Phytochemical and Antibacterial Analysis of the Formulated Cream of Black Cumin Honey

This study reports the bioactive compound of black cumin honey and investigates the antibacterial activity against Propionibacterium acnes and Pseudomonas aeruginosa. HPLC and GC-MS methods were used to analyze the bioactive compounds of the black cumin honey. The formulated cream of black cumin honey was prepared in an oil-in-water type cream with various concentrations of 1%, 5%, 10%, 15%, 20%, and 30% (w/w). Antibacterial effects against Propionibacterium acnes and Pseudomonas aeruginosa were examined by the disc diffusion method. Physical stability of formulated cream included organoleptic, homogeneity, and pH measurement test. The result of GC-MS analysis of black cumin honey showed 25 compounds: organic fatty acids, saccharides, and amino acid. HPLC analysis of flavonoid content showed quercetin as the bioactive compound at a concentration of 0.223 mg/g. Disc diffusion analysis showed great inhibitory potential against P. acnes and P. aeruginosa in 30% concentration with d = 11.2 mm and 10.8 mm, respectively. It can be concluded that the formulated cream of black cumin honey showed a great potential application as antibacterial agents of P. acnes and P. aeruginosa.

Black cumin seeds are known to have beneficial contents as antibacterial. This study aimed to determine the effectiveness of black cumin seed extract (Nigella sativa) on the growth of Klebsiella pneumoniae and Pseudomonas aeruginosa. The method used was the disc diffusion method. The ethanol extract of black cumin seeds gain by maceration method and was made into various concentration (5, 7.5, 10, 12.5, and 15)%. For positive control, ciprofloxacin disc were used and negative control used distilled water. The research model used was in vitro experimental research. The results showed that ethanol extract of black cumin seeds have antibacterial activity against Klebsiella pneumoniae with inhibition zone diameters (4.5, 6.7, 10.1, 11, 12.8) mm, Pseudomonas aeruginosa with inhibition zone diameters (7.5, 12, 12.6, 13.4, 15.6) mm, positive controls respectively (24.3 and 20.7) mm and negative controls (0). It can be concluded that black cumin seed extract has an inhibiting effect on the growth of Klebsiella pneumoniae and Pseudomonas aeruginosa.

Medicinal Plants are the good source of natural antimicrobial agents. The main aim of present study was to evaluate the antibacterial activity of stem and root of Rauwolfia serpentina against six microorganism, Powdered stem and root of plant were extracted with acetone, chloroform and methanol and streptomycin used as positive control. The antibacterial activity of Rauwolfia serpentine was detected by using disc diffusion method and agar well diffusion method on the following bacteria- Bacillus cereus, Staphylococcus aureus, Bacillus fusiformis, Escherichia coli, Pseudomonas aeruginosa and P. luminescens. The experiment reported that R. serpentina Root methanol extract shown 14.86 ± 1.11 highest antibacterial activity against Pseudomonas aeruginosa through well diffusion method. Whereas root chloroform recorded 13.46 ± 1.28 highest antibacterial activity against E. coli through disc diffusion method, maximum zone of inhibition 22.66±0.52 mm was found for the positive control, streptomycin through well diffusion method. Further studies should be undertaken to reveal the correct mechanism of action of antimicrobial effect to identify the active ingredients which can be used in drug development program.

This study aimed to test the antibacterial activity of n-hexane and methanol extracts of white cempaka flowers (Magnolia alba) against Propionibacterium acnes bacteria. The extract was obtained by using the maceration method for 72 hours. Each extract was tested for its antibacterial properties using the disc diffusion method with the variant concentration of the extract used was 0.05%; 2%; 4%; 6%; and 8%. The results showed that the methanol extract had a higher inhibition zone than the n-hexane extract. In n-hexane extract, the lowest concentration of 0.05% showed an inhibition zone of 6.6 mm and the highest concentration of 8% had an inhibition zone of 9.6 mm. Whereas in methanol extract, the inhibition zone obtained was 7.3 mm at a low concentration of 0.05%, but at the highest concentration of 8% the inhibition zone obtained was 11.6 mm. The effect of extract concentration on the diameter of the inhibition zone formed is very significant (directly proportional), where the higher the concentration used, the larger the diameter of the inhibition zone produced. Antibacterial testing for both extracts showed a weak (< 10 mm) to moderate (> 10 mm) bacterial inhibition, which was similar with the inhibition zone obtained from the positive control (11-11.3 mm; moderate bacterial inhibition).

We have studied the antimicrobial properties of 6-bromoeugenol and eugenol by three strains:Pseudomonas aeruginosa(S1),Escherichia coli(S2) andStaphylococcus aureus(S3). We have determined the minimum inhibitory concentration (MIC) for a range of concentrations using the disc diffusion method. We note that all samples present an antimicrobial activity toward the tested bacterial strains at different concentrations (1, 0.5 and 0.25 mg/ml). The 6-bromoeugenol gives modest activity with (S1) and (S3). Eugenol reacts positively with thePseudomonas aeruginosa(S1) at all concentrations and with theEscherichiacoli(S2) at 0.5 mg/ml. We remark that thePseudomonas aeruginosa(S1) is the more sensitive strain thanEscherichiacoli(S2) andStaphylococcus aureus(S3). We have estimated the activity coefficient which has confirmed the antimicrobial activity of the different samples. So, 6-bromoeugenol has shown his efficiency as antimicrobial agent.

ESBL producing Pseudomonas aeruginosa has been reported to be an important cause of nosocomial infection. The study was undertaken to determine ESBL producing Pseudomonas aeruginosa by phenotypic method and to detect bla OXA-10 ESBL gene by molecular method. A total of 288 wound infection cases attending the in-patient department of Mitford Hospital and Dhaka Medical College Hospital were enrolled for this study. Pseudomonas aeruginosa was isolated following standard procedure and subjected to antimicrobial susceptibility test by disc-diffusion method. Ceftazidime resistant strains were confirmed by MIC by agar dilution method. Confirmed ceftazidime resistant strains were tested for ESBL production by CLSI phenotypic confirmatory disc diffusion test (PCDDT) and bla OXA-10 ESBL gene was identified by employing conventional PCR. Out of 92 Pseudomonas aeruginosa, confirmed ceftazidime resistant strains were 60. PCDDT detected 49 (81.67%) ESBL producers and PCR detected 44 (73.33%) positive strains for bla OXA-10 ESBL gene among 60 ceftazidime resistant strains of Pseudomonas aeruginosa. The study showed high proportion of ESBL producers among ceftazidime resistant Pseudomonas aeruginosa. Bangladesh J Med Microbiol 2018; 12 (1): 15-19

ABSTRACTOne of the natural ingredients that are often used as a medicinal plants is Pangi (Pangium edule Reinw. Ex Blume) plant. The part of pangi that are often used are leaves, which is known have antibacterial activity. North Sulawesi people empirically use this plant as food and to cure various diseases such as treating itching on the skin caused by bacteria found on the skin. The aim of this research was to determine and study the strength of antibacterial inhibition based on the category of inhibition by Davis and Stout of ethanol extract from pangi (Pangium edule Reinw. ex Blume) leaf performed by disc diffusion Kirby and Bauer method againts Staphylococcus aureus, Escherichia coli and Pseudomonos aeruginosa bacterias. The result showed that the ethanol extract from pangi (Pangium edule Reinw. Penelitian ini bertujuan untuk mengetahui kekuatan daya hambat antibakteri berdasarkan kategori penggolongan daya hambat oleh Davis dan Stout dari ekstrak etanol daun Pangium edule Reinw. ex. Blume dengan metode difusi agar (difusi disk Kirby dan Bauer) terhadap bakteri uji Staphyloccocus aureus, Escherichia coli dan Pseudomonas aeruginosa. Hasil penelitian didapati bahwa ekstrak etanol daun Pangium edule Reinw ex. ABSTRACTSponges are multicellular metazoa animals belonging to the Porifera phylum, which has a different structure from other metazoans. The purpose of this study was to determine whether ethanol extracts from the marine organism sponge Ianthella basta have antimicrobial activity against several pathogenic microbes Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans. The extraction method used is maceration with 96% ethanol solvent. The method used is the Diffusion Method (Disc Diffusion Kirby and Bauer). The antimicrobial activity test uses a 6 mm paper disc with 50 µL absorption per disc. The results of crude ethanol extract of Ianthella basta sponge from all test microbes, namely Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans, were seen to provide the greatest inhibitory activity against Staphylococcus aureus bacteria with an average of inhibitory zone of 7.00 mm categorize as intermediate The results obtained showed that the crude extract of the sponge Ianthella basta has antimicrobial activity because it can inhibit the growth of Staphylococcus aureus, Escherichia coli and Candida albicans microbes even though the inhibition zone is categorized as intermediate. Keywords: Ianthella basta, antimicrobial, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans. ABSTRAKSpons adalah hewan metazoa multiseluler tergolong ke dalam filum Porifera, yang memiliki perbedaan struktur dengan metazoan lainnya. Tujuan penelitian ini ialah untuk mengetahui apakah ekstrak etanol dari organisme laut spons Ianthella basta memiliki aktivitas antimikroba terhadap beberapa mikroba patogen Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa dan Candida albicans. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol 96%. Metode yang digunakan yaitu Metode Difusi (Disc Diffusion Kirby and Bauer).  Pengujian aktivitas antimikroba ini menggunakan kertas cakram (paper disc) berukuran 6 mm dengan daya serap 50 µL tiap cakram. Hasil ekstrak kasar etanol Spons Ianthella basta dari semua mikroba uji yaitu Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa dan Candida albicans, terlihat yang memberikan daya hambat paling besar terdapat pada bakteri Staphylococcus aureus dengan jumlah rata – rata zona hambat yaitu 7,00 mm dengan kategori sedang. Hasil penelitian yang diperoleh menunjukkan bahwa ekstrak kasar dari Spons Ianthella basta memiliki aktivitas antimikroba karena mampu menghambat pertumbuhan mikroba uji Staphylococcus aureus, Escherichia coli dan Candida albicans walaupun dengan zona hambat yang  dikategorikan sedang. Kata Kunci : Ianthella basta, antimikroba, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans.

The aim of this study was to determine the important role of Pseudomonas aeruginosa in secondary infection of hospitalized patients, and test the sensitivity of this bacteria to some antibiotics were used in treatment of its infection .90 samples were collected from different cases in Diwanya teaching hospital ,maternity & children hospital .The results show isolation of 29 isolates 0f Pseudomonas aeruginosa (32.2%) .Microscopic examination,cultural characteristic on selective and differential media and biochemical testing were used in the diagnosis of this bacteria.The sensitivity test of this bacteria to some antibiotics was done by using disc diffusion method .Allisolates were resistant to ampicillin (100%),while it gave high sensitivity to ciprofloxacin(2.3%).

Objectives: Carbapenems are among the most powerful anti pseudomonal agents. Since meropenem and doripenem were marketed, there are limited data regarding drug susceptibility testing by routine methods (disc diffusion and Etest) for them. The aim of our study was to compare in vitro activity of the imipenem, meropenem and doripenem against Pseudomonas aeruginosa. Methods: Three hundred and eleven P. aeruginosa strains isolated from respiratory specimens in 170 patients who developed ventilator-associated pneumonia in two intensive care units were collected over a period of 31 months. The susceptibility of all of these isolates to imipenem, meropenem and doripenem were determined by Etest and disc diffusion method. Results: Considering either all of the isolates or only the first isolates recovered per patient (311 and 170 respectively) the susceptibility rate for doripenem was higher than that for meropenem and imipenem. When MICs determined by Etest were converted into interpretative categories (S, I, R) using French (CA-SFM) guidelines, agreement was poor, especially for meropenem and doripenem. The percent of agreement with the disc diffusion method were 90.6% and 89.7% for imipenem, 80.5% and 82.6% for meropenem and 80.5% and 73.3% for doripenem, for the first isolates and all of the isolates, respectively. Errors were mostly minor errors, and the rate of errors was as high as 17.7% and 16.1% for meropenem and 17.7% and 25.7% for doripenem for the first isolates and all of the isolates, respectively Conclusion: The accuracy of disc diffusion using CA-SFM guidelines appears unsatisfactory for all the three carbapenems justifying the adaptation of new guidelines for P. aeruginosa and carbapenems

Molecular docking performed to evaluate the effect of five quinoline derivatives on the MexB protein of Pseudomonas aeruginosa as a potential inhibitor by utilizing the 3D structure of each quinoline compounds (C1, C2, C3, C4, and C5), and the crystal structure of the protein, C4 showed the greatest potential with -31.4 kcal/mol binding energy, and the lowest potential was for C1 with (-18.5 kcal/mol) compared with ciprofloxacin. Fifty samples were collected from different sites from patients who are attending to the medical city of Baghdad and private Dhelal Beirut Center, Baghdad, 36 of the samples were diagnosed as P. aeruginosa by routine culture test and confirmed by VITEK2, and those isolates were subjected to the susceptibility test against carbapenems, carbenicillin, levofloxacin, and erythromycin by disc diffusion method. The isolates that showed resistance to all of four antibiotics were based to evaluate the activity of quinoline derivatives by using the agar well diffusion method, where compounds C4 and C5 showed the highest line of activity as the minimum inhibitory concentration (MIC) was 256 μg/mL, meanwhile, C1 showed the lowest activity with MIC of 1,024 μg/mL.
