Influence of Processing Schemes on Indicative Bacteria and Quality of Fresh Aquacultured Catfish Filletst†

Fresh aquacultured catfish fillets were obtained from three processors using different processing protocols in summer, autumn, winter, and spring and evaluated for microbial quality. Twenty freshly processed fillets were randomly selected and each fillet was placed in a sterile polyethylene bag. The fillets were transported on ice-pack overnight by air immediately after processing. Five fillets were randomly selected for microbial assays. Each fillet was weighed and an equal volume of sterile 0.1 % peptone water at 0 to 1°C was added aseptically. The fillet was massaged (or rinsed) for 120 s and the rinse was used to determine microbial quality. Aerobes (incubation at 35°C for 48 h) and psychrotrophs (incubation at 20°C for 96 h) were enumerated using 3M Petrifilm™ Aerobic Count plates. Escherichia coli (incubation at 35°C for 24 to 48 h) and total coliforms (incubation at 35°C for 24 to 48 h) were enumerated on 3M Petrifilm™ E. coli Count plates. Staphylococcus aureus counts were determined on Baird-Parker agar (incubation at 35°C for 48 h). Significant differences (P ≤ 0.05) in aerobic, psychrotrophic, total coliform, E. coli, and S. aureus counts due to temperature effects during production and variations in processing protocols were observed. E. coli and S. aureus counts were significantly different during the four seasons. E. coli and S. aureus counts were high during summer and low during winter weather. There was a significant difference (P ≤ 0.05) in aerobic, psychrotrophic, and total coliform counts among the three processors during warm weather; however, these differences were significantly (P≤ 0.05) reduced in cold weather.

Aquacultured channel catfish (Ictalurus punctatus) were evaluated for the presence of human pathogenic bacteria. Fresh catfish fillets procured from three catfish processors in the southeastern United States during the four annual seasons (e.g., summer, fall, winter, and fall) were screened for selected human pathogens. At each sampling time point, 20 freshly processed catfish fillets were randomly selected from each processor during each season. Five catfish fillets were randomly selected for aerobic plate counts and all 20 fillets were screened for five pathogenic bacteria viz. Campylobacter jejuni/coli, Escherichia coli O157:H7, Klebsiella pneumoniae subsp. pneumoniae, Plesiomonas shigelloides, and Vibrio cholerae. There was a significant difference (P < 0.05) in the aerobic plate counts due to differences in unit processing operations and processing season. C. jejuni/coli, E. coli O157:H7 and K. pneumoniae subsp. pneumoniae were not isolated. Only P. shigelloides and V. cholerae were isolated during the warm weather.

The study identified the quality differences of dried Chinese pomfret (Stromateus chinensis), Bombay duck (Harpodon nehereus) and ribbon fish (Trichiurus haumela) from market and drying center at Kuakata, Patuakhali. The sensory, nutritional and microbial quality of collected dried samples were evaluated following organoleptic quality index, standard AOAC and aerobic plate count methods, respectively. Organoleptically no significant difference found between the samples from market and drying center. The highest moisture content value was obtained in Bombay duck (29.78%) from the market and lowest value in Chinese pomfret (20.06%) from drying center. The highest and lowest protein content was in Chinese pomfret (60.73%) and Bombay duck (47.9%) from drying center and market, respectively. The lipid and ash content varied from 2.85% to 7.20% and 13.98% to 19.57% for all the dried fish species collected from both sites. Microbiologically, higher aerobic plate count (APC), total E. coli count (TEC) and total Salmonella count (TSC) were found in Chinese pomfret at 1.96×106 CFU/g, 3.98×105 CFU/g and 1.62×105 CFU/g from market and lowest count found in Bombay duck at 5.05×105, 2.22×105 and 0.81×105 from drying center, respectively. In ribbon fish no Salmonella sp. was found. Based on the sensory and nutritional quality the study conclude that among three species and two sites studied, the dried Bombay duck from the fish drying center had better quality characteristics than the counter parts. However, higher microbial load especially pathogenic TSC and TEC suggests to improve hygiene and sanitary practices, better market distribution and storage facilities need to be followed by the producers for safe and quality dried fishery products.

Background: In many developing countries, the risk of contracting a food-borne disease is high after consuming contaminated ready-to-eat Street-Vended Foods (SVFs). The main objective of this research was to assess the microbiological quality of SVF groups sold in the Johannesburg Metropolis, South Africa. Methods: A stratified random sampling procedure was used for collecting the ready-toeat SVF samples. Methods prescribed by the International Organization for Standardization (ISO) were used for analyses for aerobic colony count, Enterobacteriaceae count, presence of Escherichia coli O15:H7, detection of Salmonella, Staphylococcus aureus, and Listeria monocytogenes. The bacterial isolates were identified by 16S rRNA gene sequencing. Data analysis was done using IBM SPSS Statistics V25.0. Results: Of the 205 ready-to-eat SVF samples, 85.37% had aerobic growth. The vast majority (78.18%) of the 110 ready-to-eat SVF samples had Enterobacteriaceae growth. From the 110 SVF samples, the prevalence rates of L. monocytogenes, S. aureus, Salmonella spp., and E. coli O15:H7 were 46.36, 31.8, 21.8, and 1.8%, respectively. There was no statistical significant difference (p>0.05) in the prevalence rates of L. monocytogenes, S. aureus, Salmonella spp., and E. coli O15:H7 in the various SVF groups. Conclusion: Based on the findings of this study, the microbial quality and safety of ready-to-eat SVFs sold in the Johannesburg Metropolis remain a serious public health concern. Hence, it is necessary to educate street food vendors and enforce food safety legislation in the street food sector in the country.

Bacteriological characteristics of abattoir effluents (wastewater), abattoir water source, and water bodies receiving abattoir wastewater were investigated in Abuja, Nigeria using the multiple-tube fermentation technique. Source of water to the abattoirs and the usage of water bodies receiving abattoir effluents were determined using questionnaires. Bacterial counts ranged from 4.8 × 106 to 5.8 × 105 /100 mL of total coliform (TC), 8.2 × 104 to 3.2 × 104/100 mL of Fecal coliform (FC), 5.2 × 104 to 2.0 × 104/100 mL of Fecal streptococcus and 1.2 × 104 to 2.0 × 103/100 mL of Escherichia coli for abattoir effluents 6.6 × 105 to 6.0 × 105/100 mL of TC, 6.2 × 104 to 1.8 × 104/100 mL of FC, 1.8 × 104 to 6.0 × 103/100 mL of F. streptococcus, and 4.8 × 103 to 6.6 × 102/100 mL of E. coli for water bodies receiving abattoir effluents 100 m downstream. TC bacteria counts for abattoir effluents exceeded recommended limit for discharge into surface water in Nigeria. No significant difference (P<0.05) was observed between bacterial counts of abattoir effluents and receiving water bodies 100 m downstream: an indication of contamination of receiving water bodies by abattoir effluents and possible public and environmental health hazards.

Contamination with Escherichia coli O157:H7 and Salmonella have called into question the safety and microbial quality of bagged ready-to-eat leafy greens. This study expands on previous findings that these goods have high total bacteria counts (TBC) and coliform counts, variation in counts among different lots, that Escherichia coli is present, and disparities in counts when bags are top or bottom sampled. Nearly 100 bags of baby spinach and hearts of romaine lettuce from a single brand were subjected to both top and bottom sampling. Product was blended, and a portion serially diluted and plated to obtain TBC. Total coliform and E. coli levels were estimated by the most-probable-number (MPN) technique with ColiComplete discs. Top-sampled TBC from bags of baby spinach (48 bags, 13 different lots) ranged from 3.9 to 8.1 log CFU/g and bottom-sampled TBC ranged from 4.0 to 8.2 log CFU/g, with 52% of the bags (or 39% of the lots) producing TBC higher in bottom samples. For hearts of romaine (47 bags from 19 different lots), top-sampled bags had TBC ranging from 2.4 to 7.0 log, and bottom-sampled bags had TBC from 3.3 to 7.3 log, with 64% of the bags (or 63% of the lots) showing higher TBC in bottom samples. However, we are unable to reject the hypothesis that the top and bottom samples from either commodity contain the same TBC (P ≥ 0.08). No E. coli was detected and total coliform bacteria counts were, with few exceptions, ≥210 MPN/g, irrespective of TBC. In general, lots with the most number of days before the printed “use-by” date had lower TBC. However, the R2 values for either baby spinach (0.4085) or hearts of romaine (0.2946) suggest that age might not be a very good predictor of higher TBC. TBC varied widely between lots and even more so within same-lot samples, as indicated by the sum of squares results. This finding, along with higher TBC in bottom samples, suggests further consideration when a microbiological sampling scheme of bagged produce is designed.

The microbiological and nutrient quality of pool barb (Jatpunti), Puntius sophore collected from three local markets - Ananda bazaar, New Market and Polashi bazaar of Dhaka, Bangladesh were assessed. The investigation confirmed more or less higher microbial load in Ananda bazaar samples than the others. The microbial quality varied with market sources as total bacterial counts (TBC), total coliform counts (TCC) and faecal coliform counts (FCC) occurred in all storage period and that crossed the acceptable limit. All studied parameters except E. coli were significantly different at 7 days and 30 days of storage period and a significant difference (p < 0.05) was found in case of E. coli for all storage period where every 30 days storage showed one log reduction of bacterial count from every initial stage. The Salmonella shigella (SS) and Vibrio spp. were identified in all samples of studied local markets. An inverse relation was found between moisture and protein, moisture and fat, whereas the moisture content was increased after 30 days of freezing storage. This study concludes that the quality of P. sophore (Jatpunti) available in the studied three local markets of Dhaka city were neither satisfactory level nor to the standard for normal consumption.Bangladesh J. The study aims to determine the quality of clean water in the villages of Wainitu, Batumerah, Amahusu and Halong by comparing the result of water quality analysis with water quality standard. Water quality analysis includes Physiscal, Chemical, and Microbiological parameters. This research uses descriptive method, this method describes systematics, accurate about facts and characteristic of the quality of clean water of each research location. The results showed that the source of clean water in the village of Batumerah did not meet the standard of clean water quality standards indicated by the number of E. coli and the high total coliform.  Keywords: standard quality of clean water, water quality, Wainitu, Batumerah, Amahusu and Halong villages   ABSTRAK Penelitian yang bertujuan untuk menetapkan kualitas air bersih di Desa Wainitu, Batumerah, Amahusu dan Halong, dengan membandingkan hasil analisis kualitas air dengan standar baku mutu air bersih. Analisis kualitas air meliputi parameter fisika, kimia dan mikrobiologi. Penelitian ini menggunakan metode deskriptif; metode ini menggambarkan sicara sistematis, akurat, fakta dan karakteristik mengenai kualitas air bersih di masing-masing lokasi penelitian. Hasil penelitian menunjukkan bahwa sumber air bersih di Desa Batumerah tidak memenuhi standar baku mutu air bersih yang ditunjukkan oleh jumlah E. coli dan total Koliform yang tinggi. Medicinal plants may be exposed to a wide range of microbial contamination during pre- and post- harvest stages and they can present high microbial counts. In this study, the microbial quality of 44 samples of dry herbs namely: mint (Menthaspp.), lemon balm (Melissa officinalis), summer savory (Satureja hortensis), zataria (Zataria multiflora), Indian valerian (Valeriana wallichii), their brewing and extracts were analyzed. Total count using plate count agar medium (PCA), coliform count by Violet Red Bile Agar (VRBL), Enterobacteriacea by Violet Red Bile Glucose (VRBG) were evaluated. Medium Baird-Parker agar (BP) medium and Tryptone Bile X-Gluc (TBX) medium were used for the isolation and enumeration of Staphylococcus aurous and E. coli spp. respectively. Furthermore, Xylose Lysine Deoxycholate agar medium (XLD) and Bismuth Sulfite Agar medium(BSA) were used for detection of Salmonella spp. Fungal and mold contamination was assessed using yeast extract glucose chloramphenicol agar. The results showed that the contamination of the samples with total count (100%) and Enterobacteriaceae (85%), total coliform (83%), mold and yeast (98%) and E. coli ssp. (2.27) were detected, including in the study samples the absence of pathogenic bacteria like Staphylococcus aurous, Salmonella spp. Moreover, the extract had a lower microbial load in comparison to dry herb samples. Also, the lowest and the highest of contamination rates were observed for Indian valerian and zataria, respectively. According to the results, there is a need to control the environmental conditions and improve hygiene in the production process; even more, it is recommended to choose a suitable decontamination method for disinfection during packing medicinal plants and during post-packing manipulation and transport.

Abstract Background If hygiene practices along the beef processing nodes at small and medium enterprise (SME) slaughter houses and butcheries are not observed, they may pose a health risk due to microbial contamination. In SME slaughterhouses and butcheries, the risk may be higher due to transmission of foodborne pathogens. This study determined the hygienic practices and microbial quality risk among meat handlers (MH) in SME slaughterhouses and butcheries. Methods Assessment of microbiological quality of beef was carried out at slaughter houses and butcher shops in the districts of Western, Central and Eastern regions of Uganda. A cross sectional study was conducted from June 2017 to January 2018 using observation checklists to record unhygienic practices among the various actors. Microbial load at slaughter and butchery was determined from a total of 317 swab samples collected from carcass, tools, protective clothing and hands of meat handlers. The microbiological quality of beef was evaluated using standard microbiological methods. The samples were inoculated into differential and selective media. Results Butcheries had the highest microbial load on beef carcass ranging from 4.76 log 10 cfu/cm 2 to 7.90 log 10 cfu/cm 2 Total Viable Counts (TVC) while Total Coliform Counts (TCC) ranged from 1.42 log 10 cfu/cm 2 to 3.05 log 10 cfu/cm 2 , E. coli ranged from 0.68 log 10 cfu/cm 2 to 1.06 log 10 cfu/cm 2 and Staphylococcus aureus ranged from 3.25 log 10 cfu/cm 2 to 4.84 log 10 cfu/cm 2 . Salmonella was absent in all the samples analysed. Results of overall microbial quality of beef in Uganda indicated that only TCC (1.60±0.26 log 10 cfu/cm 2 ) of the beef carcass samples at slaughter houses was not significantly above the safe level (p = 0.693). Overall microbial load (TVC, TCC, E. coli and S. aureus ) at butcheries were significantly (p < 0.05) above the safe level. Butcheries of Mbale district had the highest percentage (70%) of beef carcass samples above the TCC safe levels whereas butcheries of Mbarara district had the highest percentage (40%) of beef carcass samples above the E. coli safe levels. TVC from hands and clothes at butchery across the three study districts varied significantly (p=0.007) with the highest counts (7.23 log 10 cfu/cm 2 ) recorded from personnel clothes and lowest (5.46 log 10 cfu/cm 2 ) recorded from hands. On the other hand, swab samples picked from chopping board and working table at the butchery did not show significant variation in TVC, TCC, E. coli and S. aureus microbial loads across the three study districts. Conclusion Hygienic handling of carcasses after slaughter is critical in preventing contamination and ensuring meat safety in informal meat trading sectors in Uganda. Handling practices of beef at Ugandan slaughterhouses and butcheries are not hygienic hence not up to standard and they contribute to microbial contamination of beef posing a risk to consumers. The distribution stage is the most critical period, during which the quality of meat can easily be compromised.

Universal preenrichment broth (UPB) was compared with modified Escherichia coli broth with novobiocin (mEC+n) for enrichment of Shiga toxin–producing E. coli O157 and O26, and with buffered peptone water (BPW) for preenrichment of Salmonella enterica. Ten strains each of the three pathogens were inoculated into beef and radish sprouts following thermal, freezing, or no treatment. With regard to O157 and O26, UPB incubated at 42°C recovered significantly more cells from inoculated beef than UPB at 35°C and from radish sprout samples than UPB at 35°C and mEC+n. With regard to Salmonella, UPB incubated at 42°C was as effective as UPB at 35°C and BPW at recovering cells from beef and radish sprout samples. No significant difference was noted between the effectiveness of UPB at 42°C and UPB at 35°C or BPW in the recovery of Salmonella from 205 naturally contaminated poultry samples. By using UPB at 42°C, one O157:H7 strain was isolated from the mixed offal of 53 beef samples, 6 cattle offal samples, and 50 pork samples all contaminated naturally, with no pathogen inoculation. The present study found that UPB incubated at 42°C was as effective as, or better than, mEC+n for enrichment of O157 and O26 and comparable to BPW for preenrichment of Salmonella. These findings suggest that a great deal of labor, time, samples, and space may be saved if O157, O26, and Salmonella are enriched simultaneously with UPB at 42°C.
