Proline, a well-known amino acid, has been estimated in ten different pteridophytes, including Adiantum capillus-veneris L., Adiantum caudatum L., Adiantum incisum Forssk., Adiantum philippense L., and Actiniopteris radiata (Sw.) link. Christenh. in various organs such as the root, rhizome, and leaves Hemionitis anceps (Blanf.) Christenh., Hemionitis bicolor (Roxb.) Christenh., Hemionitis formosana Christenh (Hayata). Result of study is indicate that the maximum total proline in entire plant was recorded 43.54 mg/gdw and leaves have been  recorded 24.10 mg/gdw in Pteris vittata subsp. vittata and the minimum value was recorded 3.60 mg/gdw in root of Adiantum caudatum L.

Citrulline has been recently shown to behave as a novel compatible solute in the <i>Citrullus lanatus</i> (<i>Cucurbitaceae</i>) growing under desert conditions. In the present study we have investigated some aspects of the relationship which might occur in leaves of melon seedlings, also known to produce citrulline, between the capacity to accumulate this ureido amino acid and salt tolerance. With this end in view, salt-induced changes at the citrulline level have been compared in two melon genotypes exhibiting contrasted abilities to withstand the damaging effects of high salinity. Progressive salinization of the growing solution occurred at 23 days after sowing. The final 250 mmol/l external NaCl concentration was reached within 5 days and further maintained for 16 days. In response to this treatment, it was found that the citrulline amount increased in fully expanded leaves of both genotypes according to different kinetics. The salt tolerant genotype Midyat was induced to accumulate citrulline 4 days before the salt sensitive Yuva and as a consequence the final amount of this amino acid was twice higher in the former than in the latter. Compared with citrulline, the free proline level was found to be relatively low and the changes induced in response to the salt treatment exhibited different trends according to the genotypes under study. Thus at the end of the treatment mature leaves of the salt sensitive Yuva contained higher amount of proline than those of Midyat. The changes in the calculated molar ratio between citrulline and free proline suggested that salt tolerance might be associated with high values for this ratio and vice et versa for sensitivity. The interest of citrulline as a biochemical marker for salt tolerance of melon genotypes is discussed.

During 6-day exposures of cells of Saccharomyces cerevisiae to acetate sporulation medium, the content of free amino acids declined to approximately one-third of that of vegetative cells, but proline was exceptional in that it increased conspicuously in amount. The content of combined amino acids also diminished to about one-third, ammonia was evolved, and amino acids (not including proline) passed out of the cells into the medium. When dihydroxyacetone replaced acetate in the sporulation medium, the results were similar except that the decline in content of free and combined amino acids was much greater, more ammonia was evolved, and only very small amounts of amino acids could be detected in the medium. Transfer of sporulated cells to growth medium led to an increase in the pool of free amino acids, except for proline, which declined in amount.In two other species of Saccharomyces the free proline content also increased on exposure to sporulation medium, but in Schizosaccharomyces pombe and Torulopsis famata no such increase was observed.

The amino acid proline is accumulated in plant tissues in response to a variety of stresses. The existence of two routes for its biosynthesis is well documented. However, little is known about the contribution of each pathway to the accumulation of free proline under stress conditions. In the present study young barley plants were subjected to osmotic stress by treating their roots with 25% polyethylene glycol. Prior to stress imposition roots were incubated for 24 h in nutrient solution containing proline or one of its metabolic precursors: glutamate and ornithine. Free proline quantity in the leaves was measured before and after stress. Relative water content (RWC) was used as a measure of the plant water status. Foliar proline levels showed a significant increase in ornithine- and proline-pretreated plants compared to the control. Nevertheless, no considerable changes in leaf RWC were observed. It was shown that before stress application only ornithine but not glutamate was immediately metabolized to proline. Under stress conditions, however, both precursors were converted into proline. The possible role of this amino acid in the processes of post stress recovery is discussed.

The concentration of each free amino acid present in hypocotyl tissue of sunflower seedlings was determined during the period following both sterile wounding and inoculation with either tumorigenic (strain 1025) or non-tumorigenic (strain IIBNV6) Agrobacterium tumefaciens. The concentration of the total free amino acids of hypocotyl tissue reached a maximum 68 hours after infection with strain 1025 and this level was 50% greater than that found in plants treated with strain IIBNV6. The total content of free basic amino acids increased to a peak value at 20 hours after infection by strain 1025 and this represented 1.4 to 3 times the level found in strain IIBNV6 inoculated seedlings at 20 hours.The amount of free proline decreased with time and it was essentially absent 164 hours after infection with strain 1025. At 2 months the concentration of free proline in the tumor tissue was greatly increased with 75% of the total free amino acids accounted for as proline.The free amino acid content of strain 1025 growing in either the presence or absence of an extract from wounded seedlings was measured and the results indicated no significant relationship between the plant and bacterial systems with regard to their free amino acid pools.

Knowledge of the dynamics of collagen turnover requires information regarding rates of synthesis of this group of connective-tissue proteins. The relationship of various amino acid pools to the tRNA precursor pool used for protein synthesis is known to vary between different cell types and tissues, even for essential amino acids. We studied extracellular, intracellular and tRNA-proline pools in cultured human lung IMR-90 fibroblasts to determine the relationship between them as candidate proline precursor pools for total protein and collagen synthesis. Time-course experiments showed that the three proline pools attained distinctly different steady-state specific radioactivities (extracellular greater than intracellular greater than tRNA) at the extracellular proline concentration of 0.2 mM. The kinetics of radioisotope incorporation into cell protein and collagenase-digestible protein indicated that the intracellular free proline pool could not be used reliably as a precursor for calculating synthetic rates. However, tRNA-proline behaved isotopically as if it were the precursor and provided synthesis rates 2-3-fold higher than those calculated by using either free proline pool. The incorporation of labelled lysine and leucine was constant over a wide range of extracellular proline concentrations. Fractional rates of protein synthesis based on tRNA-amino acid were the same with [3H]phenylalanine as with [3H]proline. The specific radioactivity of cell-associated hydroxyproline reached a steady-state value 8-10h after radioisotope administration which matched the mean tRNA-proline specific radioactivity, suggesting that tRNA-proline is not isotopically compartmentalized. A model of cellular proline-pool relationship is presented and discussed.

1. A method is described by which, from analysis of terminal samples, the rate constants that define the changes in specific radioactivity of free amino acids during continuous infusions can be estimated. The method involves the infusion of 3H-labelled and 14C-labelled forms of an amino acid for different, but overlapping, periods. 2. The procedure was developed for infusions of proline and tyrosine into New Zealand White rabbits and the rate constants were determined for blood and muscle. 3. The rate constant for equilibrium of radioactive free proline was much lower in muscle than in blood, and indicated that a plateau condition may not be attained in muscle by the end of a 6 h infusion. 4. Comparison of the ratio of areas under the curves of estimated specific radioactivity plotted versus time with the 3H/14C ratio of bound amino acid in muscle protein suggested that radioactive proline and tyrosine may be incorporated preferentially from an extracellular rather than an intracellular source.

Amino-acid-analog-resistant lines were selected from mutagen-treated suspension-cultured tobacco (Nicotiana tabacum L.) and carrot (Daucus carota L.) cells. Tobacco cell lines resistant to the lysine analogs aminoethyl-L-cysteine and delta-hydroxylysine and carrot lines resistant to a methionine analog and a proline analog, ethionine and hydroxyproline, respectively, were selected. The resistant lines were 10 to more than 1000 times less sensitive to the analogs than the parent strains. The resistant cells were selected with a low frequency, were generally stable during growth in the absence of the selective agent, and accumulated the corresponding natural free amino acid. The lysine-analog-resistant tobacco lines had more than 10 times the normal levels of free lysine while the methionine-analog-resistant carrot line had similar increases in free methionine. The hydroxyproline-resistant carrot line contained from 15 to 30 times the normal free proline levels and was cross resistant to certain other proline analogs.
