Aim of this study was to evaluate the effects of short-term (7 days) N-acetylcysteine (NAC) at 1,200 mg daily supplementation on muscle fatigue, maximal oxygen uptake (), total antioxidant capacity (TAC), lactate, creatine kinase (CK), and tumor necrotic factor-alpha (TNF-α). Twenty-nine sedentary men (13 controls; 16 in the supplement group) from a randomized control were included. At before and after supplementation, fatigue index (FI) was evaluated in the quadriceps muscle, and performed a graded exercise treadmill test to induce oxidative stress, and as a measure of . Blood samples were taken before exercise and 20 minutes after it at before and after supplementation, to determine TAC, CK, lactate, and TNF-αlevels. Results showed that FI and increased significantly in the supplement group. After exercise decreased the levels of TAC and increased lactate, CK, and TNF-αof both groups at before supplementation. After supplementation, lactate, CK, and TNF-αlevels significantly increased and TAC decreased after exercise in the control group. Whereas the TAC and lactate levels did not change significantly, but CK and TNF-αincreased significantly in the supplement group. Therefore, this results showed that NAC improved the muscle fatigue, , maintained TAC, controlled lactate production, but had no influence on CK and TNF-α.

AbstractPurpose. Investigate free radical production and antioxidant buffering in military pentathletes’ saliva after their performance of a standardized, running-based anaerobic sprint test (RAST). Methods. Seven members of the Brazilian Navy pentathlon team were recruited to perform a running-based anaerobic test (~90 sec). The participants provided samples of saliva before and after the test that were analyzed for biomarkers of oxidative stress such as lipid peroxidation, total antioxidant capacity and the quantity of two specific antioxidants, glutathione and uric acid. Results. The lipid peroxidation increased ~2 fold after RAST, despite an increase in total antioxidant capacity (46%). The concentration of reduced glutathione did not change, while the uric acid concentration increased by 65%. Conclusions. The evaluation in saliva following a sprint test that lasted no more than 90 sec was sensitive enough to reveal changes in redox state.

Cryopreservation-thawing of human semen was found to reduce the level of antioxidant activity surrounding the sperm, which may negatively affect post-cryopreservation (post-thaw) recovery of sperm motility. Therefore, the current manufactured cryoprotectant media have been supplemented with certain antioxidants to preserve the loss in seminal antioxidant activity. In this study, we aimed to explore the correlation between total antioxidant capacity (TAC) of human semen samples before cryopreservation and the post-thaw recovery of sperm motility. Normal semen specimens (n = 77) were recruited in this study. Sperm motility was measured for each semen sample before and after cryopreservation and the post-thaw recovery of sperm motility was calculated. Seminal TAC was measured spectrophotometrically before cryopreservation for each semen sample using the sensitive cupric ion-reducing antioxidant capacity (CUPRAC) method. The results from this study showed that the post-thaw recovery of sperm motility is negatively correlated (p = 0.0404, p = 0.0402) with the absorbance at 450 nm and the values of seminal TAC in terms of µM Trolox equivalents, as evaluated by CUPRAC, respectively. In conclusion, the total antioxidant reservoir in each ejaculated semen specimen could be a factor in determining the post-thaw recovery of sperm motility toward lower recovery for semen specimens of high antioxidant content.

Background Electronic hookah (e‐hookah) vaping has increased in popularity among youth, who endorse unsubstantiated claims that flavored aerosol is detoxified as it passes through water. However, e‐hookahs deliver nicotine by creating an aerosol of fine and ultrafine particles and other oxidants that may reduce the bioavailability of nitric oxide and impair endothelial function secondary to formation of oxygen‐derived free radicals. Methods and Results We examined the acute effects of e‐hookah vaping on endothelial function, and the extent to which increased oxidative stress contributes to the vaping‐induced vascular impairment. Twenty‐six healthy young adult habitual hookah smokers were invited to vape a 30‐minute e‐hookah session to evaluate the impact on endothelial function measured by brachial artery flow‐mediated dilation (FMD). To test for oxidative stress mediation, plasma total antioxidant capacity levels were measured and the effect of e‐hookah vaping on FMD was examined before and after intravenous infusion of the antioxidant ascorbic acid (n=11). Plasma nicotine and exhaled carbon monoxide levels were measured before and after the vaping session. Measurements were performed before and after sham‐vaping control experiments (n=10). E‐hookah vaping, which increased plasma nicotine (+4.93±0.92 ng/mL, P <0.001; mean±SE) with no changes in exhaled carbon monoxide (−0.15±0.17 ppm; P =0.479), increased mean arterial pressure (11±1 mm Hg, P <0.001) and acutely decreased FMD from 5.79±0.58% to 4.39±0.46% ( P <0.001). Ascorbic acid infusion, which increased plasma total antioxidant capacity 5‐fold, increased FMD at baseline (5.98±0.66% versus 9.46±0.87%, P <0.001), and prevented the acute FMD impairment by e‐hookah vaping (9.46±0.87% versus 8.74±0.84%, P =0.002). All parameters were unchanged during sham studies. Conclusions E‐hookah vaping has adverse effects on vascular function, likely mediated by oxidative stress, which overtime could accelerate development and progression of cardiovascular disease. Aims: Certain exercises can develop the antioxidant system. Fenugreek supplementation containing antioxidant capacity and minerals may increase the benefits of exercises. The current study aimed at investigating the effect of Pilates training with fenugreek supplement on Total Antioxidant Capacity (TAC) and minerals in active women. Methods & Materials: The current study was with a quasi-experimental design was conducted on 36 active women aged 21-28 years randomly assigned into four groups: training, supplement, training plus supplement, and control. The training groups performed six weeks of Pilates training with the intensity of 60%-80% of maximal heart rate, three times a week for six weeks. Supplement groups received 500 mg fenugreek seeds daily for six weeks. Before and after the intervention, TAC and minerals (calcium, phosphorus, and iron) were measured. Data were analyzed by two-way ANOVA, Tukey post hoc test, and paired t-test at significant levels of P<0.05. Findings: In two variables, TAC and calcium, the time effect was significant, in other words, regardless of the group factor, there were significant differences between the post-test and pre-test of TAC and calcium variables in the experimental groups. The interaction of time-group was significant in TAC variable (P<0.05). The effect of the group was not significant in any of the variables. Conclusion: The obtained results suggested that one period of Pilates training and the use of fenugreek supplement alone and in combination could possibly prevent oxidative stress induced by exercise and could be effective in enhancing TAC and increasing calcium levels in active women.
