ABSTRACT Introduction This study aimed at evaluating and comparing the antibacterial activity of six types of dental luting cements on Streptococcus mutans and Lactobacillus acidophilus using the agar diffusion test (ADT) and the direct-contact test (DCT). The antibacterial activity in ADT was measured based on the diameter of the zone of inhibition formed, whereas in DCT the density of the bacterial suspension was measured. The lower the density of the suspension, the more antibacterial activity the cement possesses. Materials and methods Agar diffusion test was carried out on the bacteria. After an incubation period of 24 hours, the plates were checked for the presence of zone of inhibition. In DCT the cement was mixed and applied. Once the cement was set, bacterial suspension and brain–heart infusion medium was poured and incubated for 24 hours. After 24 hours, the plate was placed in the enzyme-linked immunosorbent assay plate reader, which measured the optical density of the fluid. The first set of data was recorded approximately 1 hour after incubation. Overall, three sets of data were recorded. Additional experiments were performed on set test materials that were allowed to age for 24 hours, 1 week, 1, 3, and 6 months. Results When using ADT only two cements zinc oxide eugenol (ZOE) and zinc polycarboxylate (ZPC) cement showed antibacterial activity against the test organisms. When using DCT, all cements showed some amount of antibacterial activity. Zinc oxide eugenol and ZPC cement showed highest amount of antibacterial activity against S. mutans and L. acidophilus respectively. Conclusion Within the limitations of study, ZOE cement and ZPC cement were most effective against the tested microorganisms followed by the newer resin cement. The glass ionomer cement was the weakest of all. Clinical significance Patients with high caries index can be treated more effectively using the abovementioned cements. Objectives: To evaluate the antibacterial activity of black pepper, Indian bay leaf, cinnamon, and cumin against Streptococcus mutans and Lactobacillus acidophilus in-vitro and to determine their minimum inhibitory concentration (MIC). Materials and Methods: The spices (cinnamon, cumin, Indian bay leaf, and black pepper) were obtained from local market, were dried and powdered. Solvent extracts were prepared with methanol by maceration, followed by filtration and evaporation. The antimicrobial activity was assessed using cup plate diffusion method, followed by determination of MIC of the extracts. Statistical analysis was performed using one-way analysis of variance and Tukey’s post hoc test was used for pairwise comparison. P < 0.05 was considered statistically significant. Results: All the four extracts showed significant antimicrobial activity. Cinnamon demonstrated maximum activity against S. mutans (zone of inhibition of 18.1 mm ± 0.30) and L. acidophilus (zone of inhibition of 17.9 mm ± 0.44) with the least MIC against the organisms (<0.05 mg/ml). Conclusion: All the spice extracts tested demonstrated significant antibacterial activity against S. mutans and L. acidophilus. On comparison of the antibacterial activities of all the four extracts, cinnamon extract emerged as the potent agent.

To conduct a study regarding the antibacterial activity of essential oils against bacteria causing Caries. Essential oils are distillates of the volatile compounds of a plant’s secondary metabolism and may act as photoprotective agents. Their curative effect has been known since antiquity. It is based on a variety of pharmacological properties which are specific for each plant species. The mouth contains a variety of oral bacteria, but only a few species of bacteria are believed to cause dental caries. Antibacterial activity of the three essential oils, Rosemary oil, Holy basil oil, Thyme oil was screened against Streptococcus mutans, using disc diffusion technique. The rosemary oil was more effective against Streptococcus mutans with a zone of inhibition of 52 mm diameter (at concentration 200 µl), Rosemary oil showed a zone of inhibition of 44 mm diameter and with thyme oil, the zone diameter was 30 mm. The results of this study showed that the essential oils at different concentrations exhibited antibacterial activity against the bacterial species tested.

Introduction: Lactobacillus acidophilus is one of the bacteria causes dental caries. The previous study has shown that Kaempferia galanga extract has a potential to inhibit the growth of Lactobacillus acidophilus.Objective: To determine the antibacterial effect of Kaempferia galanga extract to Lactobacillus acidophilus.Methods:Kaempferia galanga is extracted in 3 different solvents:dichlormethane, ethanol, and aquades. For each solvent, 0.2 μl Kaempferia galanga extractdroped into 6 mm steril paper dics. 0.1 ml Lactobacillus acidophilus inoculated on MRS agar. Each disc contains extract were impragnated into the agar media, then incubated at 370C for 24 hours, and inhibition zone measured.Results: Mean scores of Kaempferia galanga extract in 3 different solvents are: Kaempferia galanga (dichlormethane) is 1.6400; Kaempferia galanga (ethanol) is 1.7440; Kaempferia galanga extract is 1.6600; boiled Kaempferia galanga is 1.7000. Using Mann-Whitney Test, the results are: negative controls have no inhibition effect on Lactobacillus acidophilus compaired to Kaempferia galanga extract, comparation of those 4 Kaempferia galangal treatments shows no significant difference, those 4 Kaempferia galanga treatments compaired to erythromycin antibacterial effect shows significant difference, otherwise 4 Kaempferia galanga treatments compaired to penicillin shows no significant difference except Kaempferia galanga (ethanol).Conclusions: Kaempferia galanga extract can kill Lactobacillus acidophilus. Inhibition effect of Kaempferia galanga extract has no significant difference to penicillin but lower inhibition effect than erythromycin. The Kaempferia galanga extracts showed better antibacterial activity than penicillin.

This study evaluated the effect of incorporating silver nanoparticles (AgNPs) into conventional orthodontic adhesive on its antibacterial activity and the shear bond strength (SBS) to stainless steel orthodontic brackets. Thirty-four extracted premolars were randomly allocated into two groups (n = 17). Orthodontic adhesive (Transbond XT, 3M Unitek) was blended with AgNPs (50 nm, 0.3% w/w) to form a nano-adhesive. In order to bond stainless steel twin brackets (0.022-inch, American Orthodontics), Transbond XT (n = 17) and nano-adhesive (n = 17) were used in each group, respectively, after acid etching (37% phosphoric acid, 30 s) and rinsing with water (15 s). SBS and the adhesive remnant index (ARI) scores were recorded. Antibacterial activity against Streptococcus mutans in both groups after 24 h and 30 days was assessed (Disc agar diffusion test) and the inhibition zone diameter around each specimen was measured and recorded. Adding AgNPs significantly (p = 0.009) reduced the mean (SD) SBS in the nano-adhesive group [10.51(7.15) MPa] compared to Transbond XT [17.72(10.55) MPa]. The ARI scores on the Transbond XT and nano-adhesive showed no statistically significant difference (p = 0.322). Nano-adhesive with AgNPs showed significant antibacterial activity against Streptococcus mutans at 24 h and 30 days (p < 0.001). In both groups, no significant decline in the zones of inhibition was detected after 30 days (p = 0.907). The findings suggest that SBS decreased after incorporation of AgNPs [0.3% (w/w)], but was still above the recommended SBS of 5.9–7.8 MPa. The nano-adhesive showed significant antibacterial activity which did not change much after 30 days.

AIM: This study investigated the antibacterial efficacy of five plant extracts, as well as the combinations of the two most effective plant, extracts either with or without commercial varnish (MI varnish) on the in vitro growth of Streptococcus mutans and Lactobacillus acidophilus in comparison to MI varnish using agar disk diffusion and broth dilution methods. METHODS: Methanolic extractions of five plants (Cinnamon, Turmeric, Ginger, Clove and Black seed,) were tested against the growth of the two oral pathogens. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined for the two most effective extracts, and their combinations with different ratios were evaluated against the growth of the two oral pathogens, followed by incorporating the two effective plants or each into commercial MI varnish to be assessed against the oral pathogens in comparison to MI varnish. RESULTS: Only Cinnamon and Clove produced inhibition zones against Streptococcus mutans and Lactobacillus acidophilus growth. MIC for the two plants showed equal antimicrobial activity against Streptococcus mutans, while Cinnamon had a higher sensitivity to Lactobacillus acidophilus than Clove. A mixture of Cinnamon and Clove in a ratio 1:2 exhibited the highest antibacterial activity. Integration the mixture of both plants into MI varnish in a ratio of 1:1:1 presented the highest antibacterial activity. Meanwhile, the lowest one was recorded for the MI varnish alone. CONCLUSION: Methanolic extract of Cinnamon and Clove has considerable antimicrobial activity against Streptococcus mutans and Lactobacillus acidophilus and a new tool for minimally invasive and adhesive dentistry avenues.

Abstract Introduction The main aim of this study is in vitro evaluation of the antimicrobial efficacy of different herbal products, that is, propolis, garlic, neem, aloe vera, and rosemary, against Bacillus subtilis, Staphylococcus aureus, and Enterococcus faecalis using agar diffusion test. Materials and Methods In this study, total of 42 plates were prepared, 10 each of B. subtilis, S. aureus, and E. faecalis and 12 as test control (6 as positive and 6 as negative control). The effectiveness of five herbal products was ascertained by agar diffusion method against B. subtilis, S. aureus, and E. faecalis. Cultures of these test organisms were maintained on selective media slants in a test tube to collect sufficient number of microbial colonies for evaluation. The cultures were divided into three groups based upon microbes that were lawn cultured, respectively: In group A, 10 petri plates were having growth of B. subtilis; in group B, 10 petri plates were having growth of S. aureus; and in group C, 10 petri plates were having growth of E. faecalis. In all these 30 petri plates, five different herbal product discs were placed and these discs were designated as A (aloe vera), P (propolis), N (neem), R (rosemary), and G (garlic). Among remaining 12 petri plates, 4 petri plates were used as control (2 for positive and 2 for negative) for each of the bacteria. Results Propolis and rosemary showed maximum zone of inhibition against B. subtilis. Garlic, neem, and aloe vera showed maximum zone of inhibition against S. aureus. Conclusion All the herbal products showed zone of inhibition against S. aureus, B. subtilis, and E. faecalis.

Chitosan beads with colloidal silver nanoparticles inclued in the polymer matrix have been obtained by the introduction of chitosan into an acidified nanosilver sol. Dual interconnection of drops of the resulting solution was then carried out by ionotropic gelation at the first stage and covalent crosslinking of the polymer matrix with adipic acid at the second stage. The surface morphology of the obtained beads was studied by scanning electron microscopy. Data of Fourier transform IR spectroscopy confirmed the formation of covalent bonds between chitosan and adipic acid. The antibacterial activity of obtained beads against S. aureus and E. coli was evaluated using agar diffusion test. It was shown that the сhitosan beads modified with nanostructured silver exhibited an antibacterial effect against the tested strains, and they can be used as a basis for creating biodegradable wound healing dressings with a prolonged antibacterial effect. chitosan, silver nanoparticles, antibacterial activity, wound dressings This work was supported by the "Russian Academic Excellence Project 5-100". AbstractOne of the most challenging complications of total knee arthroplasty (TKA) is periprosthetic joint infection (PJI). There is growing evidence of a good anti-infective effect of intrawound vancomycin powder in total joint arthroplasty. At the same time, various different locally applied substances have become popular in total joint arthroplasty. The objective of this study was therefore to investigate a possible inhibition of the bactericidal effect of vancomycin by tranexamic acid, adrenalin, lidocaine, or dexamethasone. The bactericidal effect of vancomycin was quantified using the established method of the agar diffusion test. The plates were incubated with Staphylococcus aureus or Staphylococcus epidermidis and four wells were stamped out. The wells were filled with vancomycin alone, the tested substance alone or a mixture of the two. The fourth well remained empty as a control. The plates were incubated overnight at 37 °C and the zone of inhibition in each field was measured on the next day. All tests were run three times for each pathogen and mean values and standard deviations of the measurements were calculated. Differences between the substances were tested using the t-test at a level of significance of 0.05. The bacterial growth was homogeneous on all plates. The baseline value for the zone of inhibition of vancomycin was on average 6.2 ± 0.4 mm for Staphylococcus aureus and 12 ± 0.3 mm for Staphylococcus epidermidis. In all other substances, no inhibition was detected around the well. The combination of vancomycin and each other substance did not show any different result compared to vancomycin alone. The bactericidal effect of vancomycin on staphylococci is not altered by tranexamic acid, adrenalin, dexamethasone, or lidocaine in vitro.
