Proanthocyanidins (PAs) are important phenolic compounds and PA biosynthesis is regulated by a ternary MBW complex consisting of a R2R3-MYB regulator, a bHLH factor and a WDR protein. In this study, a tobacco R2R3-MYB factor NtMYB330 was characterized as the PA-specific regulator in which the PA biosynthesis was promoted in the flowers of NtMYB330-overexpressing lines while decreased in the flowers of ntmyb330 mutants. NtMYB330 can interact with flavonoid-related bHLH partner NtAn1b and WDR protein NtAn11-1, and the NtMYB330-NtAn1b complex is required to achieve strong transcriptional activation of the PA-related structural genes NtDFR1, NtANS1, NtLAR1 and NtANR1. Our data reveal that NtMYB330 regulates PA biosynthesis in seeds and affects seed germination, in which NtMYB330-overexpressing lines showed higher PA accumulations in seed coats and inhibited germination, while ntmyb330 mutants had reduced seed coat PAs and improved germination. NtMYB330 affects seed germination possibly through two mechanisms: modulating seed coat PAs to affect coat-imposed dormancy. In addition, NtMYB330 regulates the expressions of abscisic acid (ABA) and gibberellin acid (GA) signaling-related genes, affecting ABA-GA crosstalk and seed germination. This study reveals that NtMYB330 specifically regulates PA biosynthesis via formation of the MBW complex in tobacco flowers and affects germination through adjustment of PA concentrations and ABA/GA signaling in tobacco seeds.

Tobacco has previously been used in investigations of metals and radionuclide uptake. This study presents determination of bioaccumulation and translocation of 60Co2+ ions in tobacco plants (Nicotiana tabacum L.) grown in Hoagland’s nutrient solution. Cobalt concentration in tobacco plants increased with increasing concentration in nutrient solution. Bioaccumulation from the initial concentration C0 = 0.96 μM Co reached 90% after 7 day cultivation. Only small amounts of Co accumulated in roots, up to 2 - 4 % were removable from roots by washing with 0.1 M CoCl2, indicating that this portion of Co is bound to the root surface in ion-exchangeable form. Tobacco roots retained approximately 2/3 of accumulated cobalt and 1/3 was transported to shoots. Autoradiography revealed that 60Co was preferentially localized in younger leaves. Prolongation of cultivation time did not change the [Co]roots : [Co]shoots ratio significantly. Relationships between growth rate, transpiration rate, uptake and distribution of cobalt in plant tissue are discussed.

The aim of the work was to characterize the samples of sewage sludge (SSL) originated from the Wastewater Treatment Plant Piešťany (TAVOS, a.s., Trnava, Slovakia) in terms of their potential application into the soils. Within the physico-chemical characterization of SSL, the samples were analysed in terms of the values of pH, cation exchange capacity (CEC), total organic carbon (TOC), water holding capacity (WHC), as well as the presence of heavy metals. It was found that SSL contained significant amounts of microelements Zn (1,269 mg.kg-1, d.w.) and Cu (224 mg.kg-1, d.w.). Laboratory lysimeter experiments involving the application of SSL into the top layer of agriculturally used soil (0 – 10 cm) forming a soil column, in which seedlings of tobacco (Nicotiana tabacum L.) were cultivated, showed that in the case of SSL application in the highest permitted amounts (15 t.ha-1) only 0.11 % Zn and 0.07 % Cu were released into the soil eluate during a 28 day of exposure, while in tobacco plants 0.13 % Zn and 0.05 % Cu were accumulated from the total amount of Zn and Cu originated from the application of SSL (Zn – 133 mg and Cu – 23.5 mg). When applying SSL in amount 30 t.ha-1 i.e., in the dose exceeding the permitted limits, only 0.02 % Zn and 0.04 % Cu were released into the soil eluate and 0.16 % Zn and 0.09 % Cu were accumulated in tobacco plants from the applied amount of SSL (Zn – 267 mg and Cu – 47.0 mg).

The traditional shallow tillage method makes the soil quality declining, and affects the efficiency of agricultural production. Taking the conventional rotary tillage(12 cm) as the control, Yunyan 87 as the test variety, and the soil type of the test site is paddy soil, we studied the effects of deep tillage(Subsoiling 30 cm) on soil nutrients, arbuscular mycorrhizal fungi (AMF) and tobacco(Nicotiana tabacum L.)growth. The results showed that deep tillage increased the contents of organic carbon, available phosphorus(AP) and available potassium(AK) in 20 ~ 40 cm soil layer. The community of AMF was changed by deep tillage. Glomus, the dominant genus in both group, increased significantly in the soil after deep tillage. The colonization rate of AMF was lower than that of conventional rotary tillage. Deep tillage was beneficial to the growth of tobacco in the middle and late stages. Root growth and nutrient content of tobacco increased. Deep tillage significantly improved the output value of tobacco. It can be seen that deep tillage is conducive to improving soil fertility, promoting the vigorous growth of root, reducing the dependence of tobacco on AMF, and promoting the high quality and yield of tobacco in drylands of Hunan.

Leaf senescence is an important process of growth and development in plant, and it is a programmed decline controlled by a series of genes. In this study, the biochemical properties and transcriptome at five maturity stages (M1∼M5) of tobacco leaves were analyzed to reveal the dynamic changes in leaf senescence of tobacco. A total of 722, 1,534, 3,723, and 6,933 genes were differentially expressed (DEG) between M1 and M2, M1 and M3, M1 and M4, and M1 and M5, respectively. Significant changes of nitrogen, sugars, and the DEGs related to metabolite accumulation were identified, suggesting the importance of energy metabolism during leaf senescence. Gene Ontology (GO) analysis found that DEGs were enriched in biosynthetic, metabolic, photosynthesis, and redox processes, and especially, the nitrogen metabolic pathways were closely related to the whole leaf senescence process (M1∼M5). All the DEGs were grouped into 12 expression profiles according to their distinct expression patterns based on Short Time-series Expression Miner (STEM) software analysis. Furthermore, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis found that these DEGs were enriched in pathways of carbon metabolism, starch and sucrose metabolism, nitrogen metabolism, and photosynthesis among these expression profiles. A total of 30 core genes were examined by Weight Gene Co-expression Network Analysis (WGCNA), and they appeared to play a crucial role in the regulatory of tobacco senescence. Our results provided valuable information for further functional investigation of leaf senescence in plants.

Basic helix-loop-helix (bHLH) transcription factor MYC2 regulates plant growth and development in many aspects through the jasmonic acid (JA) signaling pathway, while the role of MYC2 in plant carbohydrate metabolism has not been reported. Here, we generated NtMYC2a-overexpressing (NtMYC2a-OE) and RNA-interference-mediated knockdown (NtMYC2a-RI) transgenic plants of tobacco (Nicotiana tabacum L. cv. TN90) to investigate the role of NtMYC2a in carbohydrate metabolism and pollen development. Results showed that NtMYC2a regulates the starch accumulation and the starch-sugar conversion of floral organs, especially in pollen. The RT-qPCR analysis showed that the expression of starch-metabolic-related genes, AGPs, SS2 and BAM1, were regulated by NtMYC2a in the pollen grain, anther wall and ovary of tobacco plants. The process of pollen maturation was accelerated in NtMYC2a-OE plants and was delayed in NtMYC2a-RI plants, but the manipulation of NtMYC2a expression did not abolish the pollen fertility of the transgenic plants. Intriguingly, overexpression of NtMYC2a also enhanced the soluble carbohydrate accumulation in tobacco ovaries. Overall, our results demonstrated that the bHLH transcription factor NtMYC2a plays an important role in regulating the carbohydrate metabolism during pollen maturation in tobacco.

Recent studies have shown that melatonin is an important molecule in plant physiology. It seems that the most important is that melatonin effectively eliminates oxidative stress (direct and indirect antioxidant) and switches on different defence strategies (preventive and interventive actions) during environmental stresses. In the presented report, exogenous melatonin potential to protect Nicotiana tabacum L. line Bright Yellow 2 (BY-2) exposed to lead against death was examined. Analyses of cell proliferation and viability, the level of intracellular calcium, changes in mitochondrial membrane potential (ΔΨm) as well as possible translocation of cytochrome c from mitochondria to cytosol and subsequent caspase-like proteolytic activity were conducted. Our results indicate that pretreatment BY-2 with melatonin protected tobacco cells against mitochondrial dysfunction and caspase-like activation caused by lead. The findings suggest the possible role of this indoleamine in the molecular mechanism of mitochondria, safeguarding against potential collapse and cytochrome c release. Thus, it seems that applied melatonin acted as an effective factor, promoting survival and increasing plant tolerance to lead.
