilers. Assay diets contained meat and bone meal as the only source of protein, and chromic oxide as an indigestible marker for the calculation of amino acid digestibility values. Correlations of chemical composition (crude protein, ash, crude fat, and gross energy) and in vitro assays (protein solubility in 0.2% potassium hydroxide and nitrogen digestibility by 0.2% pepsin hydrolysis) with ileal amino acid digestibility were also examined. Considerable variation was observed in the contents of crude protein (38.5–67.2 g/100 g), ash (13.0– 56.5 g/100 g), crude fat (4.3–15.3 g/ 100�g), and gross energy (9.4–22.3 MJ/kg) of meat and bone meal samples. The amino acid concentrations and ileal digestibility of amino acids also varied substantially. Cystine, the first limiting amino acid in meat and bone meal, had the lowest digestibility estimates. Correlation analyses showed that the ash content was the only chemical parameter that was consistently correlated with amino acid digestibility. Digestibility of amino acids, with the exception of aspartic acid, threonine, serine, tyrosine, histidine, and cystine, was negatively correlated with ash content, with samples with high ash levels having lower digestibility. Both in vitro assay measurements were found to be insensitive indicators of variations in amino acid digestibility.

Chemical and nutritional properties of pumpkin (Curcubita pepo) seed proteins were studied. The seed was processed into defatted flour (CPF) which was further processed into Curcubita protein concentrate (CPC) and Curcubita protein isolate (CPI) by alkaline water/isoelectric precipitation. Chemical properties of the protein products were determined using standard methods of analysis. The amino acid profile was determined by an automated Technicon® liquid chromatography system. Protein digestibility was assessed in-vitro (IVPD) using trypsin-pepsin enzyme method while biological values were determined on the basis of their amino acid profile. Protein efficiency ratio (PER) was estimated according to a standard proposed regression equation. The seed proteins demonstrated high levels of crude protein (CPC=69.98% and CPI=74.15%), vitamin C (CPC=43.46 and CPI=52.36 mg/ml) and vitamin A (CPC=100.56 and CPI= 63.43 I.U/g) with low levels of thiamin and riboflavin. Both proteins showed low and similar (p>0.05) levels of sodium (0.14-0.18%), calcium (0.86-1.02%), magnesium (0.53-0.58%) and phosphorus (0.09-0.11%). Percentage ratios of essential to total amino acids obtained for CPC and CPI (44.24% and 45.50%, respectively) were greater than 36% which is considered adequate for an ideal protein. Protein biological values obtained for CPC and CPI respectively were: 95% and 53% (chemical score), 2.80 and 1.56 (PER} and 70.10% and 51.28% (essential amino acid index). CPC showed a better digestibility than CPI with IVPD value of 56.88%. Threonine and lysine were the most limiting amino acids in both protein products. All anti-nutrients evaluated were low and below allowable limits. In conclusion pumpkin seed proteins showed good biological values and could be used to improve the quality of other plant proteins or as a possible replacement for animal proteins in conventional foods.

Abstract Objectives 1) to estimate the probability of inadequate amino acid intake among infants 9–10 months of age in rural Malawi 2) to evaluate whether dietary amino acid intake or protein quality are associated with length gain from 6 to 12 months of age Methods We assessed total amino acid intake from breast milk and complementary foods in 285 infants. Breast milk intake and complementary foods were estimated using dose-to-mother deuterium oxide dilution method and repeat 4-pass interactive 24-hour recall interviews, respectively. Amino acid composition values were taken from FAO human milk profile, Tanzania Food Composition table and International Minilist. Protein quality was estimated using Digestible Indispensable Amino Acid Score (DIAAS). Probability of intake below Estimated Average Requirement (EAR) for each amino acid was estimated using National Cancer Institute (NCI) method. We estimated protein quality of complementary food using median DIAAS. We assumed a DIAAS of ≥0.75 to represent a diet or food with good protein quality. Relationships between amino acid intake or protein quality with length gain were assessed using regression models. Length was measured at 6 and 12 months of age and length for age z-score (LAZ) velocity was calculated (ΔLAZ/months). Results The probability of inadequate amino acid intake from breast milk and complementary food that included a lipid-based nutrient supplement (LNS) was 3% for lysine, 0% for tryptophan, threonine, valine, histidine, isoleucine, leucine, sulfur containing amino acids (SAA), and aromatic amino acids (AAA). Without LNS, the probability was 7% for lysine and 0–2% for the other amino acids. The median (interquartile range) DIAAS for complementary food with and without LNS was 0.70 (0.28) and 0.64 (0.32), respectively. Dietary amino acid intake and protein quality were not significantly associated with length gain velocity from 6 to 12 months even after adjusting for confounding factors. Conclusions The prevalence of inadequate amino acid intake in 9–10 months old infants in rural Malawi is very low. However, in conditions of frequent clinical or sub-clinical infections this situation may be different. Linear growth at 6–12 months does not appear to be limited by dietary amino acid intake or protein quality in this setting. Funding Sources The Bill & Melinda Gates Foundation.

The potential of bacterial cellulose as a carrier for the transport of ibuprofen (a typical example of non-steroidal anti-inflammatory drugs) through the skin was investigated. Ibuprofen and its amino acid ester salts-loaded BC membranes were prepared through a simple methodology and characterized in terms of structure and morphology. Two salts of amino acid isopropyl esters were used in the research, namely L-valine isopropyl ester ibuprofenate ([ValOiPr][IBU]) and L-leucine isopropyl ester ibuprofenate ([LeuOiPr][IBU]). [LeuOiPr][IBU] is a new compound; therefore, it has been fully characterized and its identity confirmed. For all membranes obtained the surface morphology, tensile mechanical properties, active compound dissolution assays, and permeation and skin accumulation studies of API (active pharmaceutical ingredient) were determined. The obtained membranes were very homogeneous. In vitro diffusion studies with Franz cells were conducted using pig epidermal membranes, and showed that the incorporation of ibuprofen in BC membranes provided lower permeation rates to those obtained with amino acids ester salts of ibuprofen. This release profile together with the ease of application and the simple preparation and assembly of the drug-loaded membranes indicates the enormous potentialities of using BC membranes for transdermal application of ibuprofen in the form of amino acid ester salts.

Background: E6 and E7 are oncoproteins of HPV16. Natural amino acid variation in HPV16 E6 can alter its carcinogenic potential. The aim of this study was to analyze phylogenetically E6 and E7 genes and proteins of HPV16 from Indonesia and predict the effects of single amino acid substitution on protein function. This analysis could be used to reduce time, effort, and research cost as initial screening in selection of protein or isolates to be tested in vitro or in vivo.Methods: In this study, E6 and E7 gene sequences were obtained from 12 samples of  Indonesian isolates, which  were compared with HPV16R (prototype) and 6 standard isolates in the category of European (E), Asian (As), Asian-American (AA), African-1 (Af-1), African-2 (Af-2), and North American (NA) branch from Genbank. Bioedit v.7.0.0 was used to analyze the composition and substitution of single amino acids. Phylogenetic analysis of E6 and E7 genes and proteins was performed using Clustal X (1.81) and NJPLOT softwares. Effects of single amino acid substitutions on protein function of E6 and E7 were analysed by SNAP.Results: Java variants and isolate ui66* belonged to European branch, while the others belonged to Asian and African branches. Twelve changes of amino acids were found in E6 and one in E7 proteins. SNAP analysis showed two non neutral mutations, i.e. R10I and C63G in E6 proteins. R10I mutations were found in Af-2 genotype (AF472509) and Indonesian isolates (Af2*), while C63G mutation was found only in Af2*.Conclusion: E6 proteins of HPV16 variants were more variable than E7. SNAP analysis showed that only E6 protein of African-2 branch had functional differences compared to HPV16R.

The kinetics of radioactive labelling of extra- and intra-cellular amino acid pools and protein of the extensor digitorum longus muscle were studied after incubations with radioactive amino acids in vitro. 2. The results indicated that an extracellular pool could be defined, the contents of which were different from those of the incubation medium. 3. It was concluded that amino acids from the extracellular pool, as defined in this study, were incorporated directly into protein.

The effect of cycloheximide in vitro and in vivo on the incorporation of labelled amino acids into protein by muscles, liver, kidneys, and brain of rats and pigeons was studied. In vitro incorporation of amino acids into protein by muscle microsomes, myofibrils, and myofibrillar ribosomes was not affected by cycloheximide. In contrast, administration of the antibiotic into intact animals at a concentration of 1 mg/kg body weight resulted in considerable inhibition of amino acid incorporation into protein by muscles, liver, kidneys, and brain. This inhibition was observed in all the subcellular fractions of these tissues during a period of 10–40 min after the administration of the precursor. Tissue homogenates derived from in vivo cycloheximide-treated animals did not show significant alteration in in vitro amino acid incorporation with the exception of brain, which showed a small but significant enhancement.
