Isotopic analyses and chromatographic analysis (phenolic compounds and biogenic amines), can be applied to investigate the functional and nutritional quality in different grape juices and wines. These beverages when produced exclusively from grapes contain bioactive compounds. In this way, the isotopic analysis, as well as the determination of phenolic compounds and biogenic amines were performed with the aim of verify the functional quality of juices and wines produced with Vitis vinifera and Vitis labrusca grapes. The samples that were analyzed consisted of four whole juices, two nectars, two V. vinifera wines, and two V. labrusca wines. Regarding the isotopic analyses, only one nectar, among the beverages studied, presented the addition of sugar from C4 plants. Wines from V. vinifera showed the highest content of biogenic amines and phenols; whereas, the highest content of anthocyanins were found in V. labrusca. The levels of biogenic amines and phenolic compounds were variable between samples, and recommendations for consumers should be made considering several conditions, such as physiological state, age, consumption, among others. Anthocyanins and biogenic amines, as well as isotopic analyses, can be applied as tools to measure the quality of grape derived beverages.

AbstractBackgroundRNAs to be used in transcriptome analysis must be of high quality and pure in order to ensure maximum representation of the expressed genes. RNA isolation is difficult in hazelnut tissues containing large amounts of secondary metabolite, phenolic compounds and the cell wall structure. Commonly used protocols for RNA isolation are those that require a lot of labor and time and also do not allow sufficient RNA isolation when applied to tissues rich in phenolic compounds. This study was aimed to develop an efficient method for isolation of total RNAs from bud of hazelnut to be used in RNA sequencing.Materials and methodsAn optimized new method was successfully applied on three different hazelnuts genotypes (Çakıldak, Palaz, Tombul) and about 25 times higher amount of total RNAs per mg fresh tissues were obtained compared to classical CTAB method. Different methods have been tried for the isolation of RNA from hazelnut tissues and the determination of the quality of the obtained RNAs.ResultsThe quality and quantity of isolalated total RNAs were determined by spectrophotometer, electrophoresis and PCR. This success has been caught without any compromise of purity since A260/A280 ratios ranged from 1.90 to 2.04 and A260/A230 ratios were >2.0 in all purified RNAs.ConclusionThe total RNAs isolated with new protocol was found to be suitable for RNA sequencing and other molecular applications.

The quality of drinking water in Ethiopia is an influential environmental factor of health and water can use as a medium for disease transmission in countries on all continents; all are affected from the poorest to the richest. Moreover, in our globe millions of people are unprotected to unsafe concentration of chemical pollutants in the drinking water. Dichlorohenols are considered as key water pollutants that are destructive to creatures at stumpy concentrations and many of them have been clustered as detrimental pollutants because of their probable to damage human health even in low concentration. This research aimed to determine phenolic compounds from drinking water of Jimma town which is found south west of Ethiopia, by using 4-Aminoantipyridine (AAP). The sample of drinking water before and after treatment was collected for determination of pollutants which classified as raw water (RW), treated water (TW) and system distributed water (SDW). The sample of water was preserved by phosphoric acid and sulfuric acid under pH 4 and distillation was carried out in order to remove sulfur containing compounds by addition of CuSO4 solution. In the analysis of water using 4-Aminoantipyrine the treated water and the system distributed water have been contained 2.73 ppb and 3.64 ppb which had more phenolic compounds as compared to raw water of 1.18 ppb. Therefore, the existence of phenolic compound in both treated and system distributed water of Jimma town drinking water has more phenolic compounds as compared to the permissible level of both Ethiopian Standard, 2 ppb and EPA Standards, 1 ppb.

Determination of Biogenic Amines and Endotoxin in Squid, Musky Octopus, Norway Lobster, and Mussel Stored at Room TemperatureLittle research has been published on the indicators of spoilage in Mediterranean molluscan shellfish and crustaceans. Thus is why we studied changes in the concentrations of endotoxin and four biogenic amines (histamine, putrescine, tyramine and cadaverine) in European common squid (Loligo subulata, Lamarck, 1798), musky octopus (Eledone moschata, Lamarck, 1798), Norway lobster (Nephrops norvegicus, Linnaeus, 1758), and mussel (Mytilus galloprovincialis, Lamarck, 1819) from the Adriatic Sea stored at room temperature for 24 h. Endotoxin load in fresh squid, Norway lobster, and mussel (<1 EU mg-1) indicated good microbiological quality of raw samples. Biogenic amine index (as the sum of histamine, putrescine, tyramine, and cadaverine) correlated well with endotoxin load in squid (r=0.978, p<0.001) and musky octopus (r=0.874, p<0.01). A good correlation was also found between endotoxin and putrescine in Norway lobster (r=0.777, p<0.05). The highest endotoxin load was found in decomposed mussels and was associated with histamine alone. In conclusion, increase in biogenic amine levels is species-specific. Endotoxin analysis could be used for rapid assessment of microbiological quality of cephalopods and shellfish.

One that influences human resource indicators is education. The teacher is a profession as a job of academic specialization in a relatively long time in college. Understanding related to teacher competence is very important to have by a prospective teacher because it can affect the quality of performance as a professional teacher. The teacher's competence is known as pedagogic, professional, social and personality competencies. The issue in this study is how the competency of the teacher of the Department of Geography Education FKIP Unsyiah as a prospective teacher of geography? The purpose of this study was to determine the competence of teachers in the Department of Geography Education FKIP Unsyiah as prospective geography teachers. Quantitative description approach is used in this study to find answers to the issue. The population in this study were students of the Department of Geography Education FKIP Unsyiah class of 2015 and 2016 who had been declared to have passed the Micro Teaching and Magang Kependidikan 3 course totaling 50 people. Because the population is small and can be reached, the determination of the sample using total sampling techniques so that the sample in this study is the whole population. Data collection is done by distributing test questions to respondents. The data was analyzed using the descriptive statistics percentage formula. The results of the study indicate that the level of teacher competence of Geography Education Department students as prospective teachers is in the moderate category, namely as many as 22 respondents (44%). A total of 12 respondents (24%) were in the high category, 15 respondents (30%) were in the low category and 1 respondent (2%) were in the very low category.

The article focuses on examining the influence of fluctuations in annual precipitation amount on the quality of surface waters. Water quality was estimated with data on BOD, COD and phosphate–ion concentration within five selected regions of Ukraine. Analysis of the precipitation data (1991 – 2010) showed different regional trends. Using the statistics, determination of the interconnection between precipitation amount and water resources quality were done. The obtained regularities and associated uncertainties can be used for prediction of changes in water resource quality and as a guide for future adaptation to possible climate change.
