In 2014, the National Museum of Denmark (NMD), in conjunction with the Greenland National Museum and Archives (Nunatta Katersugaasivia Allagaateqarfialu [NKA]), as well as the Museum of Cultural History, Oslo, launched the website Skin Clothing Online. The site presents the NMD’s total collection of 2,170 historic skin clothing items, dating from circa 1830 to 1950, from the circumpolar area. The clothing can be studied in minute detail due to high-resolution photos; 100 complete suits were photographed from all sides. Furthermore, 107 items of clothing were measured by means of 3D technology, which can be used to draw precise two-dimensional patterns. The documentation is made accessible to the public through a website, in compliance with creative commons licenses: CC-BY-SA for the photos and CC-BY-SA-NC for the patterns. The website uses content from the database SkinBase. Since 2017 parts of the NKA’s collection of archeological skin clothing from Qilakitsoq (circa 1475 AD), as well as historical garments and contemporary fashions, have also been made accessible, in keeping with the same copyright rules for photos. The NKA staff entered the items into the database without difficulty using a Virtual Private Connection (VPN). The Danish and Greenlandic national museums encourage international partners to contribute items to the website. The aim is to create a collaborative open forum for information and research with easy access for everybody to unique, fragile pieces of circumpolar cultural heritage. With clothing from Arctic peoples and clothing used on expeditions to Antarctica, the Polar Museum in Cambridge will be the next museum to contribute to the website.

Securing the cultural heritage is one of the primary tasks of a museum of cultural history. However, this aspect of these museums’ work rarely attracts attention nor is it subject to any substantial professional debate, despite it being of critical importance for a museum’s future opportunities for development. Since the 1990s, the cultural heritage work done at the National Museum of Military History in Denmark has moved from an object-oriented positivistic perspec- tive to a research-based, problem-oriented and constructivist perspective, as well as moving to conducting active, research-based collecting. This article surveys this process of change and argues for the necessity of departing from ideas that are cha- racteristic of 19th-century museum ideals. The overall conclusion is that collecting should always be conducted as a response to a carefully considered problem that is based on a relevant research issue. This article accounts for the model chosen by the National Museum of Military History.

Abstract The article examines varied interpretations of food heritage in contemporary Estonia, relying on the authors’ experiences of a three-year research and development project at the Estonian National Museum (ENM). The study focuses on the museum researchers’ collaboration with different stakeholders, representing small entrepreneurs and the public and non-profit sectors. The authors tackle the partners’ expectations and outcomes of diverse cooperational initiatives and the opportunities and challenges of a contemporary museum as a public forum for discussions on cultural heritage. The project revealed that diverse, complementary, and contested food heritage interpretations exist side-by-side on the Estonian foodscape. Additionally, the project enabled the authors to become better aware of the researcher’s role in the heritagisation process and of the museum as a place for negotiating the meanings and values of food culture.

Research into memory, which has been carried out in recent decades by researchers in the fields of social sciences and humanities, is also important in the field of museology.Museums collect objects that, at the time of transition, lose their original function they have in previous everyday life and acquire a new one. Objects are generators of memory, and memory works through objects. However, the stories of individual objects are necessarily less comprehensive than stories that are made up of broader semantic wholes. At some stage of the narrative a transition from the collection of individual memories or memories of individuals to a wider whole appears – a collective memory. It is not composed of a multitude of individual memories, but is processed and transformed into a whole that corresponds a particular community. Memory is connected with time, and individual memories are fixed at the points of collective time.Museums are creators of collective memory. Collective memory is connected with the concepts of historical memory, (cultural) heritage and witnessing. The collective memory generated by objects creates an identity. This can be created at every level, from personal to local, from regional to national. Structuring a particular past has an extremely important role in structuring identity. The concepts of memory, heritage, witnessing and history in the field of cultural heritage refer to national museums in the purest form. Each national museum is a guardian, researcher and promoter of a professionally and scientifically transformed collective memory, and thus a constitutive element of national consciousness.

Guaranteeing the survival of cultural heritage, increasing its accessibility, both physical and intellectual, and the creation of countless benefits for different categories of stakeholders depends both on a perfect comprehension of the interests and abilities of users to take advantage of what is offered and, above all, on identifying and analysing the various types of value that can be attributed to it. According to Montella, there are three types of value that may be analysed for this purpose: a presentation value, informative in nature and inherent in the historical, cultural and possibly artistic value implicit in the heritage; a landscape value, extended to the context, inherent in the factual information services aimed at supporting policies of preventive and programmed conservation: and a production value, commercial in nature, which concerns the external effects generated by cultural heritage management to qualify the products and the images themselves of the businesses in order to make them stand out from the competition. The aim of this article is to inquire into whether, in what way and to what extent the communication of the Paper and Watermark Museum in Faabriano and the Ascoli Piceno Papal Paper Mill Museum in Ascoli Piceno creates presentation value and therefore leads the public to understand how far paper production has influenced the economic and socio-cultural history of the area in which they are located.

In the last twenty-five years, the Swedish museum landscape has expanded and contains today several thousand museums, from the single local history museum to the merged governmental central museum, many of them organized in different networks. During the same period contemporary collecting, diversity issues and difficult matters became both attractive and urgent topics for the public cultural history museums. Also, museology and cultural heritage studies were established at several universities, with professorships as well as basic educational programmes. As a consequence, the perception of museology as research done at museums was replaced by research on museums, often with a critical view of the history of collections and exhibitions. During the last few years, however, a polarized media debate reveals that there might still be a gap between the actual, contemporary museum and the more traditional concept of the museum in (some) people’s minds.

This tutorial will discuss the methodology of low dose electron diffraction and imaging of crystalline biological objects, the problems of data interpretation for two-dimensional projected density maps of glucose embedded protein crystals, the factors to be considered in combining tilt data from three-dimensional crystals, and finally, the prospects of achieving a high resolution three-dimensional density map of a biological crystal. This methodology will be illustrated using two proteins under investigation in our laboratory, the T4 DNA helix destabilizing protein gp32*I and the crotoxin complex crystal.

Microtubules are involved in a host of critical cell activities, many of which involve transport of organelles through the cell. Different sets of microtubules appear to form during the cell cycle for different functions. Knowledge of the structure of tubulin will be necessary in order to understand the various functional mechanisms of microtubule assemble, disassembly, and interaction with other molecules, but tubulin has so far resisted crystallization for x-ray diffraction studies. Fortuitously, in the presence of zinc ions, tubulin also forms two-dimensional, crystalline sheets that are ideally suited for study by electron microscopy. We have refined procedures for forming the sheets and preparing them for EM, and have been able to obtain high-resolution structural data that sheds light on the formation and stabilization of microtubules, and even the interaction with a therapeutic drug.Tubulin sheets had been extensively studied in negative stain, demonstrating that the same protofilament structure was formed in the sheets and microtubules. For high resolution studies, we have found that the sheets embedded in either glucose or tannin diffract to around 3 Å.

SummaryThe platelet proteins of 9 thrombasthenic patients from 7 families were analysed by high resolution two-dimensional gel electrophoresis (HR-2DE) and crossed immunoelectrophoresis (CIE). In 7 patients both glycoproteins (GPs) IIb and Ilia were absent or reduced to roughly the same extent. In two related patients only a trace of GP Ilb-IIIa complex was detected in CIE, but HR-2DE revealed a glycopeptide in the position of GP Ilia in an amount comparable to type II thrombasthenia. This GP Ilia-like component was neither recognized normally by anti-GP Ilb-IIIa antibodies nor labeled by surface iodination. In unreduced-reduced two-dimensional gel electrophoresis two components were observed in the region of GP Ilia. The assumption of a structural variant of GP Ilia in the two related patients is discussed.
