Entomopathogenic nematodes (EPN) are one of the soil worms that have been widely used as a natural pest control. EPN has its pathogenic capability because of the mutualistic interactions between nematodes and symbiotic bacteria inside the digestive tract of nematodes. Symbiotic bacteria capable of producing exoenzymes that are toxic to insects. The isolation of symbiotic bacteria accomplished by infection of obtained EPN (Belik II isolate) into Tenebrio molitor larvae. Symbiotic bacteria were isolated from the hemolymph of dead larvae on NBTA media. Isolation of symbiotic bacteria was successfully obtained two morphologically distinct bacteria: B 3.1 isolate and B 4 isolate. Both bacteria were further identified using PCR analysis of the 16S rRNA gene. Based on the sequencing results, the B 3.1 isolate was in accordance with Acinetobacter pittii strain ATCC 19004, while B 4 isolate was in accordance with bacteria Enterobacter aerogenes strain KCTC 2190. The characterization of B 3.1 isolate was shown to have similarities with Acinetobacter sp., i.e.: gram-negative, non - motile, rod-shaped, and some other characteristics of biochemical tests. While the characterization of B 4 isolate was shown to have similarities with E. aerogenes i.e.: gram-negative, rod-shaped, motile, and some other characteristics of biochemical tests. These findings will be the potential to be applied as biological agents in pest control.

Abstract Background Entomopathogenic nematodes (EPNs) harboring symbiotic bacteria are one of the safest alternatives to the chemical insecticides for the control of various insect pests. Infective juveniles of EPNs locate a target insect, enter through the openings, and reach the hemocoel, where they release the symbiotic bacteria and the target gets killed by the virulence factors of the bacteria. Photorhabdus with Heterorhabditis spp. are well documented; little is known about the associated bacteria. Main body In this study, we explored the presence of symbiotic and associated bacteria from Heterorhabditis sp. (IISR-EPN 09) and characterized by phenotypic, biochemical, and molecular approaches. Six bacterial isolates, belonging to four different genera, were recovered and identified as follows: Photorhabdus luminescens, one each strain of Providencia vermicola, Pseudomonas entomophila, Alcaligenes aquatilis, and two strains of Alcaligenes faecalis based on the phenotypic, biochemical criteria and the sequencing of 16S rRNA gene. Conclusion P. luminescens is symbiotically associated with Heterorhabditis sp. (IISR-EPN 09), whereas P. vermicola, P. entomophila, A. aquatilis, and A. faecalis are the associated bacteria. Further studies are needed to determine the exact role of the bacterial associates with the Heterorhabditis sp.

Nine unidentified Gram-positive, lipophilic corynebacteria were isolated from clinical and food samples and subjected to a polyphasic taxonomic analysis. The bacteria were distinguished from Corynebacterium species with validly published names by biochemical tests, fatty acid content and whole-cell protein analysis. Comparative 16S rRNA gene sequence analysis demonstrated unambiguously that the nine strains were related phylogenetically to the species ‘Corynebacterium tuberculostearicum’ and represented a distinct subline within the genus Corynebacterium. On the basis of both phenotypic and phylogenetic evidence, the formal description of Corynebacterium tuberculostearicum sp. nov. is proposed. The type strain of C. tuberculostearicum is Medalle XT (=LDC-20T=CIP 107291T=CCUG 45418T=ATCC 35529T).

The study was intended for molecular detection of S. aureus isolated from raw cow’s milk. A total of 20 milk samples were collected from different upazila markets of Jamalpur, Tangail, Kishoreganj and Netrokona districts of Bangladesh. Milk samples were cultured onto various culture media for the isolation of bacteria. The isolated bacteria were identified by studying cultural properties on different selective media, biochemical tests, and finally by PCR. Out of 20 samples, 15 (75%) milk samples were found to be positive for S. aureus. S. aureus specific 16S rRNA gene was amplified from all isolates and identified as S. aureus. Antimicrobial sensitivity test was carried out to ascertain the susceptibility of the organism to various antibiotics. Its results showed that the S. aureus isolates were resistant to amoxicillin (100%), erythromycin (73.33%) and tetracycline (73.33%) but sensitive to azithromycin (93.33%), ciprofloxacin (93.33%), gentamicin (100%), norfloxacin (86.67%) and streptomycin (86.67%).

Sponges (Porifera) harbour diverse microorganisms which can be the potential source for microbial enzymes such as lipase. In this study, moderately halophilic lipase producing bacteria were isolated from sponges tissues collected near Balok, at Pahang coastal water. Out of 70 isolates that grew on tributyrin agar plate, only 7 isolates had produced clear zones surrounding their colonies. Out of these, 5 isolates appeared to be gram-positive rod; meanwhile, the other 2 isolates were gram-negative rod in morphologies. These isolates were subjected to several biochemical tests i.e., oxidase, gelatin hydrolysis, lactose fermentation, citrate and motility test, and 16S rRNA gene amplification and sequencing. The results from 16S rRNA sequencing showed that 2 isolates (NHTH 6B and NHTH 28A) were highly similar (>97%) with Paenibacillus illinoisensis; isolate NHTH 26A with Stenotrophomonas pavanii; and isolate NHTH 29A with Enterobacter aerogenes. Phylogenetic analysis on selected isolates (NHTH 6B, NHTH 26A, NHTH 28A and NHTH 29A) with other species from the database showed high bootstrap values of above 50%. This showed that diverse phyla of lipase producing bacteria were isolated from the sponge collected from Pahang coastal water. In the isolation of industrial important species, the presence of pathogenic group of microorganism in this sponge could indicate issues on water quality and safety in this area.

A Gram-negative, rod-shaped, motile, non-spore-forming bacterium, designated strain A62-14BT, was isolated from a constant-temperature, spring-fed, freshwater lake. On the basis of the complete 16S rRNA gene sequence, strain A62-14BT was shown to belong to the class Gammaproteobacteria, being most closely related to Rheinheimera sp. HTB082 (96.2 % sequence similarity), Rheinheimera baltica (95.01 %), Rheinheimera pacifica (96.35 %), Rheinheimera perlucida and Alishewanella fetalis (95.9 %). The major fatty acids (C16 : 1 ω7c, 38.56 %; C16 : 0, 19.04 %; C12 : 0 3-OH, 12.83 %; C18 : 1 ω7c, 7.70 %) and the motility of strain A62-14BT support its affiliation to the genus Rheinheimera. The salt intolerance of strain A62-14BT, together with the results of other physiological and biochemical tests, allowed the differentiation of this strain from the three species of the genus Rheinheimera with validly published names. Therefore strain A62-14BT represents a novel species of the genus Rheinheimera, for which the name Rheinheimera texasensis sp. nov. is proposed. The type strain is A62-14BT (=ATCC BAA-1235T=DSM 17496T). The description of the genus Rheinheimera is emended to reflect the halointolerance and freshwater origin of strain A62-14BT.

Racheal A, Kasanga CJ, Byarugaba DK. 2018. Identification and characterization of Flavobacteriaceae from farmed Oreochromis niloticusand Clarius garieoinus. Bonorowo Wetlands 2: 42-50. Bacteria under family Flavobacteriaceae (in this study were also referred to as Flavobacteria) are important pathogens of fish, people, many other animals and plants. In this study, Flavobacteria from Nile tilapia (Oreochromis niloticus) and African catfish (Clarius gariepinus) were identified and characterized from the selected farms in Uganda. Gill and skin swabs were obtained from a total of 119 fish from 19 farms and were dissected aseptically to sample internal organs. The samples were inoculated onto Sheih media and incubated at 25°C for 48 hours. The suspected isolates were identified by colon characteristics, conventional biochemical tests and API 20 NE kits. The isolates were grouped into eight based on colon characteristic similarity. One isolate was selected per group for 16S rRNA gene sequencing and identified using the EZbiocloud.net ID software. Phylogenetic analysis of selected isolates was performed using the 16S rRNA gene sequences in BioEdit and MEGA 7.0.2 software. Basing on extrapolation of sequence analysis of the selected isolates, out of the 86 isolates, Myroides marinus was the most predominant species taking up 4 of the 8 groups (60 isolates) in 13 farms. The rest of the groups comprised of; Acinetobacter pitti, one group (6 isolates) in 6 farms, Chryseobacterium gambrini 2 groups (3 isolates) in 3 farms and one isolate was unidentified, in 3 farms. However, a total of 16 isolates did not grow on sub culturing. Phylogenetic analysis indicated that M. marinus isolates grouped with other M. marinus isolates from gene bank with significant intra-species diversity which was also observed with C. gambrini isolates. All the sampled farms had at least one isolate of a Flavobacterium from Tilapia and/or Catfish. Pathogenicity studies should be conducted on the isolates to establish their importance as fish pathogens and transmission dynamics so that an appropriate control measure can be recommended.

Photorhabdus bacteria are symbiotically associated with the infective juveniles of entomopathogenic nematodes of the genus Heterorhabditis. These bacteria after infecting a susceptible host, produces a host of lethal toxins that killthe insect hosts within 24 to 72 hours. Consequently they have emerged and are trending as excellent biological control agents against insect pests of agricultural crops. Therefore, this study aim to isolate, identify and test the pathogenicity of Photorhabdus bacteria isolated from Heterorhabditis bacteriophora, entomopathogenic nematode from Kashere, Gombe State, Nigeria. To this end phenotypic and biochemical tests were conducted. The tests conducted showed that the isolate exhibited characteristics similar to those of Photorhabdus bacteria that have been identified. Last instar larvae of G. mellonella exposed to different concentrations of the bacterial isolate showed some level of susceptibility of the larvae to the bacterial isolate confirming its biological control potential.

The genus Nocardiopsis is well known to produce secondary metabolites especially antibacterial bioactive compound. Isolation and characterization of bioactive compounds producing novel isolates from unusual habitats are crucial. The present study was aimed to explore Didwana dry salt lake of Rajasthan state in India for the isolation and characterization of actinomycetes. The isolated actinomycetes isolates were characterized based on culture characteristics, biochemical tests and 16S rRNA gene sequencing. The 16S rRNA gene sequence analysis revealed that all the five isolates inhabiting soil of the said dry salt lake of Didwana, Rajasthan belonged to four species of Nocardiopsis viz., N. synnemataformans, N. potens, N. prasina and N. dassonvillei subsp. albirubida. The molecular identification based on 16S rRNA gene sequences was found accurate and robust. The phylogram generated through multiple sequence alignment of all the test isolates of Nocardiopsis revealed that the isolates aroused from a single branch and validated monophyletic association. The present study is the first report of exploring Nocardiopsis isolates from the dry salt lake. These characterized Nocardiopsis isolates isolated from Didwana dry salt lake habitat are novel stains and can be of significance in the detection and utilization of novel bioactive compounds.

Almost all regions in Nusa Tenggara Timur East Nusa Tenggara (NTT) Province Indonesia are endemic areas of Haemorragic Septicaemia (HS), which is caused by Pasteurella multocida  (P. multocida ) Serotypes B: 2. The fragment  pfhaB1gene is one of the virulence factors of P. multocida.The objective of this study was to determaine the phylogenetic, homology of P. multocidapfhaB1gene fragment of isolatedfrom Buffalo and Cattle in NTT. The P. multocida isolateswere re-cultured and further microscopic examined the biochemical tests, PCR, sequencing, homology, and phylogenetic relatedness test. P. multocida was observed as gram negative, coccobacilus, no growth on MacConkey Agar, does not produce H2S and gas, nonmotile and indole positive, does not produce urease enzymes, does not use citrate as a carbon source, does not ferment maltose and lactose but it does ferment glucose, sucrose and mannitol. ThepfhaB1gene fragmentfrom buffalo and cattle NTT isolates and also Katha strain vaccine showed DNA band 506 bp. P.multocida isolates from buffalo and cattle in NTT have 91% - 99% score homology with the comparative isolate. The isolate P. multocida from buffalo and cattle in NTT are in one cluster and their phylogenetic relatedness is close to isolates from Iran and India. It is concluded that the  pfhaB1gene fragmentof P. multocida from buffalo and cattle isolates have phylogenetic relatedness close and homolog with the other comparative isolates.    Keywords: Haemorrhagic Septicaemia; Nusa Tenggara Timur; Pasteurella multocida;  pfhaB1gene
