<p><i>trans</i>-Cyclooctenes (TCOs) are essential partners for the fastest known bioorthogonal reactions, but current synthetic methods are limited by poor diastereoselectivity. Especially hard to access are hydrophilic TCOs with favorable physicochemical properties for live cell or <i>in vivo </i>experiments. Described is a new class of TCOs, ‘a-TCOs’, that is prepared in high yield via stereocontrolled 1,2-additions of nucleophiles to trans-cyclooct-4-enone, which itself was prepared on large scale in two steps from 1,5-cyclooctadiene. Computational transition state models rationalize the diastereoselectivity of 1,2-additions to deliver a-TCO products, which were also shown to be more reactive than standard TCOs and less hydrophobic than even a <i>trans</i>-oxocene analog. Illustrating the favorable physicochemical properties of a-TCOs, a fluorescent TAMRA derivative in live HeLa cells was shown to be cell-permeable through intracellular Diels-Alder chemistry and to washout more rapidly than other TCOs.</p>

The study aimed at optimizing the extraction conditions and establishing the physicochemical properties of the mucilage obtained from root bark of Malva verticillata. Effects of temperature, time, particle size and amount of water as a solvent on the yield and composition of mucilage were established. Micrometric, physical, chemical and structural properties were used to characterise the extracted mucilage. Result showed significant correlation between extraction conditions and mucilage yield. Low temperature and short extraction time results into pharmaceutically valuable mucilage with low protein content, high viscosity and good dispersion in aqueous solvent. Large and small size particles of the plant material results into low mucilage yield without significant difference (p=0.054). The mucilage shows mixed cohesiveness with less tendencies to flow. It exhibits vibration bands in FTIR that are associated with the presence of polysaccharides and proteins. The mucilage contains 2.65% protein, 13.83% inorganics, 1.85% fats, and the rest carbohydrates. High extractive value ˃80% and moisture content ˃12% implies that mucilage has high tendency to microbial attack. Optimized extraction conditions results into high yield of the mucilage with the same physicochemical properties and mineral composition regardless of the plant material sampling sites.

The aim of the study was to assess the yield of cones and the quality of six hop varieties (“Iunga”, “Lubelski”, “Sybilla”, “Magnum”, “Lomik”, “Marynka”) in terms of their suitability for beer production, under the conditions of the Małopolska Vistula Gorge Region (21°53′ E; 51°16′ N) (2015–2017). The scope of the research included the determination of the yield of cones and their physicochemical properties, as well as determination of the contents of hop resins and essential oils. In bitter varieties, the ratio of alpha-acids to beta-acids was stable, while, in aromatic varieties, it was variable. In the essential oils of the studied hop varieties, compounds with myrcene and α-humulene were identified and dominated the profiles. “Iunga”, “Sybilla”, “Lubelski”, and “Lomik” were rich in monoterpene hydrocarbons, mainly myrcene, while there was a bit more α-humulene in the “Marynka” variety. “Magnum” was characterized by an even distribution of monoterpenes and sesquiterpenes. The “Magnum variety turned out the most useful for the brewing industry in this region. The Małopolska Vistula Gorge Region is a region where hops not only achieve a high yield of cones, but also good-quality. The bitterness and aroma content of the hops in this region is high.
